Ion Channel Nano-Diagnostics for ER+ Breast Cancer.

Ion channels are pore-forming transmembrane proteins that allow ions to move down an electrochemical gradient and across the channel pore and regulate many cell functions. Among them, are the G-protein-gated inwardly-rectifying K+ channels 1 (GIRK1) that are ubiquitously expressed with major functions in the brain and heart. Interestingly, significantly higher GIRK1 expression has been found in estrogen receptor positive (ER+) breast cancer patients compared to patients with HER2+ tumors or normal patients, and that was statistically correlated with shorter survival times and metastatic potent

Age-dependent expression of ion channel genes in rat.

Ion channels regulate a large number of cellular functions and their functional role in many diseases makes them potential therapeutic targets. Given their diverse distribution across multiple organs, the roles of ion channels, particularly in age-associated transcriptomic changes in specific organs, are yet to be fully revealed. Using RNA-seq data, we investigated the rat transcriptomic profiles of ion channel genes across 11 organs/tissues and 4 developmental stages in both sexes of Fischer 344 rats and identify tissue-specific and age-dependent changes in ion channel gene expression. Organ-

A review of the pathophysiology and the role of ion channels on bronchial asthma.

Asthma is one of the main non-communicable chronic diseases and affects a huge portion of the population. It is a multifactorial disease, classified into several phenotypes, being the allergic the most frequent. The pathophysiological mechanism of asthma involves a Th2-type immune response, with high concentrations of allergen-specific immunoglobulin E, eosinophilia, hyperreactivity and airway remodeling. These mechanisms are orchestrated by intracellular signaling from effector cells, such as lymphocytes and eosinophils. Ion channels play a fundamental role in maintaining the inflammatory res

Polycystin-2 (TRPP2): Ion channel properties and regulation.

Polycystin-2 (TRPP2, PKD2, PC2) is the product of the PKD2 gene, whose mutations cause Autosomal Dominant Polycystic Kidney Disease (ADPKD). PC2 belongs to the superfamily of TRP (Transient Receptor Potential) proteins that generally function as Ca2+-permeable nonselective cation channels implicated in Ca2+ signaling. PC2 localizes to various cell domains with distinct functions that likely depend on interactions with specific channel partners. Functions include receptor-operated, nonselective cation channel activity in the plasma membrane, intracellular Ca2+ release channel activity in the en

Mechanism of praziquantel action at a parasitic flatworm ion channel.

Praziquantel (PZQ) is an essential medicine for treating parasitic flatworm infections such as schistosomiasis, which afflicts over 250 million people. However, PZQ is not universally effective, lacking activity against liver flukes of the Fasciola genus. The reason for this insensitivity is unclear, as the mechanism of PZQ action is unknown. Here, we use ligand- and target-based methods to demonstrate that PZQ activates a transient receptor potential melastatin ion channel (TRPMPZQ) in schistosomes by engaging a hydrophobic ligand binding pocket within the voltage sensor–like domain of the ch

Controlling ion channel trafficking by targeted ubiquitination and deubiquitination.

Plasma membrane-localized ion channels are essential for diverse physiological processes such as neurotransmission, muscle contraction, and osmotic homeostasis. The surface density of such ion channels is a major determinant of their function, and tuning this variable is a powerful way to regulate physiology. Dysregulation of ion channel surface density due to inherited or de novo mutations underlies many serious diseases, and molecules that can correct trafficking deficits are potential therapeutics and useful research tools. We have developed targeted ubiquitination and deubiquitination appr

Biophysics, pathophysiology, and pharmacology of ion channel gating pores.

Voltage sensor domains (VSDs) are a feature of voltage gated ion channels (VGICs) and voltage sensitive proteins. They are composed of four transmembrane (TM) segments (S1-S4). Currents leaking through VSDs are called omega or gating pore currents. Gating pores are caused by mutations of the highly conserved positively charged amino acids in the S4 segment that disrupt interactions between the S4 segment and the gating charge transfer center (GCTC). The GCTC separates the intracellular and extracellular water crevices. The disruption of S4-GCTC interactions allows these crevices to communicate

Binding of ArgTX-636 in the NMDA receptor ion channel.

The N-methyl-d-aspartate receptors (NMDARs) constitute an important class of ligand-gated cation channels that are involved in the majority of excitatory neurotransmission in the human brain. Compounds that bind in the NMDAR ion channel and act as blockers are use- and voltage-dependent inhibitors of NMDAR activity and have therapeutic potential for treatment of a variety of brain diseases or as pharmacological tools for studies of the neurobiological role of NMDARs. We have performed a kinetic analysis of the blocking mechanism of the prototypical polyamine toxin NMDAR ion channel blocker arg

Ion channel voltage sensors: structure, function, and pathophysiology.

Voltage-gated ion channels generate electrical signals in species from bacteria to man. Their voltage-sensing modules are responsible for initiation of action potentials and graded membrane potential changes in response to synaptic input and other physiological stimuli. Extensive structure-function studies, structure determination, and molecular modeling are now converging on a sliding-helix mechanism for electromechanical coupling in which outward movement of gating charges in the S4 transmembrane segments catalyzed by sequential formation of ion pairs pulls the S4-S5 linker, bends the S6 seg

Neurological disorders caused by inherited ion-channel mutations.

Several neurological diseases-including neuromuscular disorders, movement disorders, migraine, and epilepsy-are caused by inherited mutations of ion channels. The list of these "channelopathies" is expanding rapidly, as is the phenotypic range associated with each channel. At present the best understood channelopathies are those that affect muscle-fibre excitability. These channelopathies produce a range of disorders which include: periodic paralysis, myotonias, malignant hyperthermia, and congenital myasthenic syndromes. By contrast, the mechanisms of diseases caused by mutations of ion chann

Relationship of ion channel activity to control of myometrial calcium.

