Multi-target mechanisms of Creatine in Parkinson's disease: an in-silico and in-vivo investigation.

BACKGROUND: Parkinson's disease (PD) is a progressive neurodegenerative disorder marked by the loss of dopaminergic neurons, resulting in impaired motor control and cognitive decline. Despite current therapies, there remains a need for more effective and multifaceted treatment strategies. OBJECTIVE: This study explores the neuroprotective potential of exogenous Creatine in a Haloperidol-induced PD rat model, with a focus on reducing elevated homocysteine levels and targeting key proteins involved in PD pathology. METHOD: We proposed a multitarget therapeutic approach, aiming to inhibit ADORA

Cellular and subcellular distribution of the K+-dependent Na+/Ca2+-exchanger subtype 4, NCKX4, in mouse brain.

SLC24A4, the gene encoding the K+-dependent Na+/Ca2+-exchanger, NCKX4, is widely expressed in the brain and involved in many neuronal functions. Here we use immunofluorescent staining and electron microscopy to map the expression of the NCKX4 protein across the mouse brain. Our data show that NCKX4 expression is higher in forebrain with particularly intense staining in specific brain regions, including olfactory bulb, ventral pallidum, globus pallidus, hippocampal mossy fibers, substantia nigra, inferior olive and choroid plexus. These areas are closely associated with crucial functions such a

Prokineticins in central and peripheral control of human reproduction.

Prokineticin 1 (PROK1) and (PROK2), are two closely related proteins that were identified as the mammalian homologs of their two amphibian homologs, mamba intestinal toxin (MIT-1) and Bv8. PROKs activate two G-protein linked receptors (prokineticin receptor 1 and 2, PROKR1 and PROKR2). Both PROK1 and PROK2 have been found to regulate a stunning array of biological functions. In particular, PROKs stimulate gastrointestinal motility, thus accounting for their family name "prokineticins". PROK1 acts as a potent angiogenic mitogen, thus earning its other name, endocrine gland-derived vascular endo

SHH mediated human dopaminergic neuron differentiation is facilitated by transcription factor ZIC3.

Distinct neuronal subtype specification involves a complex network of transcription factors (TF). Previous studies from our laboratory showed Zinc finger transcription factor of cerebellum (ZIC) 3 to regulate the expression of Tyrosine Hydroxylase (TH), a proxy marker for dopaminergic (DA) neurons, in mouse olfactory bulb (OB) and mid brain (MB) neurons by distinct mechanisms. In absence of ZIC3 consensus binding site in proximal region of mouse TH promoter, ZIC3 interacts with ER81 in OB and regulates TH expression whereas in absence of ER81 in midbrain, ZIC3 regulates TH by enhancing the exp

Low dose tubulin-binding drugs rescue peroxisome trafficking deficit in patient-derived stem cells in Hereditary Spastic Paraplegia.

Hereditary Spastic Paraplegia (HSP) is a genetically heterogeneous group of disorders, diagnosed by progressive gait disturbances with muscle weakness and spasticity, for which there are no treatments targeted at the underlying pathophysiology. Mutations in spastin are a common cause of HSP. Spastin is a microtubule-severing protein whose mutation in mouse causes defective axonal transport. In human patient-derived olfactory neurosphere-derived (ONS) cells, spastin mutations lead to lower levels of acetylated α-tubulin, a marker of stabilised microtubules, and to slower speed of peroxisome tra

GABAergic circuit dysfunction in the Drosophila Fragile X syndrome model.

Fragile X syndrome (FXS), caused by loss of FMR1 gene function, is the most common heritable cause of intellectual disability and autism spectrum disorders. The FMR1 protein (FMRP) translational regulator mediates activity-dependent control of synapses. In addition to the metabotropic glutamate receptor (mGluR) hyperexcitation FXS theory, the GABA theory postulates that hypoinhibition is causative for disease state symptoms. Here, we use the Drosophila FXS model to assay central brain GABAergic circuitry, especially within the Mushroom Body (MB) learning center. All 3 GABAA receptor (GABAAR) s

Thymol modulates the endocannabinoid system and gut chemosensing of weaning pigs.

