Asthma and oxidant stress: nutritional, environmental, and genetic risk factors.

A considerable body of evidence suggests that oxidant stress results in inflammation and tissue damage in the respiratory system, and later in immune damage, and that individuals with lowered cellular reducing capacity are at increased risk to develop asthma. Reducing capacity in the erythrocyte is generated through the pentose phosphate pathway and this pathway also generates a major portion of the reducing capacity in all cells of the body. Therefore, dietary, environmental, and genetic factors which diminish cellular reducing capacity will increase tissue vulnerability to oxidant stress and

Multi-omics profiling reveals coordinated metabolic and immuno-inflammatory remodeling in a high-fat diet and single-dose streptozotocin-induced mouse model of diabetic cardiomyopathy.

BACKGROUND: Diabetic cardiomyopathy (DCM) is a major myocardial complication of type 2 diabetes and a contributor to heart failure. However, coordinated remodeling across the cardiac transcriptome, proteome and metabolome in DCM remains incompletely defined. METHODS: Type 2 diabetes was induced in mice by a high-fat diet combined with single-dose streptozotocin. DCM was characterized by echocardiography, histology, transmission electron microscopy, and systemic metabolic and inflammatory assessments. Left ventricular myocardium underwent RNA sequencing, quantitative proteomics and untargeted

Hybrid Methylotrophic Pathway in Serratia marcescens for Sustainable Terpenoid Biosynthesis.

Terpenoids are valuable chemicals used across industries. Methanol, a nonsugar-based feedstock, offers an eco-friendly approach to terpenoid production. In this study, a hybrid methylotrophic pathway was engineered in Serratia marcescens (S. marcescens) HBQA7, which is tolerant to both methanol and terpenoid compounds. The pathway utilizes bacterial methanol dehydrogenase (Mdh) and yeast dihydroxyacetone synthase (Das) to produce monoterpenes and sesquiterpenes from methanol and xylose. 13C labeling experiments identified the optimal enzyme pair: Mdh from Acinetobacter gerneri and Das from Pic

Mapping the Binding Landscape of Allosteric Inhibitor G6PDi-1 on Human G6PD.

Targeting the oxidative pentose phosphate pathway by inhibiting glucose-6-phosphate dehydrogenase (G6PD) is a promising anticancer strategy, yet clinically useful inhibitors remain unavailable. A key limitation is the lack of molecular insight into how allosteric and noncompetitive inhibitors perturb enzymatic activity, limiting rational optimization. Here, we investigated the mechanism of G6PDi-1, a potent, reversible, noncompetitive G6PD inhibitor, by combining biochemical assays with molecular dynamics simulations, Markov state model, and MM/PBSA binding energy calculations. Experimentally,

The pivotal role of endoplasmic reticulum in cancer glucose metabolism.

The endoplasmic reticulum (ER) supports essential biosynthetic and quality control functions. These processes rely on sustained energy supply and precise redox control within the ER lumen. While ATP can be imported from mitochondria, pyridine nucleotides are impermeable to the ER membrane, necessitating compartment-specific mechanisms to regulate NAD(H) and NADP(H) pools. Here, we demonstrate that the ER-confined pentose phosphate pathway (ER-PPP), driven by hexose-6-phosphate dehydrogenase (H6PD), processes large amounts of glucose equivalents to preserve the local redox homeostasis in triple

G6PD deficiency triggers dopamine loss and the initiation of Parkinson's disease pathogenesis.

Loss of dopaminergic neurons in Parkinson's disease (PD) is preceded by loss of synaptic dopamine (DA) and accumulation of proteinaceous aggregates. Linking these deficits is critical to restoring DA signaling in PD. Using murine and human pluripotent stem cell (hPSC) models of PD coupled with human postmortem tissue, we show that accumulation of α-syn micro-aggregates impairs metabolic flux through the pentose phosphate pathway (PPP). This leads to decreased nicotinamide adenine dinucleotide phosphate (NADP/H) and glutathione (GSH) levels, resulting in DA oxidation and decreased total DA leve

G6PD deficiency in brain induces schizophrenia-like behaviors and synaptic dysfunction.

