Shank3 deficiency elicits autistic-like behaviors by activating p38α in hypothalamic AgRP neurons.

BACKGROUND: SH3 and multiple ankyrin repeat domains protein 3 (SHANK3) monogenic mutations or deficiency leads to excessive stereotypic behavior and impaired sociability, which frequently occur in autism cases. To date, the underlying mechanisms by which Shank3 mutation or deletion causes autism and the part of the brain in which Shank3 mutation leads to the autistic phenotypes are understudied. The hypothalamus is associated with stereotypic behavior and sociability. p38α, a mediator of inflammatory responses in the brain, has been postulated as a potential gene for certain cases of autism oc

Relationships Between Emotion Regulation, Social Communication and Repetitive Behaviors in Autism Spectrum Disorder.

The relationship between emotion regulation, social interaction and different types of restricted and repetitive behaviors is poorly understood. In the present study, structural equation modeling based on information about 239 individuals with autism was used to examine whether emotion regulation and social communication were associated with self-injury and stereotyped behaviors. Results showed that poor emotion regulation had a unique association with self-injury while difficulties with social communication was uniquely associated with stereotyped behaviors. Emotion regulation and social comm

Effects of monosialoganglioside on dopaminergic supersensitivity.

The effects of monosialoganglioside (GMl) treatment on dopaminergic supersensitivity induced by long-term haloperidol administration were studied; both general activity of rats observed in an open-field and apomorphine-induced stereotyped behavior were used as experimental parameters. GMl per se (5.0 mg/kg, twice daily, for 30 days) did not modify rat behavior, but when given in combination with haloperidol (1.0 mg/kg, twice daily, for 30 days) it increased neuroleptic withdrawal symptoms as detected in both models. When GMl (5.0 mg/kg, twice daily) was administered after abrupt withdrawal fro

The role of task preference in the effectiveness of response interruption and redirection.

Response interruption and redirection (RIRD) is a common treatment for automatically reinforced vocal stereotypy; it involves the contingent presentation of task instructions. Tasks that are included in RIRD are typically selected based on caregiver report, which may affect the efficacy of RIRD. The purpose of the current study was to evaluate the role of task preference in the efficacy of RIRD for four participants who engaged in vocal stereotypy. We conducted task-preference assessments and selected tasks of varying preferences to include in RIRD. For three out of four participants, the resu

Functional Analysis Outcomes and Comparison of Direct Observations and Informant Rating Scales in the Assessment of Severe Behavior Problems of Infants and Toddlers At-Risk for Developmental Delays.

Severe problem behaviors, like aggression, self-injury, and repetitive behaviors, in people with intellectual and developmental disabilities often appear during early development and may persist without early intervention. The frequencies of self-injurious behavior, aggression, tantrums, property destruction and stereotyped behavior among 17 infants and toddlers at risk for developmental delays and severe behavior problems were assessed using two methods: 1) direct observation of responses during 10 s partial interval recording during analogue functional analysis and 2) the Behavior Problem In

Behavioral stereotypies induced by "binge' cocaine administration are independent of drug-induced increases in corticosterone levels.

Cocaine administration causes dramatic stereotypic behavior and elevation of circulating corticosterone levels in rodents. The present study tested the possible role of increased corticosterone in mediating stereotypic behavior caused by "binge' pattern cocaine administration. Animals were administered saline or cocaine intraperitoneally for 3 days, with or without pretreatment with a D1 (SCH 23390, 2 mg/kg) or D2 (sulpiride, 50 mg/kg) dopamine receptor antagonist. Three days of cocaine "binges' significantly increased corticosterone levels in vehicle pretreated rats (P < 0.01). Both SCH 23390

Genetic mapping of fixed phenotypes: disease frequency as a breed characteristic.

Traits that have been stringently selected to conform to specific criteria in a closed population are phenotypic stereotypes. In dogs, Canis familiaris, such stereotypes have been produced by breeding for conformation, performance (behaviors), etc. We measured phenotypes on a representative sample to establish breed stereotypes. DNA samples from 147 dog breeds were used to characterize single nucleotide polymorphism allele frequencies for association mapping of breed stereotypes. We identified significant size loci (quantitative trait loci [QTLs]), implicating candidate genes appropriate to re

Reciprocal information flow between prefrontal cortex and ventral tegmental area in an animal model of schizophrenia.

The medial prefrontal cortex (PFC) is anatomically and functionally connected with the ventral tegmental area (VTA), the neuronal source of mesocorticolimbic system that is pathophysiologically related to schizophrenia-like symptoms. Methamphetamine (MAP) was applied to examine the functional relationship between PFC and VTA in an animal model of schizophrenia. Hyperactivity and stereotyped behavior were observed accompanied by a distinctive direction of information flow. In hyperactivity, information flow in the direction from PFC to VTA was dominant. Contrarily, dominant information flow fro

Enhancement of dopamine release from the striatum through metabotropic glutamate receptor activation in methamphetamine sensitized rats.

An intracerebral microdialysis technique was applied to study the effect of metabotropic glutamate receptor (mGluR) agonist on dopamine release in the striatum of methamphetamine (MAP)-sensitized rats. Rats were treated with MAP (1 mg/kg, i.p.) once daily for 6 consecutive days, followed by a 6-day withdrawal. Perfusion of 0.1 mM (1S,3R)-1-aminocyclopentane-trans-1,3-dicarboxylic acid through a microdialysis probe placed in the striatum enhanced the extracellular dopamine level, and induced stereotyped behavior in MAP-sensitized rats. The enhancement of dopamine release and the stereotyped beh

Methamphetamine-induced behavioral alterations following repeated administration of methamphetamine.

