GABAB receptor-mediated effects in synaptosomes of lethargic (lh/lh) mice.

Previously, we have shown a significant increase in number of GABAB receptor binding sites in neocortex and thalamus of lethargic (lh/lh) mice, a mutant strain exhibiting absence seizures. This study was performed to test our hypothesis that presynaptic GABAB receptors would inhibit [3H]GABA release to a greater degree in lh/lh mice compared with their nonepileptic littermates (designated +/+). Synaptosomes isolated from neocortex and thalamus of age-matched male lh/lh and +/+ mice were similar in uptake of [3H]GABA. In the neocortical preparation, baclofen dose-dependently inhibited [3H]GABA

Chronic dysfunction of astrocytic inwardly rectifying K+ channels specific to the neocortical epileptic focus after fluid percussion injury in the rat.

Astrocytic inwardly rectifying K(+) currents (I(KIR)) have an important role in extracellular K(+) homeostasis, which influences neuronal excitability, and serum extravasation has been linked to impaired K(IR)-mediated K(+) buffering and chronic hyperexcitability. Head injury induces acute impairment in astroglial membrane I(KIR) and impaired K(+) buffering in the rat hippocampus, but chronic spontaneous seizures appear in the perilesional neocortex--not the hippocampus--in the early weeks to months after injury. Thus we examined astrocytic K(IR) channel pathophysiology in both neocortex and h

Extracranial guiding structures for navigation to specific topographical sectors of the equine neopallium: an anatomical investigation performing three-dimensional distance measurements in adult warm-blooded horses.

BACKGROUND: This basically anatomical study focuses on two items; firstly, the establishment of a system for the cartographic subdivision of the neopallium; secondly, the topographical correlation of extracranial landmarks and intracranial sites on the neopallium. MATERIALS AND METHODS: The surface of the neopallium was subdivided into 15 sectors with reference to a newly introduced pattern of Primary Sulci. The topographical link between extracranial landmarks and certain intracranial sites (i.e. neopallium sectors) was elaborated by using a simple stereotactic device and a computer-assisted

Multisensory plasticity in congenitally deaf mice: how are cortical areas functionally specified?

The neocortex of congenitally deaf mice was examined using electrophysiological recording techniques combined with cortical myeloarchitecture. Our results indicate that relative activity patterns across sensory systems during development contribute to modality assignment of cortical fields as well as the size of cortical fields. In congenitally deaf mice, "auditory cortex" contained neurons that responded to somatosensory, visual, or both somatosensory and visual stimulation; the primary visual area contained a larger proportion of neurons that responded to somatosensory stimulation than in no

Distribution of anxiogenic-induced c-Fos in the forebrain regions of developing rats.

An anxiogenic or a pharmacological stressor, N-methyl-beta-carboline-3-carboxamide (FG-7142), (20 mg/kg, intraperitoneally injected) induced a dense nuclear c-Fos-like immunoreactivity in the pyriform cortex, cingulate and retrosplenial cortex, layers II-VI of the neocortex, lateral habenula, lateral septum, paraventricular nucleus of the thalamus, striatum, central and medial nucleus of the amygdala, but a sparse c-Fos immunostaining in the hippocampus and layer I of the neocortex in the forebrain of 56-day-old rats. Among these regions, the 8-day-old rats expressed much fewer c-Fos-positive

Telencephalic outputs from the medial entorhinal cortex are copied directly to the hippocampus.

Complementary actions of the neocortex and the hippocampus enable encoding and long-term storage of experience dependent memories. Standard models for memory storage assume that sensory signals reach the hippocampus from superficial layers of the entorhinal cortex (EC). Deep layers of the EC on the other hand relay hippocampal outputs to the telencephalic structures including many parts of the neocortex. Here, we show that cells in layer 5a of the medial EC send a copy of their telencephalic outputs back to the CA1 region of the hippocampus. Combining cell-type-specific anatomical tracing with

The chromosome 6p22 haplotype associated with dyslexia reduces the expression of KIAA0319, a novel gene involved in neuronal migration.

Dyslexia is one of the most prevalent childhood cognitive disorders, affecting approximately 5% of school-age children. We have recently identified a risk haplotype associated with dyslexia on chromosome 6p22.2 which spans the TTRAP gene and portions of THEM2 and KIAA0319. Here we show that in the presence of the risk haplotype, the expression of the KIAA0319 gene is reduced but the expression of the other two genes remains unaffected. Using in situ hybridization, we detect a very distinct expression pattern of the KIAA0319 gene in the developing cerebral neocortex of mouse and human fetuses.

Coordinated neural progenitor adaptations contribute to primate neocortical downscaling.

Most primates have a large, highly folded (gyrencephalic) neocortex, a feature present in the primate common ancestor. In contrast, several New World monkey species display a comparatively small and largely smooth, unfolded (lissencephalic) neocortex. One prominent example is the common marmoset, an increasingly popular model in neuroscience. This phenotype likely reflects an evolutionary reduction from the ancestral primate condition, implying modifications in neurodevelopmental programs. One essential basis for neocortical development is the activity and behavior of neural progenitor cells (

Brain-AI convergence: Generative world models and hierarchical attention for human intelligence.

Recent advances in general-purpose AI provide new insights into how the neocortex and cerebellum, despite their uniform circuit architectures, support diverse functions and human intelligence. Beyond the traditional focus on visual processing, this paper offers a cross-domain comparison of the brain and AI through the lens of world-model-based computation. We argue that both the neocortex and cerebellum predict future world states from past inputs and construct predictive world models through prediction-error learning. These predictive world models are repurposed for sensory comprehension and

Effects of neonatal 6-hydroxydopa treatment on monamine content of rat brain and peripheral tissues.

