Exploring bile acid transporters as key players in cancer development and treatment: Evidence from preclinical and clinical studies.

Bile acid transporters (BATs) are integral membrane proteins belonging to various families, such as solute carriers, organic anion transporters, and ATP-binding cassette families. These transporters play a crucial role in bile acid transportation within the portal and systemic circulations, with expression observed in tissues, including the liver, kidney, and small intestine. Bile acids serve as signaling molecules facilitating the absorption and reabsorption of fats and lipids. Dysregulation of bile acid concentration has been implicated in tumorigenesis, yet the role of BATs in this process

ABCA2 as a therapeutic target in cancer and nervous system disorders.

BACKGROUND: Overexpression of ATP-binding cassette (ABC) transporters is a major adaptive advantage used by tumor cells to evade the accumulation of cytotoxic agents. ABCA2, a transporter highly expressed in the cells of the nervous and haematopoetic systems, is associated with lipid transport and drug resistance in cancer cells, including tumor stem cells. Recently, a single nucleotide polymorphism (SNP) in Abca2 was linked to early onset Alzheimer's disease (AD). The characterization of two independent knockout mouse models has shed light on putative in vivo functions of this transporter in

The MarR family regulator RmaH underlies the trade-off between acid sensitivity and antibiotic tolerance in Lactococcus lactis.

UNLABELLED: Microorganisms exhibit remarkable adaptability to diverse environmental stresses through transcriptional reprogramming. Here, we focused on the MarR family regulator RmaH in Lactococcus lactis, which orchestrated a pivotal trade-off: enhancing tolerance to the bacteriocin nisin and the antibiotic vancomycin at the expense of acid resistance. RmaH directly activated genes for cell wall and membrane lipid biosynthesis, leading to a thicker cell wall, and repressed the arginine deiminase pathway, attenuating acid tolerance. Integrated transcriptomic, chromatin immunoprecipitation-sequ

Malaria in pregnancy regulates P-glycoprotein (P-gp/Abcb1a) and ABCA1 efflux transporters in the Mouse Visceral Yolk Sac.

Malaria in pregnancy (MiP) induces intrauterine growth restriction (IUGR) and preterm labour (PTL). However, its effects on yolk sac morphology and function are largely unexplored. We hypothesized that MiP modifies yolk sac morphology and efflux transport potential by modulating ABC efflux transporters. C57BL/6 mice injected with Plasmodium berghei ANKA (5 × 105 infected erythrocytes) at gestational day (GD) 13.5 were subjected to yolk sac membrane harvesting at GD 18.5 for histology, qPCR and immunohistochemistry. MiP did not alter the volumetric proportion of the yolk sac's histological comp

Calcium dysregulation in breast cancer progression: mechanisms of calcicoptosis and therapeutic implications.

Disruption of calcium ion (Ca2+) homeostasis represents a critical mechanism in breast cancer (BC) progression, with aberrant Ca2+ transport through store-operated Ca2+ entry pathways, plasma membrane channels, and membrane contact sites (MCSs) playing a significant role in malignant transformation, metastasis, and therapeutic resistance. This dysregulation involves complex interactions among endoplasmic reticulum Ca2+ storage systems, Ca2+-dependent signaling cascades, and specialized MCSs that facilitate abnormal Ca2+ transfer between cellular organelles. Of particular therapeutic relevance,

Neural Regulation of Urothelial Water Transport: Pelvic Nerve Stimulation Induces Aquaporin-2 and Vasopressin V1a Receptor Translocation in the Rat Urinary Bladder.

PURPOSE: Aquaporin-2 (AQP2) and vasopressin V1a receptor (AVP-V1a) are membrane proteins involved in water transport regulation in renal and extrarenal tissues. Their presence in the urinary bladder suggests a role in local water handling. This study aimed to investigate whether pelvic nerve stimulation induces translocation of AQP2 and AVP-V1a in the bladder urothelium of rats, thereby revealing a potential neural regulatory mechanism in water permeability. METHODS: Forty female Sprague-Dawley rats were assigned to either a control (n=20) or electrical stimulation group (n=20). The pelvic ne

Phosphorylation regulates the water channel activity of the seed-specific aquaporin alpha-TIP.

