Disseminated histoplasmosis in an African pygmy hedgehog.

CASE DESCRIPTION: A 2-year-old captive-bred sexually intact female African pygmy hedgehog (Atelerix albiventris) was evaluated because of vague signs of illness including inappetence, weakness, lethargy, and weight loss over a 20-day period. CLINICAL FINDINGS: Abnormalities detected via initial clinicopathologic analyses included anemia, thrombocytopenia, leukopenia, hypoproteinemia, and hypoglycemia. Results of a fecal flotation test were negative. Three weeks after the initial evaluation, splenomegaly was detected via palpation and ultrasonography. TREATMENT AND OUTCOME: The hedgehog was t

Reproductive responses of beef heifers after concurrent administration of vaccines, anthelmintic and progestogen.

A study was conducted to determine whether concurrent administration of vaccines for reproductive diseases and an anthelmintic, given immediately prior to estrus synchronization with a progestogen, would affect subsequent estrous response and fertility in beef heifers. Two hundred ninety-five yearling crossbred beef heifers on 3 ranches in Oregon and Kansas were assigned to 1 of 2 treatments. Control heifers were vaccinated and dewormed 30 d prior to the anticipated breeding. Treated heifers received the same vaccination and deworming products 9 d prior to breeding. All heifers in both groups

Chemotherapy for larval echinococcosis in animals and humans: report of a workshop.

Mebendazole, its fluorine analogue flubendazole, and other benzimidazole derivatives are active against many gastrointestinal and tissue-stage helminths. This article reviews the published literature and proceedings of a workshop on the use of benzimidazoles against larval echinococcosis (hydatid disease). Orally administered high doses (30-50 mg/kg body weight) of mebendazole given daily for 20-90 days to rodents or sheep infected with larval Echinococcus granulosus cause damage of destruction of the cyst wall, loss of cyst fluid, and death of protoscolices. Similar treatment of rodents infec

Newly identified parasitic nematode beta-tubulin alleles confer resistance to benzimidazoles.

Infections by parasitic nematodes cause large health and economic burdens worldwide. We use anthelmintic drugs to reduce these infections. However, resistance to anthelmintic drugs is extremely common and increasing worldwide. It is essential to understand the mechanisms of resistance to slow its spread. Recently, four new parasitic nematode beta-tubulin alleles have been identified in benzimidazole (BZ) resistant parasite populations: E198I, E198K, E198T, and E198stop. These alleles have not been tested for the ability to confer resistance or for any effects that they might have on organismal

Removal of adult cyathostomins alters faecal microbiota and promotes an inflammatory phenotype in horses.

The interactions between parasitic helminths and gut microbiota are considered to be an important, although as yet incompletely understood, factor in the regulation of immunity, inflammation and a range of diseases. Infection with intestinal helminths is ubiquitous in grazing horses, with cyathostomins (about 50 species of which are recorded) predominating. Consequences of infection include both chronic effects, and an acute inflammatory syndrome, acute larval cyathostominosis, which sometimes follows removal of adult helminths by administration of anthelmintic drugs. The presence of cyathosto

Disseminated Strongyloides stercoralis infection in a dog following long-term treatment with budesonide.

CASE DESCRIPTION: A 1.5-year-old 1.5-kg (3.3-lb) castrated male Pomeranian was examined because of a 10-month history of diarrhea characterized by hematochezia and weight loss and an acute onset of respiratory distress (ie, tachypnea and dyspnea). A presumptive diagnosis of inflammatory bowel disease had been made previously, and the dog had been treated with budesonide and tylosin but continued to have diarrhea and weight loss. CLINICAL FINDINGS: On initial examination, the dog was weak and slightly obtunded. Thoracic radiography revealed a moderate to severe, diffuse, unstructured interstit

Discovery of genomic intervals that underlie nematode responses to benzimidazoles.

Parasitic nematodes impose a debilitating health and economic burden across much of the world. Nematode resistance to anthelmintic drugs threatens parasite control efforts in both human and veterinary medicine. Despite this threat, the genetic landscape of potential resistance mechanisms to these critical drugs remains largely unexplored. Here, we exploit natural variation in the model nematodes Caenorhabditis elegans and Caenorhabditis briggsae to discover quantitative trait loci (QTL) that control sensitivity to benzimidazoles widely used in human and animal medicine. High-throughput phenoty

The Anti-helminthic Compound Mebendazole Has Multiple Antifungal Effects against Cryptococcus neoformans.

Cryptococcus neoformans is the most lethal pathogen of the central nervous system. The gold standard treatment of cryptococcosis, a combination of amphotericin B with 5-fluorocytosine, involves broad toxicity, high costs, low efficacy, and limited worldwide availability. Although the need for new antifungals is clear, drug research and development (R&D) is costly and time-consuming. Thus, drug repurposing is an alternative to R&D and to the currently available tools for treating fungal diseases. Here we screened a collection of compounds approved for use in humans seeking for those with anti-c

Comparison of calculation methods used for the determination of anthelmintic resistance in sheep in a temperate continental climate.

