Bioprospecting in potato fields in the Central Andean Highlands: screening of rhizobacteria for plant growth-promoting properties.

The Central Andean Highlands are the center of origin of the potato plant (Solanum tuberosum). Ages of mutualism between potato plants and soil bacteria in this region support the hypothesis that Andean soils harbor interesting plant growth-promoting (PGP) bacteria. Therefore, the aim of this study was to isolate rhizobacteria from Andean ecosystems, and to identify those with PGP properties. A total of 585 bacterial isolates were obtained from eight potato fields in the Andes and they were screened for suppression of Phytophthora infestans and Rhizoctonia solani. Antagonistic mechanisms were

Expression and characterization of recombinant fungal acetyl-CoA carboxylase and isolation of a soraphen-binding domain.

Acetyl-CoA carboxylase (ACC) catalyses the first step in fatty-acid biosynthesis. Owing to its role in primary metabolism, ACC has been exploited as a commercial herbicide target and identified as a chemically validated fungicide target. In animals, ACC is also a key regulator of fat metabolism. This function has made ACC a prime target for the development of anti-obesity and anti-Type II diabetes therapeutics. Despite its economic importance, there is a lack of published information on recombinant expression of ACC. We report here the expression of enzymically active fungal (Ustilago maydis )

Controlling wildlife fungal disease spread: in vitro efficacy of disinfectants against Batrachochytrium dendrobatidis and Mucor amphibiorum.

Chytridiomycosis in amphibians, and mucormycosis in the platypus Ornithorhynchus anatinus and amphibians, are serious fungal diseases affecting these aquatic taxa. In Tasmania, Australia, the fungi that cause these diseases overlap in range along with Phytophthora cinnamomi (Pc), an invasive fungal plant pathogen. To identify disinfectants that may be useful to reduce anthropogenic spread of these fungi to uninfected wilderness areas, for example by bush walkers and forestry or fire-fighting operations, we tested 3 disinfectants and a fire-fighting foam against Mucor amphibiorum (Ma) and teste

Glutathione deficiency of the Arabidopsis mutant pad2-1 affects oxidative stress-related events, defense gene expression, and the hypersensitive response.

The Arabidopsis (Arabidopsis thaliana) phytoalexin-deficient mutant pad2-1 displays enhanced susceptibility to a broad range of pathogens and herbivorous insects that correlates with deficiencies in the production of camalexin, indole glucosinolates, and salicylic acid (SA). The pad2-1 mutation is localized in the GLUTAMATE-CYSTEINE LIGASE (GCL) gene encoding the first enzyme of glutathione biosynthesis. While pad2-1 glutathione deficiency is not caused by a decrease in GCL transcripts, analysis of GCL protein level revealed that pad2-1 plants contained only 48% of the wild-type protein amount

Black Soldier Fly Gut Microbiota as a Green Alternative to Chemical Fungicides: Broad-Spectrum Biocontrol and Implications for Reducing Fungicide Environmental Loading.

Plant diseases cause 20%-40% of global crop losses, a problem traditionally managed by chemical fungicides whose extensive use leads to widespread environmental contamination and potential human health risks. This underscores an urgent need for sustainable, low-environmental-impact alternatives. Black soldier fly (BSF)-mediated bioconversion of organic wastes offers a circular economy solution, producing frass with reported plant disease suppression potential; however, the underlying mechanisms remain largely unexplored. Here, we demonstrate that BSF frass exhibits in vitro broad-spectrum anti

Response surface modeling and structural equation analysis reveal environmental and microbial drivers of pathogen suppression during Brassica biofumigation.

BACKGROUND: Biofumigation is increasingly recognized as an environmentally friendly strategy for controlling soil-borne pathogens. However, its efficacy is highly dependent on environmental conditions, and practical predictive frameworks remain limited. A mechanistic understanding of how environmental factors interact with microbial communities and chemical metabolites is essential for optimizing biofumigation under field conditions. RESULTS: Environmental factors significantly influenced biofumigation efficacy against Fusarium and Phytophthora. Strong interactive effects among temperature, s

Elucidating the ancestral role of Class I HD-Zip transcription factors in land plants.

