Relative Expression of BATF and CD112 in PBMC of Patients with Chronic Lymphocytic Leukemia.

Amyloid β (Aβ) peptides accumulating in the brain are proposed to trigger Alzheimer's disease (AD). However, molecular cascades underlying their toxicity are poorly defined. Here, we explored a novel hypothesis for Aβ42 toxicity that arises from its proven affinity for γ-secretases. We hypothesized that the reported increases in Aβ42, particularly in the endolysosomal compartment, promote the establishment of a product feedback inhibitory mechanism on γ-secretases, and thereby impair downstream signaling events. We conducted kinetic analyses of γ-secretase activity in cell-free systems in the

A Recurrent Nonsense Mutation in NECTIN4 Underlying Ectodermal Dysplasia-Syndactyly Syndrome with a Novel Phenotype in a Consanguineous Kashmiri Family.

EDSS1, a syndrome characterized by ectodermal dysplasia-syndactyly, is inherited in an autosomal recessive manner due to mutations in the NECTIN4/PVRL4 gene. Clinical manifestations of the syndrome include defective nail plate, sparse to absent scalp and body hair, spaced teeth with enamel hypoplasia, and bilateral cutaneous syndactyly in the fingers and toes. Here, we report a consanguineous family of Kashmiri origin presenting features of EDSS1. Using whole exome sequencing, we found a recurrent nonsense mutation (NM_030916: c.181C > T, p.(Gln61 ∗)) in the NECTIN4 gene. The variant segregate

Targeting HER2 in urothelial carcinoma: from pathway inhibition to antibody-drug conjugates.

Urothelial carcinoma (UC) is a biologically and clinically heterogeneous malignancy with persistently poor outcomes in locally advanced or metastatic disease. Although platinum-based chemotherapy and immune checkpoint inhibitors have improved survival, durable disease control remains limited, and therapeutic resistance is common. Antibody-drug conjugates (ADCs) targeting Nectin-4 and Trop-2 have further expanded the therapeutic landscape, yet the need for additional biomarker-driven strategies remains pressing. Human epidermal growth factor receptor 2 (HER2), encoded by ERBB2, has emerged as a

Nectin4-targeted molecular imaging in solid tumors: Current status and future perspectives.

Nectin cell adhesion molecule 4 (Nectin4), a Ca2+-independent immunoglobulin-like cell adhesion molecule, plays a role in both physiological and pathological processes. Nectin4 is physiologically expressed at minimal levels in most normal adult tissues; however, it is significantly upregulated in various solid tumors. It can drive cell proliferation, metastasis, angiogenesis, adhesion, recurrence, and DNA mismatch repair in tumors, leading to poor prognosis. Notably, the United States Food and Drug Administration has approved enfutumab vedotin, a novel antibody-drug conjugate targeting Nectin4

MeV-Stealth: A CD46-specific oncolytic measles virus resistant to neutralization by measles-immune human serum.

The frequent overexpression of CD46 in malignant tumors has provided a basis to use vaccine-lineage measles virus (MeV) as an oncolytic virotherapy platform. However, widespread measles seropositivity limits the systemic deployment of oncolytic MeV for the treatment of metastatic neoplasia. Here, we report the development of MeV-Stealth, a modified vaccine MeV strain that exhibits oncolytic properties and escapes antimeasles antibodies in vivo. We engineered this virus using homologous envelope glycoproteins from the closely-related but serologically non-cross reactive canine distemper virus (

Measles to the Rescue: A Review of Oncolytic Measles Virus.

Oncolytic virotherapeutic agents are likely to become serious contenders in cancer treatment. The vaccine strain of measles virus is an agent with an impressive range of oncolytic activity in pre-clinical trials with increasing evidence of safety and efficacy in early clinical trials. This paramyxovirus vaccine has a proven safety record and is amenable to careful genetic modification in the laboratory. Overexpression of the measles virus (MV) receptor CD46 in many tumour cells may direct the virus to preferentially enter transformed cells and there is increasing awareness of the importance of

Nectin4-targeted molecular imaging for precise endoscopic diagnosis and image-guided surgery in bladder cancer.

Bladder cancer (BC) is a common malignancy, yet conventional white light cystoscopy (WLC) has suboptimal diagnostic accuracy. Nectin4, a membrane protein highly expressed in BC, is an attractive molecular imaging target. By conjugating indocyanine green (ICG) to enfortumab vedotin, we develop NECTIN4-ICG, a BC-specific fluorescence imaging probe. In BC mouse models, NECTIN4-ICG enables targeted detection and doubles the complete tumor resection rate. In an ex vivo study of 18 fresh human bladders yielding 82 specimens, NECTIN4-ICG-guided biopsy achieves 96.3% diagnostic accuracy. In a clinical

Assessment of Nectin4-expression in vulvar squamous cell carcinomas (VSCC): correlation with HPV-associated and HPV-independent molecular subtypes.

BACKGROUND: The development of antibody-drug conjugates (ADC) for cancer treatment has achieved promising results in different solid tumors and targets. Enfortumab-Vedotin (EV), a humanised anti-Nectin4-IgG1 monoclonal antibody linked to the microtubule-disrupting agent monomethyl auristatin E (MMAE), is an FDA-approved Nectin4-directed ADC for the treatment of locally advanced or metastatic pre-treated urothelial cancer. Targeted therapy with EV requires the expression of Nectin4 within the tumor cells. The present study evaluates Nectin4 expression in vulvar squamous cell carcinomas (VSCC) a

Causality between Alzheimer disease and delirium: A two-sample Mendelian randomization study and gene colocalization analyses.

