Human induced pluripotent stem cells: A new source for brown and white adipocytes.

Mesenchymal stem cells (MSCs) derived from human induced pluripotent stem cells (hiPSCs) provide a novel source for generating adipocytes, thus opening new avenues for fundamental research and clinical medicine. We present the adipogenic potential of hiPSCs and the various methods to derive hiPSC-MSCs. We discuss the main characteristic of hiPSC-MSCs, which is their low adipogenic capacity as compared to adult-MSCs. Finally, we propose several hypotheses to explanation this feature, underlying a potential critical role of the micro-environment. We favour the hypothesis that the range of factor

Noradrenaline modulates the electrical activity of white adipocytes by a cAMP-dependent mechanism.

We studied the effect of the external addition of noradrenaline (NA) on the electrical activity of white adipocytes from rat epididymal tissue. NA modified the active voltage response recorded by the current clamp technique, and decreased the macroscopic membrane potassium currents measured by a whole-cell configuration patch-clamp technique in cells obtained by culturing differentiating preadipocytes. To analyse if cAMP is implicated in the effect of this hormone on potassium conductances, the changes in electrical activity caused by an increase in the intracellular concentration of cAMP were

Depot-specific adipocyte cell lines reveal differential drug-induced responses of white adipocytes--relevance for partial lipodystrophy.

Intra-abdominal (IA) fat functionally differs from subcutaneous (SC) adipose tissue, likely contributing to its stronger association with obesity-induced morbidity and to differential response to medications. Drug-induced partial lipodystrophy, like in response to antiretroviral agents, is an extreme manifestation of the different response of different fat depots, with loss of SC but not IA. Investigating depot-specific adipocyte differences is limited by the low accessibility to IA fat and by the heterogenous cell population comprising adipose tissue. Here, we aimed at utilizing immortalized

Mycobacterium tuberculosis Infects Human Visceral White Adipocytes and Expresses Dormancy Genes and Inflammatory Cytokines: The Role of Visceral Adipocytes in Latent Tuberculosis Infection.

This study explores the role of human visceral white adipocytes (hv-WAD) in latent tuberculosis infection (LTBI). While granulomas and macrophages are traditionally viewed as central to TB latency, emerging evidence highlights adipocytes as significant non-canonical host cells that may facilitate bacterial persistence by providing a protective niche. Unlike the immune-driven environment within granulomas, adipocytes can shield Mycobacterium tuberculosis (Mtb) from immune surveillance, promoting survival. In vitro experiments showed that Mtb invades approximately 39% of hv-WAD within 48-72 h po

Dimethylmalonate suppresses white adipocyte differentiation and ameliorates diet-induced obesity.

Obesity is driven by pathological expansion of white adipose tissue (WAT), reflecting both adipocyte hypertrophy and the differentiation of preadipocytes into mature white adipocytes. Arresting adipogenic differentiation is therefore a logical anti-obesity strategy. Because adipogenesis relies on extensive metabolic rewiring, blocking it by targeting mitochondrial enzymes-without collapsing cellular bioenergetics-remains an attractive but unresolved objective. Here, we investigated dimethylmalonate (DMM), a membrane-permeable diester that undergoes intracellular cleavage to yield malonate, a c

Adaptation of White Adipocytes to Cooler Temperatures: Impacts on Energy Metabolism and Protein Acetylation.

Adipocytes throughout the body reside in distinct thermal environments. Visceral adipocytes within the body core are maintained near 37°C, whereas those in bone marrow, subcutaneous, and dermal depots occupy cooler regions within the peripheral shell. Although brown and beige adipocyte responses to cold stress are well characterized, much less is known about how white adipocytes adapt to moderately reduced temperatures below 37°C. Our recent work revealed that cultured adipocytes exposed to 31°C, a temperature representative of distal adipose regions, exhibit enhanced mitochondrial function, i

Multiomics analyses reveal that PRMT5 regulates membrane transport and cholesterol synthesis in white adipocytes.