The vacuolar membrane protein alpha-TIP is a seed-specific protein of the Major Intrinsic Protein family. Expression of alpha-TIP in Xenopus oocytes conferred a 4- to 8-fold increase in the osmotic water permeability (Pf) of the oocyte plasma membrane, showing that alpha-TIP forms water channels and is thus a new aquaporin. alpha-TIP has three putative phosphorylation sites on the cytoplasmic side of the membrane (Ser7, Ser23 and Ser99), one of which (Ser7) has been shown to be phosphorylated. We present several lines of evidence that the activity of this aquaporin is regulated by phosphorylat

p.Phe508del-CFTR Trafficking: A Protein Quality Control Perspective Through UPR, UPS, and Autophagy.

Cystic fibrosis (CF) is a genetic disease due to mutations in the cystic fibrosis transmembrane conductance regulator (CFTR) gene. The most frequent mutation (p.Phe508del) results in a misfolded protein (p.Phe508del-CFTR) with an altered transport to the membrane of the cells via the conventional protein secretion (CPS) pathway. Nevertheless, it can use unconventional protein secretion (UPS). Indeed, p.Phe508del-CFTR forms a complex with GRASP55 to assist its direct trafficking from the endoplasmic reticulum to the plasma membrane. While GRASP55 is a key player of UPS, it is also a key player

Comparison of the Sulfonamide Inhibition Profiles of the α-Carbonic Anhydrase Isoforms (SpiCA1, SpiCA2 and SpiCA3) Encoded by the Genome of the Scleractinian Coral Stylophora pistillata.

The ubiquitous metalloenzymes carbonic anhydrases (CAs, EC 4.2.1.1) are responsible for the reversible hydration of CO₂ to bicarbonate (HCO₃-) and protons (H⁺). Bicarbonate may subsequently generate carbonate used in many functional activities by marine organisms. CAs play a crucial role in several physiological processes, e.g., respiration, inorganic carbon transport, intra and extra-cellular pH regulation, and bio-mineralization. Multiple transcript variants and protein isoforms exist in the organisms. Recently, 16 α-CA isoforms have been identified in the coral Stylophora pistillata. Here,

Small proteins CrgA and Rv0207c modulate the essential mycolic acid transporter MmpL3.

MmpL protein complexes are major contributors to lipid/metabolite and drug transport in Mycobacterium tuberculosis and related mycobacteria and are important targets in current drug discovery efforts. At the core of these complexes are proton-motive force driven transporters belonging to the Resistance-Nodulation-cell Division superfamily of proteins that enable export of various substrates across complex bacterial cell envelopes. This study focuses on MmpL3, which transports trehalose monomycolate in M. tuberculosis and related bacterial species and plays an essential role in the assembly of

Arsenic-sensing domain controls ACR3 transporter trafficking in Marchantia polymorpha.

Arsenic is a toxic, carcinogenic metalloid that threatens human health through contaminated water and food. Arsenite [As(III)] efflux mediated by ACR3 transporters is an ancient detoxification mechanism conserved across prokaryotes, fungi, algae, and non-angiosperm land plants, yet plant ACR3 proteins remain poorly characterized. Here, we show that MpACR3, the ACR3 ortholog from the model liverwort Marchantia polymorpha, contains an arsenic-responsive N-terminal domain that regulates intracellular trafficking. Under non-stress conditions, MpACR3 is detected in Golgi bodies as well as at the pl

Impaired intracellular trafficking of sodium-dependent vitamin C transporter 2 contributes to the redox imbalance in Huntington's disease.

Huntington's disease (HD) is a neurodegenerative disorder caused by a glutamine expansion at the first exon of the huntingtin gene. Huntingtin protein (Htt) is ubiquitously expressed and it is localized in several organelles, including endosomes. HD is associated with a failure in energy metabolism and oxidative damage. Ascorbic acid is a powerful antioxidant highly concentrated in the brain where it acts as a messenger, modulating neuronal metabolism. It is transported into neurons via the sodium-dependent vitamin C transporter 2 (SVCT2). During synaptic activity, ascorbic acid is released fr

A novel transport mechanism for MOMP in Chlamydophila pneumoniae and its putative role in immune-therapy.

