Methionine synthase and methionine synthase reductase interact with MMACHC and with MMADHC.

An increasing number of studies indicate that each step of the intracellular processing of vitamin B12 or cobalamin (Cbl) involves protein-protein interactions. We have previously described a novel interaction between methionine synthase (MS) and MMACHC and its effect on the regulation of MMACHC activity. Our goal is to further characterize the interactions of MS with other potential partners in a so-called MS interactome. We dissected the interactions and their alterations by co-immunoprecipitation and DuoLink proximity ligation assays in fibroblasts with cblG, cblE, and cblC genetic defects

Genome-Wide Interrogation of SARS-CoV-2 RNA-Protein Interactions Uncovers Hidden Regulatory Sites.

The global impact of the COVID-19 pandemic underscores the critical need for a comprehensive understanding of SARS-CoV-2 replication mechanisms. While the central roles of the RNA dependent RNA polymerase (NSP12), primase protein (NSP8), and nucleocapsid protein (N) in the virus life cycle are extensively studied, the precise nature of their interactions with the full-length viral RNA genome remain incompletely characterized. In this study, we sought to address this knowledge gap by employing enhanced crosslinking and immunoprecipitation (eCLIP) to map the binding sites of NSP8, NSP12, and N p

13q deletion in chronic lymphocytic leukemia: characterization of E4.5, a novel chromosome condensation regulator-like guanine nucleotide exchange factor.

We report the characterization of a new gene (E4.5) that maps at chromosome band 13q14.3, a chromosomal area frequently deleted in chronic lymphocytic leukemia (CLL) and in other lymphoid malignancies. E4.5 gene encodes for a 4 kb mRNA expressed in various tissues and has an open reading frame of 531 amino acids. The predicted E4.5 protein shows strong homology with the human regulator of chromosome condensation (RCC1) protein, the principal GTP exchange factor for Ran protein. The E4.5 protein contains a BTB domain in its N-terminus, a protein-protein interaction motif. Therefore, we propose

Assessing probe reliability: Functional group-specific biases revealed by interactome-wide docking of general anesthetics.

General anesthetics are widely used to induce reversible unconsciousness, yet their molecular mechanisms remain incompletely understood. Despite their low binding affinities and broad protein-binding promiscuity, general anesthetics still interact with neuronal proteins in a structurally selective manner. Experimentally, chemically modified probes have been used to map their protein targets. However, the biases introduced by the structural modifications of these remain unknown, raising the key question of how reliable such experiments are in capturing true anesthetic-protein interactions. Unexpected Importance of Aromatic-Aliphatic and Aliphatic Side Chain-Backbone Interactions in the Stability of Amyloids.

The role of aromatic and nonaromatic amino acids in amyloid formation has been elucidated by calculating interaction energies between β-sheets in amyloid model systems using density functional theory (B3LYP-D3/6-31G*). The model systems were based on experimental crystal structures of two types of amyloids: (1) with aromatic amino acids, and (2) without aromatic amino acids. Data show that these two types of amyloids have similar interaction energies, supporting experimental findings that aromatic amino acids are not essential for amyloid formation. However, different factors contribute to the

Activation and targeting of mitogen-activated protein kinases by G-protein-coupled receptors.

Over the past decade, it has become apparent that many G-protein-coupled receptors (GPCRs) generate signals that control cellular differentiation and growth, including stimulation of Ras family GTPases and activation of mitogen-activated protein (MAP) kinase pathways. The mechanisms that GPCRs use to control the activity of MAP kinases vary between receptor and cell type but fall broadly into one of three categories: signals initiated by classical G protein effectors, e.g., protein kinase (PK)A and PKC, signals initiated by cross-talk between GPCRs and classical receptor tyrosine kinases, e.g.

Protein-Capturing Microgel-Integrated Microneedle Array Patches for Enhanced Tip-Loading, Storage Stability, and Transdermal Delivery of Recombinant Proteins.

Microneedles have gained increasing attention as an ideal device for transdermal drug delivery; however, their application to proteins has been a challenging task because of their inadequate tip-loading and unsatisfactory stability. Herein, we report the development of a microgel-integrated microneedle array patch (MI-MAP) capable of not only localizing protein cargoes in the tip portion but also stabilizing them via protein-phenolic interactions. Incorporation of protein-capturing microgels allows enrichment of protein cargoes within the tip region of microneedles, resulting in a significant

Putative domain-domain interactions in the vesicular stomatitis virus L polymerase protein appendage region.

UNLABELLED: The multidomain polymerase protein (L) of nonsegmented negative-strand (NNS) RNA viruses catalyzes transcription and replication of the virus genome. The N-terminal half of the protein forms a ring-like polymerase structure, while the C-terminal half encoding viral mRNA transcript modifications consists of a flexible appendage with three distinct globular domains. To gain insight into putative transient interactions between L domains during viral RNA synthesis, we exchanged each of the four distinct regions encompassing the appendage region of vesicular stomatitis virus (VSV) India

Understanding the impact of Klebsiella pneumoniae K-Antigen based MAPS vaccine design on the immune response in animal models.

MAPS technology represents an innovative approach for the development of polysaccharide-based vaccines, relying on the affinity interaction between biotin and rhizavidin (rhavi), with potential for enhanced coverage through the incorporation of pathogen-specific proteins. Due to its flexibility, this platform is particularly attractive for the development of multivalent vaccines, as it is required for Klebsiella pneumoniae, a multidrug-resistant bacterium highlighted as prevalent cause of neonatal sepsis in low- and middle-income countries, against which no vaccines are currently available. Spatiotemporally resolved mapping of extracellular proteomes via in vivo-compatible TyroID.

Extracellular proteins play pivotal roles in both intracellular signaling and intercellular communications in health and disease. While recent advancements in proximity labeling (PL) methods, such as peroxidase- and photocatalyst-based approaches, have facilitated the resolution of extracellular proteomes, their in vivo compatibility remains limited. Here, we report TyroID, an in vivo-compatible PL method for the unbiased mapping of extracellular proteins with high spatiotemporal resolution. TyroID employs plant- and bacteria-derived tyrosinases to produce reactive o-quinone intermediates, ena

Bifenthrin exacerbates ulcerative colitis via immunotoxicity: network toxicology and experimental validation reveal novel therapeutic targets.

Bifenthrin (BF) is a widely used pyrethroid insecticide and is recognized as an endocrine-disrupting chemical (EDC). Accumulating evidence indicates that long-term exposure to BF can induce a variety of adverse health outcomes. However, its potential role in the pathogenesis of ulcerative colitis (UC) remains elusive. In this study, we integrated data from multiple databases-including the Comparative Toxicogenomics Database (CTD), TargetNet, GeneCards, SwissTargetPrediction, and STITCH-to predict potential molecular targets of BF. UC-associated genes were compiled from GeneCards, Online Mendel

Exploring the mechanism of PPCPs on human digestive system-related chronic inflammatory diseases based on network toxicology and molecular docking.

Pharmaceutical and personal care products (PPCPs), emerging pollutants, may cause chronic inflammatory and metabolic diseases by inducing metabolic disorders. To explore the underlying mechanisms, this study used network toxicology and molecular docking, focusing on four representative diseases: digestive system diseases, rheumatoid arthritis (RA), non-alcoholic fatty liver disease (NAFLD), and Parkinson's disease. By integrating data from GeneCards, OMIM, and STRING databases, we found 255, 132, 128, and 117 intersection targets between PPCPs and these diseases respectively. Protein-protein i