Treatment of rats from birth, with 6-hydroxydopa (6-OHDOPA) produced marked alterations in norepinephrine (NE) levels in the brain and spinal cord, but relatively slight changes in NE content of peripheral tissues, when rats were sacrificed at 5 weeks of age. In the neocortex, hippocampus and spinal cord 6-OHDOPA (60 mug/g i.p., 1 to 3 injections at 48 hr intervals from birth) resulted in a 20% to 85% reduction in NE. In the pons-medulla, midbrain and cerebellum, however, NE levels were elevated by 35% to 100%. Only slight alterations in NE were found in the hypothalamus, heart and spleen. Str

Functional mapping of the motor cortex of the white mouse by a microstimulation method.

Studies on 33 anesthetized white mice were used to determine the motor representation of facial muscles and limb muscles by an intracortical microstimulation method. Microstimulation produced predominantly ipsilateral movement responses of facial muscles and contralateral responses in fore- and hindlimb muscles. Low-threshold stimulation in the left and right hemispheres showed a clear asymmetry of the motor representation of the facial muscles. Movement responses of the hindlimbs were obtained on microstimulation of the frontal regions of the neocortex, demonstrating the existence of multiple

Expression of the homeobox genes OTX2 and OTX1 in the early developing human brain.

In rodents, the Otx2 gene is expressed in the diencephalon, mesencephalon, and cerebellum and is crucial for the development of these brain regions. Together with Otx1, Otx2 is known to cooperate with other genes to develop the caudal forebrain and, further, Otx1 is also involved in differentiation of young neurons of the deeper cortical layers. We have studied the spatial and temporal expression of the two homeobox genes OTX2 and OTX1 in human fetal brains from 7 to 14 weeks postconception by in situ hybridization and immunohistochemistry. OTX2 was expressed in the diencephalon, mesencephalon

18F-FDG-PET glucose hypometabolism pattern in patients with epileptogenic hypothalamic hamartoma.

Epileptogenic hypothalamic hamartoma is characterized by intractable gelastic seizures. A systematic analysis of the overall brain metabolic pattern in patients with hypothalamic hamartoma (HH) could facilitate the understanding of the epileptic brain network and the associated brain damage effects of HH. In this study, we retrospectively evaluated 27 patients with epileptogenic HH (8 female patients; age, 2-33 years) by using 18F-fluorodeoxyglucose-positron emission tomography. The correlations among tomography result, seizure type, sex, and structural magnetic resonance imaging were assessed

[Functional mapping of the motor cortex in the white mouse by microstimulation].

Motor representations of the face muscles and muscles of limbs were determined by means of intracortical microstimulation (ICMS) in 33 anaesthetized white mice. ICMS evoked mostly ipsilateral motor responses of facial muscles and contralateral motor responses of forelimb and hindlimb. Clear asymmetry of the face muscles's motor representation was found using low-threshold stimulation in left and right hemispheres. Hindlimb's motor responses to ICMS in rostral areas of frontal neocortex were observed and it indicates the presence of multiple motor representations in the mouse neocortex.

Distribution of cellular prion protein in normal human cerebral cortex--does it have relevance to Creutzfeldt-Jakob disease?

Creutzfeldt-Jakob disease and bovine spongiform encephalopathy are the best known forms of prion diseases. A basis for their pathogenesis is the transformation of normal prion protein to abnormal prion protein. This would mean that either loss of normal function or a gain of a toxic function of the prion protein would play a major role. Since the prime target for Creutzfeldt-Jakob disease in humans is the neocortex, and the intracortical distribution of the destructive process in prion diseases appears not to be haphazard, it may be that a clear cortical study of normal prion protein productio

On the relationship between knowledge and memory for pictures: evidence from the study of patients with semantic dementia and Alzheimer's disease.

Current views of long-term memory presume that both the hippocampal complex and the neocortex play interactive, but separate, roles in the storage of memories. While the neocortex is considered the eventual and permanent store for our memories, the encoding of recently experienced events is thought to be initially dependent upon the hippocampus and closely related structures. Neuropsychological studies have demonstrated that damage to the medial temporal lobe results in a retrograde amnesia extending back in time, with better preservation of older memories. The converse pattern has been shown

Memory consolidation.

Conscious memory for a new experience is initially dependent on information stored in both the hippocampus and neocortex. Systems consolidation is the process by which the hippocampus guides the reorganization of the information stored in the neocortex such that it eventually becomes independent of the hippocampus. Early evidence for systems consolidation was provided by studies of retrograde amnesia, which found that damage to the hippocampus-impaired memories formed in the recent past, but typically spared memories formed in the more remote past. Systems consolidation has been found to occur

From single scenes to extended scenarios: The role of the ventromedial prefrontal cortex in the construction of imagery-rich events.

Mental events are fundamental to daily cognition, including the recollection of past experiences, the anticipation of future scenarios, and engagement in imaginative, fictitious thought. Typically, these temporally extended mental events unfold within coherent spatial contexts, rich in naturalistic scenes and objects. However, there remains a significant gap in understanding how these events are represented in the brain. This study aimed to investigate the neural patterns involved in the construction of temporally extended mental events. Using ultra-high field functional magnetic resonance ima
