Strengthening of synchronized activity by tetanic stimulation in cortical cultures: application of planar electrode arrays.

Rat cortical neurons were cultured on planar electrode arrays with 64 embedded electrodes. Whole-cell recording from single neurons and multisite extracellular recording were carried out simultaneously in the cultured cortical networks, and the effects of focal tetanic stimulation of the culture were studied. Both the number of action potentials and the propagation velocity of stimulated bursts were increased after tetanic stimulation. These changes were associated with a marked increase in the number of late components in the synaptic current, but with little or no increase in the early peak

Long-term depression of C-fibre-evoked spinal field potentials by stimulation of primary afferent A delta-fibres in the adult rat.

Long-term potentiation (LTP) of spinal C-fibre-evoked field potentials can be induced by brief electrical stimulation of afferent C-fibres, by natural noxious stimulation of skin or by acute nerve injury. Here, we report that in urethane anaesthetized, adult rats prolonged high frequency burst stimulation of the sciatic nerve at Adelta-fibre strength produced long-term depression (LTD) of C-fibre-evoked field potentials, and also depressed the increased amplitudes of C-fibre-evoked field potentials recorded after LTP had been established (depotentiation). Electrical stimulation of Abeta-fibres

Voltage dependent allosteric modulation of IPSCs by benzodiazepines.

GABAA receptors (GABAAR) are inhibitory ion channels ubiquitously expressed in the central nervous system and play critical roles in brain development and function. Benzodiazepines are positive allosteric modulators of GABAAR, enhancing channel opening frequency when GABA is bound to the receptor. Midazolam is a commonly used benzodiazepine. It is frequently used for premature infants, but the long-term consequences of its use in this patient population are not well established. Here, we studied the acute effects of midazolam on immature synapses. Using a rodent organotypic hippocampal slice p

N-type Ca2+ channels mediate transmitter release at the electromotoneuron-electrocyte synapses of the weakly electric fish Gymnotus carapo.

The effects of omega-conotoxin-GVIA (omega-CgTX) on synaptic transmission were studied in the electromotoneuron-electrocyte synapses of the electric organ (EO) of the weakly electric fish Gymnotus carapo. omega-CgTX selectively and irreversibly blocked excitatory postsynaptic potentials (EPSPs) in a dose dependent-manner. The toxin had no effect on: (a) resting postsynaptic membrane potential and conductance; (b) postsynaptic action potentials elicited by depolarizing transmembrane current pulses; (c) the action potential conduction in the presynaptic fiber; (d) acetylcholine (ACh)-induced pos

Synaptic transmission in submucosal ganglia of guinea pig distal colon.

Intracellular electrical recording was used to investigate synaptic behavior of ganglion cells in the neural networks of the submucosal plexus of the guinea pig distal colon. Fast excitatory postsynaptic potentials (EPSPs), mediated by nicotinic receptors, were found in all S/type 1 neurons, 70% of AH/type 2, 75% of type 3, and 95% of type 4 neurons. Slow EPSPs were characterized by membrane depolarization, increased input resistance, enhanced action potential discharge, and suppression of hyperpolarizing afterpotentials in 64% of the S/type 1 neurons, 74% of AH/type 2, 31% of type 3, and 70%

Electrophysiological properties of the isolated vestibulocerebellar complex of the frog in vitro.

Parameters of the electrical activity of the isolated vestibulocerebellar complex of the frog were studied under in vitro conditions. In the region of the vestibular nucleus (nc. VIII), in the presence of stimulation of the stato-acoustic nerve (n. VIII), responses from efferent vestibular neurones and from unidentified (probably vestibulospinal) neurones were recorded. The latent periods of their excitatory postsynaptic potentials (EPSPs, 1.4-2.2 ms) were indicative of mono- and disynaptic connection. Inhibitory postsynaptic potentials (IPSPs) were also observed. Stimulation of the auricular

Functional anatomy of the inferior colliculus and the auditory cortex: current source density analyses of click-evoked potentials.

