The purified GABA/benzodiazepine/barbiturate receptor complex: four types of ligand-binding sites, and the interactions between them, are preserved in a single isolated protein complex.

A GABA/benzodiazepine/barbiturate receptor complex has been purified from bovine cerebral cortex by affinity chromatography on a benzodiazepine column. Depending on the detergent present during the isolation of the receptor (deoxycholate/Triton X-100 or CHAPS/Asolectin), and during the binding assays (Triton X-100 or CHAPS), the receptor displays different binding properties for the GABAA agonist [3H]muscimol and for the chloride ion channel blocking agent [35S]t-butylbicyclophosphorothionate (TBPS), whereas the binding properties for the benzodiazepine [3H]flunitrazepam are independent of iso

Paraventricular nucleus of the hypothalamus activation and forebrain neuroinflammation in salt-loaded rats.

Salt loading (SL) in rats for 7 days raises mean arterial pressure (MAP) and induces astrogliosis in the paraventricular nucleus of the hypothalamus (PVN). However, the exact role of the PVN in elevating MAP and sympathetic nerve activity (SNA), as well as its effects on astrocytes in forebrain osmosensitive areas, remain unclear. Male Holtzman and Wistar rats (300-320 g) received only 2% NaCl solution (SL) to drink for 7 days, while controls drank water (WT). Daily fluid and food intake, body weight, urinary volume, and MAP were recorded. On day 7, blood and cerebrospinal fluid (CSF) were col

Anterior cingulate cortex inactivation impairs rodent visual selective attention and prospective memory.

Previous studies showed that the anterior cingulate cortex (ACC) plays a role in selective visual attention. The current study further examined the role of the ACC in attention using a visual cuing task with task-relevant and task-irrelevant stimuli. On every trial, 2 stimuli were presented on the touchscreen; 1 was task-relevant and the other was task-irrelevant. Rats were trained to attend to the task-relevant stimulus over the task-irrelevant stimulus to determine which side of the touchscreen should be selected for reward. After the rats were well-trained, cannulas targeting the ACC were i

The effects of hippocampal opioidergic and septal GABAergic system interactions on anxiety-like behavior in rats.

AIMS: Previous studies have shown that the septum and hippocampus are connected and play a key role in anxiety-related behavior. In the present study, the effects of the interactions between the opioidergic system in the dorsal hippocampus (CA1) and the GABAergic system in the medial septum on anxiety in male Wistar rats were investigated. MAIN METHODS: An elevated plus maze was used to study the effects of anxiogenic and anxiolytic drugs in rodents. Male Wistar rats were used for this test. We injected morphine (2.5, 5.0, and 7.5 μg/rat) into the CA1 and GABA(A) agonist muscimol (2.5, 5.0, a

Evidence for the involvement of GABA(A) receptor blockade in convulsions induced by cephalosporins.

There is accumulating evidence that most beta-lactam antibiotics (i.e., cephalosporins and penicillins) have some degree of convulsive activity, both in laboratory animals as well as in clinical settings. The proposed mechanism is suppression of inhibitory postsynaptic responses, mainly mediated by gamma-amino butyric acid (GABA)(A)-receptors (GABA(A)-R). However, comprehensive studies on the convulsive activities of various beta-lactam antibiotics in vivo and in vitro have not been performed. We have therefore examined the convulsive activities of seven different cephalosporins using both in

Ventrolateral periaqueductal gray matter and the control of tonic immobility.

Tonic immobility is an inborn defensive behavior characterized by a temporary state of profound and reversible motor inhibition elicited by some forms of physical restraint. The periaqueductal gray matter (PAG) contains neural circuits involved in descending pain modulation, as well as in the modulation of TI. We have reported previously that the cholinergic stimulation of the ventrolateral PAG increases the duration of TI in guinea pigs. In the present study, we attempted to characterize further the modulation of TI by pharmacological alteration of the neurochemistry of the ventrolateral PAG

The relaxant effect of sex steroids in rat myometrium is independent of the gamma-amino butyric acid system.

