The bladder cancer m6A landscape is defined by global methylation dilution and focal 3'-UTR hypermethylation.

N6-Methyladenosine (m6A) is the most abundant internal modification of eukaryotic mRNAs and regulates target transcripts throughout the mRNA life cycle. Although changes in m6A have been reported in human cancers, technical limitations have hindered a comprehensive understanding of the cancer-associated m6A landscape. Here, we use GLORI-sequencing to establish the first transcriptome-wide, single-nucleotide resolution maps of m6A in bladder cancer. Comparing bladder cancer and healthy bladder samples, we discover two key m6A signatures: a global dilution of methylation and a focal hypermethyla

A novel m7G RNA methylation-related signature associated with MAPK signaling pathways in acute ischemic stroke.

OBJECTIVES: Ischemic stroke represents a major global cause of disability and death. Timely recanalization of occluded vessels to salvage the ischemic penumbra is the cornerstone of acute ischemic stroke (AIS) treatment. Nevertheless, the molecular mechanisms driving the transition from salvageable penumbra to infarct core in early AIS remain poorly understood. Recent evidence highlights the role of post-transcriptional methylation, especially N7-methylguanosine (m7G) modification, in stroke pathogenesis. This study aimed to systematically characterize transcriptome-wide m7G methylation change

METTL1 promotes hepatic steatosis by mediating m7G modification of ALOX15B mRNA.

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) is defined by aberrant hepatic lipid accumulation, yet the regulatory mechanisms underlying this process remain incompletely understood. Although epitranscriptomic modifications have emerged as key regulators of hepatic lipid homeostasis, the role of N7-methylguanosine (m7G) modification in hepatic steatosis remains unclear. METHODS: Histological and immunohistochemistry studies were used to assess lipid deposition in free fatty acids (FFAs)-incubated AML12 and HepG2 cells, high-fat diet (HFD)-fed mice, and human liv

NSUN2-Mediated m5C Methylation in Human Disease: Mechanism and Therapeutic Implications.

The epitranscriptomic landscape defined by reversible post-transcriptional RNA modifications constitutes a sophisticated layer of gene regulatory circuitry that modulates diverse biological processes. As an essential writer enzyme for RNA 5-methylcytosine (m5C), NOP2/Sun RNA methyltransferase 2 (NSUN2) catalyzes site-specific m5C deposition across an extensive repertoire of cellular transcripts spanning messenger RNAs, transfer RNAs and non-coding RNAs, and orchestrates core post-transcriptional events including transcript stabilization, nucleocytoplasmic trafficking, translational tuning and

NAT10/ac4C drives intrahepatic cholangiocarcinoma by suppressing transposable elements via chromatin remodeling.

Intrahepatic cholangiocarcinoma (ICC) poses a significant clinical challenge due to its insidious onset, aggressive biological behavior, and propensity for early metastasis, contributing to a dismal 5-y survival rate of less than 10%. Despite advances in understanding the dynamic changes in gene regulatory networks and chromatin landscapes during tumorigenesis, the functional interplay between RNA epitranscriptomic modifications and nuclear events in ICC remains poorly elucidated. Here, we identify N-acetyltransferase 10 (NAT10), the writer for N4-acetylcytidine (ac4C) mRNA modification, as a

RNA modifications in peripheral blood are associated with acute coronary syndrome.

Acute coronary syndromes (ACS) trigger inflammatory and immune reactions, yet whether this response is associated with changes in RNA modifications remains poorly understood. We investigated global epitranscriptomic patterns in peripheral blood mononuclear cells (PBMC) from patients with ST-elevation myocardial infarction (STEMI, n = 118), non-ST-elevation myocardial infarction (NSTEMI, n = 22), unstable angina pectoris (UAP, n = 10), and stable angina pectoris (SAP, n = 152). Using liquid chromatography-mass spectrometry, we quantified N1-methyladenosine (m1A), 2'-O-methyladenosine (2'-O-mA),

Integrative MeRIP-seq and RNA-seq analysis reveals m6A-mediated epigenetic regulation of host-virus interactions during HSV-2 infection.

Herpes simplex virus type 2 (HSV-2), a highly prevalent pathogen responsible for genital herpes, is characterized by neurotropism and the ability to establish lifelong latent infection. N6-methyladenosine (m6A) is a widespread epitranscriptomic modification that plays a critical role in regulating RNA metabolism and gene expression. In this study, we used methylated RNA immunoprecipitation sequencing (MeRIP-seq) and RNA sequencing (RNA-seq) to profile m6A modifications and transcriptomic changes in human foreskin fibroblasts (HFF-1) infected with HSV-2. We identified 9477 common m6A peaks and

Chronic nitric oxide mediates dual-layer gene regulation through mRNA m6A positional remodeling and parallel transcriptional reprogramming.

Nitric oxide (NO) is a pleiotropic free radical that functions as a master regulator of gene expression, and its sustained production within the tumor microenvironment reshapes the epitranscriptomic state of cancer cells. We previously demonstrated that NO inhibits the m6A mRNA demethylases FTO and ALKBH5 through dinitrosyliron complex formation while leaving the methyltransferase METTL3 intact, a demethylase-specific perturbation that increases global m6A on mRNA. Here, integrating m6A-RIP-seq and RNA-seq from triple-negative breast cancer cells, we show that chronic NO does not produce the u

NSUN2-mediated cytosine-5 methylation of FSP1 protects acute myeloid leukemia cells from ferroptosis.

