Immediate hypersensitivity (anaphylaxis) response in uninfected and Alcaligenes faecalis-infected turkey poults.

The immediate hypersensitivity (anaphylaxis) response in uninfected and Alcaligenes faecalis-infected turkey poults was studied. The optimum challenge dose of bovine serum albumin (BSA) to elicit an anaphylactic response was 40 mg/kg body weight, and the optimum poult age was 4 to 5 weeks. The response latency time was 7 days. The anaphylactic response was specific for the sensitizing antigen used. No differences were observed between A. faecalis-infected and uninfected poults in the immediate hypersensitivity response to BSA.

Alcaligenes faecalis corrects aberrant matrix metalloproteinase expression to promote reepithelialization of diabetic wounds.

Chronic wounds are a common and costly complication of diabetes, where multifactorial defects contribute to dysregulated skin repair, inflammation, tissue damage, and infection. We previously showed that aspects of the diabetic foot ulcer microbiota were correlated with poor healing outcomes, but many microbial species recovered remain uninvestigated with respect to wound healing. Here, we focused on Alcaligenes faecalis, a Gram-negative bacterium that is frequently recovered from chronic wounds but rarely causes infection. Treatment of diabetic wounds with A. faecalis accelerated healing duri

Novel 3,6-Dihydroxypicolinic Acid Decarboxylase-Mediated Picolinic Acid Catabolism in Alcaligenes faecalis JQ135.

Picolinic acid (PA), a typical C-2-carboxylated pyridine derivative, is a metabolite of l-tryptophan and many other aromatic compounds in mammalian and microbial cells. Microorganisms can degrade and utilize PA for growth. However, the precise mechanism of PA metabolism remains unknown. Alcaligenes faecalis strain JQ135 utilizes PA as its carbon and nitrogen source for growth. In this study, we screened a 6-hydroxypicolinic acid (6HPA) degradation-deficient mutant through random transposon mutagenesis. The mutant hydroxylated 6HPA into an intermediate, identified as 3,6-dihydroxypicolinic acid

Comprehensive genomic and transcriptomic analyses of the anaerobic degradation of microcystin in Alcaligenes faecalis D04.

Microcystin LR (MC-LR) pollution is a serious threat to aquatic ecosystems and public health in China and is an environmental problem that urgently needs to be solved. However, few studies have investigated the anaerobic degradation pathway and related molecular biological mechanisms of MC-LR. In this study, a bacterium capable of degrading MC-LR with a degradation efficiency of 0.303 µg/mL/d under anaerobic conditions was isolated from water. The strain was identified as Alcaligenes and named Alcaligenes faecalis D04. Two new anaerobic degradation products, one pentapeptide (Adda-Glu-Mdha-Ala

Isolation and properties of a particulate fraction for the desaturation of palmitic acid from Alcaligenes faecalis.

A particulate fraction obtained from Alcaligenes faecalis could desaturate palmitic acid to palmitoleic acid. NADPH, ATP, CoA, Fe2+ and Mg2+ were essential cofactors for the reaction. The desaturation showed an absolute requirement for O2. Metal ions like Mn2+, Mo6+ and Cu2+ did not affect the desaturation, while Zn2+ was inhibitory. Sulfhydryl agents such as cysteine, glutathione and beta-mercaptoethanol had no effect, but SH-blocking agents like HgCl2 and p-hydroxymercuribenzoate inhibited the reaction. Azide and cyanide strongly inhibited the reaction while CO had no effect. The presence of

Probiotic Properties of Alcaligenes faecalis Isolated from Argyrosomus regius in Experimental Peritonitis (Rat Model).

