Post-translational modification of proteins through lysine succinylation plays important regulatory roles in living cells. Lysine succinylation was recently identified as a novel post-translational modification in Escherichia coli, yeast, Toxoplasma gondii, HeLa cells, and mouse liver. Interestingly, only a few sites of lysine succinylation have been detected in plants to date. In this study, we identified 347 sites of lysine succinylation in 202 proteins in tomato by using high-resolution mass spectrometry. Succinylated proteins are implicated in the regulation of diverse metabolic processes,

Protein post-translational modifications (PTMs) and RNA modifications represent cutting-edge frontiers in cancer research. As a novel PTM, lactylation has been extensively implicated in tumor progression. Meanwhile, N6-methyladenosine (m6A), the most prevalent RNA modification, dictates cancer cell fate by governing RNA. Both modifications play pivotal roles in cancer initiation and progression, metabolic reprogramming, immune microenvironment remodeling, and therapeutic resistance. Accumulating evidence has revealed extensive and intricate functional crosstalk between lactylation and m6A, ser

The ryanodine receptor/Ca(2+)-release channels (RyRs) of skeletal and cardiac muscle are essential for Ca(2+) release from the sarcoplasmic reticulum that mediates excitation-contraction coupling. It has been shown that RyR activity is regulated by dynamic post-translational modifications of Cys residues, in particular S-nitrosylation and S-oxidation. Here we show that the predominant form of RyR in skeletal muscle, RyR1, is subject to Cys-directed modification by S-palmitoylation. S-Palmitoylation targets 18 Cys within the N-terminal, cytoplasmic region of RyR1, which are clustered in multipl

S-acylation is post-translational attachment of fatty acid residues, mostly palmitoyl-groups to cysteines. This lipid modification could influence protein oligomerization, localization, and topology within the membrane, and could play a regulatory role in cellular signaling. Improvements in the experimental techniques, specifically, MALDI-TOF MS, have elevated the study of S-acylation to a level of refined detail. It was possible to conclusively show that, beyond palmitate, other types of fatty acids, such as stearate and oleate could be attached to the specific binding sites. For example, fus

The activity of the phosphatase and tensin homologue (PTEN) is known to be suppressed via post-translational modification. However, the mechanism and physiological significance by which post-translational modifications lead to PTEN suppression remain unclear. Here we demonstrate that PTEN destabilization is induced by EGFR- or oncogenic PI3K mutation-mediated AKT activation in cervical cancer. EGFR/PI3K/AKT-mediated ubiquitination and degradation of PTEN are dependent on the MKRN1 E3 ligase. These processes require the stabilization of MKRN1 via AKT-mediated phosphorylation. In cervical cancer

Translational control is crucial for maintaining cellular homeostasis, yet the distinct features and regulatory requirements governing protein synthesis during erythropoiesis remain unclear. Here, we reveal that erythroid cells exhibit an extraordinarily high demand for protein synthesis, which is required for their differentiation but also implies the need for tight regulation to prevent excessive erythropoiesis. Notably, we identify significant phosphorylation of eukaryotic elongation factor 2 (eEF2) at threonine 56 during erythroid differentiation, which reduces protein synthesis and acts a

Lactylation, a recently discovered post-translational modification, links cellular metabolism to epigenetic regulation and immune function. Once considered a mere glycolytic byproduct, lactate is now recognized as a signaling metabolite that can shape gene expression and protein activity via histone and non-histone lactylation. This review synthesizes evidence on enzymatic and non-enzymatic mechanisms of lactylation, including lactoyl-CoA-dependent pathways, glyoxalase-mediated routes, and emerging functions of aminoacyl-tRNA synthetases. We highlight lactylation "writers" and "erasers," stere

PRMT5 is a member of a class of enzymes called protein arginine methyltransferases (PRMTs) that play a role in maintaining genomic stability through post-translational modification of components of the TIP60 chromatin remodeling super complex. TIP60 is required primarily for chromatin remodeling at DNA double-strand breaks. Mutations in either TIP60 or PRMT5 affect repair by homologous recombination. A recent study has shown that in leukemia and lymphoma, PRMT5 also controls mRNA expression of TIP60 and other DNA repair genes by regulating alternative splicing. This analysis utilizes publicly

MEX3A is an evolutionarily conserved RNA-binding protein characterized by two KH RNA-binding domains and a C-terminal RING domain, suggesting potential roles in both post-transcriptional regulation and protein modification. Although MEX3A has been implicated in several individual cancer types, its shared and cancer-specific roles across human cancers remain incompletely understood. In this study, we performed a comprehensive pan-cancer analysis of MEX3A by integrating publicly available datasets, including TCGA, GTEx, CPTAC, and other external resources. We systematically evaluated MEX3A expre

