Physiologically Based Pharmacokinetic Model of Magnesium Implant Absorption and Distribution in Tissue and Organs.

The long-term accumulation of magnesium (Mg-(II)) ions in human patients resulting from the biodegradation of clinical Mg (alloy) implants is investigated using a physiologically based pharmakinetic (PBPK) mathematical model. In severe cases, an excess of Mg in blood (hypermagnesemia) causes a range of health concerns and potentially death. Studies investigating clinical Mg devices generally indicate that there is little risk in healthy patients; however, there is concern that excessive Mg accumulation may occur in patients who are elderly, have osteoporosis, and/or have renal disease. The PBP

Impact of Physiological Characteristics on Chylomicron Pathway-Mediated Absorption of Nanocrystals in the Pediatric Population.

Nanocrystals exhibit significant advantages in improving the oral bioavailability of poorly soluble drugs. However, the complicated absorption properties of nanocrystals and the differences in physiological characteristics between children and adults limit pediatric applications of nanocrystals. To elucidate the absorption differences and the underlying mechanisms between children and adults, the pharmacokinetics and tissue distribution of aprepitant crystals with different particle sizes (NC200, NC500, and MC2.5) in rats and mice at different ages were studied, and their absorption mechanisms

Gastrointestinal absorption of xenobiotics in physiologically based pharmacokinetic models. A two-compartment description.

Gastrointestinal (GI) absorption in physiologically based pharmacokinetic models is typically described as first-order transfer from one compartment directly into the liver. This approach appears to be adequate for water but not for oily vehicles. In this study, a two-compartment description of GI absorption was developed in which the chemical moved from the first compartment to the second (first-order rate constant KT) and was absorbed from both compartments (first-order rate constants KAS and KAD, respectively) into the liver. Rat blood time-course data sets for methylene chloride, chlorofor

Evaluation of anti-tumor drugs by simulating physiological absorption and metabolism with a multi-organoid system.

Although patient-derived tumor organoids are widely applied for evaluating anti-tumor drugs, the lack of a physiologically relevant environment limits drug assessment. Intestinal absorption and liver metabolism are critical for drug efficacy but cannot be recapitulated in single-organoid models. Here, we establish an intestine-liver-tumor multi-organoid system to simulate in vivo absorption and metabolism, which allows for more physiologically relevant evaluations of drug efficacy and toxicity. Under fluidic conditions, the system maintains the viability of intestinal cells and liver organoids

Physiological and Transcriptome Analyses Revealed the Mechanism by Which Deferoxamine Promotes Iron Absorption in Cinnamomum camphora.

Iron deficiency causes chlorosis and growth inhibition in Cinnamomum camphora, an important landscaping tree species. Siderophores produced by plant growth-promoting rhizobacteria have been widely reported to play an indispensable role in plant iron nutrition. However, little to date has been determined about how microbial siderophores promote plant iron absorption. In this study, multidisciplinary approaches, including physiological, biochemical and transcriptome methods, were used to investigate the role of deferoxamine (DFO) in regulating Fe availability in C. camphora seedlings. Our result

Alligators and Crocodiles Have High Paracellular Absorption of Nutrients, But Differ in Digestive Morphology and Physiology.

Much of what is known about crocodilian nutrition and growth has come from animals propagated in captivity, but captive animals from the families Crocodilidae and Alligatoridae respond differently to similar diets. Since there are few comparative studies of crocodilian digestive physiology to help explain these differences, we investigated young Alligator mississippiensis and Crocodylus porosus in terms of (1) gross and microscopic morphology of the intestine, (2) activity of the membrane-bound digestive enzymes aminopeptidase-N, maltase, and sucrase, and (3) nutrient absorption by carrier-med

Pharmacokinetics of paracetamol in Göttingen minipigs: in vivo studies and modeling to elucidate physiological determinants of absorption.

PURPOSE: Onset and rate of gastric emptying are important determinants of drug absorption after oral dosing. Therefore, robust estimates of these parameters are needed in physiologically based absorption models to predict reliably plasma concentration time profiles. For human and some other laboratory animals, reasonable parameterization of gastric emptying has been established. However gastric emptying is less well characterized in minipigs, a large animal model rapidly gaining importance in pharmaceutical research. METHODS: A pharmacokinetic crossover study using different dosage forms of p

Development of physiologically-based gut absorption model for probabilistic prediction of environmental chemical bioavailability.

Absorption in the gastrointestinal tract is a key factor determining the bioavailability of chemicals after oral exposure but is frequently assumed to have a conservative value of 100% for environmental chemicals, particularly in the context of high-throughput toxicokinetics for in vitro-to-in vivo extrapolation (IVIVE). For pharmaceutical compounds, the physiologically based advanced compartmental absorption and transit (ACAT) model has been used extensively to predict gut absorption but has not generally been applied to environmental chemicals. Here we develop a probabilistic environmental c

Pathogenesis of congenital alkalosis with diarrhea. Implications for the physiology of normal ileal electrolyte absorption and secretion.

Using a triple-lumen constant perfusion system, we have studied ileal electrolyte transport in a patient with congenital alkalosis with diarrhea and made the following observations. First, chloride cannot be transported against electrochemical gradients, but can be readily absorbed or secreted down electrochemical gradients. Second, chloride secretion down an electrochemical gradient can be increased by raising lumen bicarbonate concentration. Third, sodium absorption does occur against electrochemical gradients and is associated with hydrogen ion secretion. Fourth, electrical potential differ

Complying with the physiological functions of Golgi apparatus for secretory exocytosis facilitated oral absorption of protein drugs.

