Multidrug-resistant plasmid modulates ammonia oxidation efficiency in Nitrosomonas europaea through cyclic di-guanylate and acyl-homoserine lactones pathways.

Antibiotic resistance genes present a major public health challenge and have potential implications for global biogeochemical cycles. However, their impacts on biological nitrogen removal systems remain poorly understood. In the ammonia-oxidizing bacteria Nitrosomonas europaea ATCC 19718 harboring the multidrug-resistant plasmid RP4, a significant decrease in ammonia oxidation efficiency was observed, accompanied by markedly elevated levels of cyclic di-guanylate (c-di-GMP) and acyl-homoserine lactones (AHLs), compared to plasmid-free controls. The results demonstrated that c-di-GMP facilitate

Cu(II)-catalyzed acylation by thiol esters under neutral conditions: tandem acylation-wittig reaction leading to a one-pot synthesis of butenolides.

The first catalytic acylation of alcohols with a thiol ester present in Wittig reagents under neutral conditions catalyzed by the Cu(II) salt through a push-pull mechanism is reported. Furthermore, a new methodology for the one-pot lactonization of acyloins by a copper catalyst is developed. The synthetic utility of this method for the synthesis of natural products is shown.

The Effect of Bacterial AHL on the Cyclic Adenosine Monophosphate Content in Plants According to High-Performance Liquid Chromatography.

Cyclic adenosine monophosphate (cAMP) is an important second messenger in cells, mediating various stimulation signals such as the growth and development of organisms and stress and participating in regulating various biological processes of cells. This article explores the quantitative determination of cAMP in plants using High-Performance Liquid Chromatography (HPLC) and applies this method to analyzing the changes in cAMP content during the process of plant response to the bacterial quorum sensing signal N-acyl homoserine lactone (AHL). Research has shown that the optimal detection conditio

Enhanced sensing of bacteria biomarkers by ZnO and graphene oxide decorated PEDOT film.

Developing a biofilm biomarker detector and inhibitor will immensely benefit efforts geared at curbing infectious diseases and microbiologically induced corrosion of medical implants, marine vessels and buried steel pipelines. N-Acyl homoserine lactones (AHLs) are important biomarkers gram-negative bacteria use for communication. In this work, we investigated the interactions between three AHL molecules and graphene oxide (GO) and ZnO nanomaterials embedded in conjugated poly(3,4-ethylenedioxythiophene) (PEDOT) film. The results show that PEDOT/GO/ZnO detected AHLs to a considerable extent wit

An acyl-homoserine lactone acylase found in Stenotrophomonas maltophilia exhibits both quorum quenching activity and the ability to degrade penicillin antibiotics.

Stenotrophomonas maltophilia are opportunistic, multi-drug-resistant Gram-negative pathogens increasingly prevalent in clinical settings. Quorum sensing (QS) systems play a crucial role in their pathogenesis, coordinating bacterial populations and enabling interactions within polymicrobial communities. While not the primary QS mechanism in S. maltophilia, these bacteria can respond to acyl-homoserine lactone (AHL)-type autoinducers. Some isolates exhibit AHL-quorum quenching activity, though the responsible components remain unidentified. Homology searches in S. maltophilia K279a revealed a pr

Unsaturated Fatty Acids Affect Quorum Sensing Communication System and Inhibit Motility and Biofilm Formation of Acinetobacter baumannii.

The increasing threat of Acinetobacter baumannii as a nosocomial pathogen is mainly due to the occurrence of multidrug-resistant strains that are associated with the real problem of its eradication from hospital wards. The particular ability of this pathogen to form biofilms contributes to its persistence, increases antibiotic resistance, and promotes persistent/device-related infections. We previously demonstrated that virstatin, which is a small organic compound known to decrease virulence of Vibrio cholera via an inhibition of T4-pili expression, displayed very promising activity to prevent

Quorum Quenching of Nitrobacter winogradskyi Suggests that Quorum Sensing Regulates Fluxes of Nitrogen Oxide(s) during Nitrification.

