BACKGROUND: Neutrophils (polymorphonuclear leukocytes, PMNs) have long been underestimated in the context of autoimmunity. To elucidate their role from a sex-specific perspective, an immunoendocrinological framework offers a compelling investigative approach. We hypothesize that the sex hormone 17-β-estradiol (E2) and the endocrine-disrupting chemical bisphenol A (BPA) modulate PMN functions in a sex- and concentration-dependent manner. Cadmium-induced cell death was studied in suspension-cultured tomato (Lycopersicon esculentum Mill.) cells (line MsK8) treated with CdSO(4). OBJECTIVE: To study the effects of TNF-alpha and live Streptococcus pneumoniae on human neutrophil oxidative burst and beta 2-integrin expression using flow cytometry.
METHODS: Six clinical isolates of S. pneumoniae (serotypes 3, 19A, 22F, 6A, 33F and 9N) from patients with bacteremic pneumonia or upper respiratory tract infections were studied. Whole blood was incubated either alone, with TNF-alpha or with S. pneumoniae or with both TNF-alpha and pneumococci at a ratio of one neutrophil per 1--5 bacteria. After 30 min of incubation, the tubes were put into ice, fixed and analysed.

The presence of autoantibodies to neutrophil cytoplasmic proteinase-3, usually reported as c-ANCA, is virtually diagnostic for Wegener's granulomatosis and related conditions. These antibodies are highly associated with active systemic disease, and may be implicated in pathogenesis. In vitro, c-ANCA can induce a respiratory burst and degranulation of cytokine-primed neutrophils, leading to lysis of cultured endothelial cells. This and other putative pathogenic mechanisms are reviewed.
