Severe, persistent visual impairment associated with occipital calcification and coeliac disease.

While coeliac disease is primarily a disease of the digestive system, there have been several reports of neurological effects, both motor and cognitive. Here, we present the case of a woman with coeliac disease, under dietary control, in whom there is profound long-standing visual disturbance including reduction of visual fields, loss of rapid flicker and colour sensitivity and severe deficits in acuity. Structural magnetic resonance imaging (MRI) indicates large regions of calcification and abnormal tissue that is restricted to the occipital cortex, particularly the posterior region. Function

Disorders of higher visual function and hemi-spatial neglect.

Lesions of the extrastriate cerebral cortex result in disorders of vision, which reveal two cortical processing streams extending ventrally and dorsally from the primary visual cortex. Current theories emphasize the visual perceptual and visuomotor specialization of the ventral and dorsal streams, respectively. Recent investigations of hemi-spatial neglect suggest that the inferior parietal region may lie at the junction of the visual and motor processing pathways.

Proton magnetic resonance spectroscopy revealed choline reduction in the visual cortex in an experimental model of chronic glaucoma.

Glaucoma is a neurodegenerative disease of the visual system. While elevated intraocular pressure is considered to be a major risk factor, the primary cause and pathogenesis of this disease are still unclear. This study aims to employ in vivo proton magnetic resonance spectroscopy ((1)H MRS) to evaluate the metabolic changes in the visual cortex in a rat model of chronic glaucoma. Five Sprague-Dawley female rats were prepared to induce ocular hypertension unilaterally in the right eye by photocoagulating the episcleral and limbal veins using an argon laser. Single voxel (1)H MRS was performed

Dynamic Modulation of Beta-Band Oscillations in the LGN and Their Role in Visual Processing.

Neuronal oscillations are a ubiquitous feature of thalamocortical networks and can be dynamically modulated across processing states, enabling thalamocortical communication to flexibly adapt to varying environmental and behavioral demands. The lateral geniculate nucleus (LGN), like all thalamic nuclei, engages in reciprocal synaptic interactions with the cortex, relaying retinal information to and receiving feedback input from primary visual cortex (V1). While retinal excitation is the primary driver of LGN activity, retinal synapses represent a minority of the total synaptic input onto LGN ne

Rapid effects of tryptamine psychedelics on perceptual distortions and early visual cortical population receptive fields.

N, N-dimethyltryptamine (DMT) is a psychedelic tryptamine acting on 5-HT2A serotonin receptors, which is associated with intense visual hallucinatory phenomena and perceptual changes such as distortions in visual space. The neural underpinnings of these effects remain unknown. We hypothesised that changes in population receptive field (pRF) properties in the primary visual cortex (V1) might underlie visual perceptual experience. We tested this hypothesis using magnetic resonance imaging (MRI) in a within-subject design. We used a technique called pRF mapping, which measures neural population v

PET imaging on neurofunctional changes after optogenetic stimulation in a rat model of panic disorder.

Panic disorder (PD) is an acute paroxysmal anxiety disorder with poorly understood pathophysiology. The dorsal periaqueductal gray (dPAG) is involved in the genesis of PD. However, the downstream neurofunctional changes of the dPAG during panic attacks have yet to be evaluated in vivo. In this study, optogenetic stimulation to the dPAG was performed to induce panic-like behaviors, and in vivo positron emission tomography (PET) imaging with 18F-flurodeoxyglucose (18F-FDG) was conducted to evaluate neurofunctional changes before and after the optogenetic stimulation. Compared with the baseline,

Enhancing Experience-Dependent Plasticity Accelerates Vision Loss in a Murine Model of Retinitis Pigmentosa.