Major outer membrane proteins (MOMPs) of Gram negative bacteria are one of the most intensively studied membrane proteins. MOMPs are essential for maintaining the structural integrity of bacterial outer membranes and in adaptation of parasites to their hosts. There is evidence to suggest a role for purified MOMP from Chlamydophila pneumoniae and corresponding MOMP-derived peptides in immune-modulation, leading to a reduced atherosclerotic phenotype in apoE(-/-) mice via a characteristic dampening of MHC class II activity. The work reported herein tests this hypothesis by employing a combinatio

Investigation of the Plasmodium falciparum food vacuole through inducible expression of the chloroquine resistance transporter (PfCRT).

Haemoglobin degradation during the erythrocytic life stages is the major function of the food vacuole (FV) of Plasmodium falciparum and the target of several anti-malarial drugs that interfere with this metabolic pathway, killing the parasite. Two multi-spanning food vacuole membrane proteins are known, the multidrug resistance protein 1 (PfMDR1) and Chloroquine Resistance Transporter (PfCRT). Both modulate resistance to drugs that act in the food vacuole. To investigate the formation and behaviour of the food vacuole membrane we have generated inducible GFP fusions of chloroquine sensitive an

Golvatinib (E7050) restores chemosensitivity in multidrug-resistant cancer cells by inhibiting ABCG2 transporter.

ABCG2-mediated drug efflux is a major cause of multidrug resistance (MDR), reducing the activity of various anticancer agents. This study evaluated the role of Golvatinib, originally developed as a c-MET/VEGFR2 inhibitor, as a potential ABCG2 modulator in ABCG2-expressing MDR cells. Golvatinib significantly restored cellular sensitivity to representative ABCG2 substrates, by inhibiting efflux of substrates and increasing intracellular drug accumulation. ABCG2 expression and membrane localization remained unchanged, indicating that this reversal was not due to downregulation or relocation of AB

Light-Controlled Conductive Molecular Wire in an Artificial Lipid Membrane.

In this work, we investigate a novel photochromic azobenzene derivative, TC827, which exhibits a unique photoelectric response when embedded in artificial lipid membranes. By combining spectroscopic, electrochemical, and molecular simulation analyses, we demonstrate that TC827 forms light-controlled "molecular wires" in its trans conformation, resulting in a significant increase in membrane conductivity under dark conditions, which is fully reversible, as light-induced cis isomerization suppresses the conductivity enhancement. This behavior is fundamentally different from previously studied am

Enhancing KCNQ Channel Activity Improves Neurobehavioral Recovery after Spinal Cord Injury.

Spinal cord injury (SCI) usually leads to acute neuronal death and delayed secondary degeneration, resulting in sensory dysfunction, paralysis, and chronic pain. Excessive excitation is one of the critical factors leading to secondary neural damage initiated by various insults. KCNQ/Kv7 channels are highly expressed in spinal neurons and axons and play an important role in controlling their excitability. Enhancing KCNQ channel activity by using its specific opener retigabine could thus be a plausible treatment strategy to reduce the pathology after SCI. We produced contusive SCI at T10 in adul

Polarized sphingolipid transport from the subapical compartment: evidence for distinct sphingolipid domains.

In polarized HepG2 cells, the sphingolipids glucosylceramide and sphingomyelin (SM), transported along the reverse transcytotic pathway, are sorted in subapical compartments (SACs), and subsequently targeted to either apical or basolateral plasma membrane domains, respectively. In the present study, evidence is provided that demonstrates that these sphingolipids constitute separate membrane domains at the luminal side of the SAC membrane. Furthermore, as revealed by the use of various modulators of membrane trafficking, such as calmodulin antagonists and dibutyryl-cAMP, it is shown that the fa

Lipid environment determines the drug-stimulated ATPase activity of P-glycoprotein.

P-glycoprotein (Pgp) is a multidrug transporter that uses the energy from ATP binding and hydrolysis to export from cells a wide variety of hydrophobic compounds including anticancer drugs, and mediates the bioavailability and pharmacokinetics of many drugs. Lipids and cholesterol have been shown to modulate the substrate-stimulated ATPase activity of purified Pgp in detergent solution and the substrate transport activity after reconstitution into proteoliposomes. While lipid extracts from E. coli, liver or brain tissues generally support well Pgp's functionality, their ill-defined composition
