Comparison of the effect of TAK-147 (zanapezil) and E-2020 (donepezil) on extracellular acetylcholine level and blood flow in the ventral hippocampus of freely moving rats.

The effects of zanapezil (TAK-147) and donepezil (E2020) on extracellular acetylcholine (ACh) levels were investigated by HPLC-microdialysis of ventral hippocampus (VH) in freely moving intact rats. The results showed that the basal ACh release rate in the VH is 116.7 +/- 12.4 to 158.4 +/- 22.86 fmol/20 microl. At 2, 5 and 10 mg/kg, single p.o., each drug increased ACh level by 9.4%, 106.5%, 50.8% (TAK-147) and 14.8%, 76.1%, 120.94% (E2020), respectively. The ED50 values were 4.52 mg/kg (1.43 - 14.29; R=0.52) and 4.07 mg/kg (1.77 - 9.37; R=0.985) for TAK-147 and E2020, respectively. Analysis o

The new cholinesterase inhibitors for Alzheimer's disease, Part 1: their similarities are different.

Three new cholinesterase inhibitors, donepezil, rivastigmine, and galantamine, all inhibit the enzyme AChE. Rivastigmine also inhibits BuChE, which could lead to additional benefits in late-stage Alzheimer's disease, but also cause more GI side effects at initiation of therapy. Galantamine is also an allosteric modulator of nicotinic receptors, which could lead to additional efficacy for attention and for behaviors mediated by neurotransmitters other than ACh. We are now entering an exciting era where the options for treating the devastating illness Alzheimer's disease are multiplying and crea

Improvement in sundowning in dementia with Lewy bodies after treatment with donepezil.

Sundowning, manifested as a recurring increase in restlessness and agitation in the evening, is described in a 71-year-old man with clinically diagnosed dementia with Lewy bodies. An objective measure of activity using the activity electronic monitoring technique indicated a marked increase in activity level during the evening compared to earlier in the day. After treatment with donepezil, a cholinesterase inhibitor, ratings of behavioural symptoms improved. In addition, there was a marked reduction in evening activity and an increase in daytime activity. Cognition and parkinsonism also improv

Molecular pathways: the quest for effective MAO-B inhibitors in neurodegenerative therapy.

Neurodegenerative diseases like Parkinson's and Alzheimer's are a global health challenge due to their progressive degeneration, leading to cognitive decline and motor dysfunction. Monoamine oxidase B (MAO-B) enzyme is implicated in neurodegeneration, and developing inhibitors could be a promising therapeutic strategy. This review explores MAO-B activity molecular pathways, evaluates MAO-B inhibitors in neurodegenerative therapy, identifies challenges, and suggests future research directions. This review synthesizes findings from a range of scientific literature, including experimental studies

Dissecting the Therapeutic Mechanisms of Sphingosine-1-Phosphate Receptor Agonism during Ischaemia and Reperfusion.

Sphingosine 1-phosphate (S1P) and S1P receptors (S1PR) regulate many cellular processes, including lymphocyte migration and endothelial barrier function. As neutrophils are major mediators of inflammation, their transendothelial migration may be the target of therapeutic approaches to inflammatory conditions such as ischaemia-reperfusion injury (IRI). The aim of this project was to assess whether these therapeutic effects are mediated by S1P acting on neutrophils directly or indirectly through the endothelial cells. First, our murine model of peritoneum cell recruitment demonstrated the abilit

Evaluation of the use of chemical pads to mimic latent fingermarks for research purposes.

Fingermark detection is in constant evolution, with new techniques being developed and existing ones being continuously optimised. Recently, researchers have begun to express interest in artificial fingermark secretions to overcome the issues arising from the variability of fingermark composition. Some of these artificial secretions have started to appear on the market in the form of pads that can be used to deposit fingermarks with a known and controlled composition. This study aimed at assessing the reliability of three commercially-available pads by comparing the results to those obtained b

Recent Highlights on Molecular Hybrids Potentially Useful in Central Nervous System Disorders.

Molecular hybridization is a recent strategy based on the covalent fusion of two or more pharmacophores to create a single molecule with multiple mechanisms of action, which represents an encouraging approach in the development of new drugs with potential therapeutic application in several pathologies. This review provides a comprehensive perspective of the most relevant advances in the development of hybrid molecules acting in the central nervous system. For instance, several opioid hybrids based on endogenous opioid peptides (e.g. enkephalins, deltorphins and endomorphins) have been develope

Retention time of chlorophacinone in black-tailed prairie dogs informs secondary hazards from a prairie dog rodenticide bait.