PURPOSE: Retinitis pigmentosa (RP) causes retinal degeneration and the progressive loss of vision. Here, we evaluated whether enhancing experience-dependent plasticity prolongs vision in a mouse genetic model of autosomal dominant RP. METHODS: First, we quantified the loss of visual acuity under both mesopic and photopic conditions for mice heterozygous for the P23H mutation in the Rhodopsin gene (RhoP23H/+) and littermate controls. Then, we compared acuity to the magnitude of the b-wave measured from full-field electroretinograms (ffERG) or the response properties of excitatory neurons in la

Environmental Enrichment Enhances Visual Function in Adult Amblyopia: Association With miR-132/HDAC3 Regulation.

Amblyopia is a neurodevelopmental disorder for which effective treatments in adults are limited due to reduced cortical plasticity. Environmental enrichment (EE) can partially restore this plasticity, but the underlying molecular mechanisms remain unclear. This study investigated whether the miR-132/HDAC3 pathway is associated with EE-induced visual recovery in adult amblyopic mice. An adult amblyopia model (n = 64) was established via monocular deprivation (MD). Amblyopic mice were assigned to untreated, standard housing (SH), or EE groups, with normal mice as controls. Visual function was as

N-Methyl d-aspartate receptor hypofunction reduces steady-state visual-evoked potentials.

The dynamic coordination of neural activity across populations of neurons is impaired in neuropsychiatric disorders. Here, we focused on the large-scale rhythmic responses induced by flickering light. These so-called steady-state visual-evoked potentials (SSVEPs) are reduced in people with schizophrenia (Sz). A large body of work has identified hypofunction of the N-methyl d-aspartate receptor (NMDAR) as a potential contributor to the symptoms of Sz. Here, we tested the hypothesis that NMDAR hypofunction can account for a reduced ability to generate the coordinated activity reflected in SSVEPs

Detection of visual dysfunction in optic atrophy by functional magnetic resonance imaging during monocular visual stimulation.

PURPOSE: To evaluate functional magnetic resonance imaging as an objective method for detecting visual dysfunction in various ophthalmologic disorders involving the optic nerve and the chiasm. METHODS: We performed functional magnetic resonance imaging during monocular visual stimulation on seven patients with visual field loss caused by lesions of the optic nerve and the chiasm and on three normal control subjects with no visual field loss. We correlated static threshold perimetry in the seven patients with the results of functional magnetic resonance imaging. RESULTS: In the three normal c

Loss of Postnatal Retinal Input Perturbs Cortical Lamination in the Developing Ferret Visual Cortex.

Neuroanatomical development of the visual system is influenced by activity-dependent processes. Removal of retinal input early in development in animal model experiments reproduces several structural characteristics of the primary visual cortex in humans who have been blinded early in life. Yet, many questions regarding how the loss of retinal activity affects the organization and cellular composition of the visual cortex remain to be answered. Here, we report alterations to primary visual cortex development in ferrets over the postnatal day 20 (P20) to P38 age range that arise following bilat

rTMS of the occipital cortex abolishes Braille reading and repetition priming in blind subjects.

To study the functional involvement of the visual cortex in Braille reading, we applied repetitive transcranial magnetic stimulation (rTMS) over midoccipital (MOC) and primary somatosensory (SI) cortex in blind subjects. After rTMS of MOC, but not SI, subjects made significantly more errors and showed an abolishment of the improvement in reading speed following repetitive presentation of the same word list, suggesting a role of the visual cortex in repetition priming in the blind.

A dam for retrograde axonal degeneration in multiple sclerosis?

OBJECTIVE: Trans-synaptic axonal degeneration is a mechanism by which neurodegeneration can spread from a sick to a healthy neuron in the central nervous system. This study investigated to what extent trans-synaptic axonal degeneration takes place within the visual pathway in multiple sclerosis (MS). METHODS: A single-centre study, including patients with long-standing MS and healthy controls. Structural imaging of the brain (MRI) and retina (spectral-domain optical coherence tomography) were used to quantify the extent of atrophy of individual retinal layers and the primary and secondary vis

Phosphene Perception Relates to Visual Cortex Glutamate Levels and Covaries with Atypical Visuospatial Awareness.

