Five adult dogs were used to determine whether acepromazine maleate (ACP), administered IM, decreases the maintenance requirement of halothane and to measure any decrease for the ACP dosages of 0.02, 0.04, 0.06, 0.08, 0.10, and 0.20 mg/kg. The value minimal alveolar concentration, a measure of anesthetic potency, was used as the measure of anesthetic requirement of halothane before and after ACP was administered. All dogs were randomly exposed to each dosage of ACP, as well as to control of 0.2 ml of sterile water. At all dosages of ACP, the decrease in the minimal alveolar concentration of ha

1.25 g acepromazine maleate was consumed by a young woman in an unsuccessful suicide attempt. The drug is used extensively as an anesthetic premedication in veterinary practice. The woman had worked as a lay veterinary assistant. The clinical signs of toxicity were in part consistent with phenothiazine over-dosage in the human and were inconsistent with signs usually observed in animals.

Cystometry was performed in 12 dogs under oxymorphone and acepromazine restraint. A detrusor reflex occurred in 10 of 12 dogs (83%). Mean threshold pressure and volume were 31 cm H2O and 22 ml/kg, respectively. Tonus limb II varied inversely with threshold volume. Threshold volume varied directly with body weight; threshold pressure was independent of body weight.

INTRODUCTION: Acepromazine is a phenothiazine that is used exclusively in veterinary medicine for multiple purposes. Human overdoses are rarely reported and toxicokinetic data has never been reported. We present a case of intentional acepromazine overdose resulting in central nervous system and cardiovascular toxicity with confirmatory toxicokinetic data.
CASE REPORT: A 54-year-old woman intentionally ingested 950 mg of her dog's acepromazine. Within 3 h of ingestion, she developed central nervous system and respiratory depression along with hypotension requiring non-invasive ventilation and

We evaluated the effect of ketamine-xylazine-acepromazine anesthesia (31.25, 6.25, and 1.25 mg/kg subcutaneously, respectively) on postsurgical recovery in male Sprague-Dawley (Crl:SD) rats undergoing laparotomy with and without the postoperative analgesic ketorolac. Recovery was determined by changes in body weight (BW) and water intake. The time of ketorolac administration (5 mg/kg intramuscularly), 60 min after anesthetic injection, was based on return of the pedal withdrawal reflex in Long-Evans (HsdBlu:LE) rats undergoing stereotaxic surgery in a separate experiment. Results were compared

The middle-latency auditory-evoked potential (MLAEP) has been investigated as means of monitoring anesthesia in dogs. The goals of this study were to develop a technique to record MLAEPs in awake dogs and to determine the effects of sedation. The MLAEP was recorded in 12 dogs with and without sedation with acepromazine. Three needle electrodes were inserted SC. Click stimuli were delivered biaurally. Signal acquisition, averaging, and analysis were performed by software developed in-house. Signals were recorded for 128 milliseconds, and the responses to 1,024 stimuli were averaged. The wavefor

A fatality due to ingestion of zolpidem, a recently developed nonbenzodiazepine hypnotic, and acepromazine, a phenothiazine sedative used in veterinary practice, is reported. Both drugs were identified and quantified using high-performance liquid chromatography with diode-array detection. Zolpidem and acepromazine blood concentrations were 3.29 and 2.40 micrograms/ml, respectively. It is the first report of a death involving either of these two drugs. The results are discussed in the light of the existing literature and may suggest a reciprocal potentiation of the toxic effects of zolpidem and

In this study, the degree and characteristics of sedation were assessed using the Grint scale for azaperone and acepromazine in 24 dogs, with the effects on heart rate and systemic blood pressure evaluated simultaneously using oscillometry. Posture, palpebral reflex, eye position, relaxation, response, postural resistance, attitude, systolic pressure, diastolic pressure, mean arterial pressure, and heart rate were analyzed using linear mixed-effects models to evaluate the effects of treatment, time, and their interaction. Significant effects of time were observed for all variables (p < 0.001),

Acepromazine (ACP) is one of the most used sedatives in veterinary medicine, and its cardiovascular effects are well documented. In contrast, its effects on the respiratory system remain less well understood, particularly in the context of assessment of respiratory mechanics in murine models of asthma, where its use has become more common in recent years. This study aims to investigate the effect of ACP on respiratory mechanics in BALB/c mice, specifically in an ovalbumin (OVA)-induced model of allergic asthma. Mice received ACP (2.5 mg/kg) in combination with standard ketamine (100 mg/kg) and

Sixteen New Zealand white rabbits were divided randomly into two groups of eight. Schirmer II tear tests were performed on all 16 and then eight received an intramuscular dose of 1 mg/kg acepromazine and the other eight received an intramuscular dose of 1 mg/kg diazepam. The Schirmer tests were repeated after 15 and 25 minutes. There was a significant reduction in tear production by the rabbits treated with acepromazine, but no significant change in tear production by the rabbits treated with diazepam.

Use of acepromazine (i.e., acetylpromazine) maleate in dogs with a history of seizures is reportedly contraindicated because of the risk of decreasing the seizure threshold in these animals. In this retrospective study, acepromazine was administered for tranquilization to 36 dogs with a prior history of seizures and to decrease seizure activity in 11 dogs. No seizures were seen within 16 hours of acepromazine administration in the 36 dogs that received the drug for tranquilization during hospitalization. After acepromazine administration, seizures abated for 1.5 to 8 hours (n=6) or did not rec

The effects of xylazine (an alpha 2-adrenoceptor agonist) and acepromazine (an alpha-adrenoceptor antagonist) on bronchomotor tone were investigated in seven anaesthetised, apnoeic ponies using a computer aided forced oscillation technique, which separates changes in bronchial calibre from changes in lung volume. Both agents produced bronchodilatation and a decrease in lung volume.

The sedative effect of acepromazine combined with 2 doses of tramadol [3 and 5 mg/kg body weight (BW)] was compared with the sedative effect of acepromazine alone in dogs and the effects of each sedative protocol on cardiorespiratory variables were examined. This was a prospective, randomized, blinded, crossover study. Each of 6 dogs received 3 treatments at 1-week intervals. During all anesthetic episodes, dogs received 0.05 mg/kg BW acepromazine. Approximately 25 min later, dogs were given physiological saline (control) or tramadol [3 mg/kg BW (TR3) or 5 mg/kg BW (TR5)]. All drugs were admin
