Partial to complete suppression of unilateral noise-induced tinnitus in rats after cyclobenzaprine treatment.

Some forms of tinnitus are believed to arise from abnormal central nervous system activity following a single or repeated noise exposure, for which there are no widely accepted pharmacological treatments. One central site that could be related to tinnitus awareness or modulation is the locus coeruleus, a brainstem structure associated with stress, arousal, and attention. In the present study, we evaluated the effects of cyclobenzaprine, a drug known to act on the rat locus coeruleus, on noise-induced tinnitus using Gap Prepulse Inhibition of the Acoustic Startle (GPIAS). In untreated rats, bri

Prepulse inhibition in psychiatric disorders--apart from schizophrenia.

Prepulse inhibition (PPI) is a robust operational measure of sensorimotor gating. In schizophrenic patients PPI is deficient. The aim of our review was to investigate the state of science regarding PPI and psychiatric disorders aside from schizophrenia. We used the online database PubMed in order to search for original published reports on PPI studies. The terms "prepulse inhibition", "sensorimotor gating", "blink recovery", and "blink reflex excitability" have been combined with the names of psychiatric disorders. We found that PPI is deficient in obsessive compulsive disorder (OCD) and Gille

Models and mechanisms of anxiety: evidence from startle studies.

RATIONALE: Preclinical data indicates that threat stimuli elicit two classes of defensive behaviors, those that are associated with imminent danger and are characterized by flight or fight (fear), and those that are associated with temporally uncertain danger and are characterized by sustained apprehension and hypervigilance (anxiety). OBJECTIVE: The objectives of the study are to (1) review evidence for a distinction between fear and anxiety in animal and human experimental models using the startle reflex as an operational measure of aversive states, (2) describe experimental models of anxie

Familial hyperekplexia and refractory status epilepticus: a new autosomal recessive syndrome.

Hyperekplexia is a rare disorder characterized by an exaggerated startle response to noise and handling and by neonatal hypertonia. It is predominantly an autosomal dominant disease; however, atypical cases with additional variable manifestations have been reported. We report a hitherto undescribed association of hyperekplexia and refractory status epilepticus in two siblings. Both children were born after an uneventful pregnancy to healthy unrelated Ashkenazi Jews. Both had increased startle and tone from birth and later became hypotonic. A metabolic evaluation, including a muscle biopsy, was

Changes in neurobiological markers of reactivity to alcohol-related stimuli in alcohol-dependent patients after two years of treatment.

BACKGROUND: Salience and cortisol response following exposure to alcohol-related stimuli (ARS) are considered markers of vulnerability and status in alcohol use disorders (AUD). Negative emotionality may also maintain alcohol consumption by negative reinforcement. What happens to these parameters once sustained symptom remission has been achieved is unknown. This study investigated changes in these variables after 2 years of outpatient treatment and compared them with a healthy control group. METHODS: Patients with AUD (n=154) were assessed at baseline (T0) and after 2 years of treatment (T2)

Chlorogenic Acid Improves Salicylic Acid-Induced Cochlear Injury by Reducing Oxidative Stress and Inflammatory Response Through TLR4/NF-κB Signalling Pathway.

BACKGROUND: Sodium salicylate (SS) causes hearing damage and tinnitus in humans and animals. Chlorogenic acid (CGA) has strong antioxidant and anti-apoptotic effects, but whether it can protect the cochlea is unknown. METHODS: SS was used to induce cochlear injury in rats, followed by 150, 300 and 600 mg/kg CGA treatment. The changes of tinnitus behaviour in each group were detected by auditory brainstem response (ABR), behavioural test, beam balance, Preyer reflex and auditory startle test. The levels of oxidative stress and inflammation were detected by kit. The pathological damage of cochl

Effects of carbon disulfide on motor function and responsiveness to d-amphetamine in rats.

