Abscisic Acid Lowers Postharvest Grape Acidity via PP2C-ALMT9-Dependent Malate Transport.

This study uncovers a novel concentration-dependent signaling network through which abscisic acid (ABA) modulates acid metabolism in grape berries. Treatment with 800 μM ABA significantly decreased malate and citrate synthesis and transport while enhancing glucose and fructose accumulation, optimizing the sugar-acid ratio. Multi-omics analysis revealed differential changes in 96 organic acids and identified a key malate transporter, VvALMT9. Through yeast two-hybrid screening, the VvPP2C-VvALMT9-VvSnRK2.3-VvCDPK interaction network was characterized in a dual regulatory mode. Under low-ABA con

OsNCED5 Controls Abscisic Acid-Mediated Anther Dehiscence and Male Fertility in Rice.

Proper anther dehiscence is essential for successful pollination and male fertility in flowering plants. However, many gaps remain in the regulatory mechanisms underlying anther dehiscence in plants, including a possible role for abscisic acid (ABA) in anther dehiscence. Here, we reported a member of 9-cis-epoxycarotenoid dioxygenase family (NCED), OsNCED5, which regulates proper anther dehiscence for male fertility in rice. OsNCED5 encodes a chloroplast-localized ABA biosynthetic enzyme highly expressed in anther during the late phase. Disruption of OsNCED5 by CRISPR/Cas9-mediated mutagenesis

Abscisic acid negatively interferes with basal defence of barley against Magnaporthe oryzae.

BACKGROUND: Plant hormones are well known regulators which balance plant responses to abiotic and biotic stresses. We investigated the role of abscisic acid (ABA) in resistance of barley (Hordeum vulgare L.) against the plant pathogenic fungus Magnaporthe oryzae. RESULTS: Exogenous application of ABA prior to inoculation with M. oryzae led to more disease symptoms on barley leaves. This result contrasted the finding that ABA application enhances resistance of barley against the powdery mildew fungus. Microscopic analysis identified diminished penetration resistance as cause for enhanced susce

Abscisic acid has a key role in modulating diverse plant-pathogen interactions.

We isolated an activation-tagged Arabidopsis (Arabidopsis thaliana) line, constitutive disease susceptibility2-1D (cds2-1D), that showed enhanced bacterial growth when challenged with various Pseudomonas syringae strains. Systemic acquired resistance and systemic PATHOGENESIS-RELATED GENE1 induction were also compromised in cds2-1D. The T-DNA insertion adjacent to NINE-CIS-EPOXYCAROTENOID DIOXYGENASE5 (NCED5), one of six genes encoding the abscisic acid (ABA) biosynthetic enzyme NCED, caused a massive increase in transcript level and enhanced ABA levels >2-fold. Overexpression of NCED genes re

Neuroprotective Effect of Abscisic Acid on MPTP-Induced Parkinson's Disease in Mice.

Parkinson's disease (PD) is the second largest neurodegenerative disease after Alzheimer's disease (AD), and neuroinflammation is one of its important causes. So far, there is no clear evidence that drugs can improve the onset of PD, so it is crucial to find and develop effective drugs for PD treatment. Abscisic acid (ABA) is a phytohormone with structural and medicinal functions similar to the PPAR-γ agonist thiazolidinedione drugs (TZDs). It has played therapeutic effects in a variety of inflammatory diseases, but the role and mechanism of PD have not been defined. The present study aimed to

Nitric oxide counteracts the senescence of rice leaves induced by abscisic acid.

In the present study, we evaluate the protective effect of nitric oxide (NO) against senescence of rice leaves promoted by ABA. Senescence of rice leaves was determined by the decrease of protein content. ABA treatment resulted in (1) induction of leaf senescence, (2) increase in H2O2 and malondialdehyde (MDA) contents, (3) decrease in reduced form glutathione (GSH) and ascorbic acid (AsA) contents, and (4) increase in antioxidative enzyme activities (superoxide dismutase, ascorbate peroxidase, glutathione reductase, and catalase). All these ABA effects were reduced by free radical scavengers

D27-like carotenoid isomerases: at the crossroads of strigolactone and abscisic acid biosynthesis.

Strigolactones and abscisic acid (ABA) are apocarotenoid-derived plant hormones. Their biosynthesis starts with the conversion of trans-carotenes into cis forms, which serve as direct precursors. Iron-containing DWARF27 isomerases were shown to catalyse or contribute to the trans/cis conversions of these precursor molecules. D27 converts trans-β-carotene into 9-cis-β-carotene, which is the first committed step in strigolactone biosynthesis. Recent studies found that its paralogue, D27-LIKE1, also catalyses this conversion. A crucial step in ABA biosynthesis is the oxidative cleavage of 9-cis-v

GhNAC043 and GhBPM2 interaction enhances Verticillium wilt resistance in cotton through jasmonic acid and abscisic acid pathways.

The soil-borne fungal pathogen Verticillium dahliae (V. dahliae) is the causal agent of Verticillium wilt (VW), a vascular disease that severely threatens global cotton production. Although cell wall lignification represents a cornerstone of plant immunity, the precise regulatory circuits that bridge this structural reinforcement with Verticillium dahliae resistance in cotton have yet to be fully elucidated. Here, we demonstrate that the NAC transcription factor GhNAC043 is a key positive regulator of this defense. GhNAC043 expression was rapidly induced upon V. dahliae infection. Silencing Gh

Low sucrose availability reduces basal spikelet fertility by inducing abscisic acid and jasmonic acid synthesis in wheat.

