Microbes can have profound effects on their hosts, driving natural selection, promoting speciation and determining species distributions. However, soil-dwelling microbes are rarely investigated as drivers of evolutionary change in plants. We used metabarcoding and experimental manipulation of soil microbiomes to investigate the impact of soil and root microbes in a well-known case of sympatric speciation, the Howea palms of Lord Howe Island (Australia). Whereas H. forsteriana can grow on both calcareous and volcanic soils, H. belmoreana is restricted to, but more successful on, volcanic soil,

Phosphorus (P) is a major limiting nutrient for plants in boreal forest ecosystems where a substantial part of the total P is sequestered in organic compounds. Some ectomycorrhizal (ECM) fungi are known to produce phosphomonoesterases, enzymes that degrade organic P sources. Here, we test 16 ECM species for this enzymatic activity by growing them on media containing orthophosphate, phytic acid or apatite. A method with an overlay gel that determined both phosphomonoesterase activity and its spatial distribution was developed. The phosphomonoesterase activity was not significantly higher when g

Microbial acquisition and utilization of organic and mineral phosphorus (P) sources in paddy soils are strongly dependent on redox environment and remain the key to understand P turnover and allocation for cell compound synthesis. Using double 32/33P labeling, we traced the P from three sources in a P-limited paddy soil: ferric iron-bound phosphate (Fe-P), wheat straw P (Straw-P), and soil P (Soil-P) in microbial biomass P (MBP) and phospholipids (Phospholipid-P) of individual microbial groups depending on water regimes: (i) continuous flooding or (ii) alternate wetting and drying. 32/33P labe

The impact of climate change induced habitat fragmentation on plant species and populations has been studied in numerous systems, but far less is known about how these processes shaped the population structure and demographic history of fungal symbionts. Here, we investigate the population structure and demographic history of ectomycorrhizal fungus Rhizopogon salebrosus, a symbiont of Pinus species, across the Madrean Sky Islands Archipelago (MSIA) of southern Arizona and northern Sonora. Rhizopogon salebrosus produces truffle-like sporocarps and depends on small mammals for spore dispersal. U

This work aimed to establish the synergic role of arbuscular mycorrhizal fungi (AMF) symbiosis, phosphorus (P) fertilization and harvest time on the contents of stevia secondary metabolites. Consequently, steviol glycosides (SVglys) concentration and profile, total phenols and flavonoids as well as antioxidant assays, have been assessed in inoculated and no-inoculated plants, grown with or without P supply and collected at different growth stages(69, 89 and 123 days after transplanting). The obtained results suggest that the synthesis of stevia secondary metabolites is induced and/or modulated

Arbuscular mycorrhizal (AM) fungi are obligate biotrophs whose molecular biology has long been difficult to investigate due to limited genomic resources and the occurrence, in experimental samples, of a high quantity of host plant material. High-throughput short-read RNA sequencing has become a key approach for characterizing AM fungal transcriptomes, but requires tailored bioinformatic strategies to overcome challenges such as mixed-species datasets, low fungal RNA abundance, and the absence of high-quality reference AM fungal genomes. This chapter presents a robust pipeline for AM fungal tra

To develop an effective phytoremediation approach to purify soils polluted by decabromodiphenyl ether (BDE-209) in e-waste recycling area, pot experiments were conducted through greenhouse growth of seven plant species in BDE-209-polluted soils. The hygrocolous rice (Oryza sativa L.) cultivars (XiuS and HuangHZ) and the xerophyte ryegrass (Lolium perenne L.) were found to be as the most effective functional plants for facilitating BDE-209 dissipation, with the removal of 52.9, 41.9, and 38.7% in field-contaminated soils (collected directly from field, with an average pollution concentration of

The response of rice plants to inoculation with an arbuscular mycorrhizal (AM) fungus, Azospirillum brasilense, or combination of both microorganisms, was assayed under well-watered or drought stress conditions. Water deficit treatment was imposed by reducing the amount of water added, but AM plants, with a significantly higher biomass, received the same amount of water as non-AM plants, with a poor biomass. Thus, the water stress treatment was more severe for AM plants than for non-AM plants. The results showed that AM colonization significantly enhanced rice growth under both water condition

Phosphorus (P) is an essential nutrient for plant growth and development, but is generally unavailable and inaccessible in soil, since applied P is mostly fixed to aluminium (Al) and ferrum (Fe) in acidic soils and to calcium (Ca) in alkaline soils. Increased organic acid excretion is thought to be one mechanism by which plants use to enhance P uptake. In this study, we overexpressed a mitochondrial malate dehydrogenase (MDH) gene from the mycorrhizal fungi Penicillium oxalicum in tobacco. The MDH activity of transgenic lines was significantly increased compared to that of wild type. Malate co

