Patient-Friendly Real-Time Optical Tomographic Imaging System (LOTIS) for Lupus Arthritis.

Systemic lupus erythematosus (SLE) frequently presents joint pain and stiffness, yet clinicians lack an objective, rapid method to quantify joint inflammation at the point of care. We introduce the Lupus Optical Tomography Imaging System (LOTIS), a wearable near-infrared (NIR) device that performs real-time three-dimensional tomographic imaging of hemodynamic changes in finger joints. LOTIS was developed to address key limitations of our earlier Flexible Optical Imaging System (FOIS), including mechanical fragility, high noise levels, single-joint acquisition, and slow reconstruction times. Wide-field self-referenced optical coherence tomography imaging of the corneal microlayers.

We present a technique for wide-field OCT imaging of the cornea using self-referenced interference of the anterior corneal surface with the underlying corneal structure. The method employs a custom objective lens that ensures that the probing beam remains nearly perpendicular to the anterior corneal surface, effectively utilizing it as a reference to generate interference. This approach allows for high-resolution, three-dimensional imaging of corneal layers over a 9mm optical zone. Tests on human subjects show the ability of the system to quantify the anterior corneal layer thickness.

Optical imaging of microfluidic integrated smart hydrogels for research and sensing applications.

Smart hydrogels are a versatile class of materials that are attractive for biomedical sensor applications due to their potential biocompatibility and stimuli-responsiveness. However, the integration of these hydrogels into sensors requires development and engineering efforts to optimize the volume phase transition and time response for the respective sensing applications. This work presents an optical evaluation platform for investigating hydrogel swelling properties in a liquid environment with automated flow control. It employs hydrogel features integrated in microfluidic test strips which a

Stent Graft Restenosis: Insights From Optical Coherence Tomography Imaging and Treatment Considerations.

Endovascular therapy (EVT) utilizing self-expanding stent grafts has shown promising clinical outcomes for femoropopliteal lesions. However, restenosis and thrombotic occlusion remain significant concerns with unclear underlying mechanisms. This is the case of a 53-year-old male with diabetic nephropathy requiring hemodialysis. He presented with left lower limb intermittent claudication and underwent EVT, resulting in subsequent restenosis 12 months post-implantation and recurrence of symptoms. Intravascular imaging revealed neointimal proliferation and stent-edge restenosis, with segments of

Combined Dielectric-Optical Characterization of Single Cells Using Dielectrophoresis-Imaging Flow Cytometry.

In this paper, we present a microfluidic flow cytometer for simultaneous imaging and dielectric characterization of individual biological cells within a flow. Utilizing a combination of dielectrophoresis (DEP) and high-speed imaging, this system offers a dual-modality approach to analyze both cell morphology and dielectric properties, enhancing the ability to analyze, characterize, and discriminate cells in a heterogeneous population. A high-speed camera is used to capture images of and track multiple cells in real-time as they flow through a microfluidic channel. A wide channel is used, enabl

Cross-Correlation Increases Sampling in Diffusion-Based Super-Resolution Optical Fluctuation Imaging.

Correlation signal processing of optical three-dimensional (x, y, t) data can produce super-resolution images. The second-order cross-correlation function XC 2 has been documented to produce super-resolution imaging with static and blinking emitters but not for diffusing emitters. Here, we both analytically and numerically demonstrate cross-correlation analysis for diffusing particles. We then expand our fluorescence correlation spectroscopy super-resolution optical fluctuation imaging (fcsSOFI) analysis to use cross-correlation as a postprocessing computational technique to extract both dynam

Imaging peritoneal blood vessels through optical coherence tomography angiography for laparoscopic surgery.

Laparoscopic surgery presents challenges in identifying blood vessels due to lack of tactile feedback. The image-guided laparoscopic surgical tool (IGLaST) integrated with optical coherence tomography (OCT) has potential for in vivo blood vessel imaging; however, distinguishing vessels from surrounding tissue remains a challenge. In this study, we propose utilizing an inter-A-line intensity differentiation-based OCT angiography (OCTA) to improve visualization of blood vessels. By evaluating a tissue phantom with varying flow speeds, we optimized the system's blood flow imaging capabilities in

Imaging Cerebral Blood Vessels Using Near-Infrared Optical Tomography: A Simulation Study.

Cerebral veins have received increasing attention due to their importance in preoperational planning and the brain oxygenation measurement. There are different modalities to image those vessels, such as magnetic resonance angiography (MRA) and recently, contrast-enhanced (CE) 3D gradient-echo sequences. However, the current techniques have certain disadvantages, i.e., the long examination time, the requirement of contrast agents or inability to measure oxygenation. Near-infrared optical tomography (NIROT) is emerging as a viable new biomedical imaging modality that employs near infrared light

Novel exploration of Raman microscopy and non-linear optical imaging in adenomyosis.

