The Lipoxin Receptor/FPR2 Agonist BML-111 Protects Mouse Skin Against Ultraviolet B Radiation.

Excessive exposure to UV, especially UVB, is the most important risk factor for skin cancer and premature skin aging. The identification of the specialized pro-resolving lipid mediators (SPMs) challenged the preexisting paradigm of how inflammation ends. Rather than a passive process, the resolution of inflammation relies on the active production of SPMs, such as Lipoxins (Lx), Maresins, protectins, and Resolvins. LXA4 is an SPM that exerts its action through ALX/FPR2 receptor. Stable ALX/FPR2 agonists are required because SPMs can be quickly metabolized within tissues near the site of formati

Pro-resolving mediators promote resolution in a human skin model of UV-killed Escherichia coli-driven acute inflammation.

While the treatment of inflammatory disorders is generally based on inhibiting factors that drive onset of inflammation, these therapies can compromise healing (NSAIDs) or dampen immunity against infections (biologics). In search of new antiinflammatories, efforts have focused on harnessing endogenous pathways that drive resolution of inflammation for therapeutic gain. Identification of specialized pro-resolving mediators (SPMs) (lipoxins, resolvins, protectins, maresins) as effector molecules of resolution has shown promise in this regard. However, their action on inflammatory resolution in h

Sex-based differences in conjunctival goblet cell responses to pro-inflammatory and pro-resolving mediators.

Many conjunctival inflammatory diseases differ between the sexes and altered conjunctival goblet cells (CGCs) response is often involved. Inflammation is initiated by the release of pro-inflammatory mediators and terminated by the biosynthesis of specialized pro-resolution mediators (SPMs). Herein, we determined the sex-based difference in the responses of CGCs to inflammatory stimuli or pro-resolving lipid SPMs and their interaction with sex hormones. GCs were cultured from pieces of human conjunctiva in RPMI media. CGCs were transferred 24 h before the start of experiments to phenol red-free

Microbiota and lipid mediators: from molecular crosstalk to therapeutic opportunities.

The gut microbiota constitutes a metabolically active ecosystem that dynamically interacts with the host lipid metabolism and cellular signaling networks. While dietary lipids shape the microbial composition and metabolite production, the microbiota reciprocally biotransform fatty acids, thereby regulating lipid mediator bioavailability and host cellular responses. This bidirectional molecular crosstalk plays a pivotal role in maintaining metabolic homeostasis, epithelial barrier integrity, and immune regulation. Microbiota-derived lipid signals, including short-chain fatty acids, bile acids,

Aspirin Triggered Resolvin D1 reduces inflammation and restores saliva secretion in a Sjögren's syndrome mouse model.

OBJECTIVES: SS is characterized by chronic inflammation of the salivary glands leading to loss of secretory function, thereby suggesting specialized pro-resolving mediators targeting inflammation to be a viable option for treating SS. Previous studies demonstrated that aspirin-triggered resolvin D1 (AT-RvD1) prevents chronic inflammation and enhances saliva secretion in a SS-like mouse model when applied before disease onset. However, this therapy cannot be used in SS patients given that diagnosis occurs post-disease onset and no reliable screening methods exist. Therefore, we examined whether

Evaluation of the effect of Hashimoto's Thyroiditis on fatty acids involved in inflammation in the thyroid tissue.

Hashimoto's thyroiditis (HT) is one of the most common autoimmune diseases associated with inflammation of the thyroid gland. Some fatty acids (FAs) are involved in inflammation. The aim of the study was to investigate how HT affects FA metabolism in thyroid tissue. We examined normal thyroid tissue from 92 patients with and without HT. We determined total FA panel, pro-inflammatory and specialized pro-resolving lipid mediators (SPMs) concentrations and measured the expression levels of genes encoding the corresponding enzymes of FA metabolism and mediator's synthesis. In HT tissue, we observe

Targeting inflammation and pro-resolving mediators with Anetholea anisita extract to improve scalp condition.

