With global aging, the prevalence of calcific aortic valve stenosis (CAVS) has significantly increased, and even mild CAVS elevates mortality risk, highlighting an urgent need for preventive strategies. In this study, we analyzed 153,312 participants from the UK Biobank. We found that a higher Life's Essential 8 (LE8) score was independently associated with a decreased CAVS risk [hazard ratio (HR)/standard deviation (SD): 0.72, 95% confidence interval (CI): 0.68-0.76, P < 2E-16], whereas a higher genetic risk score was independently associated with an increased CAVS risk (HR/SD: 1.63, 95% CI:

BACKGROUND: The relationship between intrinsic capacity (IC), genetic susceptibility, and CVD outcomes in individuals with diabetes remains unclear.
METHODS: Data were obtained from 18 333 participants with diabetes (58.63 ± 7.54 years). The IC deficit score was categorised into five levels: 0, 1, 2, 3, and 4+. Outcomes included: hypertension, IHD, stroke, PAD, HF, and AF. Genetic risk scores (PRS) were constructed based on previously identified single nucleotide polymorphisms (SNPs) associated with these outcomes.
RESULTS: Over a follow-up period of 9.91 to 13.43 years. Compared to those wi

BACKGROUND: To explore the relationship between family history of esophageal cancer, SNPs, and the risk of esophageal squamous cell carcinoma (ESCC), we performed a population-based case-control study and developed a genetic family history-related risk (GFR) score and non-family history-related risk (GnFR) score to quantify the cumulative number of risk genotypes carried by each individual.
METHODS: We used data of 700 patients with nonfamilial ESCC, 341 patients with familial ESCC, 1,445 controls without a family history of esophageal cancer, and 319 controls with a family history. We genoty

BACKGROUND: Hereditary factors, including single genetic variants and family history, can be used for targeting colorectal cancer (CRC) screening, but limited data exist on the impact of polygenic risk scores (PRS) on risk-based CRC screening.
METHODS: Using longitudinal health and genomics data on 453,733 Finnish individuals including 8801 CRC cases, we estimated the impact of a genome-wide CRC PRS on CRC screening initiation age through population-calibrated incidence estimation over the life course in men and women.
RESULTS: Compared to the cumulative incidence of CRC at age 60 in Finland

BACKGROUND: Genome-wide association studies have established 11 genes for late-onset Alzheimer's disease (AD). We investigated whether these genes jointly affect cognition in a nondemented population and improve prediction of AD.
METHODS: In 5171 nondemented people (age 45-99 years) from the population-based Rotterdam Study, we genotyped APOE-ε4 and single nucleotide polymorphisms from the genes CLU, PICALM, BIN1, CR1, ABCA7, MS4A6A, MS4A4E, CD2AP, EPHA1, and CD33. We constructed a genetic risk score by adding the risk alleles per individual weighted by the reported effect sizes. All people u

Prostate cancer (PCa) is the second most common cancer among men worldwide. Its etiology remains largely unknown compared to other common cancers. We have developed a risk stratification model combining environmental factors with family history and genetic susceptibility. 818 PCa cases and 1,006 healthy controls were compared. Subjects were interviewed on major lifestyle factors and family history. Fifty-six PCa susceptibility SNPs were genotyped. Risk models based on logistic regression were developed to combine environmental factors, family history and a genetic risk score. In the whole mode

Precision healthcare aims to tailor disease prevention and early detection to individual risk. Prostate cancer screening may benefit from genomics-informed approaches. We developed and validated the P-CARE model, a prostate cancer risk prediction tool combining a polygenic score, family history and genetic ancestry, using data from over 585,000 male participants in the Million Veteran Program. The model was externally validated in diverse cohorts and implemented via a blended genome-exome assay for clinical use. Here we show that the P-CARE model identifies clinically meaningful gradients of p

BACKGROUND: Clinical factors are used to estimate late complication risk in adults after atrial switch operation (AtrSO) for transposition of the great arteries (TGA), but heterogeneity in clinical course remains. We studied whether common genetic variants are associated with outcome and add value to a clinical risk score in TGA-AtrSO patients.
METHODS AND RESULTS: This multicenter study followed 133 TGA-AtrSO patients (aged 28 [IQR 24-35] years) for 13 (IQR 9-16) years and examined the association of genome-wide single-nucleotide polymorphisms (SNPs) with a composite endpoint of symptomatic

INTRODUCTION: Alzheimer's disease (AD), and more specifically late-onset Alzheimer's disease (LOAD), represents a considerable challenge in terms of early and timely diagnosis and treatment. Early diagnosis is crucial to improve the efficacy of the therapies and patients' quality of life. The current challenge is to accurately identify at-risk individuals before the manifestations of the first symptoms of AD.
METHODS AND RESULTS: Here, we present an improved model for LOAD risk prediction, which applies the k-nearest neighbors (KNN) algorithm. We have achieved a sensitivity of 0.80 and an are

