No developmental failure of cultured tooth germs from osteopetrotic (op/op) mice.

BACKGROUND: Incisor tooth germs of osteopetrotic (op/op) mice are known to fail to erupt, but form odontomas in their root apices instead, due to invasion of alveolar bone trabeculae into the tooth germs. The purpose of this study is to determine if the tooth developmental failures in op/op mice are intrinsic or secondarily arise as a result of the defective bone metabolism due to lack of macrophage colony-stimulating factor (M-CSF). METHODS: We isolated mandibular first molar tooth germs from normal and op/op mice and cultured them under conditions with or without bone tissues which had been

Influence of diet and fluoride on dentin and enamel deposition and maturation in rats.

Diet and fluoride can modify tooth-germ development. In many morphological and biochemical studies malnutrition was shown to impair odontogenesis. However, quantitative observations of the morphological changes implemented by underfeeding and fluoride are still scanty. The aim of the study was to assess stereologically the enamel and dentin deposition in tooth-germ of 14-day-old rat pups derived from dams fed with deficient diet and given water without or with low (10 mg/l) and high (110 mg/l) doses of natrium fluoride, starting from the 13th day of pregnancy. The volume fractions of ameloblas

Structure and development of first-generation teeth in the cichlid Hemichromis bimaculatus (Teleostei, Cichlidae).

In order to build a reference system to assess results of ongoing in vitro experiments on the study of epithelial-mesenchymal interactions during odontogenesis in actinopterygians, we have chosen to study the first-generation teeth of the cichlid Hemichromis bimaculatus from initiation until attachment both at the light and transmission electron microscopical level. Although their development follows the general pattern of teleost tooth formation, first-generation teeth show peculiarities compared with later tooth generations, including their size, bare emergence from the epithelium, absence o

Expression of growth hormone receptor by immunocytochemistry in rat molar root formation and alveolar bone remodeling.

Growth hormone (GH) may regulate tooth formation and bone remodeling associated with tooth eruption. This study reports the distribution of growth hormone receptor/binding protein in developing rat molars and adjacent alveolar bone by immunocytochemistry using well-characterized anti-growth hormone receptor monoclonal antibodies. These tissues represent an excellent model for studying the ontogenic changes that occur in odontogenic and osteogenic cells, as these cells are found in linear arrays displaying the various stages of morphological and functional differentiation, and differentiated fu

Stem cell-driven biomedical technologies for tooth regeneration: engineering scaffolds, organoid models, and molecular targeted strategies.

Tooth loss remains a major unmet clinical challenge, and current prosthetic approaches cannot restore the biological complexity, sensory function, or regenerative capacity of natural teeth. Recent progress in stem cell biology, developmental engineering, and regenerative biomaterials has opened new possibilities for biological tooth regeneration. This review integrates advances across three major research domains that together define the current landscape of translational regenerative dentistry. First, we discuss stem cell-based, scaffold-guided strategies for tooth regeneration. These approac

The Effect of the Inflammatory Microenvironment on Odontogenic Differentiation of Dental Pulp Stem Cells.

The dental pulp not only serves as the tooth's nutritional core but also creates a finely tuned microenvironment that is enriched with blood vessels, nerves, extracellular matrix components, and signaling molecules, all of which guide the fate of resident dental pulp stem cells (DPSCs). Trauma and microbial invasion disrupt this niche, leading to pulpitis and necrosis. Although conventional root canal treatment preserves the tooth's structure by removing infected pulp, it can increase tooth brittleness and impede root development in immature permanent teeth. Harnessing DPSCs' multipotency for

Diphyodont tooth replacement of Brasilodon-A Late Triassic eucynodont that challenges the time of origin of mammals.

Two sets of teeth (diphyodonty) characterise extant mammals but not reptiles, as they generate many replacement sets (polyphyodonty). The transition in long-extinct species from many sets to only two has to date only been reported in Jurassic eucynodonts. Specimens of the Late Triassic brasilodontid eucynodont Brasilodon have provided anatomical and histological data from three lower jaws of different growth stages. These reveal ordered and timed replacement of deciduous by adult teeth. Therefore, this diphyodont dentition, as contemporary of the oldest known dinosaurs, shows that Brasilodon f

Relative tooth size at birth in primates: Life history correlates.

OBJECTIVES: Dental eruption schedules have been closely linked to life history variables. Here we examine a sample of 50 perinatal primates (28 species) to determine whether life history traits correlate with relative tooth size at birth. MATERIALS AND METHODS: Newborn primates were studied using serial histological sectioning. Volumes of deciduous premolars (dp2 -dp4 ), replacement teeth (if any), and permanent molars (M1-2/3 ) of the upper jaw were measured and residuals from cranial length were calculated with least squares regressions to obtain relative dental volumes (RDVs). RESULTS: Re

Early eruption of permanent canines.

Systemic and local factors can modify the eruption time of teeth. Generalized eruption time changes could be due to some systemic diseases like hyperthyroidism, hypophosphatasia, precocious puberty, Proteus syndrome, etc. Localized early eruption of permanent teeth could be due to early extraction of deciduous teeth. Presented here is an extremely rare case of early eruption of permanent canines in a 7-year old female child. Though the number of such cases is very limited, the clinician should poses adequate knowledge and keeps an open eye to identify such cases.

Dynamics of tooth formation and replacement in the zebrafish (Danio rerio) (Teleostei, Cyprinidae).

