The effect of seed priming on yield and germination properties of quinoa (Chenopodium quinoa Willd).

Seed priming is a widely used pre-germination technique that enhances sprouting efficiency, synchrony, and the initial development of plantlets, particularly under suboptimal environmental conditions. This study aimed to evaluate the impact of different seed priming treatments on the germination, growth, and yield performance of quinoa (Chenopodium quinoa Willd.) through a series of laboratory, greenhouse, and field experiments conducted during the 2024 cropping season. The laboratory experiment was arranged in a completely randomized design (CRD) with four replications. Treatments included po

Low Temperature-ROS-Hormones Co-Regulation of Seed Dormancy Release and Germination in Xanthoceras sorbifolium.

Seed dormancy is an important adaptive mechanism in plants. However, some seeds, such as those of Xanthoceras sorbifolium (a valuable economic and medicinal species), exhibit strong dormancy, resulting in a very low natural germination rate. This work investigated the mechanism by which low temperatures (LT: -20 °C storage for 60 days) release seed dormancy and promote germination. This was achieved by examining seed germination conditions, applying scanning electron microscopy (SEM), and measuring physiological indicators of seeds and the hormone levels. Targeted metabolomic analysis of sugar

Coordinated Dhurrin Turnover Is Associated with HCN Potential and Salt Tolerance During Sorghum Seed Germination.

Cyanogenic glycosides function as defense compounds and nitrogen reservoirs in sorghum, but their roles during seed germination under salt stress remain unclear. Here, two sorghum cultivars differing in salt tolerance were compared using physiological, biochemical, pharmacological and transcriptomic analyses to investigate the relationship between dhurrin metabolism and germination performance under salinity. Salt stress inhibited germination and early seedling growth in both cultivars, whereas the tolerant genotype SB19 maintained higher germination rates, earlier radicle emergence and strong

Conserved storage-carbohydrate metabolic modules are rewired during germination of Trichoderma asperelloides and other Sordariomycetes.

Conidial germination requires rapid mobilization and reorganization of storage carbohydrates, yet the network architecture underlying this process remains poorly defined in filamentous fungi. Using quantitative GC-MS/MS profiling, we provide the first quantitative identification of major soluble sugar species across four germination stages of Trichoderma asperelloides T203 and compared them with four representative models in the Sordariomycetes (Metarhizium anisopliae, Cordyceps militaris, Fusarium graminearum, and Neurospora crassa). In T. asperelloides, mannitol was the most prevalent measur

Temporal transcriptomic and lipidomic analysis reveals multi-omics dynamic profiles of B. napus seed germination.

BACKGROUND: Lipids represent the major storage reserve in Brassica napus seeds. During germination, lipid mobilization delivers indispensable energy to support seedling establishment, thereby profoundly influencing germination vigor and subsequent seedling growth capacity. Although lipid mobilization is fundamentally required for successful rapeseed germination, the temporal coordination between transcriptional reprogramming and lipid metabolic conversion remains largely elusive, and the underlying molecular regulatory network remains to be systematically deciphered. RESULTS: We conducted an

Asymbiotic Seed Germination and In Vitro Seedling Development of the Endangered Orchid Species Cypripedium guttatum.

Cypripedium guttatum is a highly restricted terrestrial orchid that faces increasing endangerment owing to its habitat destruction and illegal collection. Compared to epiphytic orchids, terrestrial orchids such as C. guttatum have harder seed coats and more demanding in vitro germination conditions. This study aimed to develop an effective in vitro propagation system for C. guttatum to aid in its conservation. Seeds from mature capsules were subjected to various conditions, including sterilization using 1% sodium hypochlorite (NaOCl) and different light conditions, culture media, hormones, and

Current Advancements in the Molecular Mechanism of Plasma Treatment for Seed Germination and Plant Growth.

