Copper-catalyzed C-alkylation of secondary alcohols and methyl ketones with alcohols employing the aerobic relay race methodology.

By employing aerobic oxidation to aldehydes as a more effective alcohol activation strategy, ligand-free copper catalysts were found to be superior catalysts than other metals in aerobic dehydrative β-alkylation of secondary alcohols and α-alkylation of methyl ketones using alcohols as the green alkylating reagents. Based on our mechanistic studies and also supported by the literature, we deduce that the newly-proposed relay race process rather than the conventional borrowing hydrogen-type mechanisms should be the most possible and a more rational mechanism for the aerobic C-alkylation reactio

Effect of alcohols on protein hydration: crystallographic analysis of hen egg-white lysozyme in the presence of alcohols.

Organic solvents are known to bring about dehydration of proteins, the molecular basis of which has remained uncharacterized. The dehydration effect in many cases leads to eventual unfolding of proteins through the macroscopic solvent effect. In some cases, the organic solvent molecules also bind to protein surfaces, thereby forcing local unfolding. The X-ray structure of hen egg-white lysozyme co-crystallized in the presence of alcohols with varying hydrophobicities has been studied. It was noticed that although the alcohols have very little effect on the conformation of the overall protein s

Deoxycyanation of Alkyl Alcohols Using Photoredox Catalysis.

Cyano groups represent an important class of functional motifs in medicinal chemistry given their synthetic versatility and capacity to engage in essential interactions with biological targets. However, the synthesis of sterically hindered alkyl nitriles remains challenging, and, furthermore, traditional methods often rely on toxic cyanide sources. Herein, we report a photoredox-catalyzed, metal-free deoxycyanation of alkyl alcohols that allows rapid access to a wide array of 1°, 2°, and 3° cyanides using the easily handled, low-toxicity, tosyl cyanide reagent.

Ruthenium-Catalyzed Self-Coupling of Secondary Alcohols.

A simple catalytic method for self-coupling of secondary alcohols leading to the synthesis of β-branched ketones under mild conditions is reported. Well-defined ruthenium pincer complex catalyzed the reactions. Optimization studies revealed that sodium tert-butoxide is an appropriate base for this transformation. Functionalized aryl methanols, heteroaryl methanols, and linear and branched aliphatic secondary alcohols underwent facile catalytic self-coupling reactions. Mechanistic studies revealed that both catalyst and base are crucial to achieve dehydrogenation of secondary alcohols to ketone

Visible light photoredox-catalyzed deoxygenation of alcohols.

Carbon-oxygen single bonds are ubiquitous in natural products whereas efficient methods for their reductive defunctionalization are rare. In this work an environmentally benign protocol for the activation of carbon-oxygen single bonds of alcohols towards a reductive bond cleavage under visible light photocatalysis was developed. Alcohols were activated as 3,5-bis(trifluoromethyl)-substituted benzoates and irradiation with blue light in the presence of [Ir(ppy)2(dtb-bpy)](PF6) as visible light photocatalyst and Hünig's base as sacrificial electron donor in an acetonitrile/water mixture generall

Antimicrobial activity of alcohols from Musca domestica.

Information on the stimulatory and inhibitory effects of cuticular alcohols on growth and virulence of insecticidal fungi is unavailable. Therefore, we set out to describe the content of cuticular and internal alcohols in the body of housefly larvae, pupae, males and females. The total cuticular alcohols in larvae, males and females of Musca domestica were detected in comparable amounts (4.59, 3.95 and 4.03 μg g(-1) insect body, respectively), but occurred in smaller quantities in pupae (2.16 μg g(-1)). The major free alcohol in M. domestica larvae was C(12:0) (70.4%). Internal alcohols of M.

Oxidative Arene Ring-Opening of Benzylic Alcohols.

Arene ring-opening (ARO) reaction via C-C bond cleavage remains a formidable challenge due to the exceptional thermodynamic stability of aromatic frameworks. Inspired by enzymatic lignin degradation in nature, we report an oxidative strategy that enables arene ring-opening of benzylic alcohols under mild conditions. Using hydrogen peroxide as oxidant and 2-nitro-4-(trifluoromethyl)-benzeneseleninic acid as the catalyst, we achieved the efficient conversion of benzylic alcohols into muconolactone or muconic acid derivatives, which contain multiple functional handles enabling diverse downstream

Direct Deoxygenation of Free Alcohols and Ketones.

This work presents a feasible method for the elimination of alcohol hydroxyls through the direct activation of typical alkyl alcohols using neutral boron radicals. This transformation necessitates a proficient reagent capable of swiftly activating the alcohol hydroxyl group to produce radicals, thereby circumventing numerous alternative side reactions associated with the alcohol hydroxyl group. To implement this method, we have created an innovative photocatalytic reaction system that oxidizes sodium tetraphenylboron to produce neutral boron radicals, which subsequently enable the direct homol

Rotaxane-catalyzed aerobic oxidation of primary alcohols.