BACKGROUND: Secondary toxicity in mammals and birds that consume animals containing residues of anticoagulant rodenticides represents a persistent conflict between conservation, agriculture and environmental contamination. Chlorophacinone residues in black-tailed prairie dogs (Cynomys ludovicianus) represent a secondary exposure hazard to predatory and scavenging avian and mammalian species in the Central Plains of the United States, especially considering efforts to re-establish black-footed ferrets (Mustela nigripes). Rozol(®) Prairie Dog Bait (chlorophacinone 0.005%) is registered to contro

Donepezil in the treatment of narcolepsy.

Chronic excessive sleepiness impairs the daily life of patients suffering from narcolepsy. Antidepressants that enhance synaptic levels of noradrenaline and serotonin have been reported as having some therapeutic efficacy. For that reason, acetylcholinesterase inhibitors, which elevate acetylcholine levels, may have a protective function with respect to narcolepsy and cataplexy. The patient reported in this article received donepezil, 10 mg, for 3 consecutive months and showed an improvement of the Epworth Sleepiness Scale score from 20 up to 14.

Salicylates of intact Salix myrsinifolia plantlets do not undergo rapid metabolic turnover.

Salicylates, the main phenolic glucosides of northern willow (Salix spp.), play an important role in plant-herbivore interactions. Salicylates are labile metabolites that are thought to undergo metabolic turnover. Salicylates are synthesized from phenylalanine (Phe) via the shikimate pathway. 2-Aminoindan-2-phosphonic acid (AIP), a strong inhibitor of Phe ammonia-lyase (EC 4.3.1.5), was used to block the biosynthesis of salicylates. The aim of this study was to investigate long-term turnover of salicylates in intact micropropagated plantlets of Salix myrsinifolia Salisb. The biosynthesis of sa

Bracken carcinogens in the human diet.

The ubiquitous bracken fern (genus Pteridium) is the only higher plant known to cause cancer naturally in animals. In addition to the well-recognized syndromes of thiamine deficiency, acute haemorrhage associated with myeloid aplasia and blindness due to retinal degeneration, it causes neoplasia of the urinary bladder and in some circumstances, of the upper gut. In addition, it has been shown to cause neoplasia in a wide range of tissues in many experimental species. The major carcinogen (and the cause of the retinal degeneration and the myeloid aplasia) has been shown to be ptaquiloside (PT),

A new diuretic that does not reduce renal handling of uric acid in rats, S-8666.

The uric acid-retaining effects of diuretics were studied using sodium-restricted spontaneously hypertensive rats. Test agents were administered orally once a day for two weeks. Diuretic thiazides such as trichlormethiazide and hydrochlorothiazide and loop diuretics such as furosemide and indacrinone clearly reduced the renal function for uric acid excretion in treatment which produced major effects such as diuresis, saluresis and hypotension. However, a new diuretic with uricosuric activity, S-8666, developed in our laboratories, had no effect on the renal handling of uric acid at doses which

2-cyano-1,3-dicarbonyl compounds with antiallergic activity.

A number of 2-cyanoindan-1,3-diones and 3-cyano-4-hydroxycoumarins have been prepared and assessed for potential antiallergy activity as measured by their ability to inhibit passive cutaneous anaphylaxis in the rat, mediated by rat serum containing antigen specific IgE. The structural requirements for activity were similar not only for both series of compounds but also for the analogous 2-nitroindan-1,3-diones and 4-hydroxy-3-nitrocoumarins previously reported. The most active compounds were 2-cyano-5,6-diethylindan-1,3-dione (4e) and 3-cyano-6,7-diethyl-4-hydroxycoumarin (11h).

Interaction between rasagiline and Pueraria radix in in vitro models of parkinson's disease.

BACKGROUND: The management of Parkinson's disease (PD) remains challenging owing to the limitations of current therapies. Although rasagiline (RASA), a monoamine oxidase-B (MAO-B) inhibitor, offers advantages over levodopa, its efficacy as a monotherapy is moderate. Pueraria root (PR), a traditional East Asian medicine, has demonstrated neuroprotective potential against several neurodegenerative conditions. This study investigated the pharmacodynamic and pharmacokinetic interactions between RASA and PR to evaluate their potential as a combination therapy for PD. METHODS: We examined synergist

Targeting Lewy body dementia with neflamapimod-rasagiline hybrids.

Lewy body dementia (LBD) represents the second most common neurodegenerative dementia but is a quite underexplored therapeutic area. Nepflamapimod (1) is a brain-penetrant selective inhibitor of the alpha isoform of the mitogen-activated serine/threonine protein kinase (MAPK) p38α, recently repurposed for LBD due to its remarkable antineuroinflammatory properties. Neuroprotective propargylamines are another class of molecules with a therapeutical potential against LBD. Herein, we sought to combine the antineuroinflammatory core of 1 and the neuroprotective propargylamine moiety into a single m
