Reviewing the female accessory glands of insects: Where they come from, what they do, and where they are going.

Female accessory glands (AGs) are structures associated with the reproductive system of many arthropods. In insects, the glands are mostly related to oviposition by secreting egg coatings and adhesives. Nevertheless, the AGs are versatile concerning their function, and show a high diversity of chemical compounds. Here we aim to review the AGs in insects, highlighting their distribution among the orders, origins, location, chemical nature of the secretions, development, endocrine control, and morphological features. We also propose dividing the glands into epidermal and mesodermal, according to

A scorpion neurotoxin increases the potency of a fungal insecticide.

The low virulence of the insecticidal fungus Metarhizium anisopliae has stymied its widespread use in controlling insect pests. We show that high-level expression of an insect-specific neurotoxin from the scorpion Androctonus australis in hemolymph by M. anisopliae increases fungal toxicity 22-fold against tobacco hornworm (Manduca sexta) caterpillars and ninefold against adult yellow fever mosquitoes (Aedes aegypti) without compromising host specificity. Prelethal effects include reduced mobility and feeding of the insects targeted.

Animal models of anaphylaxis.

PURPOSE OF REVIEW: In this review we will focus on recent advances in the role of mast cells in the pathophysiology of insect allergy and the possible mechanisms of mast cell activation in anaphylaxis. RECENT FINDINGS: Anaphylactic reactions in the mouse can be induced by several independent pathways involving immunoglobulin E, immunoglobulin free light chains, or immunoglobulin G. There is considerable evidence that mast cells play a central role in anaphylactic reactions to insect stings. Mast cells can be directly activated by components of insect venom or after allergic sensitization. Of

Expression of defensin, cecropin, and transferrin in Aedes aegypti (Diptera: Culicidae) infected with Wuchereria bancrofti (Spirurida: Onchocercidae), and the abnormal development of nematodes in the mosquito.

The temporal expression of defensin, cecropin and transferrin was assessed in Aedes aegypti naturally refractory to Wuchereria bancrofti upon infection with this worm, in parallel to analysis of filarial development in the insect. Compared to controls, transcription of defensin and cecropin was higher in infected mosquitoes as soon as 2h post infection and peaked before 48h. Transferrin transcription was higher in infected mosquitoes at 24h, and at 48h was almost leveled to controls. At 72h and 7 days post infection, levels of all transcripts in infected insects decreased gradually and were si

[Individual protection against insect vectors].

Many diseases for which no vaccine is available are transmitted by insect and arthropod vectors, the main exceptions being yellow fever and Japanese encephalitis B. Treatment is less and less effective due to the development of chemoresistance to therapeutic and prophylactic drugs as is well-illustrated by malaria. One of the best methods of preventing these diseases is personal protection against insect bites. Personal protection measures can be divided into three categories which can be used separately or in combination : application of repellents to the skin, wearing clothes impregnated wit

Sulfonyl-acetohydrazide derivatives as juvenile hormone mimics to be insect growth regulators.

The need for targeted pest control strategies has led to the development of juvenile hormone (JH) mimics that selectively disrupt the life cycles of harmful insect species. Present study focuses on the synthesis, characterization and evaluation of sulfonyl-acetohydrazide derivatives (H1-H8) as novel JH mimics on two different insect species, with an emphasis on their insect-specific action. The yellow fever mosquito, Aedes aegypti and cabbage leaf borer, Spodoptera litura, were selected for this investigation. Our results indicate that while these compounds exhibit negligible effects on the de

Evaluation of Atriplex lines for selenium accumulation, salt tolerance and suitability for a key agricultural insect pest.

Thirty Atriplex lines were examined for potential habitat improvement and phytoremediation of selenium (Se) contaminated sites. Studies were conducted to determine the biomass production, Se accumulation, and resistance of each line to the beet armyworm, Spodoptera exigua, an agriculturally important insect. Plants were tested using three salinity treatments: (1) control, no Se; (2) NaCl and CaCl2 salts and 1 mg l(-1) Se (12.7 microM) added as sodium selenate; and (3) iso-osmotic to treatment 2 containing high concentrations of sulfate and I mg l(-1) Se added as sodium selenate. Isolation and Identification of Entomopathogenic Fungus GC23620 and Its Virulence and Control Efficacy Against Gynaephora qinghaiensis Larvae.

