The purpose of this study was to evaluate the effect of cynarin, a caffeoylquinic acid derivative in artichoke, on glutamate release elicited by 4-aminopyridine (4-AP) in rat cortical nerve terminals (synaptosomes). We observed that cynarin decreased 4-aminopyridine-elicited glutamate release, which was prevented by the removal of external free Ca2+ with ethylene glycol bis (β-aminoethyl ether)-N,N,N,N-tetraacetic acid (EGTA) or the blockade of P/Q-type calcium channels with ω-agatoxin IVA. Molecular docking also revealed that cynarin formed a hydrogen bond with the P/Q-type Ca2+ channel, indi

Leaves of Mangifera indica (mango) are traditionally used to alleviate inflammation and pain, yet their bioactive chemical profiles and mechanisms of action remain underexplored. Given the growing demand for safer, plant-derived therapeutics as alternatives to conventional anti-inflammatory and analgesic drugs with known adverse effects, a deeper understanding of the bioactive composition of M. indica leaf is both timely and necessary. This study investigates the metabolomic composition and therapeutic potential of leaf extracts from two cultivars-Zebda and Alphonso. Aqueous, ethanol, n-butano

BACKGROUND: Ulcerative colitis (UC) is a chronic inflammatory disease of the gastrointestinal tract. Despite the availability of traditional clinical treatments, their limitations highlight the need for alternative therapies derived from natural products that offer both strong efficacy and minimal side effects. Chaenomeles speciosa (Sweet) Nakai (CS), edible and medicinal fruit traditionally used to treat gastrointestinal disorders, has shown potential in UC management. However, the specific phytochemicals and mechanisms through which CS exerts its therapeutic effects remain poorly understood.

A feeding study was carried out in which six healthy ileostomists ingested a juice drink containing a diversity of dietary (poly)phenols derived from green tea, apples, grapes and citrus fruit. Ileal fluid and urine collected at intervals over the ensuing 24 h period were then analysed by HPLC-MS. Urinary excretions were compared with results obtained in an earlier study in which the juice drink was ingested by ten healthy control subjects with an intact colon. Some polyphenol components, such as (epi)catechins and (epi)gallocatechin(s), were excreted in urine in similar amounts in ileostomist

Pseudomonas putida DOT-T1E was used as a model to develop a "phenomics" platform to investigate the ability of P. putida to grow using different carbon, nitrogen, and sulfur sources and in the presence of stress molecules. Results for growth of wild-type DOT-T1E on 90 different carbon sources revealed the existence of a number of previously uncharted catabolic pathways for compounds such as salicylate, quinate, phenylethanol, gallate, and hexanoate, among others. Subsequent screening on the subset of compounds on which wild-type DOT-TIE could grow with four knockout strains in the global regul

Polycyclic aromatic hydrocarbons (PAHs) are widespread environmental pollutants that may impair immune function. However, the immunotoxic mechanisms of PAH exposure remain largely unexplored. The present study was designed as a mechanistic, effect-oriented investigation, in which male C57BL/6J mice were exposed to phenanthrene via intraperitoneal injection for six consecutive weeks. We assessed toxicological effects of phenanthrene exposure in mice through urinary metabolite profiling, hematological and immunological tests, and serum metabolomics. Urinary concentrations of phenanthrene metabol

Invasive fungal infections threaten immunocompromised patients, and the toxicity, resistance, and narrow spectrum of current therapies demand new agents. A chemical investigation of the medicinal plant Ligusticum chuanxiong led to the isolation of a rare pentyl-substituted cyclohexene derivative, ligustrumdiol A (LDA). Its structure was fully elucidated by spectroscopic analysis, and an enantioselective total synthesis was accomplished in 12 steps from d-quinic acid with an overall yield of 11.2%. Although LDA alone showed only weak antifungal activity against Aspergillus fumigatus (MIC = 400

BACKGROUND: Exposure to ultraviolet A (UVA) irradiation is the major cause of human skin aging. Suppression of UVA irradiation-induced skin fibroblast cell damage protects the skin against aging. An oxidative stress response transcription factor nuclear factor-(erythroid-derived 2)-related factor 2 (Nrf2) has an important role as a cytoprotective system against oxidative stress in the human skin and other organs. Propolis has been commonly used as a traditional medicine since ancient times. The water extract of propolis (WEP) mainly contains caffeoylquinic acids. In our previous study, we repo

Laetia suaveolens, known as "casinga-cheirosa", crude extract EB719 has previously shown cytotoxic activity against prostate cancer and squamous cell carcinoma. For the first time, seven molecules were isolated from its apolar-α-tocopherol (1) and sitosterol (2)-and polar-3-O-caffeoylquinic acid (3), 4-O-caffeoylquinic acid (4), 5-O-feruloylquinic acid (5), hyperoside (6), and isoquercitrin (7)-fractions. Acute toxicity was determined in a two-stage experiment: (1) a reduced number of Balb-c male mice received 5000 mg/kg of EB719 to allow evaluation of general activity and other 27 parameters,

Hibiscus species (Malvaceae) have been long used as an antihypertensive folk remedy. The aim of our study was to specify the optimum solvent for extraction of the angiotensin-converting enzyme inhibiting (ACEI) constituents from Hibiscus sabdariffa L. The 80% methanol extract (H2) showed the highest ACEI activity, which exceeds that of the standard captopril (IC50 0.01255 ± 0.00343 and 0.210 ± 0.005 µg/mL, respectively). Additionally, in a comprehensive metabolomics approach, an ultra-performance liquid chromatography (UPLC) coupled to the high resolution tandem mass spectrometry (HRMS) method

Dietary supplementation with dried plum (DP) has been shown to protect against and reverse established osteopenia in ovariectomized rodents. Based on in vitro studies, we hypothesized that DP polyphenols may be responsible for that bone-sparing effect. This study was designed to (1) analyze whether the main phenolic acids of DP control preosteoblast proliferation and activity in vitro; (2) determine if the polyphenolic content of DP or DP juice concentrate is the main component improving bone health in vivo; and (3) analyze whether DP metabolites directly modulate preosteoblast physiology ex v

Dried plum (DP), a rich source of polyphenols has been shown to have bone-preserving properties in both animal models of osteoporosis and postmenopausal women. We evaluated if DP alleviated the destruction of joints in transgenic mice (TG) that overexpress human tumor necrosis factor (TNF), a genetic model of rheumatoid arthritis (RA). A four-week treatment of 20% DP diet in TG slowed the onset of arthritis and reduced bone erosions in the joints compared to TG on a regular diet. This was associated with fewer tartrate-resistant acid phosphatase (TRAP) positive cells, suggesting decreased oste

An estimated 450 species of Dryopteris in the Dryoperidaceae family grow in Japan, North and South Korea, China, Pakistan, and Kashmir. This genus has been reported to have biological capabilities; however, research has been conducted on Dryopteris juxtapostia. Therefore, with the present study, we aimed to exploring the biological potential of D. juxtapostia root and shoot extracts. We extracted dichloromethane and methanol separately from the roots and shoots of D. juxtapostia. Antioxidant activity was determined using DPPH, FRAP, and H2O2 assays, and anti-inflammatory activities were evalua
