Relative activity of unsaturated fatty acid metabolic pathways in humans.

The four families of unsaturated fatty acids (w9, w7, w6, w3) appear to be elongated and desaturated by enzymes shared among all families. The order of relative pathway activity is hypothesized to be w3 greater than w6 greater than w9 greater than w7. In order to examine this question, we measured by high resolution capillary column gas-liquid chromatography, the relative concentrations of the various fatty acids in plasma from 20 healthy reference subjects, 33 randomly selected subjects from the Framingham Heart Study, and six patients with intestinal malabsorption and suspected severe essent

Nutritional composition and postharvest senescence control in Chaerophyllum macrospermum: a comparative study of 1-MCP, nitric oxide, and hydrogen sulfide treatments.

Chaerophyllum macrospermum is consumed in parts of Iran for its nutritional and antioxidant properties, but rapid postharvest yellowing limits its shelf-life. This study assessed its nutritional composition, bioactive compounds, and postharvest preservation strategies. Edible parts (leaf-blades, buds, petioles) were analyzed for nutrients, antioxidants, essential oils, fatty acids, amino acids, and phenolics. Buds contained the highest levels of nitrogen, zinc, and protein, while leaves were richest in iron and manganese. Fresh essential oil was dominated by α-pinene (26.35%) and α-ocimene, wh

Accumulation of (n-9)-eicosatrienoic and docosatrienoic acids in human fibroblast phospholipids.

An essential fatty acid (EFA) deficiency-like profile of fatty acids has been observed in HF-1 human skin fibroblasts cultured at clonal densities in MCDB 110 and 0.4% fetal bovine serum (FBS). The profile was characterized by an accumulation of 16:1n-7, 18:1n-9, 20:3n-9 and 22:3n-9, a reduction of n-6 fatty acids and a reduction in total polyunsaturated fatty acids (PUFA). The fatty acid composition of sphingomyelin (SPH), phosphatidylcholine (PC), phosphatidylserine (PS), phosphatidylinositol (PI) and phosphatidylethanolamine (PE) was determined and, except for SPH, each displayed an EFA def

Elovl5 is required for proper action potential conduction along peripheral myelinated fibers.

Elovl5 elongates fatty acids with 18 carbon atoms and in cooperation with other enzymes guarantees the normal levels of very long-chain fatty acids, which are necessary for a proper membrane structure. Action potential conduction along myelinated axons depends on structural integrity of myelin, which is maintained by a correct amount of fatty acids and a proper interaction between fatty acids and myelin proteins. We hypothesized that in Elovl5-/- mice, the lack of elongation of Elovl5 substrates might cause alterations of myelin structure. The analysis of myelin ultrastructure showed an enlarg

Maternal dietary fatty acids and their roles in human placental development.

Fatty acids are essential for feto-placental growth and development. Maternal fatty acids and their metabolites are involved in every stage of pregnancy by supporting cell growth and development, cell signaling, and modulating other critical aspects of structural and functional processes. Early placentation process is critical for placental growth and function. Several fatty acids modulate angiogenesis as observed by increased tube formation and secretion of angiogenic growth factors in first-trimester human placental trophoblasts. Long-chain fatty acids stimulate angiogenesis in these cells v

Chemical genetic profiling and characterization of small-molecule compounds that affect the biosynthesis of unsaturated fatty acids in Candida albicans.

The balance between saturated and unsaturated fatty acids plays a crucial role in determining the membrane fluidity. In the diploid fungal pathogen Candida albicans, the gene for fatty acid Delta9 desaturase, OLE1, is essential for viability. Using a reverse genetic approach, termed the fitness test, we identified a group of structurally related synthetic compounds that induce specific hypersensitivity of the OLE1(+/-) strain. Genetic repression of OLE1 and chemical inhibition by two selected compounds, ECC145 and ECC188, resulted in a marked decrease in the total unsaturated fatty acids and i

Polyunsaturated fatty acids increase neutrophil adherence and integrin receptor expression.

