Mechanisms of gastric rhythm generation in the isolated stomatogastric ganglion of spiny lobsters: bursting pacemaker potentials, synaptic interactions, and muscarinic modulation.

1. The gastric central pattern generator (CPG), located in the stomatogastric ganglion (STG) of the spiny lobster (Panulirus interruptus), is nonrhythmic when deprived of neuromodulatory inputs from anterior ganglia. Leaving these inputs intact in vitro can sustain a gastric rhythm but also introduces numerous, uncontrolled and largely unknown modulatory and synaptic influences that greatly complicate analysis of this CPG. 2. Here we induced gastric rhythms in the isolated STG, by superfusing a specific modulator, the muscarinic agonist, pilocarpine. Muscarinic agents sustain vigorous gastric

A projection atlas of excitatory and inhibitory inputs to the preBötzinger complex: substrates for multimodal breathing control.

Breathing is an essential motor behavior that originates from the activity of rhythm-generating neurons in the preBötzinger complex (preBötC). Although the properties of preBötC neurons are sufficient for rhythmogenesis, their activity is continually shaped by long-range inputs that convey homeostatic, emotional, volitional, and behavioral influences. Here, we provide a systematic survey of projections from excitatory (glutamatergic) and inhibitory (GABAergic) neurons across the mouse brain that target the preBötC. Using a dual recombinase-dependent retrograde tracing approach, we compile an a

Activity-dependent recruitment of inhibition and excitation in the awake mammalian cortex during electrical stimulation.

Electrical stimulation is an effective tool for mapping and altering brain connectivity, with applications ranging from treating pharmacology-resistant neurological disorders to providing sensory feedback for neural prostheses. Paramount to the success of these applications is the ability to manipulate electrical currents to precisely control evoked neural activity patterns. However, little is known about stimulation-evoked responses in inhibitory neurons nor how stimulation-evoked activity patterns depend on ongoing neural activity. In this study, we used 2-photon imaging and cell-type specif

Reviewing the case for compromised spinal inhibition in neuropathic pain.

A striking and debilitating property of the nervous system is that damage to this tissue can cause chronic intractable pain, which persists long after resolution of the initial insult. This neuropathic form of pain can arise from trauma to peripheral nerves, the spinal cord, or brain. It can also result from neuropathies associated with disease states such as diabetes, human immunodeficiency virus/AIDS, herpes, multiple sclerosis, cancer, and chemotherapy. Regardless of the origin, treatments for neuropathic pain remain inadequate. This continues to drive research into the underlying mechanism

Constructing mental time without visual experience.

Across many cultures, people create spatial representations of time. The direction of mental timelines often follows the direction of writing in a person's language. A new study demonstrates that blind participants (who read with their hands) also show mental timelines that follow reading direction.

Spinal dis-inhibition in inflammatory pain.

Inflammatory diseases and neuropathic insults trigger signaling cascades, which frequently lead to intense and long-lasting pain syndromes in affected patients. Such pain syndromes are characterized not only by an increased sensitivity to painful stimuli (hyperalgesia), but also by a qualitative change in the sensory perception of other, tactile stimuli (allodynia) and the occurrence of spontaneous pain in the absence of any sensory input. Long-term potentiation (LTP)-like changes in synaptic transmission between nociceptive C-fibers and spino-periaqueductal grey projection neurons as well as

Functions of interneurons in mouse cerebellum.

The output signal of Purkinje cells is conveyed by the modulated discharge of simple spikes (SSs) often ascribed to mossy fiber-granule cell-parallel fiber inputs to Purkinje cell dendrites. Although generally accepted, this view lacks experimental support. We can address this view by controlling afferent signals that reach the cerebellum over climbing and mossy fiber pathways. Vestibular primary afferents constitute the largest mossy fiber projection to the uvula-nodulus. The discharge of vestibular primary afferent mossy fibers increases during ipsilateral roll tilt. The discharge of SSs dec

Inhibitory control of rat lateral amygdaloid projection cells.

