• Cysteine is the metabolic precursor of essential biomolecules such as vitamins, cofactors, antioxidants and many defense compounds. The last step of cysteine metabolism is catalysed by O-acetylserine(thiol)lyase (OASTL), which incorporates reduced sulfur into O-acetylserine to produce cysteine. In Arabidopsis thaliana, the main OASTL isoform OAS-A1 and the cytosolic desulfhydrase DES1, which degrades cysteine, contribute to the cytosolic cysteine homeostasis. • Meta-analysis of the transcriptomes of knockout plants for OAS-A1 and for DES1 show a high correlation with the biotic stress series

The Arabidopsis enhanced disease susceptibility 4 (eds4) mutation causes enhanced susceptibility to infection by the bacterial pathogen Pseudomonas syringae pv. maculicola ES4326 (Psm ES4326). Gene-for-gene resistance to bacteria carrying the avirulence gene avrRpt2 is not significantly affected by eds4. Plants homozygous for eds4 exhibit reduced expression of the pathogenesis-related gene PR-1 after infection by Psm ES4326, weakened responses to treatment with the signal molecule salicylic acid (SA), impairment of the systemic acquired resistance response, and reduced accumulation of SA after

Volatile organic compounds (VOCs) emitted by certain fungi have previously been suggested to modulate plant development and their response to stresses. While some fungal species, such as Trichoderma harzianum, are known to produce VOCs, the production of VOCs in other phylogenetically divergent species, such as Mucor mucedo, was previously unknown. This study aimed to analyse the VOCs profiles emitted by these fungal species and examine their effects on the growth and defensive responses of Arabidopsis thaliana. Volatilome analysis revealed that the most abundant VOCs for both species were alc

When plants encounter biotic and abiotic stresses, they emit various volatile organic compounds to communicate with nearby plants and activate airborne defense (AD). One critical compound in this process is methyl salicylate (MeSA). Previous studies have mostly examined how stress triggers the production of MeSA at the gene level. In our study, we found that MeSA plays a key role in AD during bacterial infections and determined how plants boost MeSA production through a protein complex. Infection by Pseudomonas syringae pv. tomato DC3000 in Arabidopsis thaliana increased salicylic acid levels,

Plant responses to pathogens comprise a complex process, implying a plethora of signals and reactions. Among them, endogenous production of hydrogen cyanide (HCN) has been shown to induce resistance in Arabidopsis to the hemibiotrophic bacterium Pseudomonas syringae pv. tomato (Pst) DC3000. β-cyanoalanine synthase (CAS-C1) is responsible for the detoxification of HCN in Arabidopsis mitochondria. Here, we show that green fluorescent protein-tagged CAS-C1 is transiently reduced in leaves infected with an avirulent strain of Pst during early interactions and increased in leaves infected with a vi

Efficient communication between the pest-colonised and non-colonised organs is vital for the timely manifestation of defences that restrict systemic spread of pests. The vasculature provides an important conduit for translocation of signals that contribute to long-distance communication within a plant. Airborne signals also contribute to this process. In recent years, major advances have been made in identifying metabolites that are candidate systemic signals in plant defence against pathogens. Methyl salicylate, jasmonates, azelaic acid and a diterpenoid have been implicated as mobile signals

Papain-like cysteine proteases (PLCPs) play crucial roles in plant-pathogen/pest interactions. During these parasitic interactions, PLCPs act on non-self substrates, provoking the selection of counteracting inhibitors and other means to evade proteolysis. We review examples of PLCPs acting on molecular battlefields in the extracellular space, plant cytoplasm and herbivore gut. Examples are maize Mir1 (Maize inbred resistance 1), tomato Rcr3 (Required for Cladosporium resistance-3), Pseudomonas AvrRpt2 and AurPphB, insect DvCAL1 (Diabrotica virgifera cathepsin L-like protease-1) and nematode Mi

Plant disease resistance is a complex process that is maintained in an intricate balance with development. Increasing evidence indicates the importance of posttranscriptional regulation of plant defense by RNA binding proteins. In a genetic screen for suppressors of Arabidopsis (Arabidopsis thaliana) accelerated cell death 6-1 (acd6-1), a small constitutive defense mutant whose defense level is grossly in a reverse proportion to plant size, we identified an allele of the canonical flowering regulatory gene FLOWERING LOCUS K HOMOLOGY DOMAIN (FLK) encoding a putative protein with triple K homolo

Iron (Fe) is a poorly available mineral nutrient which affects the outcome of many cross-kingdom interactions. In Arabidopsis thaliana, Fe starvation limits infection by necrotrophic pathogens. Here, we report that Fe deficiency also reduces disease caused by the hemi-biotrophic bacterium Pseudomonas syringae and the biotrophic oomycete Hyaloperonospora arabidopsidis, indicating that Fe deficiency-induced resistance is effective against pathogens with different lifestyles. Furthermore, we show that Fe deficiency-induced resistance is not caused by withholding Fe from the pathogen but is a plan