Class I homeodomain-leucine zipper (C1HDZ) transcription factors regulate diverse developmental and stress-related processes in land plants. While extensively studied in angiosperms, their functions in other lineages are poorly understood. In the liverwort Marchantia polymorpha, the sole C1HDZ orthologue (MpC1HDZ) controls herbivore defense by regulating oil body cell formation. This suggests that the function of C1HDZ genes may have diverged during the evolution of land plants. Here, we further investigated the function of MpC1HDZ in M. polymorpha and its integration into Arabidopsis (Arabido

Fungichromin production by Streptomyces sp. WP-1, an endophyte from Pinus dabeshanensis, and its antifungal activity against Fusarium oxysporum.

In this study, we isolated an endophytic Streptomyces sp. strain, WP-1, from surface-sterilized barks of Pinus dabeshanensis, an endangered Chinese plant. WP-1 showed strong antifungal activity against diverse pathogenic fungi, such as Fusarium oxysporum, Rhizoctonia solani, Phytophthora infestan, and Candida albicans. Based on phylogenetic analyses, preliminary identification suggested that the WP-1 strain belonged to the genus Streptomyces. Column chromatogram and HPLC were employed to isolate the primary antifungal component from the culture medium of WP-1, and it was identified as the meth

Back to basics: an evaluation of NaOH and alternative rapid DNA extraction protocols for DNA barcoding, genotyping, and disease diagnostics from fungal and oomycete samples.

The ubiquity, high diversity and often-cryptic manifestations of fungi and oomycetes frequently necessitate molecular tools for detecting and identifying them in the environment. In applications including DNA barcoding, pathogen detection from plant samples, and genotyping for population genetics and epidemiology, rapid and dependable DNA extraction methods scalable from one to hundreds of samples are desirable. We evaluated several rapid extraction methods (NaOH, Rapid one-step extraction (ROSE), Chelex 100, proteinase K) for their ability to obtain DNA of quantity and quality suitable for th

Gelatin-Chitosan-PVA Hydrogels Incorporating Trichoderma and Their Application in the Control of Phytopathogens.

The utilization of microorganisms as biocontrol agents represents a sustainable alternative to agrochemicals. Trichoderma spp. has been identified as a fungus that promotes plant growth and suppresses phytopathogens. Nonetheless, conventional commercial formulations are constrained by factors such as their limited shelf life, environmental sensitivity, and inadequate carrier systems. In this study, Trichoderma harzianum (T22) and T. viride (T18) strains were encapsulated in a hydrogel composed of chitosan, gelatin, and polyvinyl alcohol, which was prepared by pH-induced gelation via alkaline p

Primary structure and expression of acidic (class II) chitinase in potato.

Infection of potato (Solanum tuberosum) leaves by the late blight fungus Phytophthora infestans or treatment with fungal elicitor leads to a strong increase in chitinase activity. We isolated cDNAs encoding acidic (class II) chitinases (ChtA) from potato leaves and determined their structures and expression patterns in healthy and stressed plants. From the total number of cDNAs and the complexity of genomic DNA blots we conclude that acidic chitinase in potato is encoded by a gene family which is considerably smaller than that encoding basic (class I) chitinase (ChtB). The deduced amino acid s

Biocontrol of Wheat Crown Rot Using Bacillus halotolerans QTH8.

Fusarium pseudograminearum causes crown rot in wheat. This study aimed to assess the effects of the bacterial strain QTH8 isolated from Cotinus coggygria rhizosphere soil against F. pseudograminearum. Bacterial strain QTH8 was identified as Bacillus halotolerans in accordance with the phenotypic traits and the phylogenetic analysis of 16S rDNA and gyrB gene sequence. Culture filtrates of bacterial strain QTH8 inhibited the mycelial growth of F. pseudograminearum and resulted in mycelial malformation such as tumor formation, protoplast condensation, and mycelial fracture. In addition, bacterial

[Anencephaly. 10 cases seen in 3 years in Brazzaville (Congo)].

