Toll-like receptors (TLRs) are critical components of the innate immune system, acting as pattern recognition receptors and interacting with oxidative stress. TLR2, TLR4, and TLR7 play distinct yet overlapping roles in immune regulation, contributing to both physiological processes and disease progression. Therefore, modulation of TLR signaling pathways represents a promising therapeutic strategy for a wide range of diseases. Natural antioxidants have attracted considerable interest as modulators of TLRs, with cinnamon-derived phytochemicals, particularly cinnamaldehyde, emerging as promising

BACKGROUND: In recent years, neuroglia has become a therapeutic target for neurodegenerative diseases. Despite the recognition of a variety of microglial morphologies associated with the neuroinflammatory process that involve diverse functionalities for this glial type, it is still unknown its beneficial or harmful role to the surrounding tissue.
METHODS: The study presented here proposes a novel approach to the neurodegenerative progression based on the reliability of its results due to the use of a natural model. Morphological alterations in microglia were assessed in cerebellar samples fro

Acute-on-chronic liver failure combines an acute deterioration in liver function in an individual with pre-existing chronic liver disease and hepatic and extrahepatic organ failures, and is associated with substantial short-term mortality. Common precipitants include bacterial and viral infections, alcoholic hepatitis, and surgery, but in more than 40% of patients, no precipitating event is identified. Systemic inflammation and susceptibility to infection are characteristic pathophysiological features. A new diagnostic score, the Chronic Liver Failure Consortium (CLIF-C) organ failure score, h

BACKGROUND: Sp4 is a zinc finger transcription factor which is closely related to Sp1 and Sp3. All three proteins recognize the same DNA elements and can act as transcriptional activators through glutamine-rich activation domains. Unlike Sp1 and Sp3, which are ubiquitous proteins, Sp4 is highly abundant in the central nervous system, but also detectable in many other tissues.
RESULTS: We have disrupted the mouse Sp4 gene by a targeted deletion of the exons encoding the N-terminal activation domains. Sp4 knockout mice show a complete absence of Sp4 expression. They develop until birth without

PURPOSE OF REVIEW: The principles of neonatal neurological protection following intrapartum hypoxia are briefly reviewed. The physiological principles behind the use of cardiotocograph patterns in defining the timing and mechanism of fetal hypoxia and injury are then demonstrated.
RECENT FINDINGS: Fetal neurological injury may result from progressive hypoxemia, acidosis, diminished cardiac output and cerebral ischemia, manifested at birth as low Apgar scores, multisystem compromise, severe acidosis and encephalopathy. More commonly, however, intrapartum injury results from often intermittent,

BACKGROUND: Beyond the well-known deleterious effects of ethanol defining Fetal Alcohol Spectrum Disorders (FASD), the notion of fetal alcohol programming has gained scientific support. This phenomenon implies early neural plasticity relative to learning mechanisms comprising ethanol´s sensory cues and physiological effects of the drug; among others, its reinforcing properties and its depressant effects upon respiration. In this study, as a function of differential ethanol exposure during gestation, we analyzed neonatal physiological and behavioral responsiveness recruited by the odor of the d

Concealing pathogen-associated molecular patterns (PAMPs) is a principal strategy used by fungi to avoid immune recognition. Surface exposure of PAMPs during germination can leave the pathogen vulnerable. Accordingly, β-glucan surface exposure during Aspergillus fumigatus germination activates an Atg5-dependent autophagy pathway termed LC3-associated phagocytosis (LAP), which promotes fungal killing. We found that LAP activation also requires the genetic, biochemical or biological (germination) removal of A. fumigatus cell wall melanin. The attenuated virulence of melanin-deficient A. fumigatu

Autophagy is a physiologically and immunologically controlled intracellular homeostatic pathway that sequesters and degrades cytoplasmic targets including macromolecular aggregates, cellular organelles such as mitochondria, and whole microbes or their products. Recent advances show that autophagy plays a role in innate immunity in several ways: (i) direct elimination of intracellular microbes by digestion in autolysosomes, (ii) delivery of cytosolic microbial products to pattern recognition receptors (PRRs) in a process referred to as topological inversion, and (iii) as an anti-microbial effec

Migraine affects the cortical physiology and may induce dysfunction both ictally and interictally. Although visual symptoms predominate during aura, other contiguous cortical areas related to less impressive symptoms are also impaired in migraine. Answers from 72.2% migraine with aura and 48.6% of migraine without aura patients on human faces and objects recognition, colour perception, proper names recalling and memory in general showed dysfunctions suggestive of prosopagnosia, dyschromatopsia, ideational apraxia, alien hand syndrome, proper name anomia or aphasia, varying in duration and seve

The current study examined the EEG of young, old and old declined adults performing a visual paired associate task. In order to examine the effects of encoding context and stimulus repetition, target pairs were presented on either detailed or white backgrounds and were repeatedly presented during both early and late phases of encoding. Results indicated an increase in P300 amplitude in the right parietal cortex from early to late stages of encoding in older declined adults, whereas both younger adults and older controls showed a reduction in P300 amplitude in this same area from early to late