Repeated administration of a large dose of methamphetamine (MA) (25 mg/kg, i.p. twice daily for 4 days) to mice enhanced locomotor activity and decreased stereotyped behavior following a subsequent injection of MA. Simultaneous determinations of catecholamines revealed a depletion of brain dopamine. The moderate doses of haloperidol significantly enhanced MA-induced locomotor activity in mice. A significant enhancement of MA-induced locomotor activity was observed in the rats pretreated with 6-hydroxydopamine into the striatum, and this effect correlated negatively with the striatal dopamine l

Stereotypes and tacit inference.

To judge another person's behavior, one often has to come to an understanding of what that behavior was in its detail. Five studies demonstrated that stereotypes influence the tacit inferences people make about the unspecified details and ambiguities of social behavior (e.g., what the behavior specifically was, what stimulus the individual reacted to, what caused the individual to act) and that these inferences occur when people encode the relevant information. One study found that participants who scored low on a measure of modern sexism were just as likely to make tacit inferences based on g

The effects of husbandry training on stereotypic pacing in captive African wild dogs (Lycaon pictus).

To examine the effects of operant conditioning on stereotypic pacing in 3 female African wild dogs located at the Franklin Park Zoo in Boston, this study made recordings of pacing behavior immediately following individual sessions of husbandry training and 2 no-training conditions. The study found significant differences in the percentage of observations spent in stereotypic pacing behaviors for all 3 dogs among the 3 different conditions. The authors discuss the data in terms of the contribution of motivated tasks to the effects and the role of food deprivation in the expression of stereotypi

Influence of the dose of ketamine used on schizophrenia-like symptoms in mice: A correlation study with TH, GAD67, and PPAR-γ.

Schizophrenia is a chronic, debilitating mental illness that has not yet been completely understood. In this study, we aimed to investigate the effects of different doses of ketamine, a non-competitive NMDA receptor antagonist, on the positive- and negative-like symptoms of schizophrenia. We also explored whether these effects are related to changes in the immunoreactivity of GAD67, TH, and PPAR-γ in brain structures. To conduct the study, male mice received ketamine (20-40 mg/kg) or its vehicle (0.9 % NaCl) intraperitoneally for 14 consecutive days. We quantified stereotyped behavior, the tim

Parallel strain-dependent susceptibility to environmentally-induced stereotypies and stress-induced behavioral sensitization in mice.

Food restriction (9 days) promoted stereotyped behavior in drug-free mice of the DBA/2 (DBA), but not in those of the C57BL/6 (C57), inbred strain. Indeed, behavior presented by food-restricted mice of the DBA strain within the home cage was characterised by a very high response rate within a single response: cage cover climbing. Moreover, enhanced climbing in food-restricted mice of the DBA strain was also observed in a test designed to detect stereotypic effects of drugs in mice. Stereotypic behavior in DBA mice did not depend on nutritional status because: 1. No stereotypies were observed i

[Effects of low frequency repetitive transcranial magnetic stimulation on Electroencephalograph rhythm of children with autism].

Autism spectrum disorders (ASD) is a complex developmental disorder characterized by impairments in social communication and stereotyped behaviors. Electroencephalograph (EEG), which can measure neurological changes associated with cortical synaptic activity, has been proven to be a powerful tool for detecting neurological disorders. The main goal of this study is to explore the effects of repetitive transcranial magnetic stimulation (rTMS) on behavioral response and EEG. We enrolled 32 autistic children, rTMS group ( n = 16) and control group ( n = 16) and calculated the relative power of the

Stereotypy in young children with autism and typically developing children.

Although stereotypy is one of the key diagnostic features of autism, few studies have compared stereotypic behavior in children with autism and typically developing children. The present study employed direct observational measurement methods to assess levels of stereotypic behavior in 2-, 3- and 4-year-old children with autism or pervasive developmental disorder - not otherwise specified (PDD-NOS) and age-matched typically developing peers. Thirty children with autism or PDD-NOS and 30 typically developing children participated. Each child's performance of several early learning and play skil

Effect of ketamine administration, alone and in combination with E-6837, on climbing behavior.

Some types of schizophrenia have been associated with repetitive movements lacking specific purpose, also known as stereotyped behavior. Dopamine agonists (D2) and noncompetitive N-methyl-D-aspartate receptor antagonists (e.g. ketamine) have been administered in rodent models to induce stereotyped behavior that resembles some motor symptoms of schizophrenia. Recently, a relationship has been found between 5-HT6 receptors (5-HT6Rs) and dopaminergic activity. The present study evaluates the effect of ketamine (5 and 10 mg/kg), alone and in combination with the 5-HT6R agonist E-6837, on the climb

The Effects of Occupational Status and Sex-Typed Jobs on the Evaluation of Men and Women.

Background: Occupational segregation by gender is one of the major problems faced by professional women in the labor market. Since the sixties, psychological explanations point out that gender stereotypes are responsible for this persistent inequality in the workforce. Nevertheless, most of research has overlooked that emotions are particularly important as the discrimination faced by professional women is better explained by the ambivalent feelings they provoke than by stereotyping. Aim: The aim of this research is to analyse from the Stereotype Content Model (SCM, Fiske et al., 2002) and the

Male isolation: a behavioral representation of the pheromonal 'female effect' in donkey (Equus asinus).

The appearance of a decisive component of the sexual response to chemosexual signals in the male donkey was investigated through a comparison of the variations in the time-span of the behavioral classes and units for the natural versus induced breeding seasons. The results demonstrate that there are significant variations in the length of the appetitive sexual behavior (ASB) and consummatory sexual behavior (CSB) under these two reproductive conditions. These differences are analyzed for the ASB, which is adaptable, compared with the stereotyped CSB. For the ASB, male isolation is the most rep
