Determination of antibiotic susceptibility is a crucial aspect of antimicrobial therapy. The objective of this study was to determine the specific susceptibility of Staphylococcus aureus (SA) to the antibiotic doripenem. A total of 180 clinical isolates from VRSA patients were tested against doripenem using minimum inhibitory concentration (MIC) and disk diffusion tests. The overall findings indicated that doripenem exhibited a high sensitivity level for SA isolates, with an MIC50 of 60 μg/mL and a MIC90 of 0.5 mg/mL. Additionally, the antimicrobial activity of doripenem, both alone and in com

Vancomycin-resistant Enterococcus faecium (VREfm) carriage in the gastrointestinal tract is a risk factor for the development of an invasive infection. The exact mechanisms underlying the transition from colonization to infection are still unclear. We conducted a longitudinal study, including 54 paired VREfm isolates, consisting of a colonization and a subsequent bloodstream isolate from the same patient. We performed whole-genome sequencing, biofilm formation assays, and spot-on-lawn assays to investigate genotypic and phenotypic characteristics of the isolates. No significant differences in

Urinary tract infections (UTIs) remain the most frequent clinical diagnosis and Vancomycin-resistant Enterococci (VRE) second-leading cause of UTIs. The aims of this study were to ascertain the patterns and prevalence of UTIs in Sindh and underlying resistance mechanisms for VRE. Bacterial colonies were identified traditionally from a total of 33272 urine samples. Enterococcus species were identified using Facklam and Collins scheme. Antibiotic susceptibility was determined by Kirby-Bauer method and minimum inhibitory concentration (MIC) by E-test. VRE phenotypes were checked using vancomycin

Vancomycin-resistant Enterococcus faecium (VREfm) is a prevalent healthcare-acquired pathogen. Gastrointestinal colonization can lead to difficult-to-treat bloodstream infections with high mortality rates. Prior studies have investigated VREfm population structure within healthcare centers. However, little is known about how and why hospital-adapted VREfm populations change over time. We sequenced 710 healthcare-associated VREfm clinical isolates from 2017-2022 from a large tertiary care center as part of the Enhanced Detection System for Healthcare-Associated Transmission (EDS-HAT) program. Due to the widespread abuse of antibiotics, drug resistance in Enterococcus has been increasing. However, the speed of antibiotic discovery cannot keep pace with the acquisition of bacterial resistance. Thus, drug repurposing is a proposed strategy to solve the crises. Lusutrombopag (LP) has been approved as a thrombopoietin receptor agonist by the Food and Drug Administration. This study demonstrated that LP exhibited significant antimicrobial activities against vancomycin-resistant Enterococcus in vitro with rare resistance occurrence. Further, LP combined with tobramycin exhibited synergist

Vancomycin-resistant enterococci (VRE) are considered one of the main nosocomial pathogens due to their increasing antibiotic resistance and ability to cause life-threatening infections in humans. This study included VRE isolates obtained from various specimens including urine, blood, faeces, wounds, sputum, and oral cavity wash. Of the 37 strains, 30 (81.1%) and 7 (18.9%) were identified by MALDI TOF as Enterococcus faecium and Enterococcus faecalis, respectively. The clinical vancomycin-resistant enterococci exhibited multi-drug resistance (MDR). Apart from vancomycin, the enterococci exhibi

Infective endocarditis (IE) is a serious condition associated with high morbidity and mortality rates. The risk factors for IE include underlying heart disease, intravenous drug use, cardiac surgery, and interventional procedures. Enterococci are a common cause of IE, and vancomycin-resistant enterococci (VRE) infections are becoming increasingly prevalent. In this report, we present the case of an 88-year-old female patient with multiple cardiac comorbidities who developed VRE endocarditis with splenic infarction and embolic stroke. The patient was successfully treated with a combination of a

Owing to the increased use of antibiotics, drug-resistant strains, including those that are resistant to the antibiotic vancomycin, have emerged, which has become a major problem. In Japan, sewage treatments consist of sterilization with chlorine; however, this may not be sufficient to inactivate these bacteria. In this study, impulse voltage was employed instead of chlorine to inactivate drug-resistant bacteria. The results showed that sterilization above 105 CFU/mL is possible with longer application times of applied voltages above 4.5 kV. The effectiveness of impulse-voltage-mediated steril

OBJECTIVE: To evaluate the in vitro antibacterial effect of vancomycin hydrogel on methicillin-resistant Staphylococcus aureus (MRSA).
METHODS: We used polylactide glycolide-polyethylene glycol-polylactide glycolide (PLGA-PEG-PLGA) copolymer as a carrier of vancomycin to prepare vancomycin hydrogel. A vancomycin hydrogel group, a PLGA-PEG-PLGA copolymer group, a phosphate-buffered saline (PBS) negative control group and a vancomycin group were set for comparison. Then, we analyzed the in vitro antibacterial effect of each group to determine the minimum inhibitory concentration (MIC) and minim

