Evaluation of the diagnostic reliability of the gill-based Ray's fluid thioglycollate medium assay for Perkinsus olseni infections in Manila clams (Ruditapes philippinarum) across Korean tidal flats.

The alveolate protozoan parasite Perkinsus olseni infects a range of marine bivalves inhabiting tidal flats and shallow subtidal zones, causing considerable damage to shellfish industries. Infection by P. olseni is typically assessed using Ray's fluid thioglycollate medium (RFTM) assay, with gill tissue often employed as a diagnostic proxy for whole-body infection. However, the diagnostic reliability of gill-based assays across diverse ecological settings-particularly under low-infection conditions-remains uncertain. In this study, we investigated tissue-specific distribution and the diagnosti

Metagenomic analysis of healthy and white plague-affected Mussismilia braziliensis corals.

Coral health is under threat throughout the world due to regional and global stressors. White plague disease (WP) is one of the most important threats affecting the major reef builder of the Abrolhos Bank in Brazil, the endemic coral Mussismilia braziliensis. We performed a metagenomic analysis of healthy and WP-affected M. braziliensis in order to determine the types of microbes associated with this coral species. We also optimized a protocol for DNA extraction from coral tissues. Our taxonomic analysis revealed Proteobacteria, Bacteroidetes, Firmicutes, Cyanobacteria, and Actinomycetes as th

Unveiling the phylogenetic position of the type species of the syndinean genus Amoebophrya and broad divergence of the A. ceratii complex.

The syndinean genus Amoebophrya comprises endoparasitic dinoflagellates that infect various marine protists. Since the description of the type species, A. sticholonchae, a parasite of the radiolarian Sticholonche zanclea, an additional six Amoebophrya species have been formally described. Over the past 2 decades, intensive high-throughput environmental sequencing has discovered widespread and extensive diversity of novel marine alveolate group II (MALV II), generally considered the Amoebophryidae. Of the large number of uncultured environmental sequences, only several clades of MALV II are ass

Protozoan Co-Infection Drives Fish Mortality Event in Crete's Karteros River.

In August 2024, a significant fish kill involving hundreds of flathead grey mullet (Mugil cephalus) was reported in the delta region of the Karteros River, Crete, Greece. The investigation identified the primary cause of mortality as severe parasitic infections, specifically from the protozoan parasites Amyloodinium ocellatum and Trichodina sp., both of which heavily affected the gills of the fish. Concurrently, poor water quality in the area, likely due to reduced water volume and limited water renewal, created favourable conditions for parasite proliferation while weakening the fish's natura

The natural alternative: protozoa as cellular models for Legionella infection.

The severe pneumonia known as Legionnaires' disease occurs following infection by the Gram-negative bacterium Legionella pneumophila. Normally resident in fresh-water sources, Legionella are subject to predation by eukaryotic phagocytes such as amoeba and ciliates. To counter this, L. pneumophila has evolved a complex system of effector proteins which allow the bacteria to hijack the phagocytic vacuole, hiding and replicating within their erstwhile killers. These same mechanisms allow L. pneumophila to hijack another phagocyte, lung-based macrophages, which thus avoids a vital part of the immu

Molecular detection of Hepatozoon canis and Babesia canis vogeli in domestic dogs from Cuiabá, Brazil.

The objective of this study was to report for the first time infection by Hepatozoon spp. and Babesia spp. in 10 dogs from the city of Cuiabá, State of Mato Grosso, central-western Brazil. A pair of primers that amplifies a 574 bp fragment of the 18S rRNA of Hepatozoon spp., and a pair of primers that amplifies a 551 bp fragment of the gene 18S rRNA for Babesia spp. were used. Six dogs were positive for Babesia spp., and 9 were positive for Hepatozoon spp. Co‑infection of Babesia spp. and Hepatozoon spp. was seen in 5 dogs. Sequenced samples revealed 100% identity with B. canis vogeli, and H.

tArp Is an Actin-Related Protein Required for Ciliary Function That Is Conserved in Ciliates and Related Eukaryotic Groups.