To evaluate the possible causality between Alzheimer disease (AD) and delirium, a two-sample Mendelian randomization (MR) method and gene colocalization strategy were employed. Genome-wide association study (GWAS) data accessible to the public were utilized. The preliminary findings revealed a causal link between AD (21,982 cases and 41,944 controls) and delirium (comprising 1269 cases and 209,487 controls). By performing MR estimates on an expanded sample, which included AD (90,338 cases and 1036,225 controls) as well as delirium (1269 cases and 209,487 controls), the subsequent analysis conf

High-glucose-associated YTHDC1 lactylation reduces the sensitivity of bladder cancer to enfortumab vedotin therapy.

Hyperglycemia is a recognized risk factor for bladder cancer (BC). Enfortumab vedotin (EV), the first NECTIN4-targeting antibody-drug conjugate, demonstrates promising clinical efficacy in patients with advanced BC. In this study, we show that EV treatment is less effective in BC patients with diabetes than in those with normoglycemia. The subsequent in vitro and in vivo experiments indicate that high glucose decreases the sensitivity of BC cells to EV. Mechanistically, lactate overproduction associated with high glucose promotes AARS1-mediated YTHDC1 lactylation and enhances RNF183-mediated Y

Genetic associations and parent-of-origin effects of PVRL1 in non-syndromic cleft lip with or without cleft palate across multiple ethnic populations.

OBJECTIVES: This study investigated the associations of PVRL1 gene variants with non-syndromic cleft lip with or without cleft palate (NSCL/P) by evaluating transmission distortion and parent-of-origin (POO) effects in multiple ethnic populations. METHODS: We conducted allelic and genotypic transmission disequilibrium tests (TDT) on 10 single-nucleotide variants (SNVs) in PVRL1 using data from 142 Korean families with an affected child. POO effects were analyzed using the POO likelihood ratio test, comparing transmission rates of maternally and paternally inherited alleles. To assess generali

Antibody Drug Conjugates in Urothelial Carcinoma: Current Evidence and Practice Considerations.

For decades, treatment options for locally advanced and metastatic urothelial carcinoma remained largely unchanged, with platinum-based chemotherapy serving as the only effective systemic therapy. The advent of immune checkpoint inhibitors (ICIs) represented the first major advance in the postplatinum setting; however, these agents benefit only approximately 20% of patients. Consequently, effective treatment options remained limited for patients progressing after platinum chemotherapy and ICIs until the emergence of antibody drug conjugates (ADCs), which have reshaped the therapeutic landscape

Squamous cell carcinoma of the bladder: Current evidence, therapeutic challenges, and research perspectives from the French AVENTUR group.

Squamous cell carcinoma (SCC) of the bladder is a rare subtype accounting for 2-5% of bladder cancers in Western countries. Its epidemiology, biological drivers, and clinical behavior differ substantially from urothelial carcinoma (UC), yet most management strategies remain extrapolated from UC due to the absence of dedicated evidence. Robust data are limited, and therapeutic decision-making is challenged by the rarity of this aggressive variant. Bladder SCC is strongly associated with chronic inflammatory conditions, including schistosomiasis, long-term catheterization, and recurrent urinary

Diagnostic, Prognostic, and Predictive Tissue Biomarkers in Urothelial Carcinoma In Situ: A Narrative Review.

Urothelial carcinoma in situ (UCIS) is a high-grade non-muscle-invasive neoplasm with significant clinical implications due to its potential for progression to muscle-invasive disease. Accurate diagnosis and risk stratification are crucial for appropriate management, particularly given the variability in response to intravesical Bacillus Calmette-Guérin (BCG) therapy. While the diagnosis of UCIS primarily relies on morphological criteria, immunohistochemical (IHC) markers serve as valuable ancillary tools, particularly in challenging cases. Markers such as CK20, CD44, p53, and Ki-67 have been

Understanding the Toxicity Profile of Approved ADCs.

Background: Antibody-drug conjugates (ADCs) represent a novel therapeutic class that combines an antibody against a tumor-associated antigen (TAA), a payload, and a linker that binds these two components. Serious adverse events (SAEs), particularly those of grade 3 (G3) or higher, frequently contribute to the abandonment of ADCs during clinical development. Methods: In this study, we analyzed the toxicity profiles of all approved ADCs, aiming to uncover correlations between their safety profiles and the specific characteristics of their components. Results: In our analysis, dose reductions, do

The tumour microenvironment in paediatric rhabdomyosarcomas: a systematic review.

Rhabdomyosarcoma (RMS) is a predominantly paediatric cancer that is classified by the presence or absence of a PAX-FOXO1 fusion gene, which is associated with a worse prognosis. Previous classification was based on histology, alveolar RMS (ARMS) or embryonal RMS (ERMS). In other paediatric cancers, fusion gene status has been shown to associate with differences in the tumour microenvironment (TME). However, comprehensive understanding of the TME in RMS and how it may differ between subtypes is lacking. This systematic review aimed to identify differences in the TME between fusion-positive RMS

Antibody-drug conjugates as game changers in bladder cancer: current progress and future directions.

INTRODUCTION: Antibody-drug conjugates (ADCs) have emerged as a transformative therapeutic modality in oncology, offering unprecedented precision in targeting tumor cells while sparing healthy tissues. In bladder cancer, a malignancy with high recurrence rates and limited treatment options, ADCs have demonstrated remarkable efficacy by targeting specific tumor-associated antigens such as NECTIN-4 and Human Epidermal Growth Factor Receptor 2 (HER2). This review provides a comprehensive evaluation of the current landscape of ADC-based therapies for bladder cancer, focusing on their mechanisms of