In June 2023, a larva of grassland caterpillar Gynaephora qinghaiensis naturally infected by an entomopathogenic fungus was collected from an alpine rangeland in Gangcha County, Haibei Tibetan Autonomous Prefecture, Qinghai Province. After laboratory isolation and cultivation, the pathogen was identified as Beauveria bassiana (designated as GC23620) based on morphological characteristics and ITS-rDNA sequence similarity analysis. The larvicidal efficacy of B. bassiana GC23620 against fourth-instar larvae of G. qinghaiensis were assessed using two inoculation methods in laboratory conditions. T

Novel selective and irreversible mosquito acetylcholinesterase inhibitors for controlling malaria and other mosquito-borne diseases.

We reported previously that insect acetylcholinesterases (AChEs) could be selectively and irreversibly inhibited by methanethiosulfonates presumably through conjugation to an insect-specific cysteine in these enzymes. However, no direct proof for the conjugation has been published to date, and doubts remain about whether such cysteine-targeting inhibitors have desirable kinetic properties for insecticide use. Here we report mass spectrometric proof of the conjugation and new chemicals that irreversibly inhibited African malaria mosquito AChE with bimolecular inhibition rate constants (k(inact)

Trypanosoma cruzi infection modulates the expression of Triatoma brasiliensis def1 in the midgut.

Antimicrobial peptides are an essential component of the insect immune system. One of the most ubiquitous is defensin, which has been identified in all examined insect orders. Triatoma brasiliensis (Heteroptera, Triatominae), the main Trypanosoma cruzi vector in semi-arid regions of north-eastern Brazil, expresses def1, a defensin encoding gene, predominantly in the anterior region (cardia and stomach) of the midgut. In the present study, we compared the transcript abundance of T. brasiliensis def1 in the anterior (stomach) and posterior midgut (small intestine) regions of naïve bugs with thos

Doru luteipes (Dermaptera: Forficulidae): Life History, Predatory Potential, and Rearing Method of Earwigs.

The earwig Doru luteipes (Scudder) (Dermaptera: Forficulidae) is recognized as the most frequently encountered predatory insect species in maize agroecosystems across maize-producing regions of Brazil and other South American countries. For over three decades, the earwig has been extensively investigated through both field surveys and controlled laboratory studies aiming at biological control of insect pests. Although augmentative biological control strategies involving the release of D. luteipes have been proposed, the currently available data on its predatory behavior and efficiency remain s

Chitosan-based strategies as eco-friendly solutions for controlling Brettanomyces bruxellensis contamination in wine production.

INTRODUCTION: This study investigates the antimicrobial activity of chitosan against Brettanomyces bruxellensis, a wine spoilage yeast responsible for producing volatile phenols that lead to undesirable sensory defects commonly referred to as "Brett" character. The most widely used antimicrobial compound in oenology is sulphur dioxide (SO2), due to its broad spectrum of action, but growing consumer demand for reduced chemical additives and evidence of Brettanomyces spp. resistance to it, have encouraged different alternative strategies. Among these, chitosan has been accepted for the control o

Impact of disinfectants on the intestinal bacterial symbionts and immunity of silkworm (Bombyx mori L.).

The insect egg surface can serve as a vehicle for vertical symbiont transmission from the maternal parent to its offspring. Hypochlorite and formaldehyde are two common disinfectants used for insect egg surface sterilization. Here, we explored the intestinal microecology and immune response profile of the silkworm Bombyx mori strain Dazao after disinfectant exposure by using high-throughput sequencing technology and real-time PCR analysis. After egg surface sterilization, no significant difference (P > 0.05) in overall body weight was observed among the control, sodium hypochlorite, and formal

Growth and Maintenance of Wolbachia in Insect Cell Lines.