Nitroxide radicals are widely utilized as catalysts for the oxidation of primary alcohols. Here, the aerobic catalytic oxidation cycle of nitroxide radicals has been implemented within a mechanically interlocked rotaxane architecture consisting of a paramagnetic crown ether, which is confined by a molecular axle containing a dialkylammonium station and a 1,2,3-triazole unit. The rotaxane is engineered to exploit the oxidation of a primary alcohol: the primary catalyst is the wheel, a nitroxide radical capable of altering its oxidation state during the catalytic cycle, while the co-oxidant is t

Fluoride Ion in Alcohols: Isopropanol vs Hexafluoroisopropanol.

The utility of alcohol as a hydrogen bonding donor is considered a providential avenue for moderating the high basicity and reactivity of the fluoride ion, typically used with large cations. However, the practicality of alcohol-fluoride systems in reactions is hampered by the limited understanding of the pertinent interactions between the OH group and F-. Therefore, this study comparatively investigates the thermal, structural, and physical properties of the CsF-2-propanol and CsF-1,1,1,3,3,3-hexafluoro-2-propanol systems to explicate the effects of the fluoroalkyl group on the interaction of

Zinc-Catalyzed Hydroalkoxylation/Cyclization of Alkynyl Alcohols.

Despite the great interest in zinc catalysis for hydroelementation reactions, the use of zinc complexes as catalysts for the hydroalkoxylation of alkynyl alcohols has not been reported to date. Scorpionate zinc complexes have been successfully designed as precatalysts for the hydroalkoxylation reaction of alkynyl alcohols under mild reaction conditions. Zinc amide complex 8 has been shown to be an excellent precatalyst for the highly selective intramolecular hydroalkoxylation process to yield the corresponding exocyclic enol ethers. Kinetic studies have been performed and confirmed that reacti

Dehydroxymethylation of Alcohols Enabled by Cerium Photocatalysis.

Dehydroxymethylation, the direct conversion of alcohol feedstocks as alkyl synthons containing one less carbon atom, is an unconventional and underexplored strategy to exploit the ubiquity and robustness of alcohol materials. Under mild and redox-neutral reaction conditions, utilizing inexpensive cerium catalyst, the photocatalytic dehydroxymethylation platform has been furnished. Enabled by ligand-to-metal charge transfer catalysis, an alcohol functionality has been reliably transferred into nucleophilic radicals with the loss of one molecule of formaldehyde. Intriguingly, we found that the d

Fluorinated Alcohols' Effects on Lipid Bilayer Properties.

Fluorinated alcohols (fluoroalcohols) have physicochemical properties that make them excellent solvents of peptides, proteins, and other compounds. Like other alcohols, fluoroalcohols also alter membrane protein function and lipid bilayer properties and stability. Thus, the questions arise: how potent are fluoroalcohols as lipid-bilayer-perturbing compounds, could small residual amounts that remain after adding compounds dissolved in fluoroalcohols alter lipid bilayer properties sufficiently to affect membranes and membrane protein function, and do they behave like other alcohols? To address t

Potent pincer-zinc catalyzed homogeneous α-alkylation and Friedländer quinoline synthesis reaction of secondary alcohols/ketones with primary alcohols.

Herein, we describe an air- and moisture-stable, homogeneous zinc catalyst stabilised using an electron deficient N^N^N pincer-type ligand. This ternary, penta-coordinated neutral molecular catalyst [Zn(N^N^N)Cl2] selectively produces α-alkylated ketone derivatives (14 examples) through a one-pot acceptorless dehydrogenative coupling (ADC) reaction between secondary and primary alcohols using the borrowing hydrogen (BH) approach in good to excellent isolated yields (up to 93%). It is worth noting that this catalyst also provides an eco-friendly route for the synthesis of quinoline derivatives

Electrochemical Dehydroxylative Thiolation of Alkyl Alcohols with Disulfides.

Herein, we report a metal-free electrochemical protocol for the direct thiolation of alkyl alcohols with odorless disulfides under mild conditions. This strategy features a broad substrate scope, excellent functional-group tolerance, and applicability to the late-stage functionalization of complex bioactive molecules, providing an efficient and sustainable platform for C-S bond formation. Furthermore, a series of control experiments highly suggest a plausible iodide/phosphine-mediated radical pathway for this transformation.

Nickel-Catalyzed Reductive Hydrolysis of Nitriles to Alcohols.

Nitriles are an abundant class of compounds that are widely used as versatile feedstocks to produce various chemicals including pharmaceuticals, and agrochemicals as well as materials. Here we report Ni-catalyzed reductive hydrolysis of nitriles to alcohols in the presence of molecular hydrogen. This conversion likely occurs in a domino reaction sequence that first involves the hydrogenation of nitrile to primary imine, then the hydrolysis of imine, and subsequent deamination to the aldehyde, which is finally hydrogenated to the desired alcohol. Crucial for this reductive hydrolysis process is

Dynamic activation of alcohols by an electrophilic phosphinine.

A bis(trifluoromethyl)-substituted phosphinine undergoes reversible oxidative addition of primary and secondary alcohols at the low-coordinate phosphorus(III) atom. This unprecedented reactivity contrasts the general inertness of uncoordinated phosphinines toward protic substrates and provides a rare example of reversible alcohol activation mediated by a main-group element compound. DFT calculations suggest that a hydrogen-bonded methanol network enables the transformation, offering a new concept for dynamic E-H bond activation by electrophilic, aromatic phosphorus heterocycles.