The mechanism of action on the uterine-relaxant effect of sex steroids has been suggested to involve an interaction with gamma-amino butyric acid (GABA) receptors. However, other lines of evidence do not seem to support this suggestion. In view of this controversy, this study was designed to investigate the potential relaxant effect of GABA, muscimol (a GABA(A) receptor agonist), testosterone, progesterone and their 5-reduced metabolites in rat uterus at different endocrine stages (pregnant, nonpregnant and estrogenized), with particular emphasis on verifying if the relaxant effect of steroids

The role of mesoaccumbens--pallidal circuitry in novelty-induced behavioral activation.

When exposed to an environment for the first time, rats express greater behavioral activation than rats which were previously habituated to that environment. The circuit containing the ventral tegmental area, nucleus accumbens and ventral pallidum is required for the expression of locomotor activity elicited by amphetamine-like psychostimulants. It was hypothesized that this circuit is necessary for the expression of novelty-induced motor activity. Dopamine is a neurotransmitter in the projection from the ventral tegmental area to the nucleus accumbens, while GABA is contained in the projectio

Accumbens GABA-ergic innervation contributes to the stressor-induced locomotor depression in rats.

The effect of intra-accumbens injections of drugs changing the function of GABA-A and GABA-B receptor systems on stressor-induced motor depression, was studied in rats. Local injections of picrotoxin and baclofen, but not of midazolam and muscimol, attenuated the inhibitory effect of inescapable footshock on locomotor activity in the open field test, examined 24 h after a single exposure of rats to the stressful event. The results obtained with picrotoxin may be related to the general disinhibitory properties of the convulsant on brain neuronal activity, in a period of time important for conso

Chronic benzodiazepine administration. IV. Rapid development of tolerance and receptor downregulation associated with alprazolam administration.

The triazolobenzodiazepine compound alprazolam may have unique clinical effects compared to other benzodiazepines, and both behavioral and neurochemical studies have indicated unusual results after acute doses of alprazolam. To determine the effects of chronic dosage in mice, alprazolam (2 mg/kg/day) was administered via osmotic pumps for 1-14 days, and open-field activity, plasma and brain concentrations, benzodiazepine receptor binding in vivo and in vitro, [35S]t-butylbicyclophosphorothionate ([35S]TBPS) binding, and muscimol-stimulated chloride uptake were determined. Responses to GABA by isolated insect neuronal somata: pharmacology and modulation by a benzodiazepine and a barbiturate.

Mechanically dissociated neuronal somata from the thoracic ganglia of Locusta migratoria and Schistocerca gregaria were viable in vitro for hours and were current- and voltage-clamped to record the responses evoked by brief pressure applications of gamma-aminobutyric acid (GABA) in the presence of various modulators. The application of GABA and muscimol, but not baclofen, produced a hyperpolarization and concurrent increase in the membrane conductance. The current underlying this response reversed at -65 mV, was evoked in all cells tested and showed outward rectification. In 6 of 74 Locusta ne

Behavioral responses of restricted-fed fowls to pharmacological manipulation of 5-HT and GABA receptor subtypes.

Effects on environmentally induced oral stereotypies (object pecking and drinker-directed activity) and other behavior (sitting, standing, pacing, preening), of preferential antagonists and agonists of central 5-HT and GABA receptor subtypes, were examined in individually caged broiler breeder fowls subjected to chronic food restriction. All drugs were injected intravenously at three doses, and their effects compared with a saline control treatment. The only significant (p < 0.05) effect of 5-HT antagonists [NAN-190 (5-HT1A), ketanserin (5-HT2), MDL-72222 (5-HT3)] was an increase in pacing wit

Differential effects mediated by GABAA receptors in thalamic nuclei in lh/lh model of absence seizures.

