JAK2 inhibition with baricitinib stunts growth and postnatal development in juvenile rats.

The Janus kinase (JAK)/signal transducer and activator of transcription (STAT) pathway has recently emerged as an important regulator of musculoskeletal development. Inhibition of the pathway with small-molecule JAK inhibitors (jakinibs) may improve tendon development and healing. The goal of the present study was to characterize the effects of blocking JAK1 and 2 during a key postnatal musculoskeletal growth phase in Sprague-Dawley rats (postnatal day (PND) 7-28). We administered vehicle or baricitinib via chow (3 mg, or 10 mg/kg BW/day), which blocks JAK1 and JAK2, for 21 days and characteri

Aromatase inhibitors induce pain-like musculoskeletal symptoms in mice: behavioural, pharmacological and pathophysiological characterization.

BACKGROUND AND PURPOSE: Breast cancer, one of the most common cancers in women, primarily manifests as estrogen receptor-positive tumours in post-menopausal women. Aromatase inhibitors (AIs) such as letrozole are the most effective standard adjuvant endocrine therapy. However, the majority of AI-treated patients experience musculoskeletal symptoms (AIMSS), which can lead to premature discontinuation of treatment. Well-characterized animal models are essential to better understand the various mechanisms of AIMSS. EXPERIMENTAL APPROACH: We studied the development of musculoskeletal symptoms in

Flatness of musculoskeletal systems under functional electrical stimulation.

Control of musculoskeletal yy system through functional electrical stimulation (FES) still remains a complex and a challenging process. Indeed, the used musculoskeletal models are often complex and highly nonlinear, which makes their control and inversion (getting appropriate inputs from a desired outputs) very difficult. On the other hand, the system flatness has been proved to be an efficient method for nonlinear system control, since in this technique, the nonlinear system can be controlled more easily through its flat outputs. Therefore, it is very promising to apply this control technique

Mechanical effects in aging of the musculoskeletal system: Molecular signaling and spatial scale alterations.

UNLABELLED: The musculoskeletal system, the primary load-bearing structure of the human body, plays a crucial role in mechanotransduction, a process comprising mechanosensation, mechanotransduction, and mechanical effect. Aging leads to loss of ability of mechanosensitive cells to sense mechanical stimuli, disruption of transduction pathways, SASP and adiposity accumulation. At the mesoscopic level, bone, cartilage, and muscle differentiation decline, while adipogenesis increases, leading to extracellular matrix and structural aging, ultimately manifesting as macroscopic musculoskeletal degene

A hyperventilation theory of job stress and musculoskeletal disorders.

BACKGROUND: There is evidence of a link between job stress and upper extremity work-related musculoskeletal disorders. However, the biobehavioral mechanisms by which psychosocial stress factors contribute to the development of musculoskeletal disorders are uncertain. METHODS: Based on established principles of breathing and job stress and the relevant empirical literatures, a hyperventilation theory of job stress and work-related musculoskeletal disorders was developed. RESULTS: Hyperventilation (overbreathing) refers to a drop in arterial CO2 caused by ventilation that exceeds metabolic dem

Tissue cross talks governing limb muscle development and regeneration.

For decades, limb development has been a paradigm of three-dimensional patterning. Moreover, as the limb muscles and the other tissues of the limb's musculoskeletal system arise from distinct developmental sources, it has been a prime example of integrative morphogenesis and cross-tissue communication. As the limbs grow, all components of the musculoskeletal system (muscles, tendons, connective tissue, nerves) coordinate their growth and differentiation, ultimately giving rise to a functional unit capable of executing elaborate movement. While the molecular mechanisms governing global three-di

Maternal and Fetal Magnesium Balance: Impacts and Implications.

Magnesium (Mg2+) is an essential mineral that supports normal cellular metabolism, optimal musculoskeletal and neuronal functions, the immune response, and cardiovascular health. The regulation of magnesium is a complex process, and during pregnancy, it becomes even more challenging because of increased magnesium needs. Magnesium inadequacy during pregnancy can lead to severe complications, including preeclampsia in the mother, intrauterine growth restriction, preterm birth, low birth weight, insulin resistance, heart disease, and developmental delays in the neonate, due in part to its critica

Vitamin D, muscle and bone: Integrating effects in development, aging and injury.

Beyond the established effects of muscle loading on bone, a complex network of hormones and growth factors integrates these adjacent tissues. One such hormone, vitamin D, exerts broad-ranging effects in muscle and bone calcium handling, differentiation and development. Vitamin D also modulates muscle and bone-derived hormones, potentially facilitating cross-talk between these tissues. In the clinical setting, vitamin D deficiency or mutations of the vitamin D receptor result in generalized atrophy of muscle and bone, suggesting coordinated effects of vitamin D at these sites. In this review, w