The adipose tissue (AT) is a main regulator of systemic energy homeostasis, and AT dysfunction leads to insulin resistance and other metabolic complications. Protein arginine methyltransferase 5 (PRMT5) catalyzes symmetrical dimethylation of arginine residues to modulate protein stability and function. Besides its well-studied oncogenic functions, PRMT5 has recently been shown to play a physiological role in AT through poorly understood mechanisms. Here, we explore the function of PRMT5 in AT through unbiased RNA sequencing and lipidomic analyses of AT in wild-type and adipocyte-specific Prmt5

BMPR2 promotes fatty acid oxidation and protects white adipocytes from cell death in mice.

Adipocyte cell death is pathologically involved in both obesity and lipodystrophy. Inflammation and pro-inflammatory cytokines are generally regarded as inducers for adipocyte apoptosis, but whether some innate defects affect their susceptibility to cell death has not been extensively studied. Here, we found bone morphogenetic protein receptor type 2 (BMPR2) knockout adipocytes were prone to cell death, which involved both apoptosis and pyroptosis. BMPR2 deficiency in adipocytes inhibited phosphorylation of perilipin, a lipid-droplet-coating protein, and impaired lipolysis when stimulated by t

White Tea extract induces lipolytic activity and inhibits adipogenesis in human subcutaneous (pre)-adipocytes.

BACKGROUND: The dramatic increase in obesity-related diseases emphasizes the need to elucidate the cellular and molecular mechanisms underlying fat metabolism. To investigate how natural substances influence lipolysis and adipogenesis, we determined the effects of White Tea extract on cultured human subcutaneous preadipocytes and adipocytes. METHODS: For our in vitro studies we used a White Tea extract solution that contained polyphenols and methylxanthines. Utilizing cultured human preadipocytes we investigated White Tea extract solution-induced inhibition of triglyceride incorporation durin

Insulin increases the density of potassium channels in white adipocytes: possible role in adipogenesis.

We studied the potassium currents in white adipocytes obtained by culturing preadipocytes from rat epididymal tissue, both with insulin (WA(i)) and without insulin (WA(o)), in order to test the role of insulin in the development of voltage-gated potassium channels (K(v)) during adipogenesis. Occasionally, very small potassium currents (I(K,V)) were present in preadipocytes; however these currents were measured in all differentiated cells (adipocytes). WA(i) exhibited greater macroscopic potassium currents than WA(o) with no apparent differences in kinetics or voltage dependence. The current de

Divergent roles of m6A in orchestrating brown and white adipocyte transcriptomes and systemic metabolism.

N6-methyladenosine (m6A) is among the most abundant mRNA modifications, yet its cell-type-specific regulatory roles remain unclear. Here we show that m6A methyltransferase-like 14 (METTL14) differentially regulates transcriptome in brown versus white adipose tissue (BAT and WAT), leading to divergent metabolic outcomes. In humans and mice with insulin resistance, METTL14 expression differs significantly from BAT and WAT in the context of its correlation with insulin sensitivity. Mettl14-knockout in BAT promotes prostaglandin secretion, improving systemic insulin sensitivity. Conversely, Mettl1

Sensory-neuron-derived CGRPα controls white adipocyte differentiation and tissue plasticity.

