Unveiling the enzymatic pathway of UMG-SP2 urethanase: insights into polyurethane degradation at the atomic level.

The recently discovered metagenomic urethanases UMG-SP1, UMG-SP2, and UMG-SP3 have emerged as promising tools to establish a bio-based recycling approach for polyurethane (PU) waste. These enzymes are capable of hydrolyzing urethane bonds in low molecular weight dicarbamates as well as in thermoplastic PU and the amide bond in polyamide employing a Ser-Ser cis -Lys triad for catalysis, similar to members of the amidase signature protein superfamily. Understanding the catalytic mechanism of these urethanases is crucial for enhancing their enzymatic activity and improving PU bio-recycling proces

Unlocking the potentials of Ustilago trichophora for up-cycling polyurethane-derived monomer 1,4-butanediol.

Plastic usage by microbes as a carbon source is a promising strategy to increase the recycling quota. 1,4-butanediol (BDO) is a common monomer derived from polyesters and polyurethanes. In this study, Ustilago trichophora was found to be an efficient cell-factory to valorize BDO. To investigate product formation by U. trichophora, we refined the traditional ion exclusion liquid chromatography method by examining eluent, eluent concentrations, oven temperatures, and organic modifiers to make the chromatography compatible with mass spectrometry. An LC-UV/RI-MS2 method is presented here to identi

Microstructure Evolution of Reactive Polyurethane Films During In Situ Polyaddition and Film-Formation Processes.

Due to the advantages of low energy consumption, no air and water pollutions, the reactive polyurethane films (RPUFs) are replacing the solvated and waterborne PUFs nowadays, which significantly promotes the green and low-carbon production of PU films. However, the microstructure evolution and in situ film-formation mechanism of RPUFs in solvent-free media are still unclear. Herein, according to time-temperature equivalence principle, the in situ polyaddition and film-formation processes of RPUFs generated by the typical polyaddition of diisocyanate terminated prepolymer (component B) and poly

Development and Characterization of Biodegradable Polyurethane-Urea-Based Hydrogels for the Prevention of Postoperative Peritoneal Adhesions.

Postoperative peritoneal adhesions occur after more than 60% of abdominal surgeries and can cause severe long-term side effects, such as chronic pain, infertility, and intestinal obstructions. However, currently available products for adhesion prophylaxis often lack efficiency or are too heavy to handle. Hydrogels are promising materials to be used for adhesion prevention as they show good mechanical stability and biocompatibility. Herein, we present a novel two-component sprayable, biodegradable, fast-curing, and shape-adaptive polyurethane urea (PUU) hydrogel system and the establishment of

Bioactive citrate-based polyurethane tissue adhesive for fast sealing and promoted wound healing.

As a superior alternative to sutures, tissue adhesives have been developed significantly in recent years. However, existing tissue adhesives struggle to form fast and stable adhesion between tissue interfaces, bond weakly in wet environments and lack bioactivity. In this study, a degradable and bioactive citrate-based polyurethane adhesive is constructed to achieve rapid and strong tissue adhesion. The hydrophobic layer was created with polycaprolactone to overcome the bonding failure between tissue and adhesion layer in wet environments, which can effectively improve the wet bonding strength.

A Novel NQO1 Enzyme-Responsive Polyurethane Nanocarrier for Redox-Triggered Intracellular Drug Release.

The design of nano-drug delivery vehicles responsive to tumor microenvironment stimuli has become a crucial aspect in developing cancer therapy in recent years. Among them, the enzyme-responsive nano-drug delivery system is particularly effective, as it utilizes tumor-specific and highly expressed enzymes as precise targets, leading to increased drug release at the target sites, reduced nonspecific release, and improved efficacy while minimizing toxic side effects on normal tissues. NAD(P)H:quinone oxidoreductase 1 (NQO1) is an important reductase associated with cancer and is overexpressed in

A main chain biodegradable polyurethane with anti-protein adsorption and anti-bacterial adhesion performances.

