Toxicological investigation of 25 aconitine-induced deaths from 2005 to 2023.

Aconitum herbs contain several highly toxic diester-diterpenoid alkaloids, including aconitine, mesaconitine, and hypaconitine. However, finding the cause of death is rather difficult for forensic pathologists during forensic autopsy of aconitine-induced death. Therefore, the ability to determine Aconitum alkaloids is important in these cases. The aim of this study was to review the data for alkaloids in postmortem specimens from 25 aconitine-induced deaths received by the Academy of Forensic Science from 2005 to 2023. Aconitum alkaloids were analyzed using an LC-MS/MS method, which was valida

The antitumor effect of the combination of aconitine and crude monkshood polysaccharide on hepatocellular carcinoma.

Aconitine, the main component in Radix Aconiti Lateralis Preparata, not only exerts the anti-tumor effect on Hepatocellular Carcinoma (HCC) but also damages on immune system. In the present study, Crude Monkshood Polysaccharide (CMP), another one natural composition component originated from the same herbal with aconitine, combined with aconitine to investigate the effects on HCC and immunity in vitro and in vivo. The combination of CMP and aconitine enhanced the ability of the immunocyte to kill the tumor cell in vitro and had an additive effect on anti-HCC in vivo. Aconitine-CMP in combinati

Synthesis and biological evaluation of aconitine-type derivatives as potential cardiotonic agents.

Eighteen C19 aconitine-type derivatives were designed and syn-thesized with the modification of core active and strong toxicity components, such as yunaconitine (YA), mesaconitine (MA), and hypaconitine (HA) in Fuzi through hydrolysis and demethylation reactions. Their structures were confirmed by MS,1H NMR and 13C NMR. Isolated frog heart perfusion assay demonstrated that compound 18 exhibited excellent anti-heart failure activity with a cardiotonic rate of 103.14%. Its cardiotonic effect could be completely antagonized by nonselective calcium channel blockers, suggesting that the pharmacolog

Antiarrhythmic effects of an aconitine-like compound, TJN-505, on canine arrhythmia models.

We examined the effects of an aconitine-like compound, TJN-505 (1alpha-16beta-dimethoxy-20-ethyl-14alpha-(4-methox ybenzoyloxy)-aconitan-8,13-diol hydrochloride), on canine arrhythmias provoked by digitalis, two-stage coronary ligation, adrenaline, programmed electrical stimulation, or aconitine. TJN-505 (2-2.5 mg/kg i.v.) suppressed digitalis-, two-stage coronary ligation- and adrenaline-induced ventricular arrhythmias. The antiarrhythmic plasma concentrations (IC50) of TJN-505 for these arrhythmia models were 1.26, 0.94 and 1.31 microg/ml, respectively. TJN-505 (2 mg/kg i.v. followed by the

Aconitine poisoning as a rare cause of pulseless diffuse ventricular arrhythmia: a case report.

A middle-aged woman presented with nausea and vomiting after consumption of dinner. Her son experienced similar symptoms after sharing the meal. The patient had a history of syncope and had been treated with a dual-chamber pacemaker years prior to the incident. While being treated by emergency medical services (EMS), the patient developed pulseless wide complex tachycardia. EMS performed cardiopulmonary resuscitation (CPR) and several defibrillation attempts. After being referred to our intensive care unit (ICU), initial workup revealed no evidence of structural heart disease, coronary artery

Ventricular Tachycardia due to an Overdose of Tibetan Drugs: A Case Report of Aconitine Poisoning.

Tibetan medicine enjoys a high recognition among Tibetans. Many Tibetan hospitals and medicine shops are dedicated to related research and prescribe Tibetan drugs to patients. Overdose intake of aconite-containing Tibetan drugs might cause severe poisoning manifesting malignant ventricular arrhythmias. Supportive measurements, including external chest compression, electric cardioversion, fluid resuscitation, and intravenous administration of amiodarone/lidocaine, aim at restoring sinus rhythm, serving as "time-buying" mechanisms to allow the body to naturally excrete the toxic alkaloids.

