Most alpha-gliadin genes of the Gli-D2 locus on the D genome of hexaploid bread wheat (Triticum aestivum) encode for proteins with epitopes that can trigger coeliac disease (CD), and several contain a 33-mer peptide with six partly overlapping copies of three epitopes, which is regarded as a remarkably potent T-cell stimulator. To increase genetic diversity in the D genome, synthetic hexaploid wheat lines are being made by hybridising accessions of Triticum turgidum (AB genome) and Aegilops tauschii (the progenitor of the D genome). The diversity of alpha-gliadins in A. tauschii has not been s

To produce a good F1 hybrid variety wheat crop, it is necessary to explore novel cytoplasmic male sterility (CMS) lines and their maintainer line. This study aimed to identify cytoplasmic variation in three isonuclear-alloplasmic male sterile lines Aegilops kotschyi (Ae.kots) -90-110, Aegilops ventricosa (Ae.ven) -90-110, and Triticum spelta (T.spelta) -90-110 and their maintainer line, A-90-110, at the molecular level. Mitochondrial DNA (mtDNA) was isolated using a combination of centrifugation and density gradient ultracentrifugation, sucrose sedimentation, lysis with sodium dodecyl sulfate

Understanding plant resistance to insects has lagged owing to a scarcity of cloned resistance genes. Here we report cloning of the greenbug resistance gene Gb3 in wheat (Triticum aestivum L.), initially introgressed into cultivars such as 'TAM112' from Aegilops tauschii. Gb3 encodes an intracellular kinase-pseudokinase protein, featuring a unique MAP3K-like domain. This gene probably originated through fusion, duplication and indel events in the Triticeae tribe, retaining its functionality in several Aegilops species. Transgenic Gb3 expression in the susceptible Fielder wheat enhanced resistan

Cold stress is a major abiotic constraint to wheat production in temperate regions, particularly in the Kashmir Valley of north-western Himalayas, India, where prolonged low temperatures delay growth, reduce yield, and disrupt the rice-wheat cropping system. To address this challenge, we evaluated a panel of 340 nested synthetic hexaploid wheat introgression lines, developed by introgressing allelic diversity from Aegilops tauschii and Triticum durum into bread wheat. The panel was phenotyped for cold stress tolerance, electrolyte leakage index, and yield-related traits across three temperate

Forager focus on wild cereal plants has been documented in the core zone of domestication in southwestern Asia, while evidence for forager use of wild grass grains remains sporadic elsewhere. In this paper, we present starch grain and phytolith analyses of dental calculus from 60 Mesolithic and Early Neolithic individuals from five sites in the Danube Gorges of the central Balkans. This zone was inhabited by likely complex Holocene foragers for several millennia before the appearance of the first farmers ~6200 cal BC. We also analyzed forager ground stone tools (GSTs) for evidence of plant pro

Wheat blast is an emerging disease caused by the highly virulent Triticum pathotype of the fungal pathogen Pyricularia oryzae that threatens wheat production worldwide. Relatively few sources of resistance to wheat blast have been identified, and no single source of resistance that protects against all strains of the pathogen has been found. We have conducted greenhouse evaluations of wheat entries submitted to the 2021 USDA-coordinated Uniform Eastern and Southern Soft Red Winter Wheat (Triticum aestivum L.) Nurseries for wheat blast resistance using an aggressive isolate (B-71) of the pathog

Alien gametocidal chromosomes cause extensive chromosome breakage prior to S-phase in the first mitotic division of gametophytes lacking the alien chromosome. The broken chromosomes may be healed either by addition of telomeric repeats in the gametophyte or undergo fusions to form dicentric or translocation chromosomes. We show that dicentric chromosomes undergo breakage fusion-bridge (BFB) cycles in the first few mitotic divisions of the sporophyte, are partially healed before the germ line differentiation regimen, and are healed completely in the ensuing gametophytic stage. The gametocidal f

Solanesyl diphosphate synthase (SPS) is essential for plastoquinone-9 (PQ-9) biosynthesis and has emerged as a promising target for herbicide discovery. The commercial SPS herbicides bind to the dimer interface of the protein; however, substrate-competitive SPS inhibitors are still rare, particularly in agronomically relevant weed and crop orthologs. Here, we show that zoledronic acid (ZOL) inhibits Arabidopsis thaliana SPS1 with an IC50 value of 2.55 μM in an Mg2+-dependent manner. Crystal structures of ZOL-bound SPS from the weed Aegilops tauschii (AeSPS) and the crop Sorghum bicolor (SbSPS)

Wheat allergies are potentially life-threatening because of the high risk of anaphylaxis. Wheats belong to four genotypes represented in thousands of lines and varieties. Monitoring changes to wheat allergens is critical to prevent inadvertent ntroduction of hyper-allergenic varieties via breeding. However, validated methods for this purpose are unavailable at present. As a proof-of-concept study, we tested the hypothesis that salt-soluble wheat allergens in our mouse model will be identical to those reported for humans. Groups of Balb/cJ mice were rendered allergic to durum wheat salt-soluble

To determine whether the Triticum urartu genome is more closely related to the A or B genome of the polyploid wheats, the amino acid sequence of its purothionin was compared to the amino acid sequences of the purothionins in Triticum monococcum, Triticum turgidum, and Triticum aestivum. The residue sequence of the purothionin from T. urartu differs by five and six amino acid substitutions respectively from the alpha 1 and alpha 2 forms coded for by genes in the B and D genomes, and is identical to the beta form specified by a gene in the A genome. Therefore, the T. urartu purothionin is either