Schizophrenia is a severe mental disorder characterized by aberrant brain development, influenced by genetic and environmental factors, with an incompletely understood etiology. Glucose-6-phosphate dehydrogenase (G6PD), a critical enzyme in the pentose phosphate pathway (PPP), facilitates NADPH production for biosynthesis and redox homeostasis. Recent studies suggests that PPP inhibition and consequent oxidative stress contribute to schizophrenia pathogenesis. While clinical investigations have proposed a link between G6PD deficiency and schizophrenia, the underlying biological mechanisms rema

Metabolic Anti-Cancer Effects of Melatonin: Clinically Relevant Prospects.

Metabolic reprogramming characterized by alterations in nutrient uptake and critical molecular pathways associated with cancer cell metabolism represents a fundamental process of malignant transformation. Melatonin (N-acetyl-5-methoxytryptamine) is a hormone secreted by the pineal gland. Melatonin primarily regulates circadian rhythms but also exerts anti-inflammatory, anti-depressant, antioxidant and anti-tumor activities. Concerning cancer metabolism, melatonin displays significant anticancer effects via the regulation of key components of aerobic glycolysis, gluconeogenesis, the pentose pho

Bridging sugar metabolism and signaling with hormones in shoot branching regulation.

Shoot branching, a major determinant of plant architecture and productivity, has long been attributed to the coordinated action of auxin, cytokinins (CKs), and strigolactones (SLs). Recent work shows that sugars act not only as metabolic resources but also as early developmental signals that regulate axillary bud outgrowth, the initial step in shoot branching. Sugar availability can rapidly trigger bud outgrowth, often preceding major hormonal changes. Mechanistically, sugars antagonize auxin and SL signaling while acting synergistically with CKs. These effects involve sugar signaling pathways

Nitrogen-modified atmosphere with re-aeration storage triggers deterioration in paddy rice by disrupting central carbon metabolism.

Nitrogen-modified atmosphere (NMA) storage is extensively adopted for the postharvest preservation of paddy rice for pest control, yet its quality tends to deteriorate at an accelerated rate during subsequent re-aeration storage (RS). This study explored the central carbon metabolic changes in indica and japonica paddy rice under conventional storage (CS), 99%-N₂ re-aeration storage (HCN-RS), and 95%-N₂ re-aeration storage (LCN-RS). Results showed that post re-aeration, HCN-RS and LCN-RS significantly reduced rice freshness, germination rate, energy charge, and activities of key metabolic enzy

Xylulose 5-phosphate fosters sustained antitumor activity of progenitor-like exhausted SLC35E2+ CD8+ T effector cells.

Metabolic adaptations involved in tumor metastasis and immune evasion merit investigation. Here, using in vivo metabolic CRISPR/Cas9 knockout screening, we identified xylulokinase (XYLB) as a tumor suppressor that impairs lung colonialization by producing xylulose 5-phosphate (Xu5P), which promotes CD8+ T cell cytotoxicity. Mechanistically, CD8+ T cells express relatively high levels of solute carrier family 35 member E2 (SLC35E2), a homolog of the plant Xu5P transporter, to facilitate Xu5P uptake and subsequently intensify the pentose phosphate pathway and glycolysis for energy/redox balance.

Hemolytic anemia in hereditary pyrimidine 5'-nucleotidase deficiency. II. Effect of pyrimidine nucleotides and their derivatives on glycolytic and pentose phosphate shunt enzyme activity.

We evaluated the glycolytic intermediate concentrations from the erythrocytes of a patient with hereditary pyrimidine 5'-nucleotidase (P5'N) deficiency. Conclusive evidence for a metabolic block was not found. We evaluated the effects of the pyrimidine (cytidine and uridine) tri- and diphosphate nucleotides (CTP, CDP, UTP, UDP) and the choline and ethanolamine derivatives of CDP (CDP-choline, CDP-ethanolamine) on the activities of key enzymes of the Embden-Meyerhof pathway. CTP and UTP inhibited fructose-6-phosphate competitively for phosphofructokinase and phosphoenolpyruvate competitively fo

Metabolic pathway competition sensitizes thioredoxin reductase-depleted Candida albicans to cell wall stress and antifungals.

Candida albicans is the most common invasive human fungal pathogen. Each of the currently available 3 antifungal drug classes is limited by toxicities, drug interactions or inadequate penetration of critical tissues. We discovered a connection between C. albicans TOR and thioredoxin reductase, Trr1. Fungal and human thioredoxin reductases diverge fundamentally and are essential for repair of oxidatively damaged macromolecules, so they may be promising drug targets. We found that Trr1 is required for C. albicans ' growth at human body temperatures, hyphal growth regulation and management of oxi

A rare metabolic cause of premature ovarian insufficiency: case report of transaldolase deficiency.