This study compared results obtained with five different fecal egg count reduction (FECR) calculation methods for defining resistance to ivermectin, fenbendazole, and levamisole in gastrointestinal nematodes of sheep in a temperate continental climate: FECR1 and FECR2 used pre- and posttreatment fecal egg count (FEC) means from both treated and control animals, but FECR1 used arithmetic means, whereas FECR2 used geometric means; FECR3 used arithmetic means for pre- and posttreatment FECs from treated animals only; FECR4 was calculated using only arithmetic means for posttreatment FECs from tre

[Biology, pathogenicity, diagnosis and control of Ancylostoma caninum].

Infections with Ancylostoma caninum are transmitted orally or percutaneously. The transmission of infectious stages with the milk of particular importance for the distribution of the species. It occurs during the dissemination of larvae that follows every infection as well as after reactivation of resting somatic larvae in the bitch at the end of the pregnancy. The galactogenic transmission of larvae occurs even when, due to existing immunity, no patent infections develop in the bitch. Immunity does not or only to a low extent influence impatient infections or the migration of reactivated soma

Reversibility of clinical and computed tomographic lesions mimicking pulmonary fibrosis in a young cat.

BACKGROUND: In humans with idiopathic pulmonary fibrosis (IPF), specific thoracic computed tomographic (CT) features in the correct clinical context may be used in lieu of histologic examination. Cats develop an IPF-like condition with similar features to humans. As few cats have invasive lung biopsies, CT has appeal as a surrogate diagnostic, showing features consistent with architectural remodeling supporting "end-stage lung". CASE PRESENTATION: A 1-year-old female spayed Domestic Shorthair cat presenting with progressive respiratory clinical signs and thoracic CT changes (reticular pattern

Survey of management practices related to parasite control from ranched North American bison (Bison bison) in the southern United States.

Anthelmintic resistance (AR) in gastrointestinal nematodes (GIN) is an emerging concern in grazing livestock, including the North American bison (Bison bison). Nevertheless, little is known about routine parasite control practices in ranched bison herds. This cross-sectional survey of bison producers in 22 states across the southern and central United States characterized herd demographics, pasture management, nutrition, animal sourcing, and anthelmintic use. Seventy-nine producers from 15 states responded, most managing herds on fewer than 300 acres. Parasite control was predominantly drug-ce

The threat of reduced efficacy of anthelmintics against gastrointestinal nematodes in sheep from an area considered anthelmintic resistance-free.

BACKGROUND: The worldwide increased difficulty to combat gastrointestinal nematode (GIN) infection in sheep, due to progressing anthelmintic resistance (AR), calls for an enhanced and standardized implementation of early detection of AR. This study provides a snapshot of the current AR status against benzimidazoles and macrocyclic lactones in southern Italy, generated with standardized techniques. METHODS: On 10 sheep farms, the efficacy of albendazole (ALB) and either eprinomectin (EPR) or ivermectin (IVM) was evaluated based on the faecal egg count reduction test (FECRT) performed with the

Spontaneous tricuspid valve chordal rupture in a dog with severe, irreversible pulmonary hypertension caused by Angiostrongylus vasorum infection.

BACKGROUND: The adult worms of Angiostrongylus vasorum reside in the pulmonary artery of dogs and can lead to cardiac, respiratory, and central neurologic signs. Due to luminal obstruction and perivascular inflammation of the pulmonary artery branches, pulmonary hypertension can arise. Pulmonary hypertension, in turn, can lead to severe damage of the right-sided cardiac structures, leading to right ventricular remodeling and tricuspid valve regurgitation. CASE PRESENTATION: An 8-year-old neutered female English Cocker Spaniel was presented to the author's institution because of abdominal dist

Real-time PCR assays for monitoring benzimidazole resistance-associated mutations in Ancylostoma caninum.

Frequent and broad application of anthelmintic drugs for treatment of intestinal parasite infection has led to drug resistance that often renders whole populations of livestock unresponsive to treatment. Therefore, it is important to detect mutations associated with drug resistance before it becomes clinically manifest. To monitor developing drug resistance against benzimidazoles (BZ), we developed real-time PCR assays and applied them to analyse the beta-tubulin isotype-1 gene of the hookworm Ancylostoma caninum, an important parasite of dogs. Previously, we developed PCR assays to monitor co

Helminth infections in alpacas (Vicugna pacos), husbandry and worm control practices in South American Camelids in Italy.

The recent global expansion of alpaca breeding, including in Italy, has driven growing interest in this species, making further research necessary to support their health and management. Gastrointestinal helminths are among the main health concerns, causing severe disease that affects welfare and productivity. In Europe, data on helminth infections in South American Camelids (SACs) are still limited. This study aimed to assess the prevalence, distribution, and risk factors for helminth infections in alpacas in Italy, and evaluate the husbandry and worm control practices through a structured qu
