Localization of bacteria in the gastrointestinal tract: a possible explanation of intestinal spirochaetosis.

The large intestine of rats and dogs was examined by light and electron microscopy. In both species localization of bacteria in specific areas of the tissue was observed. Fusiform bacteria mixed with spirochaetes were seen at the opening of the crypts into the large bowel. However, at the base of the crypts homogeneous populations of spiral organisms were found; some crypts contained Borrelia-type spirochaetes, whereas others contained spiral bacteria with other morphology. Very few spiral bacteria, including those of the spirillum type, were seen in the lumen of the large intestine. Experimen

Rapid detection of Serpulina hyodysenteriae in diagnostic specimens by PCR.

A PCR assay for the detection of Serpulina hyodysenteriae in diagnostic specimens was developed on the basis of sequence analysis of a recombinant clone designated pRED3C6. Clone pRED3C6, which contained a 2.3-kb DNA fragment unique to S. hyodysenteriae, was identified by screening a plasmid library of S. hyodysenteriae isolate B204 genomic DNA in Escherichia coli by colony immunoblot with the mouse monoclonal antibody 10G6/G10, which was produced against cell-free supernatant antigens from the same isolate. Southern blot analysis of HindIII-digested genomic DNA of S. hyodysenteriae serotypes

Borrelia mayonii induces carditis but not arthritis in Lyme-susceptible mice.

BACKGROUND: Borrelia mayonii, a Borrelia burgdorferi sensu lato (Bbsl) genospecies, is a cause of Lyme disease in the upper midwestern United States. B. mayonii infection can present with some atypical clinical manifestations, including unusually high spirochetemia. Previous studies have demonstrated the infectivity of B. mayonii in laboratory mice and found evidence of widespread dissemination to internal tissues, however, did not report evidence of high spirochetemia. In the current study, we sought to directly compare spirochetemia as well as typical Lyme disease pathology including arthrit

Fibronectin-binding protein of Borrelia hermsii expressed in the blood of mice with relapsing fever.

To identify and characterize surface proteins expressed by the relapsing fever (RF) agent Borrelia hermsii in the blood of infected mice, we used a cell-free filtrate of their blood to immunize congenic naive mice. The resultant antiserum was used for Western blotting of cell lysates, and gel slices corresponding to reactive bands were subjected to liquid chromatography-tandem mass spectrometry, followed by a search of the proteome database with the peptides. One of the immunogens was identified as the BHA007 protein, which is encoded by a 174-kb linear plasmid. BHA007 had sequence features of

Infection of mice with lyme disease spirochetes constitutively producing outer surface proteins a and B.

Outer surface protein A (OspA) of the Lyme disease spirochete is primarily produced in the tick vector. OspA, which is a receptor for attaching spirochetes to the tick gut, is down regulated as the spirochetes leave the tick and enter the mammalian host. Although OspA is not a major antigen produced in the mammal, the protein appears to be produced under some conditions and production has been linked to more severe disease. A Lyme disease vaccine based on recombinant OspA has been approved for human use. However, the vaccine is no longer available, in part because of fears that OspA causes art

Motility and chemotaxis in Serpulina hyodysenteriae.

Chemotactic- or motility-regulated mucus association appears to be the predominant mechanism of mucosal association by the causative agent of swine dysentery, Serpulina hyodysenteriae. In the present study, a modification of the Adler capillary assay was used to evaluate the chemotactic responses of S. hyodysenteriae to a variety of potential stimuli. First, however, it became necessary to study factors that influenced motility of the spirochete in vitro, since standard cultivation methods produced motility inferior to that observed for in vivo grown cells. A number of factors were found to in

Relative proportions of pathogen-related oral spirochetes (PROS) and Treponema denticola in supragingival and subgingival plaque from patients with periodontitis.

The purpose of this study was to use monoclonal antibodies to enumerate spirochetes in dental plaque, including the newly recognized pathogen-related oral spirochete (PROS) and specific serovars of Treponema denticola. Plaque was collected from control subjects with no apparent periodontal disease and from sites of moderate to severe chronic periodontitis in patients with inflammatory periodontal disease. Individual monoclonal antibodies were used to determine whether spirochetes were present and then a double-staining protocol was employed to count total spirochetes and specific treponemes in

The pathogenesis of arthritis in Lyme disease: humoral immune responses and the role of intra-articular immune complexes.

