Supplemental oxygen in premature neonates risks bronchopulmonary dysplasia (BPD). Although the metabolite itaconic acid (ITA) promotes transcription factor EB (TFEB) nuclear translocation to alleviate BPD, the upstream mechanism driving hyperoxia-induced TFEB sequestration remains unclear. Here, we investigated whether hyperoxia exploits liquid-liquid phase separation (LLPS) to trap cytoplasmic TFEB, and if ITA directly dissolves these pathological condensates. Using hyperoxia-induced BPD mice (85 % O2) and MLE12 cells, we analyzed TFEB LLPS via FRAP, fusion, and in vitro phase separation assa

Biomolecular condensates formed via liquid-liquid phase separation (LLPS) are increasingly recognised as dynamic organisers of intracellular biochemistry, particularly in neurons where spatially restricted signalling, RNA metabolism, and proteostasis are essential. Aberrant phase transitions of disease-associated proteins, including TDP-43, FUS, tau, and α-synuclein, contribute to protein aggregation and neurodegenerative pathology. Beyond protein-intrinsic sequence features, metabolic state has emerged as an important contextual regulator of condensate assembly, material properties, and liqui

PIK3CA mutations drive benign adipose overgrowth in facial infiltrating lipomatosis (FIL), but the downstream molecular mechanisms remain incompletely understood. This study investigated the role of palmitoyl-protein thioesterase 1 (PPT1)-mediated depalmitoylation in regulating aberrant adipogenesis induced by mutant PIK3CA. Using single-cell RNA-seq, molecular dynamics simulations, and functional assays in primary human FIL adipose-derived stem and progenitor cells (ASPCs), immortalized cell lines, and mouse models, we dissected the signaling pathway linking PIK3CA mutation to adipogenesis. Alpha-synuclein (αSyn) inclusions are a defining neuropathological feature of Parkinson's disease, but the cellular events that initiate their formation and promote neurotoxicity remain incompletely understood. Aberrant liquid-liquid phase separation has emerged as a potential early step in αSyn dysregulation, yet the physiological triggers and functional consequences of this process are unclear. Here, we show that lipid droplets promote the spontaneous phase separation of wild-type and E46K mutant αSyn into condensates. These condensates sequester lipid droplets and impair their turnover, ind

Phase separation is one of the mechanisms critical for protein function, and its aberrances are associated with cancer development. However, mutations that affect protein phase separation and cancer development have not been systematically identified and analyzed. In this study, we systematically identified the mutations affecting protein liquid-liquid phase separation in multiple cancers. We calculated the phase separation scores alterations for over 1,200,000 mutations across 16 cancer types using the TCGA dataset. We then performed pathway enrichment, kinase, TF enrichment, and survival ana

The aggregation and deposition of α-synuclein (α-syn) in Lewy bodies is the prominent pathological hallmark of Parkinson's disease. α-Syn aggregation proceeds via liquid-liquid phase separation (LLPS) and liquid-to-solid phase transition (LSPT) within the dense phase, leading to fibril formation. In this study, we reconstituted an instantaneous LLPS system for α-syn, which is primarily governed by electrostatic interactions, uncovering the structure of early-stage oligomers within the dense phase. LLPS promoted the α-syn aggregation process via its intricate nucleation and oligomerization step

The abnormal accumulation of α-synuclein (α-Syn) is a key feature of Parkinson's disease (PD) and other synucleinopathies. α-Syn undergoes liquid-liquid phase separation (LLPS) to accelerate the amyloid aggregation. β-synuclein (β-Syn) colocalizes with α-Syn and affects its aggregation. It remains poorly understood how the LLPS of α-Syn is regulated by β-Syn. Here, we find that β-Syn co-condenses with α-Syn, negatively regulating the LLPS of α-Syn. The mobility of α-Syn is reduced in α-Syn/β-Syn coacervates, diminishing the condensate fusion. β-Syn blocks the condensate growth and maturation o

Constructing multimetallic alloy heterostructures with the desired interface and phase separation is a promising method to optimize the electrochemical properties of noble metal nanomaterials. Nevertheless, tailoring the elemental distribution and lattice structures of such multimetallic nanomaterials still poses major difficulties. In this study, Cu-PtNi nanowires coupling PtNi with PtNiCu active sites are fabricated by seeded synthesis on a Cu template. Owing to the oxophilic nature of PtNi sites and the anti-CO poisoning capability of PtNiCu facets, the as-synthesized Cu-PtNi nanowires exhi

The effects of a series of sodium salts (Na3PO4, Na2CO3, Na2SO4, Na2SO3, Na2MO4, Na2CrO4, and Na2WO4) on the phase separation of poly(ethylene glycol) (PEG) solutions in water at PEG concentrations of 0.5 to 30 wt.% were studied. The salts' effects on phase separation are found to correlate with the change in the entropy related to the structural changes in water during anion hydration. The same salts' effects on phase separation in aqueous solutions of branched PEG and polyvinylpyrrolidone at a polymer concentration of 10 wt.% were also examined. The results obtained support the assumption th

Recent evidence has shown that the processes of liquid-liquid phase separation (LLPS) or liquid-liquid phase transitions (LLPTs) are a crucial and prevalent phenomenon that underlies the biogenesis of numerous membrane-less organelles (MLOs) and biomolecular condensates within the cells. Findings show that processes associated with LLPS play an essential role in physiology and disease. In this review, we discuss the physical and biomolecular factors that contribute to the development of LLPS, the associated functions, as well as their consequences for cell physiology and neurological disorders