In the auditory midbrain (inferior colliculus) and cortex (superior temporal gyrus) of awake squirrel monkeys profiles of click-evoked field potentials were recorded. The recording tracks were reconstructed anatomically. From the field potentials the one-dimensional current source density (CSD) distributions were calculated. By comparing the CSD profiles with the anatomical features of the reconstructed recording paths, the components of the CSDs could be attributed to certain anatomical sites. Thus a physiological method for the functional identification of recording sites was obtained. It pe

Differential bicuculline-induced epileptogenesis in rat neonatal, juvenile and adult CA3 pyramidal neurons in vitro.

The GABA(A) receptor antagonist bicuculline methiodide (BMI, 10 microM) transformed the evoked synaptic responses, recorded intracellularly from the CA3 area of neonatal (postnatal days 3-7, P3-P7), juvenile (P8-P20) and adult hippocampal slices, into long-lasting paroxysmal depolarizations (PDs), with repetitive action potentials (APs). In the same preparation, GABA(A)-mediated fast-IPSPs were depolarizing at resting membrane potential (RMP), with a reversal potential shifting to a hyperpolarizing direction with age (n=15, P6-P17). BMI provoked also spontaneous PDs in juvenile (20/30) and adu

Reorganization of synchronous and asynchronous release in developing hippocampal autaptic neurons of mice.

Presynaptic vesicle pools, particularly the readily releasable pool (RRP), are essential for synaptic transmission. The RRP comprises fast-release pool (FRP) and slow-release pool (SRP) subpopulations that mediate synchronous and asynchronous neurotransmitter release, respectively, yet their developmental regulation remains unclear. To investigate this question, we utilized a hippocampal autaptic culture system to analyze synaptic function in individual excitatory neurons at 1, 2, and 3 weeks in vitro. We observed significant enhancements in both synaptic transmission efficiency and the size o

Effects of NH4+ on reflexes in cat spinal cord.

1. In deeply barbiturate-anesthetized animals. NH4+ decreases spinal excitatory synaptic transmission by neuronal depolarization and subsequent block of conduction of action potentials into presynaptic terminals of low-threshold (presumably Ia-) afferents. Because barbiturates by themselves depress excitatory synaptic transmission and may have modified the effects of NH4+, this study examines the effect of NH4+ on excitatory synaptic transmission in the unanesthetized animal. 2. The effects of NH4+ on monosynaptic and polysynaptic excitatory reflexes as well as di- and polysynaptic inhibition

Chloride-cotransport blockade desynchronizes neuronal discharge in the "epileptic" hippocampal slice.

Antagonism of the chloride-cotransport system in hippocampal slices has been shown to block spontaneous epileptiform (i.e., hypersynchronized) discharges without diminishing excitatory synaptic transmission. Here we test the hypotheses that chloride-cotransport blockade, with furosemide or low-chloride (low-[Cl(-)](o)) medium, desynchronizes the firing activity of neuronal populations and that this desynchronization is mediated through nonsynaptic mechanisms. Spontaneous epileptiform discharges were recorded from the CA1 and CA3 cell body layers of hippocampal slices. Treatment with low-[Cl(-)

Omega-3 fatty acids can reverse the long-term deficits in hippocampal synaptic plasticity caused by prenatal ethanol exposure.

Fetal alcohol spectrum disorders result in long-lasting neurological deficits including decreases in synaptic plasticity and deficits in learning and memory. In this study we examined the effects of prenatal ethanol exposure on hippocampal synaptic plasticity in male and female Sprague-Dawley rats. Furthermore, we looked at the capacity for postnatal dietary intervention to rescue deficits in synaptic plasticity. Animals were fed an omega-3 enriched diet from birth until adulthood (PND55-70) and in vivo electrophysiology was performed by stimulating the medial perforant path input to the denta

A comparison of the presynaptic and post-synaptic actions of pentobarbitone and phenobarbitone in the neuromuscular junction of the frog.