We studied 78 patients with Lyme disease to determine how immune complexes and autoantibodies are related to the development of chronic Lyme arthritis. Circulating C1q binding material was found in nearly all patients at onset of erythema chronicum migrans, the skin lesion that marks the onset of infection with the causative spirochete. In patients with only subsequent arthritis this material tended to localize to joints where it gradually increased in concentrations with greater duration of joint inflammation. In joints, its concentration correlated positively with the number of synovial flui

A focus of Lyme disease in Monmouth County, New Jersey.

An endemic focus of Lyme disease is present in Colt's Neck, Howell, Freehold, and Wall Townships in Monmouth County, New Jersey. Cases of Lyme disease have occurred in this area from 1978 to 1982. Fifty-seven of the 117 persons (49%) who acquired their infection in New Jersey from 1978 to 1982 live or work in these four townships, whose population of 82,491 is only 1.1% of the population of the entire state. Thirty persons who contracted Lyme disease were exposed to ticks at the Naval Weapons Station, Earle, which is located within Colt's Neck and Howell Townships. The annual incidence rate fo

Current and future applications of species distribution and ecological niche modelling for the study of ticks and tick-borne pathogens.

Ecological niche models (ENMs) and species distribution models (SDMs) are essential tools for investigating the ecological requirements and geographic distributions of species at multiple spatial and temporal scales. While these modelling techniques have been employed across various taxonomic groups to explore ecology, evolution and biogeography, their application to ticks and tick-borne pathogens (TBPs) has yielded valuable-though not yet conclusive-perspectives for understanding epidemiology and pathogen transmission risk. Advances in research on these topics necessitate a review to determin

Prevalence of Brachyspira species isolated from diarrhoeic pigs in Brazil.

Pathogenic intestinal spirochaetes of pigs include Brachyspira (formerly Serpulina) hyodysenteriae, the cause of swine dysentery, and Brachyspira pilosicoli, the cause of porcine colonic spirochetosis (PCS). The purpose of this study was to assess the relative importance of Brachyspira species in diarrhoeal disease of growing pigs on farms in southern Brazil. The intensity and pattern of haemolysis, the production of indole and the hydrolysis of hippurate by reference and field porcine intestinal spirochaetes were compared with 16S-ribosomal RNA (mRNA)- and 23S-rRNA-based polymerase chain reac

Alzheimer's disease may not be a spirochetosis.

It has been reported previously that spirochetes could be one of the causes of Alzheimer's disease (AD). In this study, we have attempted to reproduce these findings by examining fresh blood samples from 22 patients diagnosed with early stage (n = 16) and late stage (n = 6) AD. The patients were participants in a clinical drug trial. Fresh necropsy brain cortical specimens from AD patients (n = 7) were also examined. Spirochetes were observed microscopically in the blood of only one late-stage AD patient. None of the brain tissues showed the presence of spirochetes. Our results suggest that sp

Reproducibility of microscopic and cultural data in repeated subgingival plaque samples.

In association studies, micro-organisms can only be recognized as suspects for playing a major rôle in the development of a pathological environment, if their destructive action goes along with a marked proportional increase of their numbers or if their first detection can be related to the clinical onset of the disease. Limitations in the reproducibility of repeated samples have to be taken into account, when changes of the microbial composition of subgingival environments are to be studied, and when local clinical changes are to be related to shifts in the composition of the pertaining micro

The antibody response in Lyme disease.

We determined the antibody response against the Ixodes dammini spirochete in Lyme disease patients by indirect immunofluorescence and an enzyme-linked immunosorbent assay (ELISA). The specific IgM response became maximal three to six weeks after disease onset, and then declined, although titers sometimes remained elevated during later disease. Specific IgM levels correlated directly with total serum IgM. The specific IgG response, often delayed initially, was nearly always present during neuritis and arthritis, and frequently remained elevated after months of remission. Although results obtain

Variation in a major surface protein of Lyme disease spirochetes.

Two monoclonal antibodies (H6831 and H5TS) differed in their indirect immunofluorescence reactivity when tested against 14 strains of Lyme disease spirochetes. Strains were bound by both antibodies, by H6831 or H5TS alone, or by neither. Western blot and immunoprecipitation studies revealed that the determinants of both antibodies were associated with abundant proteins with apparent subunit molecular weights of ca. 34,000 (34K-range proteins). The following results indicated that the 34K-range proteins were exposed on the surface of the spirochetes. (i) Antibody H6831 agglutinated the spiroche