The authors report ten cases of anencephaly diagnosed between 1989 and 1991 in the maternity units of Brazzaville (Congo). The incidence of anencephaly was 0.12 per thousand. Although there was no geographical factors to definitely identify, there is a suspicion that the consumption of potatoes, infected by phytophotora infestans at the beginning of pregnancy may have had an effect; but this hypothesis would have to be confirmed by microbiological studies. The patients were young with a mean age of 27.5 years and were of low socio-economic levels. In this study female fetuses were often affect

Phenazine 1-carboxylic acid Producing Seed Harbored Endophytic Bacteria from Cultivated Rice Variety of Kerala and Its Broad Range Antagonism to Diverse Plant Pathogens.

Endophytic microorganisms residing within the diverse parts of plants play a significant role in the plant growth and defense response. In the case of the vertically transmitted seed-borne endophytes, they form the promising initiator of the juvenile plant microbiome by supporting the growth and establishment of the seedlings. Hence, the current study emphasizes the isolation and screening of plant beneficial traits of seed endophytes from the cultivated rice variety Jyothi of Kerala, India. Among the 14 bacterial endophytes obtained in the study, the isolate S3 was found to have promising act

Genomic Selection for Late Blight and Common Scab Resistance in Tetraploid Potato (Solanum tuberosum).

Potato (Solanum tuberosum) is a staple food crop and is considered one of the main sources of carbohydrates worldwide. Late blight (Phytophthora infestans) and common scab (Streptomyces scabies) are two of the primary production constraints faced by potato farming. Previous studies have identified a few resistance genes for both late blight and common scab; however, these genes explain only a limited fraction of the heritability of these diseases. Genomic selection has been demonstrated to be an effective methodology for breeding value prediction in many major crops (e.g., maize and wheat). Ho

Endophytic bacterial communities of field-grown potato plants and their plant-growth-promoting and antagonistic abilities.

To study the effect of plant growth on potato-associated bacteria, the composition and properties of bacteria colonizing the endosphere of field-grown potato were analyzed by a multiphasic approach. The occurrence and diversity of potato-associated bacteria were monitored by a cultivation-independent approach, using terminal restriction fragment length polymorphism analysis of 16S rDNA. The patterns obtained revealed a high heterogeneity of community composition and suggested the existence of plant-specific communities. However, endophytic populations correlated to a certain extent with plant

The Impact of the Culture Regime on the Metabolome and Anti-Phytopathogenic Activity of Marine Fungal Co-Cultures.

Co-cultivation, coupled with the OSMAC approach, is considered an efficient method for expanding microbial chemical diversity through the activation of cryptic biosynthetic gene clusters (BGCs). As part of our project aiming to discover new fungal metabolites for crop protection, we previously reported five polyketides, the macrolides dendrodolides E (1) and N (2), the azaphilones spiciferinone (3) and 8α-hydroxy-spiciferinone (4), and the bis-naphtho-γ-pyrone cephalochromin (5) from the solid Potato Dextrose Agar (PDA) co-culture of two marine sediment-derived fungi, Plenodomus influorescens

Deep RNA-Seq profile reveals biodiversity, plant-microbe interactions and a large family of NBS-LRR resistance genes in walnut (Juglans regia) tissues.

Deep RNA-Seq profiling, a revolutionary method used for quantifying transcriptional levels, often includes non-specific transcripts from other co-existing organisms in spite of stringent protocols. Using the recently published walnut genome sequence as a filter, we present a broad analysis of the RNA-Seq derived transcriptome profiles obtained from twenty different tissues to extract the biodiversity and possible plant-microbe interactions in the walnut ecosystem in California. Since the residual nature of the transcripts being analyzed does not provide sufficient information to identify the e
