Are low serum levels of Vitamin D associated with dental developmental defects in primary teeth? A systematic review.

OBJECTIVE: To identify, qualify and synthesize all studies that assessed if low serum level of 25(OH)D (<50 nmol/L) is associated with dental developmental defects (DDD) in primary teeth. MATERIALS AND METHODS: Observational studies or clinical trials were included if measured 25(OH)D serum levels in pregnant women and/or in their children (up to 3 years old) and evaluated the occurrence of DDD in the primary dentition of offspring associated with the low 25(OH)D levels. Literature reviews, case reports, laboratory and/or animals' studies, conference abstracts, letters to the editor, book cha

Evaluation of the effect of a silica-enriched toothpaste on teeth with MIH: A triple-blind pilot randomized controlled clinical trial.

OBJECTIVES: To evaluate the effect of a silica-enriched remineralizing toothpaste on mineral gain in MIH-affected teeth using quantitative light-induced fluorescence (QLF). Secondary outcomes were pain sensitivity, aesthetic perception, and adverse effects after 45 days of home use. METHODS: This triple-blind, parallel-group, exploratory pilot randomized clinical trial allocated children aged 6-12 years with fully erupted MIH-affected permanent incisors to REFIX® Booster toothpaste or standard 1450-ppm fluoride toothpaste. Both groups received professional prophylaxis and a 45-day home-use ki

Elucidating the molecular nexus between dental developmental anomalies and chronic inflammatory diseases: A computational systems biology analysis of genomic sequence homology using matrix factorization.

This entirely in silico, exploratory study investigates potential sequence-level links between periodontal disease, developmental dental anomalies such as molar-incisor hypomineralization (MIH), and selected systemic conditions including leukemias, amyotrophic lateral sclerosis, and Alzheimer's disease. To identify annotation-independent signatures, we applied Non-negative Matrix Tri-Factorization (NMTF) to k-mer/tf-idf representations of disease-associated gene and protein sequences compiled from OMIM, GeneCards, and PubMed for eleven curated disease groups; similarity values denote the propo

Dental effects of enzyme replacement therapy in case of childhood-type hypophosphatasia.

BACKGROUND: Hypophosphatasia (HPP), a skeletal disease characterized by hypomineralization of bone and teeth, is caused by an ALPL gene mutation that leads to low activity of the tissue non-specific alkaline phosphatase enzyme. Although enzyme replacement therapy (ERT) was recently introduced for affected patients, no known studies have been reported regarding its dental effects related to permanent teeth and jaw bones. In the present study, we examined the dental effects of ERT in a case of childhood-type hypophosphatasia, including panoramic radiography findings used to estimate the dental a

Vitamin D status and tooth enamel hypomineralization are not associated in 4-y-old children: An Odense Child Cohort study.

Early fetal stages of tooth development are vitamin D-dependent, suggesting an impact of vitamin D status in pregnancy on tooth mineralization in human populations. We examined the association between pregnancy and cord serum 25-hydroxyvitamin D (s-25(OH)D) and hypomineralization of the second primary molars (HSPM) in the 4-year-old children in the prospective, population-based Odense Child Cohort, Denmark. S-25(OH)D was measured in early pregnancy (<20 weeks, n = 753); late pregnancy (≥20 weeks, n = 841); and in umbilical cord blood (n = 1,241) using liquid chromatography-tandem mass spectrom

Human genes for dental anomalies.

The development of the tooth at gene level is beginning to be understood. This paper reviews current knowledge and the advances in research on human genes whose defect leads to dental anomalies. Amelogenesis imperfecta (AI) is a diverse group of hereditary disorders characterized by a variety of developmental enamel defects including hypoplasia and hypomineralization, some of which have been revealed to be associated with defective amelogenin genes. The human amelogenin genes on X and Y chromosomes have been cloned and investigated extensively. Although autosomally inherited forms of AI are mo

Investigation of the most common perinatal and postnatal risk factors in children with MIH compared to healthy (non-MIH) children aged 6-12 years in Arak City.

BACKGROUND: Molar-Incisor Hypomineralization (MIH) is a widespread developmental enamel defect impacting children globally, with a multifactorial etiology that remains incompletely understood. Its rising prevalence presents significant challenges to oral health and quality of life. This study examines both perinatal and postnatal risk factors associated with MIH in a pediatric population. METHODS: This case-control study involved 426 children aged 6-12 years, with 213 diagnosed with MIH and 213 healthy controls, recruited from a university dental clinic and local schools in Arak City, Iran. The preterm child in dentistry. Behavioural aspects and oral health.

BACKGROUND: Advancements in medical care have enabled more children born preterm to survive and develop as healthy individuals alongside their full-term peers. However, a higher frequency of medical health problems, cognitive and behavioural disturbances, including problems in school, has been reported. There is limited knowledge about how this affects preterm children (PT) in dentistry, and few studies have evaluated behavioural problems related to dental treatments and oral health in PT as compared with full-term control children (C). AIMS: The overall objective was to investigate behaviour

Proof-of-principle for enhanced dentoalveolar mineralization using exogenous tissue-nonspecific alkaline phosphatase in the Hyp mouse model of X-linked hypophosphatemia.

X-linked hypophosphatemia (XLH) is caused by mutations in the PHEX gene, which leads to increased levels of fibroblast growth factor 23 and hypophosphatemia, contributing to rickets, osteomalacia, and dentoalveolar defects, including severe dentin hypomineralization, thin cementum, and alveolar bone osteomalacia. Current XLH treatment options appear to have limited efficacy on dentoalveolar tissues, suggesting underlying disease mechanisms that remain unchecked. Increased production of inorganic pyrophosphate (PPi) and osteopontin (OPN), both mineralization inhibitors, has been posited to cont

Comparison of dental findings between dentists and pediatricians using intraoral scan-based teledentistry in children.

BACKGROUND: Asynchronous transmission of health information via teledentistry offers the potential for remote diagnosis in pediatric dentistry. The aim of this study was to compare teledental findings obtained from intraoral scans (IOS) with those from conventional visual examinations (VIS) in children. Specifically, the study assessed the diagnostic accuracy of teledentistry using IOS in evaluating oral health and determining treatment needs focusing on comparisons between dentists and pediatricians. METHODS: Children (mean age 10.04 ± 2.90 years) underwent VIS during routine dental examinat
