In-situ growth of laccase-copper phosphate hybrid nanoflowers on carbon cloth for dual-mode electrochemical and colorimetric sensing of phenolic pollutants.

In this work, an electrochemical/colorimetric dual-mode biosensor based on the in-situ growth of laccase-copper phosphate hybrid nanoflowers on carbon cloth (Lac-Cu3(PO4)2 HNFs@CC) was successfully constructed for the efficient detection of phenolic pollutants. The as-prepared Lac-Cu3(PO4)2 HNFs@CC are flower-like microspheres formed by the self-assembly of nanosheets, and their unique three-dimensional hierarchical flower-like structure effectively promotes substrate diffusion and electron transfer. The as-developed dual-mode biosensor exhibits excellent sensing performance for phenol. In the

Purification, characterization and three-dimensional structure prediction of multicopper oxidase Laccases from Trichoderma lixii FLU1 and Talaromyces pinophilus FLU12.

Broad-spectrum biocatalysts enzymes, Laccases, have been implicated in the complete degradation of harmful pollutants into less-toxic compounds. In this study, two extracellularly produced Laccases were purified to homogeneity from two different Ascomycetes spp. Trichoderma lixii FLU1 (TlFLU1) and Talaromyces pinophilus FLU12 (TpFLU12). The purified enzymes are monomeric units, with a molecular mass of 44 kDa and 68.7 kDa for TlFLU1 and TpFLU12, respectively, on SDS-PAGE and zymogram. It reveals distinct properties beyond classic protein absorption at 270-280 nm, with TlFLU1's peak at 270 nm a

Effect of UVB solar irradiation on Laccase enzyme: evaluation of the photooxidation process and its impact over the enzymatic activity for pollutants bioremediation.

The multi-copper Laccase enzyme corresponds to one of the most investigated oxidoreductases for potential uses in xenobiotic bioremediation. In this work, we have investigated the photo-degradation process of Laccase from Trametesversicolor induced by UVB light and the influence on its activity over selected substrates. Laccase undergoes photo-degradation when irradiated with UVB light, and the process depends on the presence of oxygen in the medium. With the kinetic data obtained from stationary and time resolved measurements, a photo-degradation mechanism of auto-sensitization was proposed f

Laccase production by a new isolate of Irpex cf. latemarginatus using cocoa (Theobroma cacao) husk waste as substrate under solid state fermentation.

This study investigates the production and biochemical characterization of laccase from the white-rot fungus Irpex cf. latemarginatus using agro-industrial waste under solid-state fermentation. Cocoa husk waste was identified as the most suitable substrate, and sterile tap water as the optimal moisture-adjusting agent, yielding 4.4 U L-1 of laccase activity. Box-Behnken optimization approach revealed that supplementation with 30% cocoa bean shell significantly enhanced enzyme production, resulting in a 12.5-fold increase and a maximum activity of approximately 55 U L-1. The crude enzyme prepar

A cost-effective, high-efficiency, and environmentally friendly laccase-catalyzed catechol/polyethyleneimine-modified bamboo fiber/poly(butylene succinate-co-butylene adipate) composite.

Bamboo fiber-reinforced biodegradable composites have been increasingly regarded for their renewable origin, cost-effectiveness, and high specific strength. However, the inherent hydrophilicity of bamboo fibers (BFs) results in poor interfacial compatibility with poly(butylene succinate-co-butylene adipate) (PBSA), thereby limiting composite performance. Inspired by the adhesive mechanism of dopamine self-polymerization, a straightforward, cost-effective approach was proposed in this study, in which the BFs were modified through a laccase-catalyzed oxidation process employing polyethyleneimine

Non-pyrolytic synthesis of laccase-like iron based single-atom nanozymes for highly efficient dual-mode colorimetric and fluorescence detection of epinephrine.

Single-atom nanozymes have garnered significant attention due to their exceptional atom utilization and ability to establish well-defined structure-activity relationships. However, conventional pyrolytic synthesis methods pose challenges such as high energy consumption and random local environments at the active sites, while achieving non-pyrolytic synthesis of single-atom nanozymes remains a formidable technical hurdle. The present study focuses on the synthesis of laccase-like iron-based single-atom nanozymes (Fe-SAzymes) using a non-pyrolysis method facilitated by microwave irradiation. Und

Fabrication of superior laccase-mimicking enzyme with catalytic oxidative and photothermal properties for anti-bacterial and dual-mode glutathione S-transferase monitoring.

A novel laccase mimic enzyme Cu-Mn with excellent photothermal properties was firstly prepared via a combination of hydrothermal and in situ synthesis. Cu-Mn nanozymes could catalyze the typical laccase substrate 2,4-dichlorophenol (2,4-DP) to generate the red quinone imine. Further, loading the MnO2 nanosheets with photothermal properties, Cu-Mn nanozymes possessed not only excellent laccase catalytic activity, but also high photothermal conversion efficiency. The presence of glutathione S-transferase (GST) recovered the glutathione (GSH)-induced weakness of the laccase activity and photother

Genome-wide characterization of laccase gene family in Schizophyllum commune 20R-7-F01, isolated from deep sediment 2 km below the seafloor.

Laccases are ligninolytic enzymes that play a crucial role in various biological processes of filamentous fungi, including fruiting-body formation and lignin degradation. Lignin degradation is a complex process and its degradation in Schizophyllum commune is greatly affected by the availability of oxygen. Here, a total of six putative laccase genes (ScLAC) were identified from the S. commune 20R-7-F01 genome. These genes, which include three typical Cu-oxidase domains, can be classified into three groups based on phylogenetic analysis. ScLAC showed distinct intron-exon structures and conserved

Targeted synthesis of novel β-lactam antibiotics by laccase-catalyzed reaction of aromatic substrates selected by pre-testing for their antimicrobial and cytotoxic activity.

The rapidly increasing problem of antimicrobial-drug resistance requires the development of new antimicrobial agents. The laccase-catalyzed amination of dihydroxy aromatics is a new and promising method to enlarge the range of currently available antibiotics. Thirty-eight potential 1,2- and 1,4-hydroquinoid laccase substrates were screened for their antibacterial and cytotoxic activity to select the best substrates for laccase-catalyzed coupling reaction resulting in potent antibacterial derivatives. As a result, methyl-1,4-hydroquinone and 2,3-dimethyl-1,4-hydroquinone were used as parent com

A triple-modified co-immobilized laccase system with dual mediators on MnMOF: Enhanced dye degradation and wastewater detoxification for safe rice seedling cultivation.

Laccase provides a green and eco-friendly solution for the degradation of widely used anthraquinone and triphenylmethane dyes. However, its practical application remains constrained by several obstacles, including poor stability, low redox potential, and mediator leakage, which affect both dye degradation efficiency and environmental safety. To address these challenges, this study developed a novel co-immobilized laccase-mediator system (LMS), termed MnMOF@LCC5@M@PEG, through a sequential triple-modification strategy involving the in-situ immobilization of laccase (LCC5) within a manganese-bas

Quaternized chitosan enhances laccase-mimicking catalytic activity and stability of copper-based metal-organic framework for sensitive colorimetric detection of quercetin in food.

Quercetin is an important flavonoid compound in food. The existing rapid colorimetric methods for quercetin detection face many limitations including poor stability and strong substrate dependence. Herein, a copper-based metal-organic framework (Cu-MOF) possessing inherent laccase-mimicking activity was synthesized, which was further modified with quaternized chitosan (QCS) to construct a robust laccase mimic (named QCS@Cu-MOF) with enhanced catalytic activity and stability. The increased surface hydrophilicity and positive charge density promote closer contact with quercetin, thereby boosting