This article reviews the contribution of ion channels to membrane potential, the ion channels expressed in myometrium, and the effect of ion channel activity on the control of myometrial intracellular free calcium. Plasma membranes constitute barriers to permeability that establish concentration gradients of ions inside versus outside the cell. Na+, CA2+, and Cl- are normally in higher concentration outside than inside cells, whereas K+ is higher inside. In myometrium, Ca2+ entry into cells mediates myometrial membrane potential changes and serves as the internal signal for contraction. K+ eff

Familial Episodic Ataxias and Related Ion Channel Disorders.

Familial episodic ataxias are unusual hereditary disorders of early onset characterized by recurrent episodes of ataxia. Most patients recover fully between attacks, but some may develop progressive ataxia with cerebellar atrophy. There are two subtypes of episodic ataxia: type 1 (EA1), with interictal myokymia, and type 2 (EA2), with interictal nystagmus. Stress and fatigue can trigger ataxic spells, which can be responsive to acetazolamide. These clinical features are reminiscent of other channelopathies or paroxysmal neurologic disorders with progressive features caused by ion channel mutat

Activation of the Mechanosensitive Ion Channels Piezo1 and TRPV4 in Primary Human Healthy and Osteoarthritic Chondrocytes Exhibits Ion Channel Crosstalk and Modulates Gene Expression.

Osteoarthritis (OA) is the most common degenerative joint disease causing pain and functional limitations. Physical activity as a clinically relevant, effective intervention alleviates pain and promotes joint function. In chondrocytes, perception and transmission of mechanical signals are controlled by mechanosensitive ion channels, whose dysfunction in OA chondrocytes is leading to disease progression. Signaling of mechanosensitive ion channels Piezo/TRPV4 was analyzed by Yoda1/GSK1016790A application and calcium-imaging of Fura-2-loaded chondrocytes. Expression analysis was determined by qPC

The fates of internalized NaV1.7 channels in sensory neurons: Retrograde cotransport with other ion channels, axon-specific recycling, and degradation.

Neuronal function relies on the maintenance of appropriate levels of various ion channels at the cell membrane, which is accomplished by balancing secretory, degradative, and recycling pathways. Neuronal function further depends on membrane specialization through polarized distribution of specific proteins to distinct neuronal compartments such as axons. Voltage-gated sodium channel NaV1.7, a threshold channel for firing action potentials in nociceptors, plays a major role in human pain, and its abundance in the plasma membrane is tightly regulated. We have recently characterized the anterogra

Ion Channels in Cardiac Non-Excitable Cells: From Single-Cell Atlases to Disease.

Cardiovascular disease is shaped not only by cardiomyocyte excitability but also by electrophysiological and membrane-signaling programs in cardiac fibroblasts, endothelial cells, mural cells, immune cells, and adipocytes. Recent single-cell RNA sequencing, spatial transcriptomic, and disease-atlas studies have revealed broad expression of ion-channel transcripts across these non-excitable cardiac cell populations. However, transcript detection alone does not establish functional channel activity, cell-state specificity, or therapeutic relevance. This narrative Review therefore focuses on mech

Direct-Write Printing Flexible Ion Channels with Graphene Oxide/Tannic Acid/Cellulose Nanofibers.

Flexible ion channels perform directional ion transport with high mechanical strength, which could bring many important intelligent applications. In this study, composite ink of graphene oxide (GO), tannic acid (TA), and cellulose nanofiber (CNF) is direct-write printed into a GO/TA/CNF porous structure (PS) with a freeze-dried process. With the negative charge of hydroxyl (-OH), carboxyl (-COOH), and phenolic hydroxyl (Ar-OH), the GO/TA/CNF PS of 3 cm length, 3 cm width, and 7 μm thickness realizes a maximum potential of 0.13 V and a maximum current density of 26.7 A/m2 in KCl solution with a

Toxin peptides targeting acid-sensing ion channels: Opportunities for cancer diagnosis and therapy.

Acid-sensing ion channels (ASICs) are proton-gated cation channels widely expressed throughout the mammalian nervous system, where they play key roles in processes such as neuronal excitability, synaptic plasticity and pain perception. Their activation by extracellular acidification makes them highly responsive to changes in pH, which have been implicated in numerous pathologies, including cancer, where acidic microenvironments and ion imbalance are hallmark features. In recent years, toxin peptides have emerged as highly selective molecular tools to probe and modulate ASIC activity. These nat

Role of cardiac ion pumps, channels, and exchangers on regulation of mitochondrial bioenergetics.

Membrane ion pumps and channels are essential key players in intracellular function and communication with other cells, e.g., nutrient uptake and bioelectrical communication. The cation exchangers Na+, H+, Ca2+ and K+ do more than simply rebalance ions and maintain ion homeostasis; they play crucial roles in modulating overall cellular bioenergetics. This review aims to describe how these cation gradients are formed and maintained by examining the ion flux mediated by each pump, channel, and exchanger and by integrating their functions across the cell and mitochondrial membranes. Most of this

Reciprocal Coupling among Extracellular Matrix Mechanics, Mechanosensitive Ion Channels, and Cell Remodeling Behaviors.

Cells actively remodel the extracellular matrix (ECM) through a repertoire of mechanically driven behaviors, including traction force generation, migration, proteolysis, and long-term deposition. While these processes are often discussed separately, emerging evidence suggests that they are coordinated through shared mechanochemical signaling modules. In parallel, advances in hydrogel-based biomaterials have revealed that ECM mechanics, including stiffness, viscoelastic dissipation, degradability, and temporal dynamics, can systematically program cellular remodeling behaviors.