Phytopathogenic bacteria affect a wide variety of crops, causing significant economic losses. Natural biocides are the alternative to chemical methods of phytopathogens control. The goal of the present study is the evaluation of the biocidal activity of the following: 1) the extract of orange wax (EOW); 2) zinc ferrite nanoparticles (ZF-NPs); 3) the EOW adsorbed on the ZF-NPs; and 4) the EOW/ZF-NPs washed with 40% ethanol. For the biocidal activity, three phytopathogenic bacteria were used, namely, Xanthomonas axonopodis pv. Vesicatoria (Xav) Erwinia amylovora (Ew), and Pseudomonas syringae pv

During their lifetime, plants encounter numerous biotic and abiotic stresses with diverse modes of attack. Phytohormones, including salicylic acid (SA), ethylene (ET), jasmonate (JA), abscisic acid (ABA), auxin (AUX), brassinosteroid (BR), gibberellic acid (GA), cytokinin (CK) and the recently identified strigolactones (SLs), orchestrate effective defense responses by activating defense gene expression. Genetic analysis of the model plant Arabidopsis thaliana has advanced our understanding of the function of these hormones. The SA- and ET/JA-mediated signaling pathways were thought to be the b

To explore the toxicity and action mechanism of acute sulfur dioxide (SO2) on urban landscape plants, a simulated SO2 stress environment by using fumigation chamber involving increasing SO2 concentration (0, 25, 50, 100, 200 mg m-3) was carried out among three species. After 72 h of exposure, SO2-induced oxidative damage indicated by electrolyte leakage increased with higher dose of SO2. Meanwhile, SO2 decreased the contents of chlorophyll a, chlorophyll b and carotenoid and increased the contents of sulfur. Net photosynthetic rate (Pn) decreased as a result of stomatal closure when SO2 dose w

Plant-defense responses are triggered by perception of conserved microbe-associated molecular patterns (MAMPs), for example, flagellin or peptidoglycan. However, it remained unknown whether plants can detect conserved molecular patterns derived from plant-parasitic animals, including nematodes. Here we show that several genera of plant-parasitic nematodes produce small molecules called ascarosides, an evolutionarily conserved family of nematode pheromones. Picomolar to micromolar concentrations of ascr#18, the major ascaroside in plant-parasitic nematodes, induce hallmark defense responses inc

Small molecules can control plant disease either by disarming the pathogen or by priming host immunity, but single compounds that do both are rare and mechanistically unexplained. Here we show that hopeaphenol (HP), a resveratrol tetramer from stilbene-producing plants, acts on histidine kinase (HK) signaling on both sides of the Arabidopsis- Pseudomonas interaction. In Pseudomonas syringae pv. tomato DC3000, HP represses the type III secretion regulon and motility genes, restricts surface motility independently of effector delivery, and binds a defined subset of virulence-associated sensor ki

A series of novel trifluoromethylpyrimidinamine derivatives containing an amide moiety were designed and synthesized. The antimicrobial activity of the title compounds against various plant-pathogenic fungi and bacteria was evaluated. Among them, the EC50 of compound 7i against Pseudomonas syringae pv actinidiae (Psa) was 11.38 μg/mL, which was superior to that of the positive control thiodiazole copper (TC, 85.69 μg/mL), indicating excellent inhibitory activity. In vivo anti-Psa activity assays revealed that at 200 μg/mL, compound 7i conferred protective and curative effects of 69.10% and 65.

The systematic chemical investigation of the ethyl acetate extract of the rice-fermented fungus Aspergillus flavus J302-03 led to the isolation of six previously unreported compounds, including four polyketides, aspergullains A-D, and two drimane-type sesquiterpenoids, aspergullains E and F, along with sixteen known compounds. Their structures were characterized through spectroscopic data interpretation, ECD and NMR calculations, DP4+ analysis, and X-ray crystallography. Structurally, aspergullain A was a unique polyketide with a cyclohexanedione-furan hybrid skeleton, while aspergullain B pos

Chloroplast function is essential for growth, development, and plant adaptation to stress. Organelle stress and plant defence responses were examined here using noxy8 (nonresponding to oxylipins 8) from a series of Arabidopsis mutants. The noxy8 mutation was located at the CLPC2 gene, encoding a chloroplast chaperone of the protease complex CLP. Although its CLPC1 paralogue is considered to generate redundancy, our data reveal significant differences distinguishing CLPC2 and CLPC1 functions. As such, clpc1 mutants displayed a major defect in housekeeping chloroplast proteostasis, leading to a

Systemic acquired resistance is a widespread phenomenon in the plant kingdom that confers heightened and often enduring immunity to a range of diverse pathogens. Systemic immunity develops through activation of plant disease resistance protein signaling networks following local infection with an incompatible pathogen. The accumulation of the phytohormone salicylic acid in systemically responding tissues occurs within days after a local immunizing infection and is essential for systemic resistance. However, our knowledge of the signaling components underpinning signal perception and the establi

Type III secretion systems enable plant and animal bacterial pathogens to deliver virulence proteins into the cytosol of eukaryotic host cells, causing a broad spectrum of diseases including bacteremia, septicemia, typhoid fever, and bubonic plague in mammals, and localized lesions, systemic wilting, and blights in plants. In addition, type III secretion systems are also required for biogenesis of the bacterial flagellum. The HrcQ(B) protein, a component of the secretion apparatus of Pseudomonas syringae with homologues in all type III systems, has a variable N-terminal and a conserved C-termi