BACKGROUND: Previously we reported a genetic risk score significantly improved PTSD prediction among a trauma-exposed civilian population. In the current study, we sought to assess this prediction among a trauma-exposed military population.
METHODS: We examined current PTSD diagnosis and PTSD symptom severity among a random sample of 1042 community-based US military veterans. Main effects and interaction effects were assessed for PTSD genetic risk by trauma exposure using cross-product terms for PTSD x trauma exposures, including combat, lifetime trauma, and adverse childhood exposures. Psoriasis is a chronic inflammatory skin disease with substantial impact on quality of life. This study aimed to investigate the relationship between flavonoid intake and the risk of developing psoriasis, and to assess whether genetic predisposition modifies this association. This prospective study included 121 299 UK Biobank participants free of psoriasis at baseline who completed at least two Oxford WebQ dietary recalls. Incident psoriasis was identified using self-reports, primary care data, and hospital records. Intake of total flavonoids and five subclasses (anthocyanins, flavanones, flav

BACKGROUND: Glaucoma is a leading cause of worldwide irreversible blindness. Considerable uncertainty remains regarding the association between a variety of phenotypes and the genetic risk of glaucoma, as well as the impact they exert on the glaucoma development.
METHODS: We investigated the associations of genetic liability for primary open angle glaucoma (POAG) with a wide range of potential risk factors and to assess its impact on the risk of incident glaucoma. The phenome-wide association study (PheWAS) approach was applied to determine the association of POAG polygenic risk score (PRS) w

With increasing access to individual-level data from genome-wide association studies, it is now common for researchers to have individual-level data of some traits, whereas for other traits only publicly released summary statistics are available due to privacy and safety concerns. Current genetic-correlation methods require both traits to be in the same format. When one trait has individual-level data and the other has only summary statistics, researchers often convert individual-level data to summary statistics and then apply summary-based methods, which is inefficient and can lose informatio

Osteoarthritis (OA) is a heterogeneous joint disease in which patients differ widely in onset, progression, pain burden, and inflammatory activity, yet clinical tools for early risk stratification and mechanism-informed subtyping remain limited. Human genetic studies have identified many OA-associated loci, but translation into deployable biomarkers has been slow because most signals are polygenic, largely noncoding, and dependent on tissue and cell state. Despite rapid progress in OA genomics, a major remaining challenge is how to systematically convert genetic discoveries into clinically act

Dyslexia is a neurodevelopmental disorder typically diagnosed in school-aged children and associated with poor school performance and lower levels of educational attainment (EA). Here, we aimed to test the shared genetic architecture between dyslexia and EA, to dissect the polygenic contribution of dyslexia by its relationship with EA and to assess how these genetic partitions influence school performance, early manifestation of psychopathology and related traits. We first confirmed a negative genetic correlation between dyslexia and EA (rg = -0.186, SE = 0.019, P = 1.75E-22). Then, polygenic

PURPOSE: African ancestry and family history (FHx) of prostate cancer (CaP) are among the few established CaP risk factors. Few studies have evaluated the association of FHx of CaP with the risk of this disease in African men.
METHODS: Using the Men of African Descent and Carcinoma of the Prostate (MADCaP) network, we evaluated the association of self-reported FHx of CaP in fathers and brothers in a case-control study of 2,505 prostate cancer cases and 2,222 age-matched controls ascertained from seven centers across Africa. We compared the association of FHx and a 451-SNP polygenic risk score

Twenty-five years after the draft of the human genome sequence, genomics has revolutionized many areas of biology, the diagnosis of familial and early-onset diseases, and the development of targeted therapies, particularly in oncology. Genomic knowledge has had little effect on the diagnosis and treatment of most common adult-onset conditions. Genomewide association studies have revealed that for these conditions, thousands of genetic variants of small effect underpin the polygenic nature of risk. When incorporated into polygenic risk scores, these variants predict risk; however, polygenic ris

BACKGROUND AND OBJECTIVES: Genetic variants affect both Parkinson disease (PD) risk and manifestations. Although genetic information is of potential interest to patients and clinicians, genetic testing is rarely performed during routine PD clinical care. The goal of this study was to examine interest in comprehensive genetic testing among patients with PD and document reactions to possible findings from genome sequencing in 2 academic movement disorder clinics.
METHODS: In 203 subjects with PD (age = 63 years, 67% male), genome sequencing was performed and filtered using a custom panel, inclu