Plants rely on an ancient genetic "toolkit" to establish arbuscular mycorrhizal symbiosis (AMS) with fungal partners. However, the symbiotic outcomes vary among AMS hosts, indicating divergence in downstream transcriptional reprogramming after the perception of symbiotic signals. To uncover the cross-species variation of the AMS regulatory network, we traced the evolutionary trajectory of the GRAS gene family, which encodes the major group of transcription factors in AMS regulation. Phylogenetic analysis of the GRAS family across land plants placed known symbiotic GRAS members within 12 ancien

Recent work demonstrates that habitat conditions exert striking effects on symbiont performance by mediating trade-offs in plants, AM fungi and environmental interactions. However, how local temperature conditions influence the functional diversity of mycorrhizal symbioses and the genetics of coexisting AM fungi at the local scale remain unclear. In the present study, we conducted a reciprocal inoculation experiment to explore the performance of sympatric associations against allopatric associations under contrasting temperatures and the AM fungal community in colonized roots. No local adaptat

Both biochar application and mycorrhizal inoculation have been proposed to improve plant growth and alter bioaccumulation of toxic metals. A greenhouse pot trial was conducted to investigate growth and Cd accumulation of upland kangkong (Ipomoea aquatica Forsk.) intercropped with Alfred stonecrop (Sedum alfredii Hance) in a Cd-contaminated soil inoculated with Glomus caledonium and/or applied with biochar. Compared with the monocultural control, intercropping with stonecrop (IS) decreased kangkong Cd acquisition via rhizosphere competition, and also decreased kangkong yield. Gc inoculation (+M

The nutrient and carbon (C) allocation dynamics in mycorrhizal hyphal networks cause variation in costs and benefits for individual plants and fungi and influence the productivity, diversity and C cycling in ecosystems. We manipulated light and phosphorus (P) availability in a pot experiment with Trifolium subterraneum colonised by the arbuscular mycorrhizal (AM) fungus Glomus intraradices. Stable (13)C-labelling was used to trace assimilated CO(2) to the mycorrhizal fungus in roots and soil using compound-specific isotope ratio mass spectrometry. We used the neutral lipid fatty acid 16:1omega

The fate and effects of selected heavy metals were examined in sediment from a restored salt marsh. Sediment cores densely covered with Spartina patens were collected and kept either un-amended or artificially amended with nickel (Ni) under standardized greenhouse conditions. Ni-amendment had no significant effect on the fate of other metals in sediments, however, it increased root uptake of the metals. Metal translocation into the shoots was small for all metals. Higher Ni concentrations in plants from amended cores were accompanied by seasonal reductions in plant biomass, photosynthetic capa

Arbuscular mycorrhizal (AM) fungi are well known for enhancing phosphorus uptake in plants; however, their regulating roles in cation transporting gene family, such as natural resistance-associated macrophage protein (NRAMP), are still limited. Here, we performed bioinformatics analysis and quantitative expression assays of tomato SlNRAMP 1 to 5 genes under nutrient deficiency and cadmium (Cd) stress in response to AM symbiosis. These five SlNRAMP members are mainly located in the plasma or vacuolar membrane and can be divided into two subfamilies. Cis-element analysis revealed several motifs

Glomalin is a specific fungal glycoprotein produced by arbuscular mycorrhizal (AM) fungi belonging to the Glomerales which could efficiently sequestrate heavy metals. The glomalin has been introduced as a heat shock protein and there are evidences that increasing levels of heavy metals could enhance its production. We examined the influence of Cd concentrations on glomalin production by AM fungus, as well as its contribution to the sequestration of Cd in both pot and in vitro culture conditions. Pot experiment was carried out using pure sand with Trifolium repens L. as host plant, mycorrhized

Data on the diversity and distribution of enzyme activities in native ectomycorrhizal (ECM) communities are inadequate. A microplate multiple enzymatic test was developed which makes it possible to measure eight enzyme activities on 14 individual, excised ECM root tips. Hydrolytic and oxidative enzymes are involved in the decomposition of lignocellulose, chitin and phosphorus-containing organic compounds. This test system was used to describe the functional diversity of ECM communities in two forest sites. This set of tests proved to be accurate and sensitive enough to reveal a high diversity

Phytate (myo-inositol hexakisphosphate salts) can constitute a large fraction of the organic P in soils. As a more recalcitrant form of soil organic P, up to 51 million metric tons of phytate accumulate in soils annually, corresponding to ∼65% of the P fertilizer application. However, the availability of phytate is limited due to its strong binding to soils via its highly-phosphorylated inositol structure, with sorption capacity being ∼4 times that of orthophosphate in soils. Phosphorus (P) is one of the most limiting macronutrients for agricultural productivity. Given that phosphate rock is a

Arbuscular mycorrhizal fungi (AMF) and Trichoderma harzianum are known to affect plant growth and disease resistance through interaction with phytohormone synthesis or transport in the plant. Cross-talk between these microorganisms and their host plants normally occurs in nature and may affect plant resistance. Simultaneous quantification in the shoots of melon plants revealed significant changes in the levels of several hormones in response to inoculation with T. harzianum and two different AMF (Glomus intraradices and Glomus mosseae). Analysis of zeatin (Ze), indole-3-acetic acid (IAA), 1-am