Absence seizures represent synchronized burst-firing of thalamocortical neurons, which are driven by tonic GABAergic output of nucleus reticularis thalami (NRT). Activation of GABAA receptors on NRT neurons reduces NRT output and retards thalamocortical burst-firing. Although this mechanism in NRT may underlie antiabsence effects of benzodiazepines, it does not explain observations that barbiturates can worsen absence-seizures. In this study we tested the hypothesis that clonazepam and phenobarbital produce differential effects on GABAA receptors in the lh/lh genetic model of absence seizures

Basal forebrain injections of the benzodiazepine partial inverse agonist FG 7142 enhance memory of rats in the double Y-maze.

Cholinergic replacement strategies have achieved little success in the treatment of Alzheimer's disease. It has been suggested that the mnemonic function of cholinergic neurons may be enhanced by treatments that reduce GABA-ergic inhibition, while preserving the normal pattern of activity in the cholinergic neurons. Following on these suggestions, the present study investigated the mnemonic effects of intra-nucleus basalis magnocellularis (NBM) injections of the benzodiazepine receptor partial inverse agonist N-methyl-beta-carboline-3-carboxamide (FG 7142). Rats were surgically implanted with

The role of the nucleus reuniens in regulating contextual conditioning with the predator odor TMT in female rats.

RATIONALE: Experiencing intrusive distressing memories of a traumatic event(s) is a prominent symptom profile for post-traumatic stress disorder (PTSD). Understanding the neurobiological mechanisms associated with this symptom profile can be invaluable for effective treatment for PTSD. OBJECTIVES: Here, we investigated the functional role of the nucleus reuniens (RE), a midline thalamic in modulating stressor-related memory. METHODS: Female Long Evans rats were implanted with a cannula aimed at the RE. The RE was pharmacologically inactivated via muscimol (0.5 mM) prior to exposure to the pr

Local GABAA Receptor-Mediated Suppression of Dopamine Release within the Nucleus Accumbens.

Benzodiazepines make up a class of psychoactive drugs that act as allosteric co-activators of the inhibitory GABAA receptor. These drugs are useful for the treatment of several psychiatric disorders but also hold considerable abuse liability. Despite the common use and misuse of benzodiazepines, the mechanisms through which these drugs exert their reinforcing effects remain incompletely understood. Transient phasic increases in dopamine levels are believed to play an important role in defining the reinforcing properties of drugs of abuse, and we recently demonstrated that systemic administrati

The role of dorsomedial hypotalamus ionotropic glutamate receptors in the hypertensive and tachycardic responses evoked by Tityustoxin intracerebroventricular injection.

The scorpion envenoming syndrome is an important worldwide public health problem due to its high incidence and potential severity of symptoms. Some studies address the high sensitivity of the central nervous system to this toxin action. It is known that cardiorespiratory manifestations involve the activation of the autonomic nervous system. However, the origin of this modulation remains unclear. Considering the important participation of the dorsomedial hypotalamus (DMH) in the cardiovascular responses during emergencial situations, the aim of this work is to investigate the involvement of the

Topography of dyskinesias and torticollis evoked by inhibition of substantia nigra pars reticulata.

GABAergic neurons of the substantia nigra pars reticulata (SNpr) and globus pallidus pars interna (GPi) constitute the output pathways of the basal ganglia. In monkeys, choreiform limb dyskinesias have been described after inhibition of the GPi, but not the SNpr. Given the anatomical and functional similarities between these structures, we hypothesized that choreiform dyskinesias could be evoked by inhibition of an appropriate region within the SNpr. The GABAA receptor agonist, muscimol, was infused into various sites within the SNpr and the adjacent STN of freely moving macaques. The effect o

Involvement of GABAergic modulation of the nucleus submedius (Sm) morphine-induced antinociception.

Previous studies have shown that microinjection of morphine into the nucleus submedius (Sm) of the thalamus produces antinociception. The aim of the current study was to examine whether gamma-aminobutyric acid (GABA)ergic terminals in the Sm were involved in this antinociception. Under light anesthesia, the GABA(A) receptor antagonist bicuculline or agonist muscimol was microinjected into the Sm of the thalamus in Sm non-morphine-treated (control) or Sm morphine-treated (microinjection into the Sm in the thalamus) rats.