Fish oils are abundant in polyunsaturated fatty acids of the n-3 series (in particular eicosapentaenoic, 20:5 and docosahexaenoic acid, 22:6). Such fatty acids are generally considered to be beneficial in the prevention of cardiac disease and to have anti-inflammatory properties. Neutrophil adherence is an essential early event in an acute inflammatory response, and we have demonstrated that both 20:5 and 22:6 stimulate adherence in vitro. Arachidonic acid (20:4, n-6) was also stimulatory. Significant simulation of adherence was seen from 5 to 80 microM (nontoxic concentrations) 22:6, 20:5, or

RBC and WBC fatty acid composition following consumption of an omega 3 supplement: lessons for future clinical trials.

BACKGROUND: Results from increasing numbers of in vitro and in vivo studies have demonstrated that omega 3 fatty acids incorporated in cell culture media or in the diet of the animals can suppress the growth of cancers. When human clinical trials are initiated to determine the ability of omega 3 fatty acids to alter growth or response to chemotherapeutic interventions of cancers, it will be essential to determine the omega 3 intake of individuals in the trial to determine compliance with consumption of the supplement and to correlate with endpoints of efficacy. We wondered if the fatty acid co

Fatty acid composition in major depression: decreased omega 3 fractions in cholesteryl esters and increased C20: 4 omega 6/C20:5 omega 3 ratio in cholesteryl esters and phospholipids.

Recently, there were some reports that major depression may be accompanied by alterations in serum total cholesterol, cholesterol ester and omega 3 essential fatty acid levels and by an increased C20: 4 omega 6/C20: 5 omega 3, i.e., arachidonic acid/eicosapentaenoic, ratio. The present study aimed to examine fatty acid composition of serum cholesteryl esters and phospholipids in 36 major depressed, 14 minor depressed and 24 normal subjects. Individual saturated (e.g., C14:0; C16:0, C18:0) and unsaturated (e.g., C18:1, C18:2, C20:4) fatty acids in phospholipid and cholesteryl ester fractions we

Altered fatty acid metabolism in patients with angiographically documented coronary artery disease.

Plasma lipids and fatty acids have been linked to coronary artery disease (CAD), and linoleic acid deficiency has been proposed as a risk factor for cardiovascular disease, but few studies have considered their multivariate effects or found the biochemical shifts associated with abnormal fatty acid metabolism or essential fatty acid (EFA) deficiency. We studied fatty acid patterns associated with CAD using high-resolution capillary column gas-liquid chromatography to analyze fasting plasma from 47 patients with angiographically documented CAD and 56 reference subjects. CAD patients exhibited a

Effect of dietary fat on the fatty acid composition and fertilizing ability of fowl semen.

Broiler breeder roosters received two diets, containing either 5% salmon oil (SO) or 5% corn oil (CO). The diets differed essentially in their polyunsaturated fatty acid (PUFA) composition, with n-6:n-3 fatty acid ratios of 41.6 in SO and 1.5 in CO. The effects of these diets on the fatty acid composition of spermatozoa and seminal plasma, and on fertility evaluated after artificial insemination were observed. Whatever the diet, the fatty acid composition of spermatozoa showed notable amounts of 20:4n-6 (5-9%) and 22:4n-6 (15-21%). These essential fatty acids were not detected in the diets and

Glyoxylate detoxification is an essential function of malate synthase required for carbon assimilation in Mycobacterium tuberculosis.

The glyoxylate shunt is a metabolic pathway of bacteria, fungi, and plants used to assimilate even-chain fatty acids (FAs) and has been implicated in persistence of Mycobacterium tuberculosis (Mtb). Recent work, however, showed that the first enzyme of the glyoxylate shunt, isocitrate lyase (ICL), may mediate survival of Mtb during the acute and chronic phases of infection in mice through physiologic functions apart from fatty acid metabolism. Here, we report that malate synthase (MS), the second enzyme of the glyoxylate shunt, is essential for in vitro growth and survival of Mtb on even-chain

Effects of Industrial Boiling on the Nutritional Profile of Common Octopus (Octopus vulgaris).

Industrial cooking of common octopus (Octopus vulgaris) under well-established procedures is advantageous for current consumers, which demand healthy and convenient food. This work aimed to evaluate the effect of industrial water boiling, without the addition of salt, on the nutritional profile of common octopus. True retentions (TRs) were calculated for essential nutrients and toxic elements. After boiling, the moisture content decreased, resulting in a concentration of other constituents (protein, fat, fatty acids, majority of amino acids, phosphorus, zinc, and iodine). High TRs were obtaine

Acetylcholine and Royal Jelly Fatty Acid Combinations as Potential Dry Eye Treatment Components in Mice.