Testis, the only organ responsible for generating sperm, is by far the organ with the largest variety of proteins and tissue-specific proteins in humans. In testis, spermatogenesis is a multi-step complex process well-accepted that protein and mRNA are decoupled in certain stages of spermatogenesis. With the fast development of mass spectrometry-based proteomics, it is possible to systemically study protein abundances and modifications in testis and sperm to help us understand the molecular mechanisms of spermatogenesis. This review provides an overview of the recent progress of proteomics ana

Severe acute respiratory syndrome coronavirus nsp1 protein suppresses host gene expression, including type I interferon production, by promoting host mRNA degradation and inhibiting host translation, in infected cells. We present evidence that nsp1 uses a novel, two-pronged strategy to inhibit host translation and gene expression. Nsp1 bound to the 40S ribosomal subunit and inactivated the translational activity of the 40S subunits. Furthermore, the nsp1-40S ribosome complex induced the modification of the 5' region of capped mRNA template and rendered the template RNA translationally incompet

Legionella pneumophila (L.p.), the microbe responsible for Legionnaires' disease, secretes ∼300 bacterial proteins into the host cell cytosol. A subset of these proteins affects a wide range of post-translational modifications (PTMs) to disrupt host cellular pathways. L.p. has 5 conserved eukaryotic-like Ser/Thr effector kinases, LegK1-4 and LegK7, which are translocated during infection. Using a chemical genetic screen, we identified the Hsp70 chaperone family as a direct host target of LegK4. Phosphorylation of Hsp70s at T495 in the substrate-binding domain disrupted Hsp70's ATPase activity

The modifications of transfer RNA (tRNA) add an extra layer of post-transcriptional gene regulation in cancer biology. While translation initiation and termination have been extensively studied, the role of tRNA modifications in elongation remains largely unexplored. These chemical changes, particularly in the anticodon loop, enhance codon–anticodon pairing and translation fidelity, but their impact on cancer, especially breast cancer, is not well defined. We analyzed 73 tRNA modification–related genes across 33 TCGA cancer types and breast cancer subtypes by integrating transcriptomic and pro

N 6-Threonylcarbamoyladenosine at A37 (t6A37) of ANN-decoding transfer RNAs (tRNAs) is a universal modification whose functions have been well documented in bacteria and lower eukaryotes; however, its role in organellar translation is not completely understood. In this study, we deleted the mitochondrial t6A37-modifying enzyme OSGEPL1 in HEK293T cells. OSGEPL1 is dispensable for cell viability. t6A37 hypomodification selectively stimulated N1-methyladenosine at A9 (m1A9) and N2-methylguanosine at G10 (m2G10) modifications and caused a substantial reduction in the aminoacylation of mitochondria

Ebola virus (EBOV) is a highly lethal pathogen that has caused several outbreaks of severe hemorrhagic fever in humans since its emergence in 1976. The EBOV glycoprotein (GP1,2) is the sole viral envelope protein and a major component of immunogenicity; it is encoded by the GP gene along with two truncated versions: soluble GP (sGP) and small soluble GP (ssGP). sGP is, in fact, the primary product of the GP gene, and it is secreted in abundance during EBOV infection. Since sGP shares large portions of its sequence with GP1,2, it has been hypothesized that sGP may subvert the host immune respon

Pulmonary hypertension (PH) is a progressive, life-threatening cardiovascular disorder. It features irreversible pulmonary vascular remodeling and causes right ventricular failure and mortality. The underlying pathogenesis of PH is incompletely elucidated. Ubiquitination is a reversible post-translational modification. It regulates protein degradation and membrane trafficking and contributes critically to PH development and progression. The ubiquitin-proteasome system (UPS), especially E3 ubiquitin ligases and Deubiquitinases, modulates the function of pulmonary artery endothelial cells and pu

UFMylation is a conserved ubiquitin-like post-translational modification that controls protein stability and tissue homeostasis, while its role in amelogenesis remains largely uncharacterized. Here, we investigated the function of UFL1, the sole E3 ligase of the UFMylation pathway, in mammalian enamel development using K14-Cre-mediated epithelial-specific Ufl1 knockout mice. Ufl1 ablation caused severe amelogenesis imperfecta, with impaired enamel deposition, hypomineralization, and progressive tooth damage, accompanied by abnormal cervical loop development.