The obligate intracellular microbe, Wolbachia pipientis (Rickettsiales; Anaplasmataceae), is a Gram-negative member of the alpha proteobacteria that infects arthropods and filarial worms. Although closely related to the genera Anaplasma and Ehrlichia, which include pathogens of humans, Wolbachia is uniquely associated with invertebrate hosts in the clade Ecdysozoa. Originally described in Culex pipiens mosquitoes, Wolbachia is currently represented by 17 supergroups and is believed to occur in half of all insect species. In mosquitoes, Wolbachia acts as a gene drive agent, with the potential t

Trans-complementation of polyhedrin by a stably transformed Sf9 insect cell line allows occ- baculovirus occlusion and larval per os infectivity.

Oral infection of insect larvae with baculovirus is an advantageous methodology for producing high levels of recombinant proteins and for achieving plague control. However, many recombinant baculoviruses express a foreign protein in lieu of the polyhedrin and hence do not form occlusion bodies (occ-), resulting in extremely reduced per os infectivity in larvae. To overcome this limitation, stably transformed insect cell lines expressing polyhedrin capable of occluding occ- recombinant baculovirus by trans-complementation were developed to obtain oral inoculum for insect larvae infection. First

Effect of a single cold stress exposure on the reproductive behavior of male crickets.

Cold stress is an important abiotic factor that can impact insect physiology, behavior, and overall fitness. Upon exposure to cold temperature, many insects enter a reversible state of immobility called chill coma. If the cold stress is brief and mild enough, insects can recover and regain full mobility upon return to warmer temperatures. However, the long-term impact of sublethal cold stress on insect behavior has been understudied. Here, sexually naïve adult male Acheta domesticus crickets were exposed to a single 0 °C cold stress for 6 h. One week later, the ability of these males to mate w

A Conserved Planthopper MATH-BTB Protein Regulates Fecundity in Nilaparvata legens Stål.

The meprin and TRAF-C homology (MATH) family of proteins plays essential roles in diverse biological processes across eukaryotes. Fecundity is a key determinant underlying the rapid outbreaks of agricultural insect pests. Nevertheless, the potential involvement of MATH proteins in the regulation of fecundity in agriculturally important insects, particularly planthoppers, remains largely uncharacterized. This study identified key members of the MATH protein family that are conserved in planthoppers and involved in the regulation of insect fecundity. A total of 121 identified MATH proteins from

Omeprazole and its analogs exhibit insecticidal potencies as inhibitors of insect choline acetyltransferase.

Choline acetyltransferase (ChAT) is crucial for acetylcholine synthesis and regulates diverse functions in numerous biological processes. Omeprazole, an inhibitor on human ChAT, was evaluated here on insect ChAT as a potential inhibitor, as well as its insecticidal potency on Nilaparvata lugens, a major insect pest on rice. The evaluation also included omeprazole analogs and α-NETA, in order to explore a superior leading compound targeting on insect ChAT. In toxicity test, α-NETA and omeprazole exhibited insecticidal activity, among which omeprazole exhibited activity with a mortality of aroun

Cuticle immunity and susceptibility to infection in Galleria mellonella larvae envenomed by the parasitoid wasp Habrobracon hebetor.

The ectoparasitoid wasp, Habrobracon hebetor, targets a range of Lepidopteran larvae and plays an important role in biological control efforts of insect pests. The venom of H. hebetor contains a variety of components with paralytic, neurotoxic and immune-modulatory properties. Herein, we explored the interactions between the entomopathogenic fungus (EPF) Metarhizium robertsii and the integument/cuticle defence reactions of wax moth larvae, Galleria mellonella, (Lepidoptera: Pyralidae) post-envenomation by H. hebetor to gain much needed insight into the mechanisms of disease susceptibility. Behavioral responses of German cockroaches (Blattella germanica L.) to gel baits under varying environmental conditions.

BACKGROUND: Gel baits are one of the most effective tools for managing German cockroaches (Blattella germanica L.); even so, it remains unclear how German cockroaches locate baits. To investigate bait perception by German cockroaches, we evaluated differences in trap catch after applying cockroach gel baits onto glue traps and placing these baited traps at various distances away from both harborages and arena walls in large (110 cm) diameter still-air arenas. In addition, we modified various environmental factors (access to food/water, harborage size) and evaluated the impacts of these modific