OBJECTIVE: A critical and often-overlooked factor that may give rise to dandruff and oily hair is the intrinsic quality of the scalp stratum corneum (SC), which is often unbalanced and susceptible to external aggressions. Addressing the inflammation element of unhealthy scalp plays an important role in promoting healthy-looking and feeling hair. Although specialized pro-resolving lipid mediators (SPMs) have been studied in the skin to end the inflammation process and promote tissue regeneration, no studies have been provided in the scalp. This study aims to investigate SPMs expression and its

Influence of physico-chemical properties of two lipoxin emulsion-loaded hydrogels on pre-polarized macrophages: a comparative analysis.

Inflammation, a crucial defense mechanism, must be rigorously regulated to prevent the onset of chronic inflammation and subsequent tissue damage. Specialized pro resolving mediators (SPMs) such as lipoxin A4 (LXA4) have demonstrated their ability to facilitate the resolution of inflammation by orchestrating a transition of M1 pro-inflammatory macrophages towards an anti-inflammatory M2 phenotype. However, the hydrophobic and chemically labile nature of LXA4 necessitates the development of a delivery system capable of preserving its integrity for clinical applications. In this study, two types

Low-dose pro-resolving mediators temporally reset the resolution response to microbial inflammation.

BACKGROUND: Specialized pro-resolving mediators (SPMs) promote resolution of inflammation, clear infections and stimulate tissue regeneration. These include resolvins, protectins, and maresins. During self-resolving acute inflammation, SPMs are produced and have key functions activating endogenous resolution response for returning to homeostasis. Herein, we addressed whether infections initiated with ongoing inflammation alter resolution programs, and if low-dose repetitive SPM regimen re-programs the resolution response. METHODS: Inflammation was initiated with zymosan (1 mg/mouse) followed

Oxidized Lipids in Persistent Pain States.

Chemotherapy, nerve injuries, or diseases like multiple sclerosis can cause pathophysiological processes of persistent and neuropathic pain. Thereby, the activation threshold of ion channels is reduced in peripheral sensory neurons to normally noxious stimuli like heat, cold, acid, or mechanical due to sensitization processes. This leads to enhanced neuronal activity, which can result in mechanical allodynia, cold allodynia, thermal hyperalgesia, spontaneous pain, and may initiate persistent and neuropathic pain. The treatment options for persistent and neuropathic pain patients are limited; f

Serum Maresin-1 and Resolvin-D1 Levels as Non-Invasive Biomarkers for Monitoring Disease Activity in Ulcerative Colitis.

Background: Specialized pro-resolving lipid mediators (SPMs), such as maresins and resolvins, play a key role in resolving inflammation and repairing tissues. This study aimed to evaluate whether maresin-1 (MaR1) and resolvin-D1 (RvD1) could serve as serum non-invasive biomarkers for monitoring disease activity in ulcerative colitis (UC). Methods: This cross-sectional study included 60 UC patients (30 active, 30 remission) and 30 healthy controls. Disease activity was assessed using the Mayo Endoscopic Subscore (MES). Inflammatory indices, including the neutrophil-lymphocyte ratio (NLR), monoc

Lipoxygenase drives lipidomic and metabolic reprogramming in ischemic heart failure.

BACKGROUND: After myocardial infarction (MI), delayed progression or reversal of cardiac remodeling is a prime target to limit advanced chronic heart failure (HF). However, the temporal kinetics of lipidomic and systemic metabolic signaling is unclear in HF. There is no consensus on metabolic and lipidomic signatures that influence structure, function, and survival in HF. Here we use genetic knock out model to delineate lipidomic, and metabolic changes to describe the role of lipoxygenase in advancing ischemic HF driven by leukocyte activation with signs of non-resolving inflammation. Lower plasma resolvin D1 level and greater sputum eosinophilia characterize a cluster of severe asthma patients.

BACKGROUND: Severe asthma is a chronic disease of airway inflammation with substantial morbidity. Deficient specialized pro-resolving mediators (SPMs) are associated with persistent airway inflammation and impaired lung function in some patients with severe asthma. Resolvin D1 (RvD1) is an SPM agonist for inflammation resolution. METHODS: Plasma RvD1 was measured longitudinally over 5 years in 23 severe asthma patients in the Severe Asthma Research Program (SARP) to identify relationships between clinical parameters, type 2 inflammation, and sputum gene expression. Periodontitis: Microbial Dysbiosis, Non-Resolving Inflammation, or Both?