A strain of Alcaligenes faecalis A12C (A. faecalis A12C) isolated from Argyrosomus regius is a probiotic in fish. Previous experiments showed that A. faecalis A12C had inhibitory effects on the growth of multidrug-resistant bacteria. We aimed to confirm whether A. faecalis A12C is safe and has adequate intestinal colonization in experimental rats, and evaluate its efficacy in an animal model of peritonitis. We used 30 male rats, randomly divided into 6 groups (n = 5): three groups (HA7, HA15, HA30) received A. faecalis A12C in drinking water (6 × 108 CFU/mL) for 7 days, and three control group

Genomic and resistome analysis of Alcaligenes faecalis strain PGB1 by Nanopore MinION and Illumina Technologies.

BACKGROUND: Drug-resistant bacteria are important carriers of antibiotic-resistant genes (ARGs). This fact is crucial for the development of precise clinical drug treatment strategies. Long-read sequencing platforms such as the Oxford Nanopore sequencer can improve genome assembly efficiency particularly when they are combined with short-read sequencing data. RESULTS: Alcaligenes faecalis PGB1 was isolated and identified with resistance to penicillin and three other antibiotics. After being sequenced by Nanopore MinION and Illumina sequencer, its entire genome was hybrid-assembled. One chromo

Isolation and identification of Alcaligenes faecalis W2-3 with high-yield production of dimethyl disulfide.

Dimethyl disulfide (DMDS) is an important volatile organic sulfur compound with diverse applications and substantial demand in the chemical, agricultural, and food industries. The challenges associated with their chemical synthesis have spurred exploration of biological synthesis as an alternative route. Nevertheless, there is limited research on the biosynthesis of DMDS, and the yield remains low. In this study, strain W2-3, exhibiting high DMDS production, was isolated from aged tobacco leaves collected in Hubei Province, China. Based on morphological characteristics, 16S rRNA gene sequencin

Crystallization and X-ray structure analysis of a thermostable penicillin G acylase from Alcaligenes faecalis.

The enzyme penicillin G acylase (EC 3.5.1.11) catalyzes amide-bond cleavage in benzylpenicillin (penicillin G) to yield 6-aminopenicillanic acid, an intermediate chemical used in the production of semisynthetic penicillins. A thermostable penicillin G acylase from Alcaligenes faecalis (AfPGA) has been crystallized using the hanging-drop vapour-diffusion method in two different space groups: C222(1), with unit-cell parameters a = 72.9, b = 86.0, c = 260.2 , and P4(1)2(1)2, with unit-cell parameters a = b = 85.6, c = 298.8 . [Bacterial endotoxins produced by Alcaligenes faecalis and Erwinia herbicola as potential occupational hazards for agricultural workers].

Laboratory animals (guinea pigs and rabbits) were exposed to the inhalation of aerosolized endotoxins derived from the cell mass of Alcaligenes faecalis and Erwinia herbicola, the Gram-negative bacteria commonly occurring in organic dusts. Single 1 hour-lasting exposure caused the significant increase in the number of free lung cell, mostly lymphocytes, compared to the control group exposed to saline (P < 0.001). Prolonged exposure to tested endotoxins (Fifteen 1 hour-lasting exposures every second day) caused both specific and non specific immunological changes: enhanced inhibition of leukocy

Alcaligenes faecalis, Stenotrophomonas maltophilia, and Pseudomonas aeruginosa in a Mixed Microbial Contact Lens Keratitis: A Case Report.

We present a case of mixed microbial keratitis in an otherwise healthy contact lens wearer. The microbes detected on microscopy included Pseudomonas aeruginosa, Acanthamoeba, Alcaligenes faecalis, and Stenotrophomonas maltophilia. P. aeruginosa and Acanthamoeba are well-recognised corneal pathogens, although S. maltophilia is uncommon, and A. faecalis is extremely rare. We were only able to find one previously reported case of A. faecalis keratitis, in a cornea that was otherwise compromised by ocular cicatricial pemphigoid. To the best of our knowledge, this case represents the second reporte

Right Lower Extremity Osteomyelitis With Alcaligenes faecalis in a Patient With Poorly Controlled Type 2 Diabetes Mellitus.