RNA 5-methylcytosine (m5C), a prevalent epitranscriptomic modification that critically regulates gene expression and cellular homeostasis. While its roles in solid tumors have been increasingly recognized, the functional landscape of m5C in acute myeloid leukemia (AML) remains unexplored. Here, we identified NSUN2, the principal RNA m5C methyltransferase, as a key regulator of AML progression. NSUN2 was aberrantly upregulated in AML patient samples and correlated with poor prognosis. Functional studies demonstrated that NSUN2 promoted leukemic cell proliferation, enhanced tumor growth in xenog

METTL8 links mt-tRNA m3C modification to the HIF1α/RTK/Akt axis to sustain GBM stemness and tumorigenicity.

Epitranscriptomic RNA modifications are crucial for the maintenance of glioma stem cells (GSCs), the most malignant cells in glioblastoma (GBM). 3-methylcytosine (m3C) is a new epitranscriptomic mark on RNAs and METTL8 represents an m3C writer that is dysregulated in cancer. Although METTL8 has an established function in mitochondrial tRNA (mt-tRNA) m3C modification, alternative splicing of METTL8 can also generate isoforms that localize to the nucleolus where they may regulate R-loop formation. The molecular basis for METTL8 dysregulation in GBM, and which METTL8 isoform(s) may influence GBM

Targeting G3BP1 Condensate Topology Promotes Stress Granule Assembly via m6A-IGF2BP1 for Ischemic Stroke Rescue.

Ras-GTPase-activating protein SH3 domain-binding protein 1 (G3BP1) mediates stress granules (SGs) via phase separation. However, there is limited understanding of the allosteric mechanism and the identification of regulatory molecules. Here, we identify icariin (ICA), a small-molecule inducer that promotes G3BP1-driven biomolecular condensate formation, which effectively restructures SGs architecture. Moreover, we demonstrate that ICA interacts with the N-terminal nuclear transport factor 2-like (NTF2L) domain of G3BP1, inducing a conformational switch from "closed-to-open" that facilitates G3

Unscheduled m6A Deposition in RNA via m6ATP Incorporation by DNA Polymerases.

N6-methyladenosine (m6A) is the most abundant modification of mRNA and plays a crucial role in mediating cellular functions, and it is associated with cancer and neurodegenerative diseases. Studies have shown that m6A is predominantly deposited on its consensus motif by the m6A writer proteins RNA methyltransferase METLL3/METLL14. However, it was found that nonconventional m6A deposition by other alternative pathways may also exist and can modulate epitranscriptomic regulation in cells. Thus, understanding the molecular mechanisms underlying nonconventional m6A deposition outside the canonical

An Association between Decreased Small Intestinal RNA Modification and Disturbed Glucagon-like Peptide-1 Secretion under High-Fat Diet Stress.

Unhealthy diets rich in fats and/or sugar are considered as the major external cause of the obesity epidemic, which is often accompanied by a significant decrease in gut hormone glucagon-like peptide-1 (GLP1) levels. Numerous studies have demonstrated notable contributions of the gut microbiota in this process. Nevertheless, the underlying mechanism still needs further investigation. The role of epigenetic modifications in gene expression and metabolism has been well demonstrated, with m6A methylation on RNAs being the most prevalent modification throughout their metabolism. Fat mass and obesity-associated protein alleviates Aβ1-40 induced retinal pigment epithelial cells degeneration via PKA/CREB signaling pathway.

Amyloid-β (Aβ) is thought to be a critical pathologic factor of retinal pigment epithelium (RPE) degeneration in age-related macular degeneration (AMD). Aβ induces inflammatory responses in RPE cells and recent studies demonstrate the N6-methyladenosine (m6A) regulatory role in RPE cell inflammation. m6A is a reversible epigenetic posttranslational modification, but its relationship with Aβ-induced RPE degeneration is yet to be thoroughly investigated. The present study explored the role and mechanism of m6A in Aβ-induced RPE degeneration model. This model was induced via intravitreally inject

Malaria Parasite Stress Tolerance Is Regulated by DNMT2-Mediated tRNA Cytosine Methylation.

Malaria parasites need to cope with changing environmental conditions that require strong countermeasures to ensure pathogen survival in the human and mosquito hosts. The molecular mechanisms that protect Plasmodium falciparum homeostasis during the complex life cycle remain unknown. Here, we identify cytosine methylation of tRNAAsp (GTC) as being critical to maintain stable protein synthesis. Using conditional knockout (KO) of a member of the DNA methyltransferase family, called Pf-DNMT2, RNA bisulfite sequencing demonstrated the selective cytosine methylation of this enzyme of tRNAAsp (GTC)

M6A methylation in thyroid cancer: Functions, mechanisms, and clinical significance.

Thyroid cancer (TC), the most prevalent endocrine malignancy, exhibits marked biological heterogeneity and a persistent therapeutic gap in aggressive, treatment-refractory subsets. N6-methyladenosine (m6A), pervasive across eukaryotic RNA processing-splicing, maturation, stability, translation, and localization-constitutes a central layer of epigenetic regulation. By reshaping post-transcriptional programs, m6A modifications govern tumor initiation and progression, sustaining stemness, accelerating proliferation and invasion, and mediating therapy resistance. A growing body of evidence implica

METTL3: a multifunctional regulator in diseases.

N6-methyladenosine (m6A) methylation is the most prevalent and abundant internal modification of mRNAs and is catalyzed by the methyltransferase complex. Methyltransferase-like 3 (METTL3), the best-known m6A methyltransferase, has been confirmed to function as a multifunctional regulator in the reversible epitranscriptome modulation of m6A modification according to follow-up studies. Accumulating evidence in recent years has shown that METTL3 can regulate a variety of functional genes, that aberrant expression of METTL3 is usually associated with many pathological conditions, and that its expr