Phosphenes are illusory visual percepts produced by the application of transcranial magnetic stimulation to occipital cortex. Phosphene thresholds, the minimum stimulation intensity required to reliably produce phosphenes, are widely used as an index of cortical excitability. However, the neural basis of phosphene thresholds and their relationship to individual differences in visual cognition are poorly understood. Here, we investigated the neurochemical basis of phosphene perception by measuring basal GABA and glutamate levels in primary visual cortex using magnetic resonance spectroscopy. We

Behavioral measures of cortical hyperexcitability assessed in people who experience visual snow.

OBJECTIVE: To determine whether visual perceptual measures in people who experience visual snow are consistent with an imbalance between inhibition and excitation in visual cortex. METHODS: Sixteen patients with visual snow and 18 controls participated. Four visual tasks were included: center-surround contrast matching, luminance increment detection in noise, and global form and global motion coherence thresholds. Neuronal architecture capable of encoding the luminance and contrast stimuli is present within primary visual cortex, whereas the extraction of global motion and form signals requir

Feedback inhibition derived from the posterior parietal cortex regulates the neural properties of the mouse visual cortex.

Feedback regulation from the higher association areas is thought to control the primary sensory cortex, contribute to the cortical processing of sensory information, and work for higher cognitive functions such as multimodal integration and attentional control. However, little is known about the underlying neural mechanisms. Here, we show that the posterior parietal cortex (PPC) persistently inhibits the activity of the primary visual cortex (V1) in mice. Activation of the PPC causes the suppression of visual responses in V1 and induces the short-term depression, which is specific to visual st

Transcranial direct current stimulation enhances recovery of stereopsis in adults with amblyopia.

Amblyopia is a neurodevelopmental disorder of vision caused by abnormal visual experience during early childhood that is often considered to be untreatable in adulthood. Recently, it has been shown that a novel dichoptic videogame-based treatment for amblyopia can improve visual function in adult patients, at least in part, by reducing inhibition of inputs from the amblyopic eye to the visual cortex. Non-invasive anodal transcranial direct current stimulation has been shown to reduce the activity of inhibitory cortical interneurons when applied to the primary motor or visual cortex. In this do

Functional magnetic resonance imaging of binocular interactions in visual cortex in strabismus.

PURPOSE: To observe changes in primary visual cortical activation associated with perceptual suppression in individuals with strabismus, using functional magnetic resonance imaging (fMRI). METHODS: In Phase 1, pilot data were collected from 1 control and 8 strabismic participants, including 5 with amblyopia. In Phase 2, results were collected from 7 participants with strabismus (2 recalled and 5 new), including 2 with amblyopia. fMRI compared primary visual cortex activation in two conditions: visual stimuli presented to both eyes, to evoke perceptual suppression of one eye in individuals wit

Comparative Neuroplasticity in Frontal- and Lateral-Eyed Mammals With Induced-Binocular Vision Dysfunction: Insights From Monocular Deprivation Models.

Visual cortical plasticity during early postnatal life is profoundly shaped by species-specific ocular anatomy and ecological demands. This review synthesizes comparative evidence on how monocular deprivation (MD)-a classical model of amblyopia-affects visual system development in frontal- versus lateral-eyed mammals. Frontal-eyed species, including cats and primates, exhibit extensive binocular field overlaps and columnar architecture in the primary visual cortex (V1), making them highly susceptible to MD-induced shifts in ocular dominance and synaptic remodeling. In contrast, lateral-eyed sp

Cross-modal plasticity for sensory and motor activation patterns in blind subjects.

Experimental data on cortical reorganization in blind subjects using H(2)(15)O positron emission tomography and functional magnetic resonance imaging (fMRI) showed activation of the visual cortex related to Braille reading and tactile discrimination tasks in congenitally and early blind subjects. The purpose of our study was to differentiate whether occipital activation of blind subjects during Braille reading is task specific or only triggered by sensory or motor area activation. Twelve congenitally and early-onset blind subjects were studied with fMRI during Braille reading, discriminating n