1. Pentobarbitone or phenobarbitone, in increasing concentrations up to 0-5 mM, progressively reduced the amplitude of miniature end-plate potentials (min.e.p.p.s). Pentobarbitone was the more potent of the two barbiturates in this regard. 2. Both barbiturates produced a monotonic increase in mean quantum content of the end-plate potential (e.p.p.) with increasing concentrations up to 0-5 mM. Pentobarbitone and phenobarbitone were equally potent in their action on evoked transmitter release. 3. The effect, if any, of increasing concentrations of barbiturates on the e.p.p. amplitude was depress

[The effects of the pathogenic influence of botulin toxin on different types of spinal motor neurons].

The motor neuron cells of the lumbosacral region were investigated in the spinal cord of cat with the local botulin paralysis. Development of this paralysis was followed by reduction of the membrane potential, of the amplitude of antidromic AP, mono- and polysynaptic EPSP, a fall of the input resistance and by an increase in the level of the critical depolarization of the somatic membrane of the phasic motor neurons of the damaged segments in the spinal cord. Excitation of the tonic motor neurons was not greatly altered in the dynamics of local botulism.

Computation of frequency-to-spatial transform by olfactory bulb glomeruli.

A physiological simulation of 2.5% of the input and inhibitory neurons and 25% of the primary mitral/tufted cells in a single mammalian olfactory bulb glomerulus was constructed. This physiological simulation used the integrate-and-fire paradigm with realistic activation curves and synaptic delays. The dendritic integration incorporated non-linear interactive effects of individual cell excitatory and inhibitory post-synaptic potentials (PSPs) from both axodendritic and dendro-dendritic synaptic contacts. Refractory periods for granule-cell inhibition of mitral/tufted cell activity lead to rela

Changes in spinal inhibitory networks induced by furosemide in humans.

During neural development in animals, GABAergic and glycinergic neurons are first excitatory, and then become inhibitory in the mature state. This developmental shift is due mainly to strong expression of the cation-chloride K-Cl cotransporter 2 (KCC2) and down-regulation of Na-K-Cl cotransporter 1 (NKCC1) during maturation. The down-regulation of co-transporter KCC2 after spinal cord transection in animals leads to the depolarising (excitatory) action of GABA and glycine and thus results in a reduction of inhibitory synaptic efficiency. Furosemide, a loop diuretic, has been shown to selective

Effects of L-DOPA on nigral dopamine neurons and local field potential: comparison with apomorphine and muscimol.

L-DOPA is more effective than direct dopamine (DA) agonists in relieving the motor deficits in Parkinson's disease. Using in vivo recording, we compared the effect of l-DOPA and the direct DA agonist apomorphine on DA neurons in rat substantia nigra (SN). L-DOPA (50-100 mg/kg i.v.) decreased the firing rate as well as the variability and slow oscillation (SO) of firing. All effects were blocked by raclopride and mimicked by quinpirole, suggesting that they are mediated through D2-like receptors. Autoreceptor-selective doses of apomorphine (5-20 μg/kg i.v.) also inhibited all three parameters.

Long-term potentiation of silent synapses in substantia gelatinosa neurons.

In this study, we observed quantal changes in single synapse excitatory postsynaptic currents to characterize N-methyl-D-aspartate receptor-mediated silent synapses between primary afferents and spinal substantia gelatinosa neurons. The failure rate of primary afferent quantal excitatory postsynaptic currents was lower at depolarized holding potentials than at hyperpolarized potentials. This lower failure rate at depolarized potentials was due to the activation of N-methyl-D-aspartate receptor-mediated excitatory postsynaptic currents. Pairing primary afferents with the postsynaptic depolariza

At immature mossy-fiber-CA3 synapses, correlated presynaptic and postsynaptic activity persistently enhances GABA release and network excitability via BDNF and cAMP-dependent PKA.

In the adult rat hippocampus, the axons of granule cells in the dentate gyrus, the mossy fibers (MF), form excitatory glutamatergic synapses with CA3 principal cells. In neonates, MF release into their targets mainly GABA, which at this developmental stage is depolarizing. Here we tested the hypothesis that, at immature MF-CA3 synapses, correlated presynaptic [single fiber-evoked GABA(A)-mediated postsynaptic potentials (GPSPs)] and postsynaptic activity (back propagating action potentials) may exert a critical control on synaptic efficacy. This form of plasticity, called spike-timing-dependen