Low-temperature atmospheric pressure plasma has been used in various fields such as plasma medicine, agriculture, food safety and storage, and food manufacturing. In the field of plasma agriculture, plasma treatment improves seed germination, plant growth, and resistance to abiotic and biotic stresses, allows pesticide removal, and enhances biomass and yield. Currently, the complex molecular mechanisms of plasma treatment in plasma agriculture are fully unexplored, especially those related to seed germination and plant growth. Therefore, in this review, we have summarized the current progress

Cytosolic copper is a major modulator of germination, development and secondary metabolism in Streptomyces coelicolor.

Streptomycetes are important biotechnological bacteria with complex differentiation. Copper is a well-known positive regulator of differentiation and antibiotic production. However, the specific mechanisms buffering cytosolic copper and the biochemical pathways modulated by copper remain poorly understood. Here, we developed a new methodology to quantify cytosolic copper in single spores which allowed us to propose that cytosolic copper modulates asynchrony of germination. We also characterised the SCO2730/2731 copper chaperone/P-type ATPase export system. A Streptomyces coelicolor strain muta

In vitro germination and biochemical profiling of Brassica napus in response to biosynthesised zinc nanoparticles.

With the progression of nanotechnology, the use of nanoparticles (NPs) in consumer products has increased dramatically and green synthesis is one of the cheapest and eco-friendly methods to obtain non-hazardous NPs. In the current research zinc (Zn) NPs synthesis was carried out by using the fresh and healthy leaves of Mentha arvensis L. followed by characterisation through ultraviolet (UV)-visible spectroscopy, X-ray diffraction (XRD) and scanning electron microscopy (SEM). UV-visible spectroscopy confirmed the green synthesis of ZnNPs, while XRD confirmed the size of NPs, which was 30-70 nm.

Tracking celiac disease-triggering peptides and whole wheat flour quality as function of germination kinetics.

Germination is already a well-accepted process by consumers with many products made from sprouted seeds or containing limited amounts of flour form sprouted grains. The present work aimed assessing the usefulness of germination in reducing gluten peptides associated with celiac disease, at the same time evaluating some technological features of the obtained germinated wheat. In the first part of the work, celiac disease (CD)-triggering peptides were tracked as a function of germination kinetics (from day 1 to day 6). Using simulated gastrointestinal digestion and liquid chromatography coupled

Germination, Outgrowth, and Vegetative-Growth Kinetics of Dry-Heat-Treated Individual Spores of Bacillus Species.

DNA damage kills dry-heated spores of Bacillus subtilis, but dry-heat-treatment effects on spore germination and outgrowth have not been studied. This is important, since if dry-heat-killed spores germinate and undergo outgrowth, toxic proteins could be synthesized. Here, Raman spectroscopy and differential interference contrast microscopy were used to study germination and outgrowth of individual dry-heat-treated B. subtilis and Bacillus megaterium spores. The major findings in this work were as follows: (i) spores dry-heat-treated at 140°C for 20 min lost nearly all viability but retained th

Effectiveness of Germination on Protein Hydrolysis as a Way To Reduce Adverse Reactions to Wheat.

In this work, the aim is to study the effectiveness of germination on wheat protein degradation, with a specific focus on proteins involved in adverse reactions to wheat. The effects of 8 days of germination at 25 °C on the chemical composition and the protein profile were determined. Germination did not have a significant effect on starch, protein, lipid, and ash contents. General protein profile, as indicated by SDS-PAGE analysis, revealed that germination induced a relevant degradation in protein fraction. After in vitro gastrointestinal digestion, gluten peptides involved in celiac disease

Rice (Oryza sativa L.) Seed Sterilization and Germination Enhancement via Atmospheric Hybrid Nonthermal Discharge Plasma.

We designed a system to produce atmospheric hybrid cold-discharge plasma (HCP) based on microcorona discharge on a single dielectric barrier and applied it to inactivate microorganisms that commonly attach the rice seed husk. The cold-plasma treatment modified the surface of the rice seeds, resulting in accelerated germination and enhanced water imbibition. The treatment can operate under air-based ambient conditions without the need for a vacuum. The cold-plasma treatment completely inactivated pathogenic fungi and other microorganisms, enhancing the germination percentage and seedling qualit

Effect of germination and autoclaving of sprouted finger millet and kidney beans on cyanide content.