UNLABELLED: Quorum sensing (QS) is a widespread process in bacteria used to coordinate gene expression with cell density, diffusion dynamics, and spatial distribution through the production of diffusible chemical signals. To date, most studies on QS have focused on model bacteria that are amenable to genetic manipulation and capable of high growth rates, but many environmentally important bacteria have been overlooked. For example, representatives of proteobacteria that participate in nitrification, the aerobic oxidation of ammonia to nitrate via nitrite, produce QS signals called acyl-homoser

Two Novel AHL-Degrading Enzymes, PoAcy1 and PoLac, Attenuate Pectobacterium carotovorum and Pseudomonas aeruginosa Virulence through Quorum-Sensing Disruption.

Ensuring food safety remains a significant challenge due to the persistent contamination and spoilage caused by biofilm-forming and resistant pathogenic bacteria. Quorum quenching presents an ecofriendly control strategy. We identified a novel acylase (PoAcy1) and a broad-spectrum lactonase (PoLac) from marine-derived bacterium Pseudooceanicola onchidii XY-99. Both the XY-99 strain and purified PoLac effectively suppressed Pectobacterium carotovorum subsp. carotovorum (Pcc)-induced soft rot in potato tubers. Heterologous expression of either enzyme in Pseudomonas aeruginosa PAO1 significantly

Combinatorial Drug Therapy for Controlling Pseudomonas aeruginosa and Its Association With Chronic Condition of Diabetic Foot Ulcer.

Diabetic foot ulcer (DFU) is a major complication of diabetes mellitus, major observations of DFU cases have reported on amputation of foot region, and microbial bioburden during DFU is a major cause that affects healing of the wound regions. Pathogenic microbes are routinely isolated from these wound regions, especially Staphylococcus, Pseudomonas, Klebsiella, and Escherichia coli have been reported, whereas higher prevalence of Pseudomonas species during chronic condition in the deeper part of the wound, when left untreated, leads to gangrene. Multiple drug-resistant Pseudomonas strains are

The presence of acylated homoserine lactones and diffusible signal factor in bronchoalveolar lavage fluid from horses with clinical exacerbation of severe equine asthma.

Several bacteria associated with chronic lung pathology use quorum sensing (QS) signaling molecules to regulate their virulence in pure cultures and poly-microbial communities. Their excessive growth and biofilm formation in the respiratory tract increase the morbidity and mortality of inflammatory airway diseases in humans, such as chronic obstructive pulmonary disease (COPD), asthma and cystic fibrosis (CF). In horses, severe equine asthma (SEA) has many parallels to these human diseases. We hypothesized that QS molecules associated with the most common biofilm-forming lung pathogens in huma

Growth and Toxin Production of Gambierdiscus spp. Can Be Regulated by Quorum-Sensing Bacteria.

Gambierdiscus spp. are the major culprit responsible for global ciguatera fish poisoning (CFP). At present, the effects of microbiological factors on algal proliferation and toxin production are poorly understood. To evaluate the regulatory roles of quorum-sensing (QS) bacteria in the physiology of Gambierdiscus, co-culture experiments with screened QS strains were conducted in this study. Except for the growth-inhibiting effect from the strain Marinobacter hydrocarbonoclasticus, the algal host generally displayed much higher growth potential and toxin production ability with the existence of

CytoSorb in burn patients with septic shock and acute kidney injury on continuous kidney replacement therapy efficiently reduces the levels of Gram-negative-derived quorum-sensing molecules associated with lactate increases.

INTRODUCTION: Quorum-sensing (QS) molecules produced by Gram-negative bacteria regulate virulence and host-pathogen interactions, but their systemic role in human septic shock remains largely unexplored. We hypothesized that QS molecules are detectable in the circulation of burn patients with septic-shock-associated acute kidney injury (SA-AKI), are associated with disease severity, and are amenable to removal by extracorporeal treatment. METHODS: In this observational pilot study, plasma concentrations of eight Gram-negative-derived QS molecules were quantified by HPLC-MS/MS in 12 burn patie

MrtR of Mesorhizobium tianshanense reveals both activation and inhibition mechanisms of a LuxR-type quorum sensing receptor.