Cells exhibit a wide range of behaviors due to their diverse molecular systems, which have been established through the molecular evolution of individual species. Protein families generated through gene duplications may be involved in this process. The actin cytoskeleton is no exception. The majority of eukaryotes express not only actin, but also several types of actin-related proteins (Arps). The ciliate Tetrahymena thermophila possesses a species-specific Arp, named tArp, which localizes to cilia. However, the evolutionary origin of the gene and its detailed cellular functions remain unclear

A conserved GTPase complex gates exocytosis for defence and parasitism across alveolates.

Regulated protein secretion is a fundamental process by which eukaryotic cells respond to and interact with their dynamic environments, yet its mechanism outside metazoans remains largely unknown. Here we uncover an atypical small GTPase complex (Nd-GTPase-Nd9-NdP1) conserved across the Alveolata-a major eukaryotic clade encompassing ciliates and apicomplexan parasites-that gates organelle exocytosis for diverse functions ranging from cellular defence to host invasion. Using Tetrahymena thermophila and Toxoplasma gondii as comparative models, we combine functional genetics, cryo-electron tomog

Evolutionary analysis identifies a Golgi pathway and correlates lineage-specific factors with endomembrane organelle emergence in apicomplexans.

The organelle paralogy hypothesis (OPH) aims to explain the evolution of non-endosymbiotically derived organelles. It predicts that lineage-specific pathways or organelles should result when identity-encoding membrane-trafficking components duplicate and co-evolve. Here, we investigate the presence of such lineage-specific membrane-trafficking machinery paralogs in Apicomplexa, a globally important parasitic lineage. We are able to identify 18 paralogs of known membrane-trafficking machinery, in several cases co-incident with the presence of new endomembrane organelles in apicomplexans or thei

Stable endocytic structures navigate the complex pellicle of apicomplexan parasites.

Apicomplexan parasites have immense impacts on humanity, but their basic cellular processes are often poorly understood. Where endocytosis occurs in these cells, how conserved this process is with other eukaryotes, and what the functions of endocytosis are across this phylum are major unanswered questions. Using the apicomplexan model Toxoplasma, we identified the molecular composition and behavior of unusual, fixed endocytic structures. Here, stable complexes of endocytic proteins differ markedly from the dynamic assembly/disassembly of these machineries in other eukaryotes. We identify that

DAPI staining and DNA content estimation of nuclei in uncultivable microbial eukaryotes (Arcellinida and Ciliates).

Though representing a major component of eukaryotic biodiversity, many microbial eukaryotes remain poorly studied, including the focus of the present work, testate amoebae of the order Arcellinida (Amoebozoa) and non-model lineages of ciliates (Alveolata). In particular, knowledge of genome structures and changes in genome content over the often-complex life cycles of these lineages remains enigmatic. However, the limited available knowledge suggests that microbial eukaryotes have the potential to challenge our textbook views on eukaryotic genomes and genome evolution. In this study, we develo

Identification of MMV Malaria Box inhibitors of Perkinsus marinus using an ATP-based bioluminescence assay.

"Dermo" disease caused by the protozoan parasite Perkinsus marinus (Perkinsozoa) is one of the main obstacles to the restoration of oyster populations in the USA. Perkinsus spp. are also a concern worldwide because there are limited approaches to intervention against the disease. Based on the phylogenetic affinity between the Perkinsozoa and Apicomplexa, we exposed Perkinsus trophozoites to the Medicines for Malaria Venture Malaria Box, an open access compound library comprised of 200 drug-like and 200 probe-like compounds that are highly active against the erythrocyte stage of Plasmodium falc

Diversity of extracellular proteins during the transition from the 'proto-apicomplexan' alveolates to the apicomplexan obligate parasites.