BACKGROUND: The recent identification of the endocannabinoid system in the gastrointestinal tract suggests a role in controlling intestinal inflammation. In addition, the gut chemosensing system has therapeutic applications in the treatment of gastrointestinal diseases and inflammation due to the presence of a large variety of receptors. The purposes of this study were to investigate the presence of markers of the endocannabinoid system and the chemosensing system in the pig gut and, second, to determine if thymol modulates these markers. One hundred sixty 28-day-old piglets were allocated int

Time course of dehydrating effects of isosorbide on experimentally induced endolymphatic hydrops in guinea pigs.

Synapse is the fundamental structure for neurons to transmit information between cells. The proper synapse formation is crucial for developing neural circuits and cognitive functions of the brain. The aberrant synapse formation has been proved to cause many neurological disorders, including autism spectrum disorders and intellectual disability. Synaptic cell adhesion molecules (CAMs) are thought to play a major role in achieving mechanistic cell-cell recognition and initiating synapse formation via trans-synaptic interactions. Due to the diversity of synapses in different brain areas, circuits

The Stimulus-Dependent Gradient of Cyp26B1+ Olfactory Sensory Neurons Is Necessary for the Functional Integrity of the Olfactory Sensory Map.

UNLABELLED: Stimulus-dependent expression of the retinoic acid-inactivating enzyme Cyp26B1 in olfactory sensory neurons (OSNs) forms a dorsomedial (DM)-ventrolateral (VL) gradient in the mouse olfactory epithelium. The gradient correlates spatially with different rates of OSN turnover, as well as the functional organization of the olfactory sensory map, into overlapping zones of OSNs that express different odorant receptors (ORs). Here, we analyze transgenic mice that, instead of a stimulus-dependent Cyp26B1 gradient, have constitutive Cyp26B1 levels in all OSNs. Starting postnatally, OSN diff

An Alzheimer's Disease Patient-Derived Olfactory Stem Cell Model Identifies Gene Expression Changes Associated with Cognition.

An early symptom of Alzheimer's disease (AD) is an impaired sense of smell, for which the molecular basis remains elusive. Here, we generated human olfactory neurosphere-derived (ONS) cells from people with AD and mild cognitive impairment (MCI), and performed global RNA sequencing to determine gene expression changes. ONS cells expressed markers of neuroglial differentiation, providing a unique cellular model to explore changes of early AD-associated pathways. Our transcriptomics data from ONS cells revealed differentially expressed genes (DEGs) associated with cognitive processes in AD cells

Microencapsulated Schwann cell transplantation inhibits P2X2/3 receptors overexpression in a sciatic nerve injury rat model with neuropathic pain.

Transplantation of Schwann cells (SCs) can promote axonal regeneration and formation of the myelin sheath, reduce inflammation, and promote repair to the damaged nerve. Our previous studies have shown that transplantation of free or micro-encapsulated olfactory ensheathing cells can relieve neuropathic pain. There are no related reports regarding whether the transplantation of micro-encapsulated SCs can alleviate neuropathic pain mediated by P2X2/3 receptors. In the present study, we micro-encapsulated SCs in alginic acid and transplanted them into the region surrounding the injured sciatic ne

Environmental neurotoxicants as accelerators of brain ageing: exposomic mechanisms linking pollution to amyloidogenesis and Alzheimer's disease.

Alzheimer's disease (AD) is a multifactorial neurodegenerative disorder characterised by progressive cognitive decline, amyloid-β (Aβ) deposition, tau hyperphosphorylation, synaptic dysfunction, and chronic neuroinflammation. While rare familial cases are genetically driven, most AD is sporadic, underscoring the critical role of environmental determinants. Emerging evidence from epidemiological, experimental, and exposomic studies identifies environmental neurotoxicants, including air pollutants, heavy metals, pesticides, and emerging contaminants such as micro- and nanoplastics, as significan

Comprehensive review on adult neurogenesis through immunological studies in the brain regions of vertebrate classes.

Adult neurogenesis is the process in which new neurons are produced, mature, and integrated into the neural circuits of adult vertebrate brains- fish, amphibians, reptiles, aves, and mammals. All vertebrate groups exhibit a great capacity for neurogenesis, which is dependent upon the number of neurogenic zones in the brain. The fish brain is known to have the highest neurogenic rate because they acquire the maximum number of neurogenic zones, such as the olfactory bulb, telencephalon, preoptic area, cerebellum, thalamus, hypothalamus, optic tectum, and medulla oblongata. The amphibian brain is

Cutaneous α-Synuclein Pathology as a Differential Marker: A Histological and Statistical Comparison across Neurodegenerative Disease Groups.