Aims Ireland has the highest vancomycin-resistant Enterococcus faecium (VRE) bloodstream infection prevalence in Europe. Two patterns of VRE carriage are recognised. European, with widespread community prevalence and North American, where carriage is predominantly nosocomial. It is unclear which pattern is dominant in Ireland. This uncertainty limits infection control measures. This study sought to explore this issue via a cross sectional point prevalence study. Methods Asymptomatic community volunteers, represented by patients undergoing elective outpatient colonoscopy testing, were opportuni

WHAT IS KNOWN AND OBJECTIVE: The rise of vancomycin-resistant enterococci (VRE) has been a major health problem in most countries of the world including Asia, since its discovery. There is a paucity of data on VRE in many countries of Asia as well as limited pooled estimates. Therefore, we performed a systematic review and meta-analysis to estimate a pooled prevalence of VRE in Asia.
METHODS: A literature search in electronic databases like PubMed, Embase and Google Scholar and manual searching of references and grey literature, comprising the information on the prevalence of VRE with at leas

This study introduces a spinning and fully integrated paper-based microdevice that can perform multiple functions, including DNA extraction, amplification, and colorimetric detection, for monitoring two major vancomycin-resistant Enterococci (VREs), which carry the vanA and vanB genes. The spinning microdevice is composed of a stationary part and a spinning part. The square-shaped stationary part has two zones: the lysis and reaction zones. The spinning part, which has a spin wheel-like shape, was inserted perpendicularly into the stationary part so that its two semicircles remained on the upp

OBJECTIVES: The global dissemination of vancomycin-resistant enterococci (VRE) has become a serious public-health concern. Although outbreaks are typically caused by nosocomial transmission, contaminated food and water may contribute to the spread of VRE. The aim of this study was to assess the presence of VRE in flowing surface water bodies in Switzerland and to characterise the isolates.
METHODS: Surface water was sampled from rivers, streams and canals throughout Switzerland and was screened for the presence of VRE. Whole-genome sequencing was used to identify antimicrobial resistance gene

BACKGROUND: Linezolid, an oxazolidinone antibiotic, is recommended for vancomycin-resistant enterococci (VRE). However, 100% free-drug concentration above the minimum inhibitory concentration (fT>MIC) and an area under the curve of free drug to MIC ratio (fAUC24/MIC) >100 were associated with favorable clinical outcome with less emerging resistance. A plasma trough concentration (Ctrough) of linezolid ≥9 μg/mL was also related to hematologic toxicity. Thus, linezolid dose optimization is needed for VRE treatment. The study aimed to determine the in vitro linezolid activity against clinical VRE

Vancomycin is a standard drug for the treatment of multidrug-resistant Gram-positive bacterial infections. Albeit, development of resistance (VRE, VRSA) and its inefficacy against persistent infections is a demerit. It is also intrinsically inactive against Gram-negative bacteria. Herein, we report a vancomycin derivative, VanQAmC10, that addresses these challenges. VanQAmC10 was rapidly bactericidal against carbapenem-resistant A. baumannii (6 log10 CFU/mL reduction in 6 h), disrupted A. baumannii biofilms, and eradicated their stationary phase cells. In MRSA infected macrophages, the compoun

The aim of this study was to evaluate the antibacterial properties of methylsulfonylmethane (MSM) on vancomycin-resistant Enterococcus faecium (VRE). Bacterial proliferation was measured spectrophotometrically during growth in brain heart infusion broth with 0%, 3%, 5%, 7%, 10%, 12%, and 16% MSM. To assess the mechanism of inhibition, VRE was grown overnight with 0-16% MSM and enumerated on unmedicated and medicated (3-16% MSM) brain heart infusion agar (BHIA). Viability studies were performed to evaluate the impact of 10-16% MSM on VRE over 7 days. Absorbance data indicated a dose-dependent i

BACKGROUND: "MIC creep" is a phenomenon that describes an increase of an organism MICs over time and have been reported from different parts of the world. High MIC in MRSA has been theoretically liked to treatment failure and may be a precursor to hVISA and VISA. This study was conducted to assess presence of vancomycin minimum inhibitory concentration (MIC) creeps among clinical isolates of methicillin-resistant Staphylococcus aureus (MRSA) in Saudi Arabia.
METHODS: Minimum inhibitory concentration (MIC) of vancomycin by E test of all MRSA isolates of from 2013 to 2018 were reviewed.