Communication between the brain and adipose tissue is mediated in part by the peripheral nervous system. The sympathetic branch transmits lipolytic and thermogenic signals toward adipose tissue, while sensory nerves relay information to the central nervous system. Importantly, sensory nerve activation also triggers local neuropeptide release. Here, we show that sensory-neuron-derived α-calcitonin gene-related peptide (CGRPα) inhibits white, but not brown, preadipocyte differentiation in a cell-autonomous manner. In vivo, CGRPα expression in mouse subcutaneous adipose tissue during cold exposur

Altered miRNA processing disrupts brown/white adipocyte determination and associates with lipodystrophy.

miRNAs are important regulators of biological processes in many tissues, including the differentiation and function of brown and white adipocytes. The endoribonuclease dicer is a major component of the miRNA-processing pathway, and in adipose tissue, levels of dicer have been shown to decrease with age, increase with caloric restriction, and influence stress resistance. Here, we demonstrated that mice with a fat-specific KO of dicer develop a form of lipodystrophy that is characterized by loss of intra-abdominal and subcutaneous white fat, severe insulin resistance, and enlargement and "whiten

An acute temperature rise to 40°C inhibits free fatty acid uptake into white adipocytes.

Fever reflects a physiological rise in body temperature accompanied by elevated production of adrenaline. The increased body temperature in fever is caused by shivering thermogenesis in skeletal muscle and non-shivering thermogenesis in brown adipose tissue (BAT), the latter being mediated by uncoupled oxidation of free fatty acids (FFAs). We hypothesized that an acute temperature rise to 40°C increases adrenalin-induced lipolysis in white adipocytes, thereby potentially providing FFAs as an energy substrate to sustain fever-induced thermogenesis in skeletal muscle and BAT. In 3T3-L1 and prima

Resveratrol promotes white adipocytes browning and improves metabolic disorders in Sirt1-dependent manner in mice.

Obesity has become an epidemic concern in modern society. The chronic obesity is associated with metabolic disorders, such as hyperglycemia, hyperlipidemia, fatty liver, and cadiovascular disease, which cause high risk for mortality. The novel potential strategy to overcome obesity is to "burn out" the extra fat via "browning" of the white adipose tissues. The phytochemical resveratrol (Res) has attracted substantial attention due to its powerful amelioratory effects in metabolic diseases. However, how Res regulates the browning of adipose tissues remains largely elusive. Our data show that th

Development of an Optimized Clearing Protocol to Examine Adipocyte Subpopulations in White Adipose Tissue.

Organic solvent dibenzyl ether (DBE)-based protocols have been widely used in adipose tissue clearing. However, benzyl alcohol/benzyl benzoate (BABB)-based clearing has been shown to offer better transparency in other tissues. The addition of diphenyl ether (DPE) to BABB (BABB-D4) is often included to preserve fluorescent signals, but its effects on adipose tissue transparency and shrinkage have not been explored. Distinct adipocyte subpopulations contribute to its cellular composition and biological activity. Here, we compared clearing solvents to create an optimized clearing methodology for

YAP and TAZ protect against white adipocyte cell death during obesity.

The expansion of the white adipose tissue (WAT) in obesity goes along with increased mechanical, metabolic and inflammatory stress. How adipocytes resist this stress is still poorly understood. Both in human and mouse adipocytes, the transcriptional co-activators YAP/TAZ and YAP/TAZ target genes become activated during obesity. When fed a high-fat diet (HFD), mice lacking YAP/TAZ in white adipocytes develop severe lipodystrophy with adipocyte cell death. The pro-apoptotic factor BIM, which is downregulated in adipocytes of obese mice and humans, is strongly upregulated in YAP/TAZ-deficient adi

Oleoylethanolamide enhances β-adrenergic-mediated thermogenesis and white-to-brown adipocyte phenotype in epididymal white adipose tissue in rat.

β-adrenergic receptor activation promotes brown adipose tissue (BAT) β-oxidation and thermogenesis by burning fatty acids during uncoupling respiration. Oleoylethanolamide (OEA) can inhibit feeding and stimulate lipolysis by activating peroxisome proliferator-activating receptor-α (PPARα) in white adipose tissue (WAT). Here we explore whether PPARα activation potentiates the effect of β3-adrenergic stimulation on energy balance mediated by the respective agonists OEA and CL316243. The effect of this pharmacological association on feeding, thermogenesis, β-oxidation, and lipid and cholesterol m