Biofilms are initially formed by substances such as proteins secreted by bacteria adhering to a surface. To achieve a durable antibacterial material, biodegradable dihydroxyl-terminated poly[(ethylene oxide)-co-(ethylene carbonate)] (PEOC(OH)2) with anti-protein adsorption properties was synthesized in this study. Further polycondensation of PEOC(OH)2 and isophorone diisocyanate (IPDI) led to biodegradable polyurethane (PEOC-PU) with PEOC as the soft segment. For comparison, polyurethanes with polyethylene glycol (PEG-PU) and polypropylene glycol (PPG-PU) as soft segments were also synthesized

Strong Thermo-tolerant Silicone-Modified Waterborne Polyurethane/Polyimide Pressure-Sensitive Adhesive.

A waterborne polyurethane pressure-sensitive adhesive (WPUPSA) has the advantages of low pollution and good viscoelasticity. However, its poor thermo-tolerance limits its application in the field of high temperatures. Hence, a novel silicone-modified strong thermo-tolerant waterborne polyurethane/polyimide pressure-sensitive adhesive is developed as a way to remedy this problem. The single-chain structure of waterborne polyurethane (WPU) is transformed into a network structure by introducing the three-position network structure to increase the cohesive energy and heat resistance of the WPUPSA.

Study on Temperature-Dependent Uniaxial Tensile Tests and Constitutive Relationship of Modified Polyurethane Concrete.

Modified polyurethane concrete (MPUC) is a new material for steel deck pavements. In service, the pavement is often cracked due to excessive tensile stress caused by temperature changes. In order to study the tensile properties of MPUC in the diurnal temperature range of steel decks, uniaxial tensile tests of MPUC were carried out at five temperatures. Three kinds of specimens and a novel fixture were designed and fabricated to compare the results of four different tensile test methods. The deformation of the specimen was collected synchronously by two methods: pasting strain gauge and digital

Antibacterial Host-Guest Intercalated LDH-Adorned Polyurethane for Accelerated Dermal Wound Healing.

Curcumin has a limited clinical application because of its extremely poor accessibility. In the present study, improved curcumin bioavailability within a castor oil polyurethane/layered double hydroxide (LDH) wound cover was achieved by preparing a curcumin p-sulfonic acid calix[4]arene (SC4A) inclusion complex. Then, it was utilized to intercalate MgAl-layered double hydroxide (MgAl-LDH) nanosheets. The incorporation of the nanostructure into a PU/Cur-SC4A-LDH film provided bacteria-killing performance against both Gram-positive Staphylococcus aureus and Gram-negative Escherichia coli bacteri

Fast Curing Multifunctional Tissue Adhesives of Sericin-Based Polyurethane-Acrylates for Sternal Closure.

The use of wire cerclage after sternal closure is the standard method because of its rigidity and strength. Despite this, they have many disadvantages such as tissue trauma, operator-induced failures, and the risk of infection. To avoid complications during sternotomy and promote tissue regeneration, tissue adhesives should be used in post-surgical treatment. Here, we report a highly biocompatible, biomimetic, biodegradable, antibacterial, and UV-curable polyurethane-acrylate (PU-A) tissue adhesive for sternal closure as a supportive to wire cerclage. In the study, PU-As were synthesized with

High-Strength Heat-Elongated Thermoplastic Polyurethane Elastomer Consisting of a Stacked Domain Structure.

We found that a high-strength elastomer was obtained by the heat elongation of a thermoplastic polyurethane (TPU) film consisting of a high content of crystalline hard segments (HS). The stress upturn continuously increased with the elongation ratio without a decrease in the strain recovery by heat elongation, i.e., the stress at break of a quenched TPU film was increased from 55 to 136 MPa by heat elongation at an elongation ratio of 300%. The results of small-angle X-ray scattering, DSC, and AFM observations revealed that: (1) anisotropically shaped HS domains were stacked at a nanometer sca

Anisotropic elastic behavior of a hydrogel-coated electrospun polyurethane: Suitability for heart valve leaflets.

Although xenograft biomaterials have been used for decades in replacement heart valves, they continue to face multiple limitations, including limited durability, mineralization, and restricted design space due to their biological origins. These issues necessitate the need for novel replacement heart valve biomaterials that are durable, non-thrombogenic, and compatible with transcatheter aortic valve replacement devices. In this study, we explored the suitability of an electrospun poly(carbonate urethane) (ES-PCU) mesh coated with a poly(ethylene glycol) diacrylate (PEGDA) hydrogel as a synthet