Within a spike of wheat, the central spikelets usually generate three to four fertile florets, while the basal spikelets generate zero to one fertile floret. The physiological and transcriptional mechanism behind the difference in fertility between the basal and central spikelets is unclear. This study reports a high temporal resolution investigation of transcriptomes, number and morphology of floret primordia, and physiological traits. The W6.5-W7.5 stage was regarded as the boundary to distinguish between fertile and abortive floret primordia; those floret primordia reaching the W6.5-W7.5 st

Nitrate attenuates abscisic acid signaling via NIN-LIKE PROTEIN8 in Arabidopsis seed germination.

Abscisic acid (ABA) suppresses Arabidopsis (Arabidopsis thaliana) seed germination and post-germinative growth. Nitrate stimulates seed germination, but whether it directly regulates ABA signaling and the associated underlying molecular mechanisms remain unknown. Here, we showed that nitrate alleviates the repressive effects of ABA on seed germination independently of the nitric oxide (NO) pathway. Moreover, nitrate attenuates ABA signaling activated by ABSCISIC ACID INSENSITIVE3 (ABI3) and ABI5, two critical transcriptional regulators of the ABA pathway. Mechanistic analyses demonstrated that

Abscisic acid and aloe-emodin against NS2B-NS3A protease of Japanese encephalitis virus.

There are no specific drugs for the treatment of Japanese Encephalitis. Thus, new chemical entities or exploration of existing molecules is required. We have tested the antiviral potential of abscisic acid and aloe-emodin against protease of the Japanese encephalitis virus (JEV) using the computational and target-based assay. Maestro Schrödinger glide suite 2019 was used for molecular docking and dynamic studies, and NS2B-NS3A JEV protease kit was used to confirm protease inhibitory activity of abscisic acid and aloe-emodin. The abscisic acid and aloe-emodin have shown optimum binding affinity

Abscisic acid metabolizing rhizobacteria decrease ABA concentrations in planta and alter plant growth.

Although endogenous phytohormones such as abscisic acid (ABA) regulate root growth, and many rhizobacteria can modulate root phytohormone status, hitherto there have been no reports of rhizobacteria mediating root ABA concentrations and growth by metabolising ABA. Using a selective ABA-supplemented medium, two bacterial strains were isolated from the rhizosphere of rice (Oryza sativa) seedlings grown in sod-podzolic soil and assigned to Rhodococcus sp. P1Y and Novosphingobium sp. P6W using partial 16S rRNA gene sequencing and phenotypic patterns by the GEN III MicroPlate test. Although strain

Abscisic acid in the xylem: where does it come from, where does it go to?

Abscisic acid is a hormonal stress signal that moves in the xylem from the root to the different parts of the shoot where it regulates transpirational water loss and leaf growth. The factors that modify the intensity of the ABA signal in the xylem are of particular interest because target cells recognize concentrations. ABA(xyl), will be decreased as radial water flow through the roots is increased, assuming that radial ABA transport occurs in the symplast only. Such dilutions of the plant hormone concentration can be compensated in different ways, which help to keep the ABA-concentrations in

Abscisic Acid accumulation in spinach leaf slices in the presence of penetrating and nonpenetrating solutes.

Abscisic acid (ABA) accumulated in detached, wilted leaves of spinach (Spinacia oleracea L. cv Savoy Hybrid 612) and reached a maximum level within 3 to 4 hours. The increase in ABA over that found in detached turgid leaves was approximately 10-fold. The effects of water stress could be mimicked by the use of thin slices of spinach leaves incubated in the presence of 0.6 molar mannitol, a compound which causes plasmolysis (loss of turgor). About equal amounts of ABA were found both in the leaf slices and in detached leaves, whereas 2 to 4 times more ABA accumulated in the medium than in the sl

Development of an in vitro assay using abscisic acid to study Toxoplasma gondii infectivity.

The common parasite Toxoplasma gondii can infect all warm-blooded animals, including humans. Although most infections in humans remain asymptomatic, clinical toxoplasmosis can develop into a fatal disease. Infections are usually contracted by oral ingestion of tissue cysts or oocysts contained in cat feces. Currently, the mouse bioassay is applied as a final experiment to evaluate meat infectivity. This study aims to establish an alternative cell culture and quantitative polymerase chain reaction (qPCR)-based in vitro infectivity assay for tissue cysts. The phytohormone abscisic acid (ABA) is

Integrative transcriptomics reveals association of abscisic acid and lignin pathways with cassava whitefly resistance.

BACKGROUND: Whiteflies are a global threat to crop yields, including the African subsistence crop cassava (Manihot esculenta). Outbreaks of superabundant whitefly populations throughout Eastern and Central Africa in recent years have dramatically increased the pressures of whitefly feeding and virus transmission on cassava. Whitefly-transmitted viral diseases threaten the food security of hundreds of millions of African farmers, highlighting the need for developing and deploying whitefly-resistant cassava. However, plant resistance to whiteflies remains largely poorly characterized at the gene
