Urinary tract infections (UTIs) are common bacterial infections. The global emergence of multidrug-resistant uropathogens in the last decade underlines the need to search for new antibiotics with novel mechanisms of action. In this regard, exploring endophytic fungi inhabiting medicinal plants used locally against urinary tract infections could be a promising strategy for novel drug discovery. This study investigates crude metabolites from endophytic fungi isolated from Annona muricata as potential sources of antibiotic drugs to fight against uropathogens and reduce related oxidative stress. INTRODUCTION: Penicillium marneffei is one of the most common opportunistic infectious fungi associated with AIDS (Acquired Immunodeficiency Syndrome). It is prevalent in Southeast Asia and southern China (such as Guangdong), but rare in inland provinces of China. Penicillium marneffei infections are often misdiagnosed.
CASE REPORT: Here, we report a patient with Penicillium marneffei infection from Shaanxi Province, China, who was previously repeatedly diagnosed as Kala-azar in two hospitals. The patient received medical treatment due to fever and multiple papules, worked as a tour guide, an

BACKGROUND: Talaromyces marneffei (T. marneffei) is a thermal dimorphic pathogenic fungus that often causes fatal opportunistic infections in human immunodeficiency virus (HIV)-infected patients. Although T. marneffei-infected cases have been increasingly reported among non-HIV-infected patients in recent years, no cases of T. marneffei infection have been reported in pulmonary sarcoidosis patients. In this case, we describe a T. marneffei infection in an HIV-negative patient diagnosed with pulmonary sarcoidosis.

An 80-year-old woman from rural Guangxi with post-tuberculosis lung disease (PTLD) (hereinafter referred to as PTLD)presented with one month of cough and fever. One month prior, she had ingested raw rodent meat-a known exposure for Talaromyces marneffei. Chest HRCT showed bilateral tree-in-bud opacities superimposed on prior left lung destruction. Conventional microbiological tests, including acid-fast bacilli smears, were negative. A nasopharyngeal swab was positive for SARS-CoV-2 (cycle threshold 17). Metagenomic next-generation sequencing (mNGS) of bronchoalveolar lavage fluid identified bo

BACKGROUND Talaromyces marneffei (TM) infection is an opportunistic infection that typically affects individuals with compromised immune systems. TM is an invasive fungal infection associated with high mortality in patients with advanced human immunodeficiency virus (HIV) disease. Syphilis, caused by Treponema pallidum, is a re-emerging infectious disease with diverse clinical manifestations. While primary syphilis often goes unnoticed, secondary syphilis is characterized by systemic symptoms and variable skin lesions, earning it the epithet "the great imitator." In areas where TM is non-endem

Adult-onset immunodeficiency associated with anti-interferon-γ (IFN-γ) autoantibodies (AIGA) is a rare immune disorder that neutralizes IFN-γ activity and compromises defense against intracellular pathogens. It is predominantly reported in Southeast Asia and predisposes affected individuals to recurrent and multiple intracellular bacterial infections, which can be life-threatening. As a "silent disruptor," AIGA often go unnoticed due to their insidious nature, leading to a lack of sufficient clinical awareness. This report presents the case of a 59-year-old male patient from Wenzhou, Zhejiang

The influences of combination of garbage enzyme and biochar on total organic carbon (TOC) degradation, humification and the fungal succession during sewage sludge (SS) composting were established. Results showed that the GE and BC + GE treatments significantly increased the enzyme activity of fluorescein diacetate hydrolase (FDA) and increased the TOC degradation rate by 9.8% and 21.9% relative to control. The excitation-emission matrix (EEM) combined with the percentage fluorescence response (Pi, n) also proved that the combination of BC and GE promoted fulvic acid-like and humic-like substan

An endophytic fungus isolated from the rhizomes of Rosmarinus officinalis was identified as a Talaromyces species. Five distinct compounds were successfully isolated from the crude extract obtained from the fermentation culture, and identified as (1) thymol, (2) phloretin, (3) hesperetin, (4) dihydroquercetin, and (5) 6,7-dihydroxycoumarin. The antimicrobial activities of the five compounds were evaluated against Candida albicans, Escherichia coli, and Staphylococcus aureus, revealing varied efficacy. Compound 2 was the most effective inhibitor of C. albicans. Activity against E. coli was obse

BACKGROUND: Talaromyces marneffei causes talaromycosis, a life-threatening fungal disease with limited treatment options. The standard treatment of amphotericin B (AmB) induction followed by itraconazole consolidation still results in 15% to 30% mortality. This study aimed to investigate the potential of AmB and flucytosine (5FC) combination therapy to enhance antifungal activity.
METHODS: The in vitro antifungal activity of AmB and 5FC alone and in combination against 60 T. marneffei clinical isolates was evaluated using a validated colorimetric antifungal susceptibility assay and the checke

BACKGROUND: Talaromycosis, caused by the thermally dimorphic fungus Talaromyces marneffei, is a life-threatening opportunistic infection in individuals with advanced immunodeficiency, particularly people living with HIV in endemic regions of Southeast Asia. Early diagnosis is essential to reduce morbidity and mortality; however, conventional diagnosis relies mainly on fungal culture, which is time-consuming and has limited sensitivity.
METHODS: We developed and clinically evaluated a direct sandwich enzyme-linked immunosorbent assay (ELISA) using a biotin-labeled monoclonal antibody (4D1) for

BACKGROUND: Autoantibodies against interferon-γ (IFN-γ) can inhibit IFN-γ-dependent signal transducer and activator of transcription 1 phosphorylation and thus increase the risk of infection with intracellular pathogens, such as Talaromyces marneffei (TM), nontuberculous mycobacteria (NTMs), and Mycobacterium tuberculosis (TB). Here, we report a rare case of triple infection caused by TM, NTM, and TB in a human immunodeficiency virus-negative patient.
CASE PRESENTATION: A middle-aged female was admitted to our hospital after experiencing recurrent rash, cough, and expectoration for 4 months.

Haemophagocytic lymphohistiocytosis (HLH) is a life-threatening hyperinflammatory syndrome that may complicate advanced HIV infection, most commonly triggered by opportunistic infections. We report a man with advanced HIV who presented with prolonged fever and pancytopenia without an identifiable source despite extensive investigation. Bone marrow examination demonstrated haemophagocytosis, while blood mycobacterial culture yielded Penicillium marneffei, now classified as Talaromyces marneffei. Together with prolonged fever, pancytopenia, marked hyperferritinaemia and a retrospective HScore of

The initial host response to fungal pathogen invasion is critical to infection establishment and outcome. However, the diversity of leukocyte-pathogen interactions is only recently being appreciated. We describe a new form of interleukocyte conidial exchange called "shuttling." In Talaromyces marneffei and Aspergillus fumigatus zebrafish in vivo infections, live imaging demonstrated conidia initially phagocytosed by neutrophils were transferred to macrophages. Shuttling is unidirectional, not a chance event, and involves alterations of phagocyte mobility, intercellular tethering, and phagosome

The effect of endophytic fungal biotransformation on the formation of metabolic substances has become a non-negligible factor in assessing the quality of medicinal plants. However, the relevant evidences are still particularly lacking. In this study, an endophytic fungus CA3-A with biotransformation activity was screened and identified as Talaromyces coprophilus from root of Astragalus mongholicus. Its biotransformation effect on host metabolism was studied by co-culture with fungus and host root. The results showed that in addition to the significant changes in primary metabolites, secondary