Quorum sensing (QS) enables common gram-negative bacteria to coordinate collective behaviors through small molecule signals, yet how these signals tune receptor activity remains incompletely understood. Here, we define a mechanism by which ligand structure controls function in a LuxR-type QS receptor. Using structural and biochemical analyses, we investigate MrtR from Mesorhizobium tianshanense and show that ligand acyl-chain length governs receptor assembly and activity. We present full-length structures of MrtR bound to activating and inhibitory ligands, revealing a switch in oligomeric stat

The chemotaxis disruption of Aeromonas veronii by Lactococcus lactis and its probiotic effect on loach (Misgurnus anguillicaudatus).

Aeromonas veronii is a pathogen that is widely found in aquaculture environments. It has been observed to colonize the gastrointestinal tract of fish and other aquatic animals, with the potential to cause severe infections. Previous studies have demonstrated that probiotics, such as Lactococcus lactis 01, Lactococcus lactis 02, Enterococcus pseudoavium 03, and Bacillus sp. 04, can reduce the colonization capability of A. veronii. In this study, we sought to ascertain whether these four probiotics could affect chemotaxis by interfering with N-acyl-homoserine lactones (AHLs), thereby regulating

Biochar-induced quorum sensing enhances methane production by strengthening direct interspecies electron transfer.

Biochar addition promotes direct interspecies electron transfer (DIET) between syntrophic bacteria and methanogens and increases the secretion of N-acyl homoserine lactones (AHLs); however, the specific role of AHLs in biochar-promoted DIET remains unclear. In this study, biochar accelerated the degradation of volatile fatty acids, increasing the maximum methanation rate by 24.97 ± 0.23 %. Compared to the control system, biochar significantly enhanced the secretion of AHLs, resulting in an approximate 2.7-fold increase. The increase in AHLs subsequently promoted the synthesis of c-type cytochr

Membrane-enclosed Pseudomonas quinolone signal attenuates bacterial virulence by interfering with quorum sensing.

Outer membrane vesicle (OMV)-delivered Pseudomonas quinolone signal (PQS) plays a critical role in cell-cell communication in Pseudomonas aeruginosa. However, the functions and mechanisms of membrane-enclosed PQS in interspecies communication in microbial communities are not clear. Here, we demonstrate that PQS delivered by both OMVs from P. aeruginosa and liposome reduces the competitiveness of Burkholderia cenocepacia, which usually shares the same niche in the lungs of cystic fibrosis patients, by interfering with quorum sensing (QS) in B. cenocepacia through the LysR-type regulator ShvR. Quorum sensing molecules enhance cadmium detoxification and cellular removal in the marine diatom Phaeodactylum tricornutum.

Acyl-homoserine lactones (AHLs) are ubiquitous bacteria-derived quorum-sensing molecules in marine environments and play important roles in regulating microbial physiological processes. However, their influence on metal detoxification and handling in microalgae remains poorly understood. Here, we investigated the effects of exogenous AHLs on cell growth, metabolic responses, and cadmium (Cd) removal in the marine diatom Phaeodactylum tricornutum under both Cd-free and Cd-contaminated conditions. It was demonstrated that AHLs slightly inhibited the growth of P. tricornutum and increased extrace

Quorum sensing and self-quorum quenching in the intracellular pathogen Brucellamelitensis.

Brucella quorum sensing has been described as an important regulatory system controlling crucial virulence determinants such as the VirB type IV secretion system and the flagellar genes. However, the basis of quorum sensing, namely the production of autoinducers in Brucella has been questioned. Here, we report data obtained from the use of a genetic tool allowing the in situ detection of long-chain N-acyl-homoserine lactones (AHL) activity at single bacterium level in Brucella melitensis. These data are consistent with an intrinsic production of AHL by B. melitensis in low concentration both d