Intestinal epithelial cells are the primary biological barriers for orally administrated nano-formulations and the delivered protein drugs. Thereinto, besides the cellular uptake, intracellular trafficking pathway and the related exocytosis are of great importance to the trans-epithelial transport of drug-loaded NPs. Herein, inspired by the physiological functions of Golgi apparatus for secreting proteins out of cells, Golgi localization-related amino acid l-cysteine (Cys) was modified on the surface of NPs to see whether and how this modification could guide the Golgi pathway-related transpor

Mechanistic embedding of concise physiological absorption models for enhanced prediction in population pharmacokinetic modeling of oral ibuprofen.

Ibuprofen, a commonly used non-steroidal anti-inflammatory drug (NSAID), exhibits considerable variability in pharmacokinetics following oral administration, particularly during the absorption phase. This study aimed to enhance the mechanistic understanding of ibuprofen absorption by developing and integrating physiologically-informed models of absorption within a population pharmacokinetic (PopPK) framework. Using existing datasets of ibuprofen plasma concentrations, two one-compartment PopPK models with linear elimination were developed: the Sequential Absorption Pharmacokinetic (SAPK) model

Chronic alcohol consumption and intestinal thiamin absorption: effects on physiological and molecular parameters of the uptake process.

Thiamin is essential for normal cellular functions, and its deficiency leads to a variety of clinical abnormalities. Humans and other mammals obtain the vitamin via intestinal absorption. The intestine is exposed to two sources of thiamin, a dietary and a bacterial (i.e., normal microflora of the large intestine) source. Chronic alcohol consumption is associated with thiamin deficiency, which is caused (in part) by inhibition in intestinal thiamin absorption. However, little is known about the physiological and molecular aspects of the intestinal thiamin uptake process that are affected by chr

Comparative toxicity of copper and zinc contaminated wastewater irrigation on growth, physiology, and mineral absorption of wheat.

A comparative pot study was performed to assess the toxic effects of copper (Cu) and/or zinc (Zn) contaminated wastewater (WW) irrigation on the growth, physiology, and element concentration of wheat grown for two months. The treatments included irrigation with uncontaminated wastewater (WW) as control, Cu-contaminated WW (CuWW), Zn-contaminated WW (ZnWW), and Cu + Zn contaminated WW (CuZnWW) in a completely randomized design. Compared to ZnWW, irrigation with CuWW or CuZnWW had severe effects on growth, physiology, and mineral absorption by wheat. Irrigation with CuWW or CuZnWW reduced shoot

Physiologically based absorption modeling to predict bioequivalence of controlled release and immediate release oral products.

Physiologically based absorption modeling was conducted to predict bioequivalence (BE) for immediate release (IR) and controlled release (CR) formulations. In case of the CR formulation of a BCS class 1 drug, sensitivity analyses were conducted to investigate the impact of gastrointestinal (GI) transit time and absorption scaling factors in caecum and colon on formulation PK. The regional absorption profiles of the test and reference formulations were compared to provide additional confidence on the BE predictions. For IR formulation of BCS class 2b drug, the sensitivity of dissolution rate, p

Physiologically Based In vitro Models to Predict the Oral Dissolution and Absorption of a Solid Drug Delivery System.

To understand the sophisticated dynamic behaviors of drug elution and permeation in the gastrointestinal tract (GIT), researchers have tried to reemerge it by employing various in vitro experimental models. However, official in vitro apparatuses routinely used for quality control purposes, employ simple, non-physiologic buffers, and hydrodynamics conditions, and can not accurately perform continuous, dynamic in vivo pharmacokinetics (PK) behaviors. Therefore, different angles of GI physiology information are incorporate into novel models to forecast the dissolution and permeation of drug solid

Potential Applications of Chitosan-Based Nanomaterials to Surpass the Gastrointestinal Physiological Obstacles and Enhance the Intestinal Drug Absorption.

The small intestine provides the major site for the absorption of numerous orally administered drugs. However, before reaching to the systemic circulation to exert beneficial pharmacological activities, the oral drug delivery is hindered by poor absorption/metabolic instability of the drugs in gastrointestinal (GI) tract and the presence of the mucus layer overlying intestinal epithelium. Therefore, a polymeric drug delivery system has emerged as a robust approach to enhance oral drug bioavailability and intestinal drug absorption. Chitosan, a cationic polymer derived from chitin, and its deri

Mechanism for Fe(III) to decrease cadmium uptake of wheat plant: Rhizosphere passivation, competitive absorption and physiological regulation.

The presence of dissolved Fe(III) and Fe(III)-containing minerals has been found to alleviate cadmium (Cd) accumulation in wheat plants grown in Cd-contaminated soils, but the specific mechanism remains elusive. In this work, hydroponic experiments were conducted to dissect the mechanism for dissolved Fe(III) (0-2000 μmol L-1) to decrease Cd uptake of wheat plants and study the influence of Fe(III) concentration and Cd(II) pollution level (0-20 μmol L-1) on the Cd uptake process. The results indicated that dissolved Fe(III) significantly decreased Cd uptake through rhizosphere passivation, com