Cyanide contents of locally purchased brown finger millet (Eleusine corocana L. Gaertner) and brown speckled kidney bean seeds (Phaseolus vulgaries var. Rose Coco) were determined using raw, germinated and autoclaved samples. The aim was to establish the extent of cyanide content increase resulting from the germination process and the effectiveness of the autoclaving process on the reduction of cyanide levels in the samples, for safety considerations. Autoclaving was carried out at 121degree C for 20 minutes. It was found that germination increased the cyanide content by 2.11 to 2.14 fold in f

Factors involved in the germination and inactivation of Bacillus anthracis spores in murine primary macrophages.

Since macrophages have been implicated in inhalation anthrax either for defense and/or as enablers for spore trafficking, their function has been further defined. Spores were efficiently taken up by primary mouse bone marrow-derived macrophages even in the absence of serum but a minimal amount was required for spore germination and subsequent inactivation. With 10% fetal bovine serum (FBS) virtually all of the spores germinated but when the concentration of FBS was lowered to 1.0% or less, or when 10% horse serum replaced FBS, only 50% of the spores were inactivated within 1 h with no subseque

Both trivalent and hexavalent chromium strongly alter in vitro germination and ultrastructure of kiwifruit pollen.

Due to its widespread industrial use, chromium is considered a dangerous environmental pollutant. It is known to inhibit plant growth and development. The present study provides the first evidence of the toxicity of this metal on the male haploid generation of a higher plant. Both Cr(III) and Cr(VI) species, supplied as CrCl(3) and CrO(3), respectively, exerted a strong dose-dependent inhibitory effect on kiwifruit pollen tube emergence and growth. Cr(III) resulted more effective than Cr(VI) in the 16-75microM interval; moreover, complete inhibition of germination was attained at much lower do

Expression of proteins and protein kinase activity during germination of aerial spores of Streptomyces granaticolor.

Dormant aerial spores of Streptomyces granaticolor contain pre-existing pool of mRNA and active ribosomes for rapid translation of proteins required for earlier steps of germination. Activated spores were labeled for 30 min with [35S]methionine/cysteine in the presence or absence of rifamycin (400 microg/ml) and resolved by two-dimensional electrophoresis. About 320 proteins were synthesized during the first 30 min of cultivation at the beginning of swelling, before the first DNA replication. Results from nine different experiments performed in the presence of rifamycin revealed 15 protein spo

Identification and characterization of a germination operon on the virulence plasmid pXO1 of Bacillus anthracis.

The spores of Bacillus anthracis, the agent of anthrax disease, germinate within professional phagocytes, such as murine macrophage-like RAW264.7 cells and alveolar macrophages. We identified a cluster of germination genes extending for 3608 nucleotides between the pag and atxA genes on the B. anthracis virulence plasmid pXO1. The three predicted proteins (40, 55 and 37 kDa in size) have significant sequence similarities to B. subtilis, B. cereus and B. megaterium germination proteins. Northern blot analysis of total RNA from sporulating cells indicated that the gerX locus was organized as a t

Action spectra for phytochrome A- and B-specific photoinduction of seed germination in Arabidopsis thaliana.

We have examined the seed germination in Arabidopsis thaliana of wild type (wt), and phytochrome A (PhyA)- and B (PhyB)-mutants in terms of incubation time and environmental light effects. Seed germination of the wt and PhyA-null mutant (phyA) was photoreversibly regulated by red and far-red lights of 10-1,000 micromol m-2 when incubated in darkness for 1-14 hr, but no germination occurred in PhyB-null mutant (phyB). When wt seeds and the phyB mutant seeds were incubated in darkness for 48 hr, they synthesized PhyA during dark incubation and germinated upon exposure to red light of 1-100 nmol