The recent completion of high-coverage draft genome sequences for several alveolate protozoans - namely, the chromerids, Chromera velia and Vitrella brassicaformis; the perkinsid Perkinsus marinus; the apicomplexan, Gregarina niphandrodes, as well as high coverage transcriptome sequence information for several colpodellids, allows for new genome-scale comparisons across a rich landscape of apicomplexans and other alveolates. Genome annotations can now be used to help interpret fine ultrastructure and cell biology, and guide new studies to describe a variety of alveolate life strategies, such a

Chromerid genomes reveal the evolutionary path from photosynthetic algae to obligate intracellular parasites.

The eukaryotic phylum Apicomplexa encompasses thousands of obligate intracellular parasites of humans and animals with immense socio-economic and health impacts. We sequenced nuclear genomes of Chromera velia and Vitrella brassicaformis, free-living non-parasitic photosynthetic algae closely related to apicomplexans. Proteins from key metabolic pathways and from the endomembrane trafficking systems associated with a free-living lifestyle have been progressively and non-randomly lost during adaptation to parasitism. The free-living ancestor contained a broad repertoire of genes many of which we

Navigating Clinical Utilization of Direct-from-Specimen Metagenomic Pathogen Detection: Clinical Applications, Limitations, and Testing Recommendations.

BACKGROUND: Metagenomic next generation sequencing (mNGS) is becoming increasingly available for pathogen detection directly from clinical specimens. These tests use target-independent, shotgun sequencing to detect potentially unlimited organisms. The promise of this methodology to aid infection diagnosis is demonstrated through early case reports and clinical studies. However, the optimal role of mNGS in clinical microbiology remains uncertain. CONTENT: We reviewed studies reporting clinical use of mNGS for pathogen detection from various specimen types, including cerebrospinal fluid, plasma

Genome assembly and gene annotation of Babesia ovis Israeli strain (Piroplasmida: Babesiidae) using PacBio HiFi sequencing.

Babesia ovis is a unicellular parasite of sheep and goats transmitted by the tick Rhipicephalus bursa. B. ovis infection is widespread and can be lethal, and therefore there is need for a deeper understanding of its biology and pathogenesis. There is currently a single genome available for a clinical isolate of B. ovis, and no laboratory strains have been sequenced so far. In this study we have sequenced and assembled the genome of the Israeli laboratory strain of B. ovis using highly accurate long-read sequencing (PacBio HiFi). The resulting assembly has high contiguity, with the four largest

Distribution and Evolution of Peroxisomes in Alveolates (Apicomplexa, Dinoflagellates, Ciliates).

The peroxisome was the last organelle to be discovered and five decades later it is still the Cinderella of eukaryotic compartments. Peroxisomes have a crucial role in the detoxification of reactive oxygen species, the beta-oxidation of fatty acids, and the biosynthesis of etherphospholipids, and they are assumed to be present in virtually all aerobic eukaryotes. Apicomplexan parasites including the malaria and toxoplasmosis agents were described as the first group of mitochondriate protists devoid of peroxisomes. This study was initiated to reassess the distribution and evolution of peroxisom

Atypical RanGAP drives nucleocytoplasmic transport in a parasitic Alveolate.

Transport of macromolecules between the nucleus and cytoplasm requires a gradient of the small GTPase, Ran. Ran:GTP marks the nucleus because Ran activity requires a cytoplasmic GTPase activating protein (RanGAP) for GTP hydrolysis. As expected for such central and essential cellular machinery, both Ran and RanGAP are conserved across the vast majority of eukaryotes. Many Alveolates, including apicomplexan parasites, however, lack a canonical RanGAP. Here, we biochemically purify RanGAP activity from the model Alveolate Toxoplasma gondii. We demonstrate that this activity is provided by a neof

Parasitic Infections of Wild Boars (Sus scrofa) in Iran: A Literature Review.

BACKGROUND: Swine species are an important source of meat production worldwide, except in Islamic countries where pig breeding and pork consumption are forbidden. Hence, they are often neglected in these regions. A considerable number of wild boars (Sus scrofa) inhabit Iranian territories, particularly in dense forests of north, west and southwest of the country, but our knowledge regarding their parasites is very limited. OBJECTIVE: The lack of a comprehensive record in this connection encouraged us to review the whole works of literature in the country. METHODS: The current review presents