Male, albino rats of the Fisher strain were exposed to 2 mg carbon disulfide/l of air for 4 hours per day, 5 days per week for 6 weeks. Neurobehavioral effects were not observed after 3 weeks of dosing, but hindlimb extensor responses and performance on the inclined screen (motor coordination) were impaired after 6 weeks of exposure. Forelimb grip strength and exploratory locomotor activity were not affected. Three weeks after cessation of dosing, recovery of function was observed. Rats exposed to CS2 for 6 weeks were found to be stimulated less than air ventilated controls by 3 mg/kg of d-amp

Anesthetic management of a parturient with hyperekplexia.

Hyperekplexia is a hereditary disorder characterized by exaggerated startle reflex in response to unexpected acoustic, tactile, and other stimuli. Neonates with hyperekplexia may present with hypertonia, developmental delays, apnea, and sudden death. The diagnosis is based on published clinical criteria. In some cases, a mutation encoding the postsynaptic inhibitory glycine receptors (GLRA1, GLRB) or presynaptic glycine transporter (SLC6A5) resulting in abnormal glycinergic neurotransmission is present. We report the case of a 38-year-old gravida 6 para 1 (G6P1) parturient with hyperekplexia w

Two Japanese families with hyperekplexia who have a Arg271Gln mutation in the glycine receptor alpha 1 subunit gene.

We report two Japanese patients from two families with hyperekplexia who have a Arg271Gln mutation in the glycine receptor alpha 1 subunit gene. The clinical course of both patients was typical for hyperekplexia, characterized by neonatal hypertonia and exaggerated startle response, and which improved gradually with age. One was associated with umbilical hernia and hip dislocation, diagnosed at 11 months, while the other was diagnosed at 1 month. Both showed positive head retraction reflex. Four Japanese families have been reported as having hyperekplexia including our cases, of which three ha

GLRB allelic variation associated with agoraphobic cognitions, increased startle response and fear network activation: a potential neurogenetic pathway to panic disorder.

The molecular genetics of panic disorder (PD) with and without agoraphobia (AG) are still largely unknown and progress is hampered by small sample sizes. We therefore performed a genome-wide association study with a dimensional, PD/AG-related anxiety phenotype based on the Agoraphobia Cognition Questionnaire (ACQ) in a sample of 1370 healthy German volunteers of the CRC TRR58 MEGA study wave 1. A genome-wide significant association was found between ACQ and single non-coding nucleotide variants of the GLRB gene (rs78726293, P=3.3 × 10-8; rs191260602, P=3.9 × 10-8). We followed up on this findi

Effects of perinatal asphyxia on the neurobehavioral and retinal development of newborn rats.

Perinatal asphyxia during delivery produces long-term deficits and represents a major problem in both neonatal and pediatric care. Several morphological, biochemical and behavioral changes have been described in rats exposed to perinatal asphyxia. The aim of the present study was to evaluate how perinatal asphyxia affects the complex early neurobehavioral development and retinal structure of newborn rats. Asphyxia was induced in ready-to-deliver mothers by removing the pups by cesarian section after 15 min of asphyxia. Somatic and neurobehavioral development was tested daily during the first 3

A non-invasive biomechanical model of mild TBI in larval zebrafish.

A mild traumatic brain injury is a neurological dysfunction caused by biomechanical forces transmitted to the brain in physical impacts. The current understanding of the neuropathological cascade resulting in the manifested clinical signs and symptoms is limited due to the absence of sensitive brain imaging methods. Zebrafish are established models for the reproduction and study of neurobiological pathologies. However, all available models mostly recreate moderate-to-severe focal injuries in adult zebrafish. The present work has induced a mild brain trauma in larval zebrafish through a non-inv

Aversive imagery in posttraumatic stress disorder: trauma recurrence, comorbidity, and physiological reactivity.

BACKGROUND: Posttraumatic stress disorder (PTSD) is characterized as a disorder of exaggerated defensive physiological arousal. The novel aim of the present research was to investigate within PTSD a potential dose-response relationship between past trauma recurrence and current comorbidity and intensity of physiological reactions to imagery of trauma and other aversive scenarios. METHODS: A community sample of principal PTSD (n = 49; 22 single-trauma exposed, 27 multiple-trauma exposed) and control (n = 76; 46 never-trauma exposed, 30 trauma exposed) participants imagined threatening and neut

Maturation and transformation of reflexes that protect the laryngeal airway from liquid aspiration from fetal to adult life.