Liquid-liquid phase separation (LLPS) is a complex physicochemical phenomenon mediated by multivalent transient weak interactions among macromolecules like polymers, proteins, and nucleic acids. It has implications in cellular physiology and disease conditions like cancer and neurodegenerative disorders. Many proteins associated with neurodegenerative disorders like RNA binding protein FUS (FUsed in Sarcoma), alpha-synuclein (α-Syn), TAR DNA binding protein 43 (TDP-43), and tau are shown to undergo LLPS. Recently, the tau protein responsible for Alzheimer's disease (AD) and other tauopathies i

During meiotic prophase I, X and Y chromosomes in mammalian spermatocytes only stably pair at a small homologous region called the pseudoautosomal region (PAR). However, the rest of the sex chromosomes remain largely unsynapsed. The extensive asynapsis triggers transcriptional silencing - meiotic sex chromosome inactivation (MSCI). Along with MSCI, a special nuclear territory, sex body or XY body, forms. In the early steps of MSCI, DNA damage response (DDR) factors, such as BRCA1, ATR, and γH2AX, function as sensors and effectors of the silencing signals. Downstream canonical repressive histon

Amyloidoses (misfolded polypeptide accumulation) are among the most debilitating diseases our aging societies face. Amyloidogenesis can be catalyzed by hydrophobic-hydrophilic interfaces (e.g., air-water interface in vitro [AWI]). We recently demonstrated hydrogelation of the amyloidogenic type II diabetes-associated islet amyloid polypeptide (IAPP), a hydrophobic-hydrophilic interface-dependent process with complex kinetics. We demonstrate that human IAPP undergoes AWI-catalyzed liquid-liquid phase separation (LLPS), which initiates hydrogelation and aggregation. Insulin modulates these proce

Amide-induced phase separation of hexafluoro-2-propanol (HFIP)-water mixtures has been investigated to elucidate solvation properties of the mixtures by means of small-angle neutron scattering (SANS), (1)H and (13)C NMR, and molecular dynamics (MD) simulation. The amides included N-methylformamide (NMF), N-methylacetamide (NMA), and N-methylpropionamide (NMP). The phase diagrams of amide-HFIP-water ternary systems at 298 K showed that phase separation occurs in a closed-loop area of compositions as well as an N,N-dimethylformamide (DMF) system previously reported. The phase separation area bec

Chromosomes with various degrees of centromere spreading and completely separated chromatids, as well as metaphasic chromosomes, were observed simultaneously in the same Burkitt's lymphoma cell. In another study, a pair of prematurely and completely separated chromatids was found in a metaphase of nasopharyngeal carcinoma cell. Based on these and other observations, the possible significance of centromere spreading and out-of-phase sister chromatid separation in the origin of chromosome nondisjunction in tumor cells is suggested and discussed.

Rosin-based chromatographic columns are widely used for separation purposes, but, to date, their phase ratios (Φ) have been imprecisely measured. This affects the understanding of their separation mechanism and the calculation of related thermodynamic parameters. In this study, a stationary phase was synthesized by bonding dehydroabietic acid (DA) to silica gel (Si-DO) and applied for reversed-phase liquid chromatography. The distribution coefficient (Kdm) of methyl dehydroabietate (MD), which has the same structure as the bonded phase of Si-DO, was used as a surrogate for the determination of

This study introduces a new in-syringe homogeneous liquid-phase microextraction method for the rapid on-site extraction of chloroanilines from water samples. Extraction was performed using a plastic syringe, eliminating the use of any electrical power source. Di-(2-ethylhexyl) phosphoric acid (DEHPA) served as the extractant. The process initially involved dissolving DEHPA in an alkaline solution to obtain a homogeneous solution. Subsequently, the sodium salt of DEHPA was precipitated by salting-out, and the resulting heterogeneous mixture was filtered using a syringe filter. The precipitate c

The distribution of fatty acid species at the sn-1/3 position or the sn-2 position of triacylglycerols (TAGs) in natural fats and oils affects their physical and nutritional properties. In fats and oils, determining the presence of one or two regioisomers and the identification of structure, where they do have one, as well as their separation, became a problem of fundamental importance to solve. A variety of instrumental technics has been proposed, such as MS, chromatography-MS or pure chromatography. A number of studies deal with the optimization of the separation, but very often, they are ex

Heterochromatin protein 1α (HP1α) is an intrinsic component of heterochromatin domains where it is involved in a diverse set of functions including heterochromatin spreading and organization, chromatin compaction and transcriptional silencing. It has been suggested that HP1α functions through a phase separation mechanism, a process that has been observed in vitro in the presence of N-terminal phosphorylation, nucleic acids and nucleosome arrays. HP1α can also interact with numerous binding partners that contain a specific motif called an HP1 access code (HAC). HACs recognize and bind to an int

Biomolecular condensates are condensed assemblies of biomolecules that form thrwough the process of liquid phase separation. Condensates in biology typically function as membraneless organelles, providing compartmentalization in the absence of a dividing lipid membrane. The molecular make-up of different condensates includes diverse multivalent proteins and nucleic acids as the primary drivers, many of them including significant fractions of intrinsically disordered regions (IDRs). To date, the sequence to phase separation relationship of IDRs has focused largely on one protein at a time, stud