There is an urgent need for early and accurate biomarkers of neurodegenerative disorders. Due to the high innervation and accessibility of the skin, a skin biopsy is a minimally invasive method of detecting phosphorylated α-synuclein (p-α-syn) and assessing intraepidermal nerve fiber density (IENFD). We analyzed biopsies taken from the back and the leg of patients with parkinsonian syndromes (Park.sy.), α-synucleinopathies, multiple system atrophies (MSA), tauopathies, and other neurological disorders, as well as from healthy controls. Double immunofluorescence was performed for p-α-syn (Ser12

Suppressors of Break-Induced Replication in Human Cells.

Short tandem DNA repeats are drivers of genome instability. To identify suppressors of break-induced mutagenesis human cells, unbiased genetic screens were conducted using a lentiviral shRNA library. The recipient cells possessed fragile non-B DNA that could induce DNA double-strand breaks (DSBs), integrated at an ectopic chromosomal site adjacent to a thymidine kinase marker gene. Mutagenesis of the thymidine kinase gene rendered cells resistant to the nucleoside analog ganciclovir (GCV). The screen identified genes that have established roles in DNA replication and repair, chromatin modifica

Association of OBPIIa genetic variants with prodromal Parkinsonian phenotypes and dopaminergic biomarkers in the PPMI cohort.

BACKGROUND: Odorant-binding protein IIa (OBPIIa) gene variants are linked to Parkinson's disease (PD)-related olfactory loss, but their role in clinical subtypes and mechanisms remains unclear. This study examined associations with clinical scales, neuroimaging, and cerebrospinal fluid (CSF) biomarkers. METHODS: Data from the Parkinson's Progression Markers Initiative (PPMI) cohort were analyzed. Multinomial logistic regression was employed to investigate the role of OBPIIa SNPs in PD prodromal or clinically established PD status. Additionally, the associations of OBPIIa SNPs with clinical sc

Curcumin-activated olfactory ensheathing cells attenuate astroglial scar to facilitate nerve repair via the IL-10/NF-κB axis after spinal cord injury.

Excessive astrocyte activation following spinal cord injury (SCI) can trigger the substantial deposition of inhibitory extracellular matrix components, particularly chondroitin sulfate proteoglycans (CSPGs) at the lesion core. This astroglial response typically leads to the formation of dense glial scars, which profoundly impede neuronal regeneration and axonal extension. In this study, we show that transplantation of curcumin-activated olfactory ensheathing cells (aOECs) markedly promotes nervous tissue reorganization in SCI rats, as evidenced by reduced lesion cavitation and aligned GFAP-pos

Global gene expression profile of proliferative verrucous leukoplakia and its underlying biological disease mechanisms.

BACKGROUND: Proliferative verrucous leukoplakia (PVL) is a rare and enigmatic oral potentially malignant disorder which almost invariably results in oral squamous cell carcinoma (OSCC). The aims of this project were to use transcriptome profiling to characterise PVL gene expression patterns for biomarker identification and gain insight into the molecular aetiopathogenesis of PVL. METHODS: Forty-three oral cavity mucosal biopsies from 32 patients with oral lesions clinically compatible with either PVL or non-PVL conventional oral leukoplakia (OLK) underwent transcriptome profiling by RNA seque

Evaluation of Fluoro-Jade C Staining: Specificity and Application to Damaged Immature Neuronal Cells in the Normal and Injured Mouse Brain.

Fluoro-Jade C (FJC) staining is widely used for the specific detection of all degenerating mature neurons, including apoptotic, necrotic, and autophagic cells. However, whether FJC staining can detect degenerating immature neurons and neural stem/precursor cells remains unclear. In addition, some conflicting studies have shown that FJC and its ancestral dyes, Fluoro-Jade (FJ) and FJB, can label resting/activated astrocytes and microglia. In the present study, we examined the validity of FJC staining for the detection of neuronal cells in adult and embryonic mouse brains under normal and injure