Several reflexes are initiated in the fetus and newborn when hypochloremic or strongly acidic solutions contact the epithelium that surrounds the entrance to the laryngeal airway. These reflexes, known collectively as the laryngeal chemoreflex (LCR), include startle, rapid swallowing, apnea, laryngeal constriction, hypertension, and bradycardia. Many studies have shown that prolonged apnea associated with the LCR may be life threatening and might conceivably be a cause of sudden infant death syndrome. This certainly may be true, but the concept of a lethal LCR paradoxically contrasts with the

Selective immunolesioning of cholinergic neurons in nucleus basalis magnocellularis impairs prepulse inhibition of acoustic startle.

Information processing and attentional abnormalities are prominent in neuropsychiatric disorders. Since the cholinergic neurons located in the nucleus basalis magnocellularis have been shown to be involved in attentional performance and information processing, recent efforts to analyze the significance of the basal forebrain in the context of schizophrenia have focused on this nucleus and its projections to the cerebral cortex. We report here that bilateral selective immunolesioning of the cholinergic neurons in the nucleus basalis magnocellularis is followed by significant deficits in sensori

The effect of an alerting acoustic stimulus on the heart rate, sympathetic activity and aortic baroreceptor discharge in conscious rabbits before and after naloxone administration.

Blood pressure, heart rate, aortic nerve activity and cervical sympathetic discharge were recorded simultaneously in 10 rabbits. Chronic recordings were made with electrodes implanted to the uncut aortic and cervical sympathetic nerves. 1. The alerting acoustic stimulus produced a short lasting decrease in sympathetic activity with a transient bradycardia. 2. In 6 out of 8 rabbits i.v. administration of naloxone chloride (100 mg/kg) diminished or abolished early inhibitory effects evoked by acoustic stimulus. 3. The sympatho-inhibitory system involved in the startling response appears to be in

The cholinergic system-dependent myoclonus of the baboon Papio papio is a reticular reflex myoclonus.

Neurophysiological studies were performed on four Papio papio baboons presenting with nonepileptic myoclonus (a startle response resembling stimulus-sensitive jerk). Investigations of the EEG, back-averaged EEG, and somatosensory evoked potentials revealed the absence of cortical correlates preceding the jerks, and exclusion of cerebral cortex involvement. No long-latency reflexes could be recorded in these animals. The jerks were symmetric when evoked by unilateral stimulation in normal baboons as well as in a split-brain animal. Polymyographic records showed that the first muscle involved du

The clinical features and prognosis of chronic posthypoxic myoclonus.

The clinical and neurophysiologic features of 14 patients with chronic posthypoxic myoclonus are presented. Patients were first seen a mean of 2.5 years (range, 2 to 105 months) after the hypoxic event and followed up for 3.7 years (range, 7 to 84 months) thereafter. All patients had had a cardiorespiratory arrest, most caused by an acute asthmatic attack (11 cases). All patients had multifocal action myoclonus. Eleven patients had additional stimulus-sensitive myoclonus. There was late improvement in the myoclonic syndrome and the level of disability in all but one patient. Three patients wer

Positive allosteric modulation of GABAB receptors ameliorates sensorimotor gating in rodent models.

BACKGROUND: Converging evidence points to the involvement of γ-amino-butyric acid B receptors (GABABRs) in the regulation of information processing. We previously showed that GABABR agonists exhibit antipsychotic-like properties in rodent models of sensorimotor gating deficits, as measured by the prepulse inhibition (PPI) of the acoustic startle reflex. The therapeutic potential of these agents, however, is limited by their neuromuscular side effects; thus, in this study, we analyzed whether rac-BHFF, a potent GABABR-positive allosteric modulator (PAM), could counter spontaneous and pharmacolo
