BACKGROUND: Pamamycins are a family of highly bioactive macrodiolide polyketides produced by Streptomyces alboniger as a complex mixture of derivatives with molecular weights ranging from 579 to 705 Daltons. The large derivatives are produced as a minor fraction, which has prevented their isolation and thus studies of chemical and biological properties.
RESULTS: Herein, we describe the transcriptional engineering of the pamamycin biosynthetic gene cluster (pam BGC), which resulted in the shift in production profile toward high molecular weight derivatives. The pam BGC library was constructed

BACKGROUND: Lifestyle modification delivered through digital self-management is central to metabolic dysfunction-associated steatotic liver disease (MASLD) care, yet long-term engagement remains the threshold beyond which clinical benefit is realized. Understanding attrition requires examining both retention (dropout) and adherence (usage quality), which are often evaluated in isolation. Existing systematic reviews of digital interventions for MASLD have focused predominantly on clinical effectiveness, leaving less attention on attrition.
OBJECTIVE: This study aimed to integrate quantitative

Anthropometry offers noninvasive measurements of body size, proportions, and composition that can inform musculoskeletal assessment and rehabilitation. Traditional anthropometric measures continue to be widely used owing to their accessibility, whereas digital technologies provide further approaches for objective body and movement assessment. However, evidence on the applications and measurement properties of traditional and digital anthropometric measures across orthopaedic and rehabilitation settings remains inconsistent. This systematic review assessed the measurement methods, clinical appl

BACKGROUND: Panfacial fractures are among the most technically demanding injuries in craniomaxillofacial trauma surgery. Digital workflows - including virtual surgical planning (VSP), three-dimensional printing, and intraoperative navigation - have demonstrated benefit in elective and delayed surgical settings, yet their integration into acute panfacial fracture management remains poorly characterized.
PURPOSE: This systematic review evaluated the evidence for digital workflow integration in acute primary panfacial fracture management, characterizing achievable outcomes, assessing implementat

BACKGROUND: Intrinsic capacity (IC) has become a central concept in healthy aging because it emphasizes functional ability across the aging trajectory rather than disease alone. However, current IC assessment primarily relies on episodic clinical evaluations, which are insufficient for continuous monitoring and early identification of functional decline. Recent advances in AI and digital health technologies have created new opportunities for objective, continuous, and real-world assessment of IC. However, existing evidence remains fragmented across AI-enabled devices, digital biomarkers (DBs),

BACKGROUND: Bipolar disorder (BD) features episodic shifts among (hypo)mania, depression, mixed states, and euthymia. Timely detection of mood transitions is difficult due to infrequent clinical touchpoints. Digital health technologies, including wearables and smartphones, offer a unique opportunity to passively and continuously monitor behavior and physiology that could reflect underlying mood dynamics in real-world settings.
OBJECTIVE: We aim to systematically review passively collected digital biomarkers for BD mood states, characterize devices/modalities and analytic approaches, appraise

BACKGROUND: Digital health technology (DHT) plays an increasingly vital role in managing chronic diseases by enabling patients to actively manage their health. These tools have been shown to improve self-management and adherence to medical advice. However, for DHT to be fully effective, patients with chronic conditions must be digitally literate. The eHealth Literacy Scale (eHEALS), an 8‑item tool with scores ranging from 8 to 40, was developed to assess individuals' perceived ability to find, evaluate, and apply digital health information. Assessing patients' digital health literacy (DHL) and

BACKGROUND: Digitally delivered physical exercise interventions are becoming increasingly popular in addressing the obesity epidemic. However, there remains uncertainty on their efficacy regarding the reduction of body weight (BW) and body fat, which may, at least partly, be due to variations in study designs and inconsistent adherence to international physical activity (PA) guidelines.
OBJECTIVE: This study aimed to evaluate the effectiveness of digital exercise interventions based on PA guidelines in reducing BW and fat in adults with overweight or obesity, as well as their impact on PA-rel

INTRODUCTION: Inequities in systemic lupus erythematosus (SLE) care affect outcomes (i.e., flares, damage, mortality), interventions (referrals, advanced therapies), and populations (gender, race/ethnicity, and geographic region). This systematic review highlights themes related to access to care for SLE patients and explores potential solutions to address these disparities.
METHODS: In adherence to PRISMA guidelines, we conducted a systematic review of the literature. We searched for articles published in PubMed, Scopus, Web of Science, Cochrane Library, and EBSCO. Publications related to ec