Cardiovascular changes produced by the injection of aconitine at the area of the locus coeruleus in unanesthetized rats.

Blood pressure, heart rate and electrocardiogram were monitored in unanesthetized, unrestrained rats while the area of the locus coeruleus was stimulated by the injection of 0.03 to 10 micrograms of aconitine to pharmacologically evaluate its cardiovascular action. Aconitine elicited a dose-dependent elevation of mean blood pressure to 200 mmHg; at doses of 1-10 micrograms, it increased heart rate by more than 100 beats/min and produced arrhythmias. Phentolamine and the serotonergic antagonist, 2-bromo-lysergic acid diethylamide, prevented aconitine-induced cardiovascular changes. Diminution o

Mechanism of aconitine mediated neuronal apoptosis induced by mitochondrial calcium overload caused by MCU.

Aconitine is a crucial toxic component in Chinese herbal medicines such as Aconitum, Aconitum coreanum, and Aconitum soongaricum. The poisoning symptoms of the central nervous system and cardiovascular system caused by it are relatively common in China, and there are many studies on cardiovascular system diseases caused by aconitine. However, the specific mechanism of neurotoxicity induced by aconitine is still unclear. This study explored the effect and mechanism of mitochondrial calcium uniporter on mitochondrial energy metabolism disorder in aconitine poisoning hippocampal neurons. The resu

Left Stellate Ganglion Blockade for Refractory Ventricular Arrhythmias With Aconitine Poisoning: A Case Report.

Aconitine poisoning causes refractory ventricular arrhythmias (VAs). In a 20-year-old man, VAs of unknown etiology did not respond to drugs and electrical defibrillation. However, left stellate ganglion blockade (SGB) dramatically decreased arrhythmias without complications. At a later date, we found that refractory VAs were caused by aconitine poisoning. Left SGB is effective for treating refractory VAs with aconitine poisoning and can be easily performed with few complications for VAs of unknown etiology even if patients are receiving anticoagulant therapy. Also, left SGB can be performed to

Evidence for aconitine-induced inhibition of delayed rectifier K(+) current in Jurkat T-lymphocytes.

Aconitine (ACO) is a highly toxic diterpenoid alkaloid and known to exert the immunomodulatory action. However, whether it has any effects on ion currents in immune cells remains unknown. The effects of ACO and other related compounds on ion currents in Jurkat T-lymphocytes were investigated in this study. ACO suppressed the amplitude of delayed-rectifier K(+) current (I(K(DR))) in a time- and concentration-dependent manner. Margatoxin (100 nM), a specific blocker of K(V)1.3-encoded current, decreased the I(K(DR)) amplitude in these cells and the ACO-induced inhibition of I(K(DR)) was not reve

Uncovering the poisonous aconitine containing plants in homemade herbal liquor using a convergent approach.

Human exposure to toxic plants is a global concern, with numerous reported cases of accidental poisoning. In this study, a patient experienced poisoning after inadvertently consuming an herbal preparation preserved in alcohol. The patient exhibited characteristic electrocardiogram abnormalities, prompting further investigation into the toxic plant responsible. A relative provided the suspected herbal sample for identification. Symptomatic treatment was administered, successfully stabilizing the patient. Given the suspicion of aconitine toxicity-despite the absence of naturally occurring Aconit

Gating current experiments on frog nodes of Ranvier treated with Centruroides sculpturatus toxins or aconitine.

(1) Gating currents were recorded from frog nodes of Ranvier treated either with toxins III or IV from the venom of the scorpion Centruroides sculpturatus or with the alkaloid toxin aconitine. (2) Toxins III or IV from Centruroides sculpturatus (which drastically reduce the sodium permeability PNa and slightly shift its voltage dependence in the depolarizing direction) caused a small depolarizing shift of the relation between charge (Qon) and membrane potential (E) without affecting the maximum charge Qon max. (3) On nodes treated with toxins III or IV from Centruroides sculpturatus, a depolar