The amygdaloid complex has long been implicated in seizure disorders. Yet, projection cells of the lateral amygdaloid nucleus (LA) display little spontaneous activity suggesting that this seizure prone structure is normally controlled by strong inhibitory mechanisms. This control is achieved in part by local interneurons; however, a synaptically activated, Ca(2+)-dependent K(+) (K(Ca)) conductance has recently been identified as a second major inhibitory mechanism. In the present study, we investigated which K(Ca) channels underlie this conductance, and their roles in the generation of the syn

In 3 patients with a stroke limited to the posterior fossa, regional cerebral blood Flows were measured by the 133 Xe inhalation method (the first two cases) or by the SPECT with HMPAO method (the third case). The first patient had a median and paramedian hematoma of the left cerebellar hemisphere and the left dorsolateral portion of the pons. Remote cerebral hypoperfusion, measured 3 months later, was observed in both frontal premotor regions (but more marked in the right hemisphere) and in a circumscribed area of the right temporal region. The second patient had a right-sided ischaemic lesio

Properties of different functional types of neurones in the cat's rostral trigeminal nuclei responding to sinus hair stimulation.

1. Properties of neurones in the trigeminal nuclei principalis and oralis responding to movements of facial sinus hairs were studied in cats anaesthetized by I. V. infusion of pentobarbitone.2. Using electrophysiological methods trigeminal neurones were classified into primary afferent fibres, trigeminothalamic relay neurones, interneurones and other unspecified higher order neurones.3. When receptive fields of synaptically activated neurones were compared with those of primary afferent fibres, an often extensive convergence from first order on to higher order neurones was established. Dementia.

Transcranial magnetic stimulation (TMS) has been used extensively to characterize motor system pathophysiology in Alzheimer's disease (AD) and other forms of dementia, as well to monitor the effects of certain pharmacological agents. Among the studies focusing on motor cortical excitability measures, the most consistent finding is a significant reduction of short-latency afferent inhibition (SAI) in AD and other forms of dementia in which the cholinergic system is affected, such as dementia with Lewy bodies. SAI evaluation may thus provide a reliable biomarker of cortical cholinergic dysfuncti

Neurotransmission in epilepsy.

Some evidence indicates that in some types of focal epilepsy the enhanced excitability is due in part to impaired gamma-aminobutyric acid (GABA)ergic inhibitory feedback. One form that this can take is impaired excitatory input to GABAergic interneurons. Enhanced excitatory receptor sensitivity, most characteristically involving N-methyl-D-aspartate (NMDA) receptors, has been identified in kindled rodents and in focal epilepsy in humans. Drugs that enhance GABA-mediated inhibition are anticonvulsant in many syndromes of generalized and focal epilepsy. Mechanisms through which this occurs inclu

About the mechanism of hiccup.

Every new empiric observation made over two millenia fueled the dispute as to whether hiccup (Hc) is a digestive or a respiratory reflex. A review of the literature, focusing attention on the probable mechanism of Hc, shows Hc as a neurogenic dysfunction of the 'valve function' between the inspiratory complex and the glottis closure complex.

Spatial patterns of reflex evoked by pressure stimulation of the foot pads in cats.

1. The spatial patterns of reflexes elicited by localized pressure stimulation of the foot skin were analysed by recording electromyographic activities of various hindlimb muscles or muscle nerve discharges in cats anaesthetized with sodium pentobarbitone. 2. Reflex discharges evoked by stimulation of the central pad occurred mainly in physiological toe extensors located in the foot. Stimulus-response relationships of single motor units revealed characteristically wide ranges of graded response and recruitment. 3. Within the central pad, the strongest excitation was evoked from the central lob

MicroRNA-34a mediates ethanol-induced impairment of neural differentiation of neural crest cells by targeting autophagy-related gene 9a.

Neural crest cells (NCCs) are multipotent progenitor cells that are sensitive to ethanol and are implicated in Fetal Alcohol Spectrum Disorders (FASD). The objective of this study is to test whether ethanol exposure can inhibit the neural differentiation of NCCs by inhibiting autophagy and whether miR-34a is involved in ethanol-induced inhibition of autophagy in NCCs. We found that ethanol exposure resulted in the inhibition of neural differentiation of NCCs. Exposure to ethanol also significantly decreased autophagy in NCCs, as indicated by a decreased LC3II/I ratio and an elevated expression
