An application of the hydrodynamic pore theory to percutaneous absorption of drugs.

Skin permeability of drugs was evaluated based on the hydrodynamic pore theory. Four polar solutes were used, with differing molecular sizes--ethylene glycol, 1,3-butylene glycol, antipyrine and sucrose--and isosorbide dinitrate was also selected as a lipophilic drug. The skin permeations of solvent (D2O) and one of these drugs were measured simultaneously under various osmotic pressures to calculate the reflection coefficient. The clearance of isosorbide dinitrate was independent of the solvent flux, whereas a linear relationship was obtained between the solvent flux and the clearance of each

Skin Barrier Modulation by Oleyl-Based Co-Formulants: Mechanistic Insights from Molecular Simulations.

Understanding and predicting dermal absorption is important for many applications including transdermal drug delivery, personal care product development, and chemical risk assessment. Real-world dermal exposures typically involve chemically complex formulations that contain not only the active ingredient but also coformulants such as solvents, thickeners, humectants and emulsifiers, which can significantly alter skin permeability and dermal absorption. In the case of personal care and pharmaceutical formulations, penetration enhancers are often added to improve delivery of active ingredients,

Occlusion vs. skin barrier function.

BACKGROUND/AIMS: Skin occlusion may increase percutaneous absorption of applied chemicals, with some exceptions. It also obstructs the normal ventilation of the skin surface and increases stratum corneum hydration and hence compromises skin barrier function. METHODS/RESULTS: This review focuses the effects of occlusion on skin barrier function, in particularly, as defined with objective skin bioengineering technology. CONCLUSIONS: The effects of occlusion on skin barrier function have been defined with various techniques. Optimal hydrocolloid materials can absorb excess water and reduce the

Why and by How Much is Insulin Absorption Reduced by Insulin-derived Amyloidosis? A Scoping Review.

Insulin therapy is one of the central treatments for diabetes mellitus. Insulin-derived localized amyloidosis (IDLA) is a known skin-related complication of insulin injection. This is one of the causes of poor glycemic control in diabetic patients on insulin therapy. The aim of this study was to review and update the findings on the extent and mechanism of reduced insulin absorption in IDLA. A literature search was conducted on decreased insulin absorption and its mechanisms, and nine references were selected, with seven of these on decreased insulin absorption and four on mechanisms. Insulin

Spectroscopic observations on human pigmentation.

BACKGROUND: The main chromophores of human skin are melanins and hemoglobins along with carotenoids, bilirubin, and other compounds. In an effort to study the spectral signatures of skin melanin, we measured absorption spectra in a variety of situations, including a method to show early signs of re-pigmentation in vitiligo. METHODS: To measure skin in vivo, the essential component was a "Bifurcated Optical Fiber" with one end connected to the light source and the second end connected to the spectrometer while the common end was placed on the skin. RESULTS: In a typical in situ "melanin in sk

An assessment of the phototoxic hazard of a personal product ingredient using in vitro assays.

Where substances are intended for use in personal products applied to the skin an assessment of potential phototoxic hazard is required. This report describes a tiered testing strategy involving in vitro assays used for the phototoxic hazard assessment of a personal product ingredient (Ingredient X). The initial assay was measurement of a UV/visible absorption spectrum to identify absorption at relevant wavelengths. This was followed by in vitro assays for phototoxicity (3T3 cell neutral red uptake phototoxicity test) and photoallergy (photobinding to human serum albumin). These in vitro scree

Optimized cutaneous delivery of anti-wrinkle dipeptide KT via molecular modification: Preformulation, permeation, and the importance of conjugate chain length.

INTRODUCTION: Anti-aging peptides, such as dipeptide KT, are promising rejuvenating agents and have recently received significant attention. However, their hydrophilic nature makes skin absorption therapeutically inadequate. The excessive hydrophilicity of peptides is partially solved by lipoidal conjugates, however, the increased molecular weight due to conjugation creates a new obstacle to skin permeation. METHODS: In an attempt to concurrently solve these limitations, here we have studied different short-mid chain fatty acids (C6-C18) conjugates of dipeptide KT. Different fatty acid chain

Dermal bioavailability of perfluoroalkyl substances using in vitro 3D human skin equivalent models.

Perfluoroalkyl substances (PFAS) have been identified in various products that come in contact with human skin, ranging from school uniforms to personal care products. Despite this, knowledge on human dermal uptake of PFAS is lacking. Thus, the human dermal absorption of 17 PFAS was assessed, for the first time, using in vitro 3D-human skin equivalent models exposed to 500 ng/cm2 PFAS dissolved in methanol over 24-36 h. The distribution of target PFAS is presented, based on three fractions: absorbed, un-absorbed, and retained within skin tissue (absorbable dose). Perfluoropentanoic acid (PFPeA

Skin decontamination of Carfentanil in vitro.

Skin decontamination of Chemical Biological Radioactive and Nuclear (CBRN) materials involves the timely and effective removal of the contaminants from the skin surface. The current work evaluated Fuller's Earth & The Reactive Skin Decontaminant Lotion Kit (RSDL®) to investigate whether they were as efficacious against free base Carfentanil skin contamination as they are against chemical warfare agents. The in vitro methodology used allowed for evaluation of decontamination regimens as specified by the decontaminant manufacturer rather than as an application of a bolus dose left in situ for th

Tuning lipid structure by bile salts: Hexosomes for topical administration of catechin.

The delivery of bio-active molecules through the skin is challenging given the complex structure of its outer layer, the stratum corneum. Here we explore the possibility to encapsulate natural compounds into nanocarriers containing permeation enhancers that can affect the fluidity of the stratum corneum lipids. This approach is expected to facilitate dermal or transdermal release. For this purpose, the application of bile salts, which are natural surfactants involved in vivo in lipid digestion, was exploited. Bile salts were added to lipid liquid crystalline nanoparticles (NPs) made of monoole