Part I reviewed wound healing biology and a pragmatic reconstructive ladder for interventional radiology (IR) access and postprocedural wounds. Part II focuses on flap and tissue transfer options: why and when they are needed, how they work physiologically, and how to think about anatomy and risk in a way that is clinically useful to non-surgeons. The emphasis is on common wound scenarios relevant to IR (groin and thigh access, upper limb access, device- or graft-adjacent breakdown, radiated tissue, infected hematomas, and exposed critical structures). We describe local, regional pedicled, and

CONTEXT: Femoroacetabular impingement (FAI) is a common cause of intra-articular hip pathology and secondary osteoarthritis. It affects athletes at a young age as they push their bodies beyond the diminished physiologic limits imposed by the altered joint morphology. Understanding the cause of this condition and its assessment in athletes is important.
EVIDENCE ACQUISITION: The scientific literature was reviewed to reflect the current understanding of hip joint pathology among athletic individuals. Focus is given to the literature since 2003, when FAI was first reported as a cause of joint da

BACKGROUND: The giant panda (Ailuropoda melanoleuca) is a well-known, rare and endangered species. Baylisascaris schroederi is a pathogenic ascarid. Infection with B. schroederi may cause death in giant pandas. At present, the immune evasion mechanism of B. schroederi is little known. Cysteine protease inhibitors (CPI) play important roles in the regulation of host immune responses against certain nematodes. In this study, we focused on the analysis of the regulation of B. schroederi migratory larvae CPI (rBsCPI-1) on mice immune cells.

Pentraxin 3 (PTX3) is an inflammatory mediator acting as a fluid-phase pattern recognition molecule and playing an essential role in innate immunity and matrix remodeling. Inflammatory mediators also contribute to skeletal homeostasis, operating at multiple levels in physiological and pathological conditions. This study was designed to investigate the role of PTX3 in physiological skeletal remodeling and bone healing. Micro-computed tomography (μCT) and bone histomorphometry of distal femur showed that PTX3 gene-targeted female and male mice (ptx3-/- ) had lower trabecular bone volume than the

Flexible hydrogel sensors show great utilization potential in the development of wearable devices. Nevertheless, issues such as unsatisfactory sensitivity, intricate fabrication processes, and substantial cost continue to impede their widespread application. Furthermore, a significant challenge resides in developing a convenient method to mold the flexible hydrogel into a variety of shapes, thereby fulfilling the different scenario requirements. Herein, we report a cost-effective, multifunctional polyvinyl alcohol (PVA)/multilayer MXene composite PMI hydrogel, which is crosslinked by the addit

An extraordinary variety of experimental (e.g., flicker, magnetic fields) and clinical (epilepsy, migraine) conditions give rise to a surprisingly common set of elementary hallucinations, including spots, geometric patterns, and jagged lines, some of which also have color, depth, motion, and texture. Many of these simple hallucinations fall into a small number of perceptual geometries-the Klüver forms-that (via a nonlinear mapping from retina to cortex) correspond to even simpler sets of oriented stripes of cortical activity (and their superpositions). Other simple hallucinations (phosphenes a

Several studies have reported that olfactory dysfunction is an early neuropathological manifestation of Parkinson's disease (PD). Reduced cardiac meta-iodobenzylguanidine ((123)I-MIBG) uptake may be one of the earliest signs of PD. We studied the relation of olfactory dysfunction to cardiovascular dysautonomia in patients with PD. The study group comprised 66 patients with PD (70.5 years) and 26 controls (70.3 years) for olfactory assessment, 21 controls (72.1 years) for cardiac (123)I-MIBG scintigraphy and heart rate variability (HRV), assessed using the coefficient of variation for RR interv

Primary sclerosing cholangitis (PSC) is a chronic cholestatic disease marked by fibrosis, strictures, and a high risk for developing cholangiocarcinoma (CCC). Recognition of CCC in PSC remains a diagnostic challenge. PET/CT using fibroblast activation protein inhibitors (FAPI) has emerged as a promising imaging modality in CCC, but its potential in the specific profibrotic setting of PSC has not yet been explored to the best of our knowledge. Methods: PET/CT with 68Ga-FAPI-46 was performed in 18 patients (n = 9 with PSC, without proven CCC; n = 7 without PSC, with histologically confirmed CCC;

The mesenteric whirl sign on computed tomography (CT) should be approached as a surgical decision signal for potentially bowel-threatening mechanical disease rather than as an isolated radiologic label. Not every whirl pattern represents fixed active volvulus; however, underestimating a true whirl sign can delay recognition of closed-loop obstruction, internal hernia, strangulation, bowel necrosis, perforation, sepsis, and death. This review reframes the sign around the management question most relevant to general surgeons: when should imaging evidence of mesenteric rotation alter care before