OBJECTIVES: Transaldolase deficiency (TALDO-D) is a rare autosomal recessive disorder of the pentose phosphate pathway, characterized by growth restriction, dysmorphism, cytopenia, with variable liver involvement. CASE PRESENTATION: Herein, we report a rare case of TALDO-D who presented with primary amenorrhea, elevated gonadotrophins, and low/undetectable estradiol levels, consistent with premature ovarian insufficiency. Molecular genetic analysis identified a homozygous frameshift variant in TALDO1 (c.345dup), caused by a duplication. CONCLUSIONS: This case expands the clinical spectrum of

mTORC1-Dependent Metabolic Reprogramming Underlies Escape from Glycolysis Addiction in Cancer Cells.

Although glycolysis is substantially elevated in many tumors, therapeutic targeting of glycolysis in cancer patients has not yet been successful, potentially reflecting the metabolic plasticity of tumor cells. In various cancer cells exposed to a continuous glycolytic block, we identified a recurrent reprogramming mechanism involving sustained mTORC1 signaling that underlies escape from glycolytic addiction. Active mTORC1 directs increased glucose flux via the pentose phosphate pathway back into glycolysis, thereby circumventing a glycolysis block and ensuring adequate ATP and biomass producti

6-phosphogluconate dehydrogenase: a target for drugs in African trypanosomes.

New drugs are urgently required for Human African Trypanosomiasis (sleeping sickness), a disease which has re-emerged as a major health threat in Sub-Saharan Africa. The third enzyme of the pentose phosphate pathway, 6-phosphogluconate dehydrogenase, has been shown to be a good target for drugs. The enzyme is essential to the trypanosomes that causes sleeping sickness and structural differences when compared to its mammalian counterpart allow for selective inhibition. Three series of inhibitors have been designed, these include phosphorylated carbohydrate substrate and transition state analogu

Characterization of microbial intolerances and ruminal dysbiosis towards different dietary carbohydrate sources using an in vitro model.

AIM: This study aimed to characterize the critical points for determining the development of dysbiosis associated with feed intolerances and ruminal acidosis. METHODS AND RESULTS: A metabologenomics approach was used to characterize dynamic microbial and metabolomics shifts using the rumen simulation technique (RUSITEC) by feeding native cornstarch (ST), chemically modified cornstarch (CMS), or sucrose (SU). SU and CMS elicited the most drastic changes as rapidly as 4 h after feeding. This was accompanied by a swift accumulation of d-lactate, and the decline of benzoic and malonic acid. A con

Dexamethasone improves redox state in ataxia telangiectasia cells by promoting an NRF2-mediated antioxidant response.

Ataxia telangiectasia (A-T) is a rare incurable neurodegenerative disease caused by biallelic mutations in the gene for ataxia-telangiectasia mutated (ATM). The lack of a functional ATM kinase leads to a pleiotropic phenotype, and oxidative stress is considered to have a crucial role in the complex physiopathology. Recently, steroids have been shown to reduce the neurological symptoms of the disease, although the molecular mechanism of this effect is largely unknown. In the present study, we have demonstrated that dexamethasone treatment of A-T lymphoblastoid cells increases the content of two

Cutaneous Thermal Injury Modulates Blood and Skin Metabolomes Differently in a Murine Model.

As the field of metabolomics develops further, investigations of how the metabolome is affected following thermal injury may be helpful to inform diagnostics and guide treatments. In this study, changes to the metabolome were tested and validated in a murine burn injury model. After a 30% total body surface scald injury or sham procedure sera and skin biopsies were collected at 1, 2, 6, or 24 hr. Burn-specific changes in the metabolome were detected compared to sham animals. The sera metabolome exhibited a more rapid response to burn injury than that of the skin and it peaked more proximal to

Conditional impairment of Coxiella burnetii by glucose-6P dehydrogenase activity.

Coxiella burnetii is a bacterial obligate intracellular parasite and the etiological agent of query (Q) fever. While the C. burnetii genome has been reduced to ∼2 Mb as a likely consequence of genome streamlining in response to parasitism, enzymes for a nearly complete central metabolic machinery are encoded by the genome. However, lack of a canonical hexokinase for phosphorylation of glucose and an apparent absence of the oxidative branch of the pentose phosphate pathway, a major mechanism for regeneration of the reducing equivalent nicotinamide adenine dinucleotide phosphate (NADPH), have be