The central question addressed in this review revisits the historical chicken-and-egg debate: "In periodontitis, does microbial dysbiosis drive inflammation, or does inflammation shape the subgingival microbiome?" This question is reframed through the lens of inflammation resolution. Specialized pro-resolving mediators (SPMs) provide a mechanistic framework for understanding how inflammation intersects with microbial dysbiosis. Derived from omega-3 and omega-6 fatty acids, SPMs actively promote the resolution of inflammation through binding of specific cell surface receptors rather than nonspe

Resolution of experimental malaria-associated acute respiratory distress syndrome is Alox12 independent and shows residual inflammation.

BACKGROUND: Malaria-associated acute respiratory distress syndrome (MA-ARDS) is a lung complication affecting 5-25% of adults with severe malaria and has a mortality rate of ~ 40% despite mechanical ventilation. Effective recovery requires the resolution of inflammation, an active process involving pathogen clearance, suppression of inflammation, and tissue repair. While antimalarial drugs eliminate the parasite, they do not adequately promote resolution. METHODS: Using Nanostring nCounter technology, 840 inflammation- and metabolism-related genes were profiled in lung tissue from Plasmodium

Exploring receptors for pro-resolving and non-pro-resolving mediators as therapeutic targets for sarcopenia.

Sarcopenia is defined by a reduction in both muscle strength and mass. Sarcopenia may be an inevitable component of the aging process, but it may also be accelerated by comorbidities and metabolic derangements. The underlying mechanisms contributing to these pathological changes remain poorly understood. We propose that chronic inflammation-mediated networks and metabolic defects that exacerbate muscle dysfunction are critical factors in sarcopenia and related diseases. Consequently, utilizing specialized pro-resolving mediators (SPMs) that function through specific G-protein coupled receptors

Whole-Cell Production of Resolvin E2 and 17R-Resolvin D5 by a Double-Oxygenating 15S-Lipoxygenase With 5S-Lipoxygenase Activity From Pseudobdellovibrionaceae bacterium.

Resolvin E2 (RvE2; 5S,18R-dihydroxyeicosapentaenoic acid) and 17R-resolvin D5 (17R-RvD5; 7S,17R-dihydroxydocosahexaenoic acid) are specialized pro-resolving mediators (SPMs) that actively promote the resolution of inflammation. However, their efficient biocatalytic production remains challenging because of the limited catalytic performance and poor stability of previously reported 5S-lipoxygenases (5S-LOXs). Here, we identified and characterized a novel double-oxygenating 15S-LOX from Pseudobdellovibrionaceae bacterium exhibiting 5S-LOX activity and evaluated its potential as a whole-cell bioc

Beyond cartilage degeneration: osteoarthritis as a systems failure of inflammation regulation.

Osteoarthritis (OA) is traditionally framed as a degenerative cartilage disease caused by age-related mechanical wear. However, this paradigm does not adequately account for the synovitis, metabolic disturbance, and pain that commonly precede overt structural loss. In this Hypothesis and Theory article, we propose a testable systems model in which OA reflects a failure of interdependent synovial regulatory networks rather than a purely structural endpoint. These networks include: (1) energy-redox imbalance driven by declining NAD+ availability and mitochondrial inefficiency, which may prime sy

Protectin DX resolves fracture-induced postoperative pain in mice via neuronal signaling and GPR37-activated macrophage efferocytosis.

Protectin DX (PDX) is a member of the superfamily of specialized pro-resolving mediators (SPMs) and exerts anti-inflammatory actions in animal models; but the specific signaling pathways involved in its actions have not been fully elucidated. Here we demonstrate the analgesic actions of PDX in a mouse model of tibial fracture-induced postoperative pain (fPOP) via activation of GPR37. Intravenous early-phase and late-phase treatment of PDX (100 ng/mouse) effectively alleviated fPOP. Compared to protectin D1/neuroprotectin D1, DHA, steroid, and meloxicam, PDX produced superior pain relief. While