Dry eye is a multifactorial disease characterized by ocular discomfort and visual impairment. Our previous studies have shown that royal jelly (RJ) has restored the capacity for tear secretion by modulating muscarinic calcium signaling. RJ contains acetylcholine, which is a major cholinergic neurotransmitter, and a unique set of fatty acids with C 8 to 12 chains, which are expected to be associated with health benefits. The purpose of the present study was to investigate the active components involved in tear secretion capacity, focusing on acetylcholine and fatty acids in RJ. Using the stress

Dietary effects on brain fatty acid composition: the reversibility of n-3 fatty acid deficiency and turnover of docosahexaenoic acid in the brain, erythrocytes, and plasma of rhesus monkeys.

Rhesus monkeys given pre- and postnatal diets deficient in n-3 essential fatty acids develop low levels of docosahexaenoic acid (22:6 n-3, DHA) in the cerebral cortex and retina and impaired visual function. This highly polyunsaturated fatty acid is an important component of retinal photoreceptors and brain synaptic membranes. To study the turnover of polyunsaturated fatty acids in the brain and the reversibility of n-3 fatty acid deficiency, we fed five deficient juvenile rhesus monkeys a fish oil diet rich in DHA and other n-3 fatty acids for up to 129 weeks. The results of serial biopsy sam

Chronic effects of perchloroethylene on the composition of lipid and acyl groups in cerebral cortex and hippocampus of the gerbil.

Continuous exposure of Mongolian gerbils to perchloroethylene (PCE) (120 ppm) for 12 months in an inhalation chamber caused no changes in body or brain weights. The protein content, the concentration of lipid phosphorus or cholesterol were unaltered in the cerebral cortex and hippocampus. However, a small change in the fatty acid pattern of phospholipid was observed. In the phosphatidylethanolamine of cerebral cortex and hippocampus a decrease was found among the long-chain, linolenic acid-derived, fatty acids. The ratio 22:4 (N-6)/22:5 (N-3) was increased, indicating a shift towards the corre

Delta6-desaturase (FADS2) deficiency unveils the role of omega3- and omega6-polyunsaturated fatty acids.

Mammalian cell viability is dependent on the supply of the essential fatty acids (EFAs) linoleic and alpha-linolenic acid. EFAs are converted into omega3- and omega6-polyunsaturated fatty acids (PUFAs), which are essential constituents of membrane phospholipids and precursors of eicosanoids, anandamide and docosanoids. Whether EFAs, PUFAs and eicosanoids are essential for cell viability has remained elusive. Here, we show that deletion of delta6-fatty acid desaturase (FADS2) gene expression in the mouse abolishes the initial step in the enzymatic cascade of PUFA synthesis. The lack of PUFAs an

Rewriting Inflammation in IBD: Lipidomics from Pathogenesis to Clinical Application.

Lipids (sphingolipids, fatty acids, phospholipids, and lipoproteins) are vital to intestinal barrier integrity, as precursors for pro-inflammatory and pro-resolving mediators and undergo remodelling through host microbiome interactions. Accumulating evidence implicates the Western diet-high in long-chain saturated and omega-6 polyunsaturated fatty acids and low in omega-3-in both the onset and progression of IBD. In contrast, microbiota derived lipid metabolites, including short-chain fatty acids and secondary bile acids, contribute to mucosal homeostasis and immune regulation. This review is

Raman Spectroscopic Analysis Reveals Abnormal Fatty Acid Composition in Tumor Micro- and Macroenvironments in Human Breast and Rat Mammary Cancer.

Fatty acids play essential roles in the growth and metastasis of cancer cells. To facilitate their avid growth and proliferation, cancer cells not only alter the fatty acid synthesis and metabolism intracellularly and extracellularly, but also in the macroenvironment via direct or indirect pathways. We report here, using Raman micro-spectroscopy, that an increase in the production of polyunsaturated fatty acids (PUFAs) was identified in both cancerous and normal appearing breast tissue obtained from breast cancer patients and tumor-bearing rats. By minimizing confounding effects from mixed che