The pathogen cereal cyst nematode (CCN) is deleterious to Triticeae crops and is a threat to the global crop yield. Accession no. 1 of Aegilops variabilis, a relative of Triticum aestivum (bread wheat), is highly resistant to CCN. Our previous study demonstrated that the expression of the phenylalanine ammonia lyase (PAL) gene AevPAL1 in Ae. variabilis is strongly induced by CCN. PAL, the first enzyme of phenylpropanoid metabolism, is involved in abiotic and biotic stress responses. However, its role in plant-CCN interaction remains unknown. In the present study, we proved that AevPAL1 helps t

Chromosome 2C of Aegilops cylindrica induces chromosomal rearrangements in alien chromosome addition lines, as well as in euploid lines, of common wheat. To induce chromosomal rearrangements in barley chromosome 7H, reciprocal crosses were made between a mutation-inducing common wheat line that carries a pair of 7H chromosomes and one 2C chromosome and a 7H disomic addition line of common wheat. Many shrivelled seeds were included in the progeny, which was an indication of the occurrence of chromosome mutations. The chromosomal constitution of the viable progeny was examined by FISH (fluoresce

Septoria nodorum blotch (SNB), caused by the necrotrophic fungal pathogen Parastagonospora nodorum, is a major foliar disease of wheat (Triticum aestivum) that significantly reduces grain yield and quality. In this study, we evaluated a modified nested association mapping (NAM) population for SNB resistance. This population consisted of 276 BC1F8 progenies derived from crosses between four synthetic hexaploid wheat (SHW) and ten hard red spring wheat (HRSW) varieties/lines. We screened the population using two P. nodorum isolates (Sn4 and Sn2000) and three necrotrophic effectors (SnToxA, SnTox

BACKGROUND: Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) is a key glycolytic enzyme that also plays important roles in plant responses to abiotic stresses. However, the evolutionary relationships of the GAPDH genes among wheat species with different ploidy levels remain poorly understood, and the key TaGAPDH members involved in the drought response of wheat have yet to be identified.
RESULTS: In this study, we identified 5, 6, 13, and 19 GAPDH proteins in Triticum urartu, Aegilops tauschii, Triticum turgidum, and Triticum aestivum with different ploidy levels. All identified GAPDH protein

Lignin is a crucial component of the cell wall, providing mechanical support and protection against biotic and abiotic stresses. However, little is known about wheat lignin-related mutants and their roles in pathogen defence. Here, we identified an ethyl methanesulfonate (EMS)-derived Aegilops tauschii mutant named brown glume and internode 1 (bgi1), which exhibits reddish-brown pigmentation in various tissues, including internodes, spikes and glumes. Using map-based cloning and single nucleotide polymorphism (SNP) analysis, we identified AET6Gv20438400 (BGI1) as the leading candidate gene, en

Recent research on the wheat curl mite species complex has revealed extensive genetic diversity that has distinguished several genetic lineages infesting bread wheat (Triticum aestivum L.) and other cereals worldwide. Turkey is the historical region of wheat and barley (Hordeum vulgare L.) domestication and diversification. The close relationship between these grasses and the wheat curl mite provoked the question of the genetic diversity of the wheat curl mite in this region. The scope of the study was to investigate genetic differentiation within the wheat curl mite species complex on grasses

Homeotic transformation of stamens into pistil-like structures (pistillody) has been observed in a cytoplasmic substitution (alloplasmic) line of wheat (Triticum aestivum L.) cv. Norin 26, which has the cytoplasm of a wild relative species, Aegilops crassa L. On the other hand, an alloplasmic line of wheat cv. Chinese Spring (CS) with Ae. crassa cytoplasm has normal flowers. This is due to the presence in the CS nucleus of a fertility-restoring gene, Rfd1. Deletion mapping analysis revealed that Rfd1 is located on the middle part of the long arm of chromosome 7B. To investigate the function of

The gametocidal factor on the Aegilops cylindrica chromosome 2Cc was used to induce and analyze the nature of chromosomal rearrangements in rye chromosomes added to wheat. For this purpose we isolated plants disomic for a given rye chromosome and monosomic for 2Cc and analyzed their progenies cytologically. Rearranged rye chromosomes were identified in 7% of the progenies and consisted of rye deficiencies (4.6%), wheat rye dicentric and rye ring chromosomes (1.8%), and terminal translocations (0.6%). The dicentric and ring chromosomes initiated breakage-fusion-bridge cycles (BFB) that ceased w

INTRODUCTION: The success of allopolyploids is partly attributable to their ability to resolve genomic conflicts during the early generations following allopolyploidization. Codon usage patterns represent important genomic signatures, and codon usage bias can influence gene expression and cellular function through multiple processes. However, little is known about how subgenomic conflicts in codon usage are resolved during allopolyploidization.
METHODS: We characterized changes in codon usage from the wild progenitors of bread wheat to modern bread wheat. In addition, we generated full-length

Bread wheat serves as an important staple crop in the human diet, largely because of the physicochemical properties of its dough and its protein content. Gluten is the main and complex component of wheat proteins. Despite the significant importance in breadmaking properties, wheat gluten contains some immunogenic peptides capable of triggering a T cell reaction in celiac disease (CD) patients, leading to inflammation in the small intestine. Among gluten proteins, α-gliadins are the most immunogenic components because they possess the primary T-cell stimulating epitopes (DQ2.5-Glia-α1, DQ2.5-Gl