BACKGROUND: Adenomyosis is a common gynecological disease in women. A relevant literature search found that approximately 82% of patients with adenomyosis chose to undergo hysterectomy. However, women of childbearing age are more likely to undergo surgery to preserve the uterus. Because it is difficult to determine the extent of adenomyosis, it is almost impossible to resect adenomyotic tissue and retain the uterus at the same time. MATERIALS AND METHODS: Following ethics approval and patient consent, tissue samples were resected and prepared to create frozen slices for analysis. One slice wa

Hypotony maculopathy captured with vertical rasters on optical coherence tomography (OCT) imaging.

PURPOSE: To remind eye care providers of the importance of obtaining vertical rasters in OCT evaluation of patients with suspected hypotony maculopathy. OBSERVATIONS: OCT with vertical rasters may identify chorioretinal folds that are missed by ophthalmoscopic examination, fundus photography, and traditional OCT with horizontal rasters alone. CONCLUSIONS AND IMPORTANCE: In patients with low IOP and decreased vision, OCT imaging with horizonal and vertical rasters should be obtained to diagnose hypotony maculopathy and monitor response to treatment.

Standardization of Optical Coherence Tomography Angiography Imaging Biomarkers in Diabetic Retinal Disease.

Optical coherence tomography Angiography (OCT-A) represents a revolution in the noninvasive evaluation of retinal and choroidal circulation especially in detecting early clinical signs of diabetic retinal disease (DRD). With appropriate use, OCT-A characteristics and measurements have the potential to become new imaging biomarkers in managing and treating DRD. Major challenges include (a) provision of standardized outputs from different OCT-A instruments providing standardized terminology to correctly interpret data; (b) the presence of artifacts; (c) the absence of standardized grading or int

Rational design of genetically encoded reporter genes for optical imaging of apoptosis.

Apoptosis is a process in which cells are genetically regulated to cause a series of changes in morphology and metabolic activity, which ultimately lead to cell death. Apoptosis plays a vital role in the entire life cycle of an organism. Too much or too little apoptosis can cause a variety of diseases. Therefore, efficient and convenient methods for detecting apoptosis are necessary for clinical treatment and drug development. Traditional methods for detecting apoptosis may cause damage to the body during sample collection, such as for flow cytometry analysis. So it is necessary to monitor apo

Noncontact optical imaging of brain hemodynamics in preterm infants: a preliminary study.

Extremely preterm infants' hemodynamic instability places them at high risk of brain injury. Currently there is no reliable bedside method to continuously monitor cerebral hemodynamics in the neonatal intensive care unit (NICU). This paper reports a feasibility study to adapt and test an innovative speckle contrast diffuse correlation tomography (scDCT) device for noncontact, high-density, 3D imaging of cerebral blood flow (CBF) in preterm infants. The scDCT scans a focused point near-infrared illumination to multiple source positions for deep tissue penetration, and controls an electron multi

Optical Redox Imaging of Fixed Unstained Muscle Slides Reveals Useful Biological Information.

PURPOSE: Optical redox imaging (ORI) technique images cellular autofluorescence of nicotinamide adenine dinucleotide (NADH) and oxidized flavoproteins (Fp containing FAD, i.e., flavin adenine dinucleotide). ORI has found wide applications in the study of cellular energetics and metabolism and may potentially assist in disease diagnosis and prognosis. Fixed tissues have been reported to exhibit autofluorescence with similar spectral characteristics to those of NADH and Fp. However, few studies report on quantitative ORI of formalin-fixed paraffin-embedded (FFPE) unstained tissue slides for dise

In vivo imaging in the oral cavity by endoscopic optical coherence tomography.

The common way to diagnose hard and soft tissue irregularities in the oral cavity is initially the visual inspection by an experienced dentist followed by further medical examinations, such as radiological imaging and/or histopathological investigation. For the diagnosis of oral hard and soft tissues, the detection of early transformations is mostly hampered by poor visual access, low specificity of the diagnosis techniques, and/or limited feasibility of frequent screenings. Therefore, optical noninvasive diagnosis of oral tissue is promising to improve the accuracy of oral screening. Consider

Tomographic optical imaging of cortical responses after crossing nerve transfer in mice.

To understand the neural mechanisms underlying the therapeutic effects of crossing nerve transfer for brachial plexus injuries in human patients, we investigated the cortical responses after crossing nerve transfer in mice using conventional and tomographic optical imaging. The distal cut ends of the left median and ulnar nerves were connected to the central cut ends of the right median and ulnar nerves with a sciatic nerve graft at 8 weeks of age. Eight weeks after the operation, the responses in the primary somatosensory cortex (S1) elicited by vibratory stimulation applied to the left forep