Alcaligenes faecalis (A. faecalis) is a Gram-negative, rod-shaped, oxidase(+) and catalase(+), obligate aerobe commonly found in soil or water. It has also been found in human intestinal microbiota and more rarely in hospital settings and is typically associated with opportunistic infections. Although it has intrinsic resistance to many commonly used antibiotics, it is increasingly found to have developed antibiotic resistance. We present a rare case of A. faecalis osteomyelitis in a patient with a chronic diabetic foot ulcer.

Genetic diversity and horizontal transfer of genes involved in oxidation of reduced phosphorus compounds by Alcaligenes faecalis WM2072.

Enrichment was performed to isolate organisms that could utilize reduced phosphorus compounds as their sole phosphorus sources. One isolate that grew well with either hypophosphite or phosphite was identified by 16S rRNA gene analysis as a strain of Alcaligenes faecalis. The genes required for oxidation of hypophosphite and phosphite by this organism were identified by using transposon mutagenesis and include homologs of the ptxD and htxA genes of Pseudomonas stutzeri WM88, which encode an NAD-dependent phosphite dehydrogenase (PtxD) and 2-oxoglutarate-dependent hypophosphite dioxygenase (HtxA

Degradation characteristics and mechanisms of ammonium and phenol by the direct ammonia oxidation bacterium Alcaligenes faecalis AP3-14.

Direct ammonia oxidation (Dirammox) is an emerging nitrogen removal process that exhibits strong environmental adaptability, a simple flow, and can directly convert ammonium into N₂. However, no Dirammox bacterium has been reported for efficient treatment of antibiotic pharmaceutical wastewater co-contaminated with phenol and nitrogen. In this study, a simultaneous nitrogen- and phenol-removing strain, Alcaligenes faecalis AP3-14, was isolated from activated sludge. The strain harbors the key Dirammox gene cluster dnfABC, suggesting its direct ammonia oxidation potential. With sodium acetate a

Fed batch bioconversion of 2-propanol by a solvent tolerant strain of Alcaligenes faecalis entrapped in Ca-alginate gel.

A gram-negative, rod-shaped, aerobe, capable of converting 2-propanol (isopropanol, IPA) to acetone was isolated from an oil/sump, and identified by 16 S rDNA analysis as Alcaligenes faecalis. Investigations showed this strain to be extremely solvent-tolerant and it was subsequently named ST1. In this study, A. faecalis ST1 cells were immobilized by entrapment in Ca-alginate beads (3 mm in diameter), and used in the bioconversion of high concentration IPA. The biodegradation rates and the corresponding microbial growth inside the beads were measured at four different IPA concentration ranges f

Efficient cascade synthesis of ampicillin from penicillin G potassium salt using wild and mutant penicillin G acylase from Alcaligenes faecalis.

To avoid isolation and purification of the intermediate 6-aminopenicillanic acid (6-APA), a two-enzyme two-step cascade synthesis of ampicillin from penicillin G was established. In purely aqueous medium, penicillin G hydrolysis and ampicillin synthesis were catalyzed by immobilized wild-type and mutagenized penicillin G acylases from Alcaligenes faecalis (Af PGA), respectively (Fig. 1). The βF24 G mutant Af PGA (the 24th Phenylalanine of the β-subunit was replaced by Glycine) was employed for its superior performance in enzymatic synthesis of ampicillin. Lipopolysaccharide Derived From the Lymphoid-Resident Commensal Bacteria Alcaligenes faecalis Functions as an Effective Nasal Adjuvant to Augment IgA Antibody and Th17 Cell Responses.

Alcaligenes spp., including A. faecalis, is a gram-negative facultative bacterium uniquely residing inside the Peyer's patches. We previously showed that A. faecalis-derived lipopolysaccharides (Alcaligenes LPS) acts as a weak agonist of toll-like receptor 4 to activate dendritic cells and shows adjuvant activity by enhancing IgG and Th17 responses to systemic vaccination. Here, we examined the efficacy of Alcaligenes LPS as a nasal vaccine adjuvant.