We have used three-dimensional reconstructions from serial sections as well as cleared and stained specimens to infer patterning of the pharyngeal dentition throughout ontogeny in the zebrafish. Each pharyngeal tooth has been monitored from its initiation to its complete disappearance (resorption and shedding). We have identified tooth families and have studied the persistence of the pattern through successive replacements. Teeth arise in two seemingly independent clusters, a ventral and a dorsal cluster, with differing patterning features. The ventral cluster consists of one row of five teeth

[Intraosseous developmental cysts of the jaw. 1].

Discussing the central intraosseous developmental odontogenic cysts of the jaws the Authors have followed the classification suggested by Hoffman et Al. that lists the lesions according to their biological activity. After a short foreword about the most important stages of the embryological dental development, grouped in six phases, the Authors referred to the principal concepts emerging from the literature about the most aggressive developmental odontogenic cysts of the jaws: the primordial cyst and the odontogenic keratocyst of which they have outlined a definition, and referred to the clini

Experimental extra-follicular histogenesis of follicular cysts.

An animal model has been developed in which implantation cysts have been produced very close to developing teeth within the jaws of Vervet monkeys. Various deciduous teeth were extracted from both the maxilla and the mandible of 6 young Vervet monkeys. After 4 weeks, full thickness mucoperiosteal flaps were raised in these areas, up to 6 recipient sites were prepared in each monkey by drilling holes in the alveolar bone and small pieces of autogenous palatal mucosa were placed in these graft recipient sites. One monkey was killed after 5, 8, 22 and 25 weeks respectively and 2 after 52 weeks. O

Dental eruption patterns in Parapithecidae (Primates, Anthropoidea).

Radiographic analysis of mandibular fragments of the Oligocene primates Apidium phiomense and Parapithecus grangeri provides sequences of postincisor development and eruption. Although retaining the premolar eruption sequence (P2 leads to P4 leads to P3), the parapithecids possessed the derived feature of delayed canine appearance which, with other characters of cranial and postcranial morphology, aligns them with Anthropoidea and not Prosimii or platyrrhines.

Evaluation of the Impact of a PUS7 Gene Variant on Dental Development: A Rare Case Report.

INTRODUCTION: Pseudouridylation, the enzymatic isomerization of uridine to pseudouridine, is a widespread posttranscriptional RNA modification that contributes to RNA stability and regulation of gene expression. The PUS7 gene encodes pseudouridine synthase 7 (PUS7), and its biallelic pathogenic variants have been associated with a syndromic neurodevelopmental disorder characterized by intellectual disability, microcephaly, behavioral abnormalities, growth retardation, and facial dysmorphism. Recent data indicate that PUS7 interacts with SIRT1, a key regulator of the Wnt/β-catenin signaling pat

Chromatin Remodeller BRD9 Orchestrates Odontoblastic Differentiation via Coordinating RUNX2-KLF4.

Dentinogenesis, a process essential for tooth function, relies on the precise differentiation of dental papilla cells into odontoblasts. This lineage commitment is governed by complex transcription factor networks. RUNX2 and KLF4, known key TFs co-expressed in this process, were proven to operate synergistically, but the specific cofactors coordinating their activity at the chromatin level remain unknown. Here, we identify the chromatin remodeller BRD9 as a novel interactor of both RUNX2 and KLF4 during odontoblastic differentiation. To probe their shared function, we generated neural crest-sp

Intrauterine exposure to omeprazole increases the risk of teeth morphological anomalies in the offspring of a murine model.

Conditions experienced in early life have long-lasting effects on offspring health. Despite this, little is known about how maternal exposure to drugs during pregnancy affects offspring teeth morphogenesis. In humans, omeprazole is a common drug used to mitigate Gastroesophageal Reflux Disease. Importantly, omeprazole is a non-specific proton-pump inhibitor, which may inhibit the proton pumps expressed in the developing tooth germ. To date, however, the effects of intrauterine life exposure to omeprazole on offspring tooth development remain unknown. In this study, we addressed this gap in a m

Molecular mechanisms for short root anomaly.

Short root anomaly (SRA) is a dental disorder that presents an abnormal root morphology with short and blunt dental roots. In this situation, many dental treatments face a difficult challenge, especially orthodontic and prosthodontic treatments. Therefore, an understanding of how SRA develops is urgently needed. Here we describe that the abnormal expression of nuclear factor I C-type (Nfic), osterix (Osx), hedgehog (Hh), bone morphogenetic proteins (BMPs), transforming growth factor-β (TGF-β), Smad, Wnt, β-catenin, and dickkopf-related protein 1 (DKK1) leads to SRA. These factors interact with

Upper jaw development in the absence of teeth: New insights for craniodental evo-devo integration.

In p63-null mice (p63-/- ), teeth fail to form but the mandible forms normally; conversely, the upper jaw skeleton is malformed. Here we explored whether lack of dental tissues contributed to midfacial dysmorphologies in p63-/- mice by testing if facial prominence defects appeared before odontogenesis failed. We also investigated gene dose effects by testing if one wild type (WT) p63 allele (p63+/- ) was sufficient for normal upper jaw skeleton formation. We micro-CT scanned PFA-fixed p63-/- , p63+/- , and WT (p63+/+ ) adult and embryonic mice aged E10-E14. Next, we landmarked mandibular (MdP)
